An alcohol dehydrogenase (ADH) was purified from dry baker’s yeast. This is a key enzyme of the primary short-chain alcohol metabolism in many organisms. In the present study, the obtained enzymatic preparation of baker’s yeast, containing 2.7 U/mg of ADH, was used in the reactions. The purified extract of the ADH obtained from Fermix commercial dry yeast, presented the highest activity and purification factor when ammonium sulfate was added in the precipitation of protein, in the range 35-60% (w/v). The enzymatic preparation was maintained for 2 months in the lyophilized form at 4ºC (retention of 96.2% of activity) in the presence of 1 mmol/L of sodium azide, and it maintained 47% of activity for 30 days at 30°C in the presence of 15% PEG. The assays of ethanol (detection range 5 mM -150 mM or 2.3 x 10-4 – 6.91 x 10-3g/L) in different samples in alcoholic beverages, presented a maximum deviation of only 2.1%. Assays of recovery of the substrate (99.25%) added in the wine showed that the methodology is viable for this sample type. The standard curve and the analytic curve of this method meet the conditions of precision, sensitivity, simplicity, and low cost, required for a useable analytical method.

A series of anti-inflammatory derivatives containing an N-acyl hydrazone subunit (4a–e) were synthesized and characterized. Docking studies were performed that suggest that compounds 4a–e bind to cyclooxygenase (COX)-1 and COX-2 isoforms, but with higher affinity for COX-2. The compounds display similar anti-inflammatory activities in vivo, although compound 4c is the most effective compound for inhibiting rat paw edema, with a reduction in the extent of inflammation of 35.9% and 52.8% at 2 and 4 h, respectively. The anti-inflammatory activity of N-acyl hydrazone derivatives was inferior to their respective parent drugs, except for compound 4c after 5 h. Ulcerogenic studies revealed that compounds 4a–e are less gastrotoxic than the respective parent drug. Compounds 4b–e demonstrated mucosal damage comparable to celecoxib. The in vivo analgesic activities of the compounds are higher than the respective parent drug for compounds 4a–b and 4d–e. Compound 4a was more active than dipyrone in reducing acetic-acid-induced abdominal constrictions. Our results indicate that compounds 4a–e are anti-inflammatory and analgesic compounds with reduced gastrotoxicity compared to their respective parent non-steroidal anti-inflammatory drugs.

The determination of loin eye area (LEA) is one of the most common methods to assess carcass quality and it is possible to adjust calculations to predict meat amount in the carcass which is one of the important features in the classification process. The aim of this study was to compare different methodologies used to determine LEA in swine. Fifteen crossbred pigs, Landrace x Large White, males and females, were slaughtered at 140 days of age. After 24 hours of cooling at 4°C, the Longissimus dorsi muscle was removed from the left half part of carcasses for evaluation of LEA between the 12th and 13th rib. The methods were: point counting on plastic grid of 1 cm2 and 0.25 cm2 (PCGP 1 cm2 and PCGC 0.25 cm2 ), circumference method by Echo Image Viewer (ECHO), circumference method using AutoCad ® program (CAD) and method of weighing paper (WP). The design was completely randomized with 15 replications per treatment and the data were subjected to Pearson correlation and variance analysis. Averages were compared by Tukey test at a significance level of 5%. LEA values were: 35.84, 35.69, 33.86, 34.22 and 36.71 cm2 , for the PCGP 1 cm2 , PCGP 0.25 cm2 , ECHO, CAD and WP methods, respectively. The LEA values determined by methods PCGP 1 cm2 , PCGP 0.25 cm2 and WP were similar; however they were higher than those obtained by the ECHO and CAD methodologies (P<0.0001). PCGP 1 cm2 , PCGP 0.25 cm2 and WP methods may overestimate LEA values. The choice of method to be used should be based on its practicality and availability of resources, since the difference obtained between them is low.

