Estudar o perfil patogênico e de resistência aos antimicrobianos em amostras de Staphylococcus haemolyticus, Staphylococcus warneri, Staphylococcus lugdunensis e Staphylococcus hominis. Foram estudadas 65 amostras isoladas de pacientes do Hospital das Clínicas da FMB, Botucatu, sendo 23 S. haemolyticus, 23 S. hominis, 10 S. warneri e 9 S. lugdunensis. Foram pesquisados por PCR os genes responsáveis pela produção de biofilme (icaA, icaC, icaD), genes de enterotoxinas (sea, seb, sec, sed), Toxina 1 da Síndrome do Choque Tóxico (tst) e resistência à oxacilina (mecA). Das 65 amostras estudadas, 83% apresentaram ao menos um dos genes das toxinas pesquisadas, 87,7% um dos genes ica e 63,1% o gene mecA. O SCCmec foi tipado por PCR-Multiplex, sendo o tipo I o mais prevalente (34,1%). A heterorresistência à vancomicina foi pesquisada através da triagem em ágar BHI com 4 μg ml-1, encontrada em 36,9% das amostras, e com 6 μg ml-1 de vancomicina, encontrada em 15,4%. Todas as espécies estudadas foram altamente toxigênicas. A presença do SCCmec I apresentou relação com a heterorresistência à vancomicina. Ainda, S. hominis e S. haemolyticus se revelaram mais virulentos e resistentes, levando em conta os fatores de virulência, resistência à oxacilina e heterorresistência à vancomicina. A evidência e a necessidade de maior preocupação com as espécies S. hominis e S. haemolyticus ficou clara, o que ainda não havia sido relatado, bem como a relação entre a presença de SCCmec I e heterorresistência à vancomicina

Whereas the assembly torque calculation has great importance on efficiency and life of bolts, the objective of this study was to analyze the combined stresses during assembly, the friction coefficient, compare the rupture torque results with the indicated torques by standards and to present a new torque method without the stress combination (shear stress and normal stresses). For this study were carried eighteen tests with different diameters and materials, this bolts were undergoing five ranges torque until the breaking torque. With the experimental rupture torque values for each test and with the bolt quality certificate was possible to calculate the friction coefficient so compared with the theory. Another comparison was the experimental rupture torque with assembly torque indicated by NBR-8800 and ASME-PCC. The main conclusion are related to combined stress, because for this factor can fail at the assembly time, so for in some cases must use the tensioning method, because in this method won't be present torsional stresses

Infections Related to Healthcare (IRAS) is a problem of worldwide concern, responsible for much morbidity and lethality. The incidence of IRAS associated with resistant microorganisms has increased worldwide. It is also known that the etiology of bacterial resistance is multifactorial, thus controlling the spread of resistant microorganisms requires the implementation of control measures that involve the performance of standard and contact precautions, plus the use of antimicrobials. To diagnose the problem as to the likely accession of the measures of infection control was prepared in a form to inspect the practices of the health staff acting with patients in contact precautions. The inspections had an educational and awareness, no punitive measures were taken to staff as warnings, suspensions or complains of a breach of ethics. If any irregularities were found, the professional standards of care countershaft contact and the nurse responsible for the head nurse or unit area were notified and counseled about the correct practice of isolation. While an undergraduate student initiated precautionary inspections of contact in order to verify the actual use of isolation measures, since the professionals were unaware of my active participation in the Committee on Infection Control. Facilities and difficulties were encountered in the process, and from the inspections was possible interventions to facilitate compliance with standards for isolation imposed by contact. Since the instrument has significantly improved adherence to contact isolation practices, the members of the Infection Control regulated inspections and intend to use it as a permanent method