The extrapolation chamber is a parallel-plate ionization chamber that allows variation of its air-cavity volume. In this work, an experimental study and MCNP-4C Monte Carlo code simulations of an ionization chamber designed and constructed at the Calibration Laboratory at IFEN to be used as a secondary dosimetry standard for low-energy X-rays are reported. The results obtained were within the international recommendations, and the simulations showed that the components of the extrapolation chamber may influence its response up to 11.0%. OBJETIVO: Comparar os achados histopatológicos e de apoptose em pulmões de ratos preservados em soluções low-potassium dextran (LPD, baixo potássio dextrana), histidine-tryptophan-ketoglutarate (HTK, histidina-triptofano-cetoglutarato) ou salina normal (SN) em 6 h e 12 h de isquemia pela utilização de um modelo experimental de perfusão pulmonar ex vivo. MÉTODOS: Sessenta ratos Wistar foram anestesiados, randomizados e submetidos à perfusão anterógrada pela artéria pulmonar com uma das soluções preservadoras. Após a extração, os blocos cardiopulmonares foram preservados por 6 ou 12 h a 4ºC, sendo então reperfundidos com sangue homólogo em um sistema de perfusão ex vivo durante 60 min. Ao final da reperfusão, fragmentos do lobo médio foram extraídos e processados para histopatologia, sendo avaliados os seguintes parâmetros: congestão, edema alveolar, hemorragia alveolar, hemorragia, infiltrado inflamatório e infiltrado intersticial. O grau de apoptose foi avaliado pelo método TdT-mediated dUTP nick end labeling. RESULTADOS: A histopatologia demonstrou que todos os pulmões preservados com SN apresentaram edema alveolar após 12 h de isquemia. Não houve diferenças em relação ao grau de apoptose nos grupos estudados. CONCLUSÕES: No presente estudo, os achados histopatológicos e de apoptose foram semelhantes com o uso das soluções LPD e HTK, enquanto a presença de edema foi significativamente maior com o uso de SN.

The first cleavage divisions and preimplantation embryonic development are supported by mRNA and proteins synthesized and stored during oogenesis. Thus, mRNA molecules of maternal origin decrease and embryonic development becomes gradually dependent on expression of genetic information derived from the embryonic genome. However, it is still unclear what the role of the sperm cell is during this phase and whether the absence of the sperm cell during the artificial oocyte activation affects subsequent embryonic development. The objective of this study was to determine, in bovine embryos, changes in cell cycle-associated transcript levels (cyclin A, cyclin B, cyclin E, CDC2, CDK2, and CDK4) after oocyte activation in the presence or absence of the sperm cell. To evaluate that, in vitro-produced (IVP) and parthenogenetically activated (PA) embryos (2-4 cells (2-4C), 8-16 cells (8-16C) and blastocysts) were evaluated by real-time PCR. There was no difference in cleavage and blastocyst rates between IVP and PA groups. Transcript level was higher in oocytes than in IVP and PA embryos. Cleaved PA embryos showed higher expression of cyclin A, cyclin B, cyclin E, and CDK2 and lower expression of CDC2 when compared with that from the IVP group. At the time of activation, all transcripts were expressed less in PA than in IVP embryos, whereas at the blastocyst stage, almost all genes were expressed at a higher level in the PA group. These results suggest that in both groups there is an initial consumption of these transcripts in the early stages of embryonic development. Furthermore, 8-16C embryos seem to synthesize more cell cycle-related genes than 2-4C embryos. However, in PA embryos, activation of the cell cycle genes seems to occur after the 8- to 16-cell stage, suggesting a failure in the activation process.

The conversion coefficients from air kerma to ICRU operational dose equivalent quantities for ENEA’s realization of the X-radiation qualities L10-L35 of the ISO “Low Air Kerma rate” series (L), N10-N40 of the ISO “Narrow spectrum” series (N) and H10-H60 of the ISO “High Air-kerma rate” (H) series and two beams at 5 kV and 7.5 kV were determined by utilising X-ray spectrum measurements. The pulse-height spectra were measured using a planar high-purity germanium spectrometer (HPGe) and unfolded to fluence spectra using a stripping procedure then validate with using Monte Carlo generated data of the spectrometer response. HPGe portable detector has a diameter of 8.5 mm and a thickness of 5 mm. The entrance window of the crystal is collimated by a 0.5 mm thick Aluminum ring to an open diameter of 6.5 mm. The crystal is mounted at a distance of 5 mm from the Berillium window (thickness 25.4 micron). The Monte Carlo method (MCNP-4C) was used to calculate the efficiency, escape and Compton curves of a planar high-purity germanium detector (HPGe) in the 5-60 keV energy. These curves were used for the determination of photon spectra produced by the X-ray machine SEIFERT ISOVOLT 160 kV in order to allow a precise characterization of photon beams in the low energy range, according to the ISO 4037. The detector was modelled with the MCNP computer code and validated with experimental data. To verify the measuring and the stripping procedure, the first and the second half-value layers and the air kerma rate were calculated from the counts spectra and compared with the values measured using an a free-air ionization chamber. For each radiation quality, the spectrum was characterized by the parameters given in ISO 4037-1. The conversion coefficients from the air kerma to the ICRU operational quantities Hp(10), Hp(0.07), H’(0.07) and H*(10) were calculated using monoenergetic conversion coefficients. The results are discussed with respect to ISO 4037-4, and compared with published results for low-energy X-ray spectra. The main motivation for this work was the lack of a treatment of the low photon energy region (from a few keV up to about 60 keV).

Seyfert galaxies are the closest active galactic nuclei. As such, we can use
them to test the physical properties of the entire class of objects. To investigate
their general properties, I took advantage of different methods of data analysis. In
particular I used three different samples of objects, that, despite frequent overlaps,
have been chosen to best tackle different topics: the heterogeneous BeppoS AX
sample was thought to be optimized to test the average hard X-ray (E above 10 keV)
properties of nearby Seyfert galaxies; the X-CfA was thought the be optimized to
compare the properties of low-luminosity sources to the ones of higher luminosity
and, thus, it was also used to test the emission mechanism models; finally, the
XMM–Newton sample was extracted from the X-CfA sample so as to ensure a
truly unbiased and well defined sample of objects to define the average properties
of Seyfert galaxies.
Taking advantage of the broad-band coverage of the BeppoS AX MECS and
PDS instruments (between ~2-100 keV), I infer the average X-ray spectral propertiesof nearby Seyfert galaxies and in particular the photon index (

Nel lavoro di tesi qui presentato si indaga l'applicazione di tecniche di apprendimento mirate ad una più efficiente esecuzione di un portfolio di risolutore di vincoli (constraint solver). Un constraint solver è un programma che dato in input un problema di vincoli, elabora una soluzione mediante l'utilizzo di svariate tecniche. I problemi di vincoli sono altamente presenti nella vita reale. Esempi come l'organizzazione dei viaggi dei treni oppure la programmazione degli equipaggi di una compagnia aerea, sono tutti problemi di vincoli. Un problema di vincoli è formalizzato da un problema di soddisfacimento di vincoli(CSP). Un CSP è descritto da un insieme di variabili che possono assumere valori appartenenti ad uno specico dominio ed un insieme di vincoli che mettono in relazione variabili e valori assumibili da esse. Una tecnica per ottimizzare la risoluzione di tali problemi è quella suggerita da un approccio a portfolio. Tale tecnica, usata anche in am- biti come quelli economici, prevede la combinazione di più solver i quali assieme possono generare risultati migliori di un approccio a singolo solver. In questo lavoro ci preoccupiamo di creare una nuova tecnica che combina un portfolio di constraint solver con tecniche di machine learning. Il machine learning è un campo di intelligenza articiale che si pone l'obiettivo di immettere nelle macchine una sorta di `intelligenza'. Un esempio applicativo potrebbe essere quello di valutare i casi passati di un problema ed usarli in futuro per fare scelte. Tale processo è riscontrato anche a livello cognitivo umano. Nello specico, vogliamo ragionare in termini di classicazione. Una classicazione corrisponde ad assegnare ad un insieme di caratteristiche in input, un valore discreto in output, come vero o falso se una mail è classicata come spam o meno. La fase di apprendimento sarà svolta utilizzando una parte di CPHydra, un portfolio di constraint solver sviluppato presso la University College of Cork (UCC). Di tale algoritmo a portfolio verranno utilizzate solamente le caratteristiche usate per descrivere determinati aspetti di un CSP rispetto ad un altro; queste caratteristiche vengono altresì dette features. Creeremo quindi una serie di classicatori basati sullo specifico comportamento dei solver. La combinazione di tali classicatori con l'approccio a portfolio sara nalizzata allo scopo di valutare che le feature di CPHydra siano buone e che i classicatori basati su tali feature siano affidabili. Per giusticare il primo risultato, eettueremo un confronto con uno dei migliori portfolio allo stato dell'arte, SATzilla. Una volta stabilita la bontà delle features utilizzate per le