The Brazilian livestock stands out for having the world largest commercial herd of cattle and leads meat exportation and production of bovine embryos. The in vitro production (IVP) of embryos is considered an effective option to overcome problems such as infertility in cows with high economic value and also for genetic improvement of cattle. The in vitro oocyte maturation is an essential step to the success of IVP, but is still considered poor when compared to in vivo maturation. Recent studies have suggeested an important role of Fibroblast Growth Factor 10 (FGF10) on the in vitro maturation of oocytes, which favored the expression of genes related to oocyte maturation and cumulus cell expansion. Aware that maturity stage influences the final production of blastocysts, we aimed study to verify if the addition of FGF10 into the maturation medium is able to affect positively the IVP of bovine embryos. Hence, FGF10 was added to maturation in five different concentrations: 0.5 ng/mL (group 0.5), 2.5 ng/mL (group 2.5), 5 ng/mL (group 5), 10 ng/mL (group 10) and 50 ng/mL (group 50). Additionally, two other maturation groups were used, group BSA (Bovine Serum Albumin, 4 mg/mL) and group FCS (Fetal Calf Serum, 10%). The rates of cleavage, morula and blastocyst were analyzed by Analysis of Variance (ANOVA), differences of P<0.05 were considered significant. Cleavage rates did not differ between the seven groups. On the other hand, morula rate on FCS group was higher than groups BSA, 0.5, 10 and 50 (P<0.05), but did not differ among groups treated with intermediate doses of FGF10 (2.5 and 5). FCS group presented higher blastocyst rate compared to all other groups that were well below the FCS group (P<0.0001). Therefore, the use of FGF10 during oocyte maturation did not affect positively embryo development on the IVP of bovine embryos

As infecções do trato urinário (ITU) são definidas como a invasão e posterior multiplicação de microorganismos nos tecidos das vias urinárias, sendo a bactéria gram-negativa E.coli o principal uropatógeno envolvido nessa infecção. A avaliação para a presença de ITU é feita através do quadro clínico característico desse tipo de paciente e o exame de cultura de urina. As amostras positivas para as culturas são avaliadas quanto ao perfil de sensibilidade aos antimicrobianos dos agentes etiológicos encontrados. Descrever a porcentagem de ocorrência dos agentes etiológicos e o perfil de sensibilidade aos antimicrobianos das Infecções do Trato Urinário em pacientes ambulatoriais de Botucatu e região. Foram incluídas neste estudo culturas de urinas de pacientes atendidos em UBS de Botucatu e região Pólo Cuesta, no período de Janeiro de 2008 a Maio de 2010. As culturas positivas foram avaliadas quanto a porcentagem de ocorrência dos uropatógenos segundo sexo, faixa etária e origem. Foi utilizado o teste do qui-quadrado para verificar associação entre cada antimicrobiano e as porcentagens de sensibilidade e resistência encontradas. Foram avaliadas 11.768 culturas de urina provenientes de 11 municípios da região, sendo consideradas as culturas positivas (28%). Quanto à ocorrência dos agentes etiológicos verificou-se que a E.coli é responsável 58,76% dos casos. A distribuição dos uropatógenos segundo sexo revelou maior ocorrência de E.coli nos dois sexos. O segundo agente etiológico de maior ocorrência foi, nos homens o Proteus mirabilis e nas mulheres, o S. saprophyticus. Dentre as faixas etárias, a E.coli foi o patógeno de maior ocorrência. O perfil de sensibilidade aos antimicrobianos revelou que a E.coli é mais sensível a Ertapenem, Imipenem, Meropenem, Fosfomicina, Amicacina, Ceftriaxona, Nitrofurantoína e Norfloxacina; e mais resistente... Preeclampsia is a specific disorder of pregnancy, characterized by arterial hypertension and proteinuria detected after 20 weeks of gestation. This pathology is associated with hyperuricemia, higher levels of pro-inflammatory cytokines, enhanced leukocyte activation and oxidative stress. Adenosine deaminase (ADA) is an enzyme present in all human tissues and, it is involved with the maturation of the immune system. Although its function is not fully understood, ADA is considered an indicator of cellular inflammation and, its increased serum concentration is observed in inflammatory diseases, such as tuberculosis and rheumatoid arthritis. This study aimed to assess serum ADA levels in preeclamptic patients (PE) compared with normotensive pregnant (NT) and non-pregnant women (NP), and to correlate these values with TNF-α and IL-1β production. Ninety pregnant women were included: 60 were pre-eclamptic and 30 were normotensive matched for gestational age. As control group 20 healthy non-pregnant women matched with pregnant for age were included. Peripheral blood mononuclear cells (PMMC) obtained from the three groups studied were cultured with or without