classicazioni, andremo a risolvere i problemi simulando uno scheduler. Tali simulazioni testeranno diverse regole costruite con classicatori precedentemente introdotti. Prima agiremo su uno scenario ad un processore e successivamente ci espanderemo ad uno scenario multi processore. In questi esperimenti andremo a vericare che, le prestazioni ottenute tramite l'applicazione delle regole create appositamente sui classicatori, abbiano risultati migliori rispetto ad un'esecuzione limitata all'utilizzo del migliore solver del portfolio. I lavoro di tesi è stato svolto in collaborazione con il centro di ricerca 4C presso University College Cork. Su questo lavoro è stato elaborato e sottomesso un articolo scientico alla International Joint Conference of Articial Intelligence (IJCAI) 2011. Al momento della consegna della tesi non siamo ancora stati informati dell'accettazione di tale articolo. Comunque, le risposte dei revisori hanno indicato che tale metodo presentato risulta interessante.

Bread dough and particularly wheat dough, due to its viscoelastic behaviour, is probably the most dynamic and complicated rheological system and its characteristics are very important since they highly affect final products’ textural and sensorial properties. The study of dough rheology has been a very challenging task for many researchers since it can provide numerous information about dough formulation, structure and processing. This explains why dough rheology has been a matter of investigation for several decades. In this research rheological assessment of doughs and breads was performed by using empirical and fundamental methods at both small and large deformation, in order to characterize different types of doughs and final products such as bread. In order to study the structural aspects of food products, image analysis techniques was used for the integration of the information coming from empirical and fundamental rheological measurements. Evaluation of dough properties was carried out by texture profile analysis (TPA), dough stickiness (Chen and Hoseney cell) and uniaxial extensibility determination (Kieffer test) by using a Texture Analyser; small deformation rheological measurements, were performed on a controlled stress–strain rheometer; moreover the structure of different doughs was observed by using the image analysis; while bread characteristics were studied by using texture profile analysis (TPA) and image analysis. The objective of this research was to understand if the different rheological measurements were able to characterize and differentiate the different samples analysed. This in order to investigate the effect of different formulation and processing conditions on dough and final product from a structural point of view. For this aim the following different materials were performed and analysed: - frozen dough realized without yeast; - frozen dough and bread made with frozen dough; - doughs obtained by using different fermentation method; - doughs made by Kamut® flour; - dough and bread realized with the addition of ginger powder; - final products coming from different bakeries. The influence of sub-zero storage time on non-fermented and fermented dough viscoelastic performance and on final product (bread) was evaluated by using small deformation and large deformation methods. In general, the longer the sub-zero storage time the lower the positive viscoelastic attributes. The effect of fermentation time and of different type of fermentation (straight-dough method; sponge-and-dough procedure and poolish method) on rheological properties of doughs were investigated using empirical and fundamental analysis and image analysis was used to integrate this information throughout the evaluation of the dough’s structure. The results of fundamental rheological test showed that the incorporation of sourdough (poolish method) provoked changes that were different from those seen in the others type of fermentation. The affirmative action of some ingredients (extra-virgin olive oil and a liposomic lecithin emulsifier) to improve rheological characteristics of Kamut® dough has been confirmed also when subjected to low temperatures (24 hours and 48 hours at 4°C). Small deformation oscillatory measurements and large deformation mechanical tests performed provided useful information on the rheological properties of samples realized by using different amounts of ginger powder, showing that the sample with the highest amount of ginger powder (6%) had worse rheological characteristics compared to the other samples. Moisture content, specific volume, texture and crumb grain characteristics are the major quality attributes of bread products. The different sample analyzed, “Coppia Ferrarese”, “Pane Comune Romagnolo” and “Filone Terra di San Marino”, showed a decrease of crumb moisture and an increase in hardness over the storage time. Parameters such as cohesiveness and springiness, evaluated by TPA that are indicator of quality of fresh bread, decreased during the storage. By using empirical rheological tests we found several differences among the samples, due to the different ingredients used in formulation and the different process adopted to prepare the sample, but since these products are handmade, the differences could be account as a surplus value. In conclusion small deformation (in fundamental units) and large deformation methods showed a significant role in monitoring the influence of different ingredients used in formulation, different processing and storage conditions on dough viscoelastic performance and on final product. Finally the knowledge of formulation, processing and storage conditions together with the evaluation of structural and rheological characteristics is fundamental for the study of complex matrices like bakery products, where numerous variable can influence their final quality (e.g. raw material, bread-making procedure, time and temperature of the fermentation and baking).

Der Transferrin-Zyklus ist ein wichtiges Modell für denintrazellulären Transport, daher sollten in der vorliegendenArbeit einzelne, immer noch unverstandene Prozesse desvesikulären, intrazellulären Transportes durch dieCharakterisierung einen in vitro-Transportassay untersuchtwerden. Der Ansatz eines in vitro-Systems wurde deshalbgewählt, um mit Experimenten in denen einzelne Faktoren undbestimmte Konditionen untersucht werden sollten, diese unterdefinierten, reproduzierbaren Konditionen durchzuführen, diein einem in vivo-System kaum zu gewährleisten sind. Ohne denEinfluss von störenden, weil unkontrollierten Faktoren,wie es bei in vivo-Systemen der Fall ist, konnte imvorliegende Ansatz der Transport zu immunisoliertenRecycling-Endosomen (die Isolierung erfolgte hierbei mitanti-Rab11-Antikörpern, einem Marker fürRecycling-Endosomen) unter bestimmten Bedingungen untersuchtwerden. Dabei wurde als Marker Acridinium-markiertesTransferrin gewählt, welches in Zellen internalisiert wurde.Die Spezifität des Transportes in dem zellfreien System warhierbei sehr hoch, wie Kontrollexperimente inImmunisolierungsansätzen ohne Rab11-Antikörper zeigten. ImRahmen einer ersten Charakterisierung des Transportassayswurden essentielle, für den in vivo-Transport essentielleParameter auch in den in vitro-Experimenten untersucht.Hierbei wurde zum einen der Faktor Temperatur gewählt, daTransport in Zellen bei 4°C in der Regel zum Erliegen kommt.Dies konnte auch in dem vorgestellten System gezeigt werden.Ein weiterer, essentieller Faktor ist Energie in Form vonATP. ATP-Depletion wurde in den Experimenten durch Hinzugabeeines ATP-erschöpfenden Systems erzielt. Auch hier zeigteder Transport von Ac-Tfn zu Recycling-Endosomen eine starkeInhibierung. Mit Hilfe des so charakterisierten Assayskonnten anschließend weitere Experimente durchgeführtwerden, die den Einfluss von bestimmten Reagenzien undKonditionen auf den Transport untersuchten. So zeigte derTransport in Zeitverlaufsexperimenten einen Anstieg desTransportes bis 30 Minuten, bei 30 Minuten wurde ein Maximumerreicht. Nach Erreichen dieses Maximums war nachfolgendeine leichte Abnahme des Transfers von Ac-Tfn zu denRecycling-Endosomen zu beobachten. Da Rab-Proteine alsSchlüsselregulatoren für den intrazellulären, vesikulärenTransport gelten, und die Immunisolierungen mitanti-Rab11-Antikörpern durchgeführt wurden, wurde somit auchder Einfluss dieser GTPasen auf das Transportsystemuntersucht. Zugegebenes GDI, welches in der Lage istRab-Proteine in GDP-gebundener Form von spezifischenMembranen zu extrahieren, und daher ein gut untersuchterInhibitor von Rab-Funktionen ist, konnte auch in diesemTransportassay den Transport von Transferrin inhibieren. Einweiterer Aspekt war die Rolle des Cytoskelettes imintrazellulären Transport. Da in früheren Untersuchungen(Trischler et al., 1999) Aktin auf Recycling-Endosomengefunden wurde, erfolgte in