lipopolysaccharide (LPS) for 18h at 37oC, and TNF-α and IL-1β production was assessed in the supernatant of cultures by enzyme immunoassay (ELISA). ADA plasmatic concentration was determined by colorimetric method. The results show that ADA plasma levels were significantly higher in PE group compared with NT and NP groups. A positive correlation between ADA and uric acid levels was detected in preeclamptic women. There was no significant difference in relation to ADA levels when PE patients were classified in early and late-onset PE. The endogenous production of IL-1β and TNF-α by PBMC was significantly higher in PE group than in NT and NP women, showing the activation state of these cells in PE. Pre-eclampsia (PE) is a complication of human pregnancy characterized by hypertension and proteinuria after 20 weeks of gestation. Its incidence varies from 5% to 7% of pregnancies and is a major cause of morbidity and maternal and fetal mortality. This is a multisystemic disease, with focus on vascular dysfunction and is closely related to the exacerbated activation of the immune system. In addition to increased activation of monocytes and granulocytes, there is an elevated production of proinflammatory cytokines in pregnant women with PE. The nuclear transcription factor-kB (NF-kB) is present in the cells of the immune system and is responsible for transcription of genes related to inflammation. Whereas the PE is associated with intense inflammatory response, the use of substances modulating the activity of NF-kB factor could be useful in alleviating the inflammation present in these patients. Silibinin is the main component of silymarin, a polyphenolic extract obtained from fruits and seeds of Sylibum marianum with potent hepatoprotective, anti-inflammatory and anti-fibrotic activities. The silibinin mechanism of action includes the ability to inhibit NF-kB activation and, consequently, its migration to the nucleus. The objective of this study was to assess whether silibinin modulates the activity of NF-kB and the production of inflammatory cytokines in mononuclear cells of patients with PE. We evaluated 34 pregnant women with PE, 20 normotensive pregnant women (GN) and 15 non-pregnant women (NG). The semidecidual forests, as tropical forests, presents zoochoric dispersal syndrome in the most of their plant species, with approximately one third of frugivorous species responsible for the seeds dispersal are birds. The bills width is a limiting factor, it must be equal or bigger than the seeds width, because the viable seeds dispersal depends on leaving the digestive system after been swallowed hole. Birds and plants inventorys were realized in a semidecidual Forest fragment, in Botucatu – SP, called Mata da Bica. After this, the birds consumers of fuits/seeds were selected (frugivorous, granivorous and onivorous) and also the zoochoric plants, both for building a possibilities matrix, to verify the potential seed dispersers according to the seeds and bills width. Going to the matrix end we can observe a decrease in the interaction possibillities, because the seeds width increases and the bills width decreases. It has been observed that 50,7% seeds could be dispersed by all listed birds, because the seeds were narrower than the narrowest bill. In the other way large seeded species, as Holocalix balansae and Hymemaea courbaril has shown only two possible dispersers: Trogon surrucura and Piaya cayana. There are no possible dispersers to Syagrus oleraceae in the matrix. The need for large birds or other animals that works as large seed disperses demand conservation efforts, since most of these animals need big and conserved areas for their life habits

As a result of the 20th-century industrial development, and particularly after World War II, cars have become an important consumer good, and this has led to a large number of circulating vehicles. To this day, the number of cars has increased exponentially worldwide, which results in a high number of traffic accidents (TAs) every day. TAs are responsible for 1.2 million deaths worldwide each year, and in Brazil, they are considered to be a public health problem, considering that its traffic is regarded as one of the worst and most dangerous in the world. In addition to temporary or permanent physical disabilities that bring great harm to individuals, accidents also cause psychological disorders, such as the post-traumatic stress disorder. Hence, considering the importance of TAs in the country and their outcomes, this study aimed at investigating