diesen Arbeiten eineKonzentration auf die Rolle des Aktin in diesenTransportprozessen. Durch die Zugabe von Cytochalasin D, daseinen Aufbau von Aktingerüsten verhindert, wurde derTransport ebenfalls inhibiert. Durchaffinitätschromatographische Aufreinigungen konnte einestarke Interaktion von Aktin an immobilisiertes Rab11gezeigt werden. Die eluierten Fraktionen, die neben Aktinnoch weitere, jedoch unbekannte Proteine enthielten, konntenin dem in vitro-Fusionsassay eingesetzt werden und führtendort zu einer Stimulation des Transportes. Neben demgefundenen Aktin, könnten somit noch weitere, unbekannteProteine in dem Proteingemisch wichtige Funktionen imintrazellulären, vesikulären Transport übernehmen. EineIdentifizierung dieser Proteine ist dabei für weiterführendeArbeiten essentiell.Caveolin-1, Markerprotein für die Caveolae-Membrandomänewird überraschenderweise von verschiedenen Zellensekretiert. Da Caveolin-1 normalerweise ein integralesMembranprotein ist, wird von einer Sekretion alsLipoproteinpartikel ausgegangen. Die Rolle diesessekretierten Partikels ist unbekannt, wobei einige Autoreneine Funktion als autokrinen/ parakrinen Faktor vorschlagen(Tahir et al., 2001). In der vorliegenden Arbeit solltendiese Partikel daher aufgereinigt und erstmalscharakterisiert werden. Die Partikel wurden aus transienttransfizierten LNCaP-Zellen gewonnen, die Cav-1 in dasserumfreie Medium abgaben. In einer erstenGrößenuntersuchung durch FPLC konnte ein Molekulargewichtzwischen 2.000.000 Da und 660.000 Da bestimmt werden. DieseResultate konnten durch den Ansatz der nativenBlau-Gelelektrophorese bestätigt werden. In einem weiterenAnsatz, der die Dichte der Partikel charakterisieren sollte,wurde in zwei unterschiedlichen Ansätzen (CsCl-, sowieOptiprep Dichtezentrifuagtion) eine ähnliche Dichte desPartikels wie HDL ermittelt. Um eine stärkere Aufreinigungder Partikel zu erzielen, wurde eine Aufreinigung mit Hilfevon Ni-NTA-Agarose durchgeführt. Dies war möglich, denn diebei der Transfektion verwendete C-DNA trug einen His6-tag.Die so aufgereingten Partikel verloren auch nach derNi-NTA-Chromatographie nicht ihre biochemischenEigenschaften, wie in überprüfenden CsCl-Gradienten zu sehenwar. Die Partikel konnten anschließend zum ersten Mal inelektronenmikroskopischen Aufnahmen (Negativkontrastierung)visualisiert werden. Ein weiteres Ziel dieser Arbeit war es,zu untersuchen ob auf Cav-1 Lipoproteinpartikeln nochweitere Proteine zu finden waren. Durch eine kombinierteAufreinigung über Ni-NTA Chromatographie undCsCl-Dichtezentrifugation und im Vergleich mit demAusgangsmaterial konnten in der Silberfärbung Proteinbandenerkannt werden, die wie Cav-1 in den Fraktionen angereichertvorlagen. Eine massenspektroskopische Identifikation einerder Banden ergab, dass es sich hierbei um nm 23(Nukleosid-diphosphat-kinase) handelte, einem Protein dasebenfalls von verschiedenen Tumoren sekretiert wird.

L’imballaggio alimentare si può definire come un sistema coordinato per disporre i beni per il trasporto, la distribuzione, la conservazione, la vendita e l’utilizzo. Uno dei materiali maggiormente impiegati, nell’industria alimentare, per la produzione di imballaggi sono le materie plastiche. Esse sono sostanze organiche derivanti da petrolio greggio, sono composti solidi allo stato finito, ma possono essere modellate allo stato fluido. Un imballaggio alimentare deve svolgere determinate funzioni tra cui: - contenimento del prodotto - protezione del prodotto da agenti esterni - logistica - comunicativa - funzionale - ecologica L'ultimo punto sopracitato è il principale problema delle materie plastiche derivanti dal petrolio greggio. Questi materiali sono difficilmente riciclabili perché spesso un imballaggio è composto da più materiali stratificati o perché si trova a diretto contatto con gli alimenti. Inoltre questi materiali hanno un lungo tempo di degradazione (da 100 a 1000 anni) che ne rendono difficile e costoso lo smaltimento. Per questo nell’ultimo decennio è cominciata la ricerca di un materiale plastico, flessibile alle esigenze industriali e nel contempo biodegradabile. Una prima idea è stata quella di “imitare la natura” cercando di replicare macromolecole già esistenti (derivate da amido e zuccheri) per ottenere una sostanza plastico-simile utilizzabile per gli stessi scopi, ma biodegradabile in circa sei mesi. Queste bioplastiche non hanno preso piede per l’alto costo di produzione e perché risulta impossibile riconvertire impianti di produzione in tutto il mondo in tempi brevi. Una seconda corrente di pensiero ha indirizzato i propri sforzi verso l’utilizzo di speciali additivi aggiunti in minima misura (1%) ai classici materiali plastici e che ne permettono la biodegradazione in un tempo inferiore ai tre anni. Un esempio di questo tipo di additivi è l’ECM Masterbatch Pellets che è un copolimero di EVA (etilene vinil acetato) che aggiunto alle plastiche tradizionali rende il prodotto finale completamente biodegradabile pur mantenendo le proprie caratteristiche. Scopo di questo lavoro di tesi è stato determinare le modificazioni di alcuni parametri qualitativi di nettarine di Romagna(cv.-Alexa®) confezionate-con-film-plastici-tradizionali-e-innovativi. I campioni di nettarine sono stati confezionati in cestini in plastica da 1 kg (sigillati con un film flow-pack macroforato) di tipo tradizionale in polipropilene (campione denominato TRA) o vaschette in polipropilene additivato (campione denominato BIO) e conservati a 4°C e UR 90-95% per 7 giorni per simulare un trasporto refrigerato successivamente i campioni sono stati posti in una camera a 20°C e U.R. 50% per 4 giorni al fine di simulare una conservazione al punto vendita. Al tempo 0 e dopo 4, 7, 9 e 11 giorni sono state effettuate le seguenti analisi: - coefficiente di respirazione è stato misurata la quantità di CO2 prodotta - indice di maturazione espresso come rapporto tra contenuto in solidi solubili e l’acidità titolabile - analisi di immagine computerizzata - consistenza della polpa del frutto è stata misurata attraverso un dinamometro Texture Analyser - contenuto in solidi totali ottenuto mediante gravimetria essiccando i campioni in stufa sottovuoto - caratteristiche sensoriali (Test Accettabilità) Conclusioni In base ai risultati ottenuti i due campioni non hanno fatto registrare dei punteggi significativamente differenti durante tutta la conservazione, specialmente per quanto riguarda i punteggi sensoriali, quindi si conclude che le vaschette biodegradabili additivate non influenzano la conservazione delle nettarine durante la commercializzazione del prodotto limitatamente ai parametri analizzati. Si ritiene opportuno verificare se il processo di degradazione del polimero additivato si inneschi già durante la commercializzazione della frutta e soprattutto verificare se durante tale processo vengano rilasciati dei gas che possono accelerare la maturazione dei frutti (p.e. etilene), in quanto questo spiegherebbe il maggiore tasso di respirazione e la più elevata velocità di maturazione dei frutti conservati in tali vaschette. Alimentary packaging may be defined as a coordinate system to dispose goods for transport, distribution, storage, sale and use. Among materials most used in the alimentary industry, for the production of packaging there are plastics materials. They are organic substances deriving from crude oil, solid compounds in the ended state, but can be moulded in the fluid state. Alimentary packaging has to develop determinated functions such as: - Product conteniment - Product protection from fieleders agents - logistic - communicative - functional - ecologic This last term is the main problem of plastic materials deriving from crude oil. These materials are hardly recyclable because a packaging is often composed by more stratified materials or because it is in