the epidemiological aspects of car accidents occurring in 2010 who were assisted in the emergency room of a university hospital in São Paulo state. By means of records made by the nursing team and medical records analysis, it was possible to extract a stratified sample of 88 cases involving adults and eight cases involving children. The sample was analyzed by the SAS software for Windows, version 9.2. As regards the accidents involving adults, there was a predominance of males (67%), with an approximate 2:1 ratio in relation to females. The victims’ most frequent age range was 20-29 years (39%), and they were mostly single (56%). The month showing the largest number of occurrences was July, with 17%, and the weekday was Sunday, with 25%. Showing 32%, the time period when most accidents occurred was from 06:00 o’clock p.m. to 11:59 p.m. As regards vehicles types involved, motorcycles were in first place, with 47% of cases, and the most common accident type was crashing between vehicles, with 43%... The steel type AISI 4130 (ultra-high strength steel) is an alloy of low carbon and its main alloying elements are chromium and molybdenum, which improves the toughness of the weld metal. It has numerous applications, especially where the need for high mechanical strength. It is widely used in equipment used by the aviation industry, such as cradle-tomotor, and this is the motivation for this study. Cots are of fundamental importance, because the engine supports and maintains balance in the fixed landing gear. This equipment is subjected to intense loading cycles, whose fractures caused by fatigue are constantly observed. Will be determined the effects caused by re-welding the structure of aeronautical equipment, and will also study the microstructure of the metal without welding. The studies will be done on materials used in aircraft, which was given to study. The results provide knowledge of microstructure to evaluate any type of fracture that maybe caused by fatigue. Fatigue is a major cause of aircraft accidents and incidents occurred, which makes the study of the microstructure of the metal, weld and re-solder the knowledge essential to the life of the material. Study the semi-quantitative and quantitative technique in the diagnosis of catheter-related infections in newborns and to determine oxacillin resistance in Staphylococcus isolated. It was analyzed 353 catheter tips from 273 newborns in the Neonatal Unit of Hospital FMB. To confirm the diagnosis of infection, were analyzed the clinical data of newborns, the presence of at least one positive blood culture and growth of ≥ 1000 CFU/mL on quantitative culture and/or ³15 UFC on semiquantitative culture, with the same microorganism isolation (species and drug sensitivity) in blood culture and no other focus of infection except the catheter. The disk diffusion technique was used to check similarity of strains and resistance to oxacillin. Of the 353 tips analyzed, 39 were included in this study as the inclusion criteria. The semiquantitative culture was positive in 26 (66.7%) catheters and quantitative culture was positive in 24 (61.5%). Of 273 patients, 19 (6.9%) had a diagnosis of catheter-related bloodstream Infection (CR-BSI). Of the 19 episodes of CR-BSI, S. epidermidis was the predominant etiological agent (84.2%). The resistance to the antibiotic methicillin was found in 14 (73.7%) strains of Staphylococcus. The semiquantitative method was more sensitive (79%) compared with the quantitative method (63%). The use of antibiotics may have influenced the sensitivity of the quantitative method as the microorganisms present in the lumen are exposed to higher concentrations of antibiotics administered via the catheter

O fator de início de tradução 5A (eukaryotic translation iniciation factor 5A, eIF5A) é altamente conservado entre arqueas a eucariotos, sendo que as proteínas eIF5A de Saccharomyces cerevisiae e de mamíferos são 63% idênticas. eIF5A sofre uma modificação pós-traducional única na célula, a hipusinação de um resíduo de lisina. Essa proteína já foi relacionada ao início da tradução, transporte nucleocitoplasmático, decaimento de mRNA e proliferação celular, mas a função crítica de eIF5A ainda não foi esclarecida. A depleção deste fator em S. cerevisiae leva a uma diminuição (30%) da taxa de síntese protéica, sugerindo que eIF5A seja um fator envolvido na tradução de um grupo específico de mensageiros. Dados do laboratório demonstram interação física entre eIF5A e proteínas ribossomais bem como com o fator de elongação 2 da