direct contact with aliments. Beside these materials have a long degradation time(from 100 to 1000 years) that make disposal difficult and expensive. For this reason in the last decade the research for a new plastic material is begin, to make industrial demands more flexible and, at the same time, to make this material biodegradable: At first, the idea to “imitate the nature” has been thought, trying to reply macromolecules already existents (derived from amid and sugars) to obtain a similar-plastic substance that can be used for the same purposes, but it has to be biodegradable in about six months. These bioplastics haven’t more success bacause of the high production cost and because reconvert production facilities of all over the wolrd results impossible in short times. At second, the idea to use specials addictives has been thought. These addictives has been added in minim measure (1%) to classics plastics materials and that allow the biodegradation in a period of time under three years. An example of this kind of addictives is ECM Masterbatch Pellets which is a coplymer of EVA (Ethylene vinyl acetate) that, once it is added to tradizional plastics, make final product completely biodegradable however maintaining their own attributes. The objective of this thesis work has been to determinate modifications of some Romagna’s Nectarines’ (cv. Alexa®) qualitatives parameters which have been packaged-with traditional and innovative-plastic film. Nectarines’ samples have been packaged in plastic cages of 1 kg (sealed with a macro-drilled flow-pack film) of traditional type in polypropylene (sample named TRA) or trays in polypropylene with addictives (sample named BIO) and conservated at 4°C and UR 90-95% for 7 days to simulate a refrigerated transport. After that, samples have been put in a camera at 20°C and U.R. 50% for 4 days to simulate the conservation in the market point. At the time 0 and after 4, 7, 9 and 11 days have been done the following analaysis: - Respiration coefficient wherewith the amount CO2 producted has been misurated - Maturation index which is expressed as the ratio between solid soluble content and the titratable acidity - Analysis of computing images - Consistence of pulp of the fruit that has been measured through Texture Analyser Dynanometer - Content in total solids gotten throught gravimetry by the drying of samples in vacuum incubator - Sensorial characteristic (Panel Test) Consequences From the gotten results, the two samples have registrated no significative different scores during all the conservation, expecially about the sensorial scores, so it’s possible to conclude that addictived biodegradable trays don’t influence the Nectarines’ conservation during the commercialization of the product qualifiedly to analized parameters. It’s advised to verify if the degradation process of the addicted polymer may begin already during the commercialization of the fruit and in particular to verify if during this process some gases could be released which can accelerate the maturation of fruits (p.e. etylene), because all this will explain the great respiration rate and the high speed of the maturation of fruits conservated in these trays.

A series of imidazolium salts of the type [BocNHCH2CH2ImR]X (Boc = t-Bu carbamates; Im = imidazole) (R = Me, X = I, 1a; R = Bn, X = Br, 1b; R = Trityl, X = Cl, 1c) and [BnImR’]X (R’ = Me, X = Br, 1d; R’ = Bn, X = Br, 1e; R’ = Trityl, X = Cl, 1g; R’ = tBu, X = Br, 1h) bearing increasingly bulky substituents were synthetized and characterized. Subsequently, these precursors were employed in the synthesis of silver(I)-N-heterocyclic (NHC) complexes as transmetallating reagents for the preparation of rhodium(I) complexes [RhX(NBD)(NHC)] (NHC = 1-(2-NHBoc-ethyl)-3-R-imidazolin-2-ylidene; X = Cl; R = Me, 4a; R = Bn, 4b; R = Trityl, 4c; X = I, R = Me, 5a; NHC = 1-Bn-3-R’-imidazolin-2-ylidene; X = Cl; R’ = Me, 4d, R’ = Bn, 4e, R’ = Trityl, 4g; R’ = tBu, 4h). VT NMR studies of these complexes revealed a restricted rotation barriers about the metal-carbene bond. While the rotation barriers calculated for the complexes in which R = Me, Bn (4a,b,d,e and 5a) matched the experimental values, this was not true for the complexes 4c,g, bearing a trityl group for which the values are much smaller than the calculated ones. Energy barriers for 4c,g, derived from a line shape simulation, showed a strong dependence on the temperature while for 4h the rotational energy barrier is stopped at room temperature. The catalytic activity of the new rhodium compounds was investigated in the hydrosilylation of terminal alkynes and in the addition of phenylboronic acid to benzaldehyde. The imidazolium salts 1d,e were also employed in the synthesis of new iron(II)-NHC complexes. Finally, during a six-months stay at the University of York a new ligand derived from Norharman was prepared and employed in palladium-mediated cross-coupling.