tradução (eEF2). A interação com eEF2, sugere que eIF5A atua na etapa de elongação da tradução, ao invés do início da tradução, como proposto inicialmente. Com o objetivo de avaliar a relação de eIF5A com a etapa de elongação, foram realizadas análises de interações genéticas entre o gene codificador de eIF5A (TIF51A) e diversos genes codificadores de proteínas envolvidas na tradução. Através de análises de interações genéticas, foi observado que o mutante estável de eIF5A, tif51AK56A, apresenta um defeito de crescimento quando o mutante de eEF2, eft2H699K, está expresso em alto número de cópias, enquanto que o mutante tif51AQ22H/L93F não apresenta defeitos nesta condição. Foi observado também que o mutante tif51AQ22H/L93F apresenta um defeito de crescimento mais severo quando ocorre superexpressão de EFT2, gene codificador de eEF2. Foi observado ainda que não há complementação alélica entre os mutantes estáveis de eIF5A e que a reversibilidade do fenótipo de sensibilidade a temperatura... Propomos avaliar o potencial de uma abordagem imunoterápica que tem o fator de crescimento do endotélio vascular (VEGF) como alvo. A angiogênese é necessária para nutrir o tumor e propicia sua metastatização. Sabe-se que VEGF é produzido pela maioria dos tumores sólidos metastatizantes. A eficiência do anticorpo anti-VEGF, BEVACIZUMAB, já foi demonstrada. Entretanto, efeitos adversos limitam sua utilização em alguns casos e, com base na teoria da rede idiotípica e na experiência do grupo com anticorpos anti-idiotípicos (anti-Id ou AB2), anticorpos AB2 capazes de mimetizar VEGF foram obtidos. Assim, com o objetivo de explorar o rationale, dois anticorpos monoclonais (MAb) anti-Id de Bevacizumab, 10.D7 e 3.E3, foram produzidos em nosso laboratório. O controle da purificação de anticorpos foi feito através da avaliação do material purificado em géis de poliacrilamida. Immunoblots comprovaram a especificidade do reconhecimento do idiotipo de Bevacizumab pelos MAbs anti-Id. Foi mostrado que os AB2 obtidos se ligam a moléculas da superfície de células endoteliais, que expressam receptores de VEGF, sugerindo que esses anticorpos mimetizam VEGF. Entretanto, não foi possível verificar inibição do crescimento dessas células quando cultivadas em presença desses MAbs. Será necessária a otimização dos protocolos utilizados para uma avaliação mais definitiva. Uma vez demonstrada a atividade anti-idiotípica dos AB2 in vitro, 10.D7 e 3.E3 foram utilizados como imunógenos em camundongos Balb/c, com o objetivo de avaliar a resposta AB3 ou anti-anti-Id. Por meio de ensaio imunoenzimático (ELISA) indireto foi verificada a presença de anticorpos anti-anti-Id ligantes de VEGF nos soros dos camundongos imunizados. Staphylococcus aureus é um patógeno ubíquo capaz de causar uma variedade de infecções em humanos. A resistência desse microrganismo a antibióticos como oxacilina e meticilina é um problema sério, de crescimento significativo para a terapêutica antimicrobiana clínica em pacientes acometidos por infecções estafilocócicas. A resistência à meticilina em S. aureus é decorrente da alteração do sítio de ação dos antibióticos β-lactâmicos, os quais agem através da inibição de enzimas que catalisam a síntese da parede celular. Essas enzimas são o sítio de ação das penicilinas, e por isso, passaram a ser chamadas de proteínas ligadoras de penicilinas (PBPs). O gene mecA codifica a PBP2a, que substitui a função das outras PBPs neste patógeno e confere resistência a β-lactâmicos. A PBP2a exibe uma afinidade reduzida pelo anel β-lactâmico e permite que a bactéria continue a sintetizar a parede bacteriana. Este gene faz parte de um elemento genético móvel encontrado em isolados de MRSA, designado cassete cromossômico estafilocócico mec (SCCmec), integrado ao cromossomo de S. aureus. O objetivo do estudo foi a caracterização molecular de 139 isolados de S.aureus provenientes de pacientes pediátricos com bacteremia durante o período de 1991 a 2010. Métodos moleculares foram utilizados para a determinação do perfil genético das amostras, incluindo, identificação do tipo de SCCmec, detecção do gene codificador de leucocidina de panton valentine (PVL) e similaridade clonal em gel de eletroforese em campo pulsado (PFGE). O gene mecA foi detectado em 32 (23%) amostras e houve predomínio do SCCmec IV (68,8%) em relação ao SCCmec III (31,2%). A presença de PVL foi encontrada em 18 amostras (12,9%), todas sensíveis à oxacilina. O clone epidêmico brasileiro, relacionado ao SCCmec tipo III, esteve presente na unidade neonatal do hospital...
