Tantalum coatings are of particular interest today as promising candidates to replace potentially hazardous electrodeposited chromium coatings for tribological and corrosion resistant applications, such as the internal lining on large-caliber gun barrels. Tantalum coatings have two crystalline phases, α-Ta (body-centered-cubic) and β-Ta (metastable tetragonal) that exhibit relatively different properties. Alpha-Ta is typically preferred for wear and corrosion resistant applications and unfortunately, is very difficult to deposit without the assistance of substrate heating or post-annealing treatments. Furthermore, there is no general consensus on the mechanism which causes α or β to form or if there is a phase transition or transformation from β → α during coating deposition. In this study, modulated pulsed power (MPP) magnetron sputtering was used to deposit tantalum coatings with thicknesses between 2 and 20 μm without external substrate heating. The MPP Ta coatings showed good adhesion and low residual stress. This study shows there is an abrupt β → α phase transition when the coating is 5–7 μm thick and not a total phase transformation. Thermocouple measurements reveal substrate temperature increases as a function of deposition time until reaching a saturation temperature of ~ 388 °C. The importance of substrate temperature evolution on the β → α phase transition is also explained.

Aquaporins and Rh proteins can function as gas (CO2 and NH3) channels. The present study explores the urea, H2O, CO2, and NH3 permeability of the human urea transporter B (UT-B) (SLC14A1), expressed in Xenopus oocytes. We monitored urea uptake using [14C]urea and measured osmotic water permeability (Pf) using video microscopy. To obtain a semiquantitative measure of gas permeability, we used microelectrodes to record the maximum transient change in surface pH (∆pHS) caused by exposing oocytes to 5% CO2/33 mM HCO3- (pHS increase) or 0.5 mM NH3/NH4+ (pHS decrease). UT-B expression increased oocyte permeability to urea by >20-fold, and Pf by 8-fold vs. H2O-injected control oocytes. UT-B expression had no effect on the CO2-induced ∆pHS but doubled the NH3-induced ∆pHS. Phloretin reduced UT-B-dependent urea uptake (Jurea * ) by 45%, Pf * by 50%, and (- ∆pHS * )NH3 by 70%. p-Chloromercuribenzene sulfonate reduced Jurea * by 25%, Pf * by 30%, and (∆pHS * )NH3 by 100%. Molecular dynamics (MD) simulations of membrane-embedded models of UT-B identified the monomeric UT-B pores as the main conduction pathway for both H2O and NH3 and characterized the energetics associated with permeation of these species through the channel. Mutating each of two conserved threonines lining the monomeric urea pores reduced H2O and NH3 permeability. Our data confirm that UT-B has significant H2O permeability and for the first time demonstrate significant NH3 permeability. Thus the UTs become the third family of gas channels. Inhibitor and mutagenesis studies and results of MD simulations suggest that NH3 and H2O pass through the three monomeric urea channels in UT-B.

Epithelial cells are mainly responsible for the formation of tissues that cover the external and internal surfaces of organs like skin, lining of the lungs and intestines. The cells must adhere to substrates and to each other in compliance with certain stimulus. In this way, adhesion properties can be regulated by the cell which simultaneously senses the chemical and mechanical properties of its environment. Their adhesion and growth on biomaterials depends on substrate properties such as surface wettability, topography and chemistry. The aim of this study is to investigate cell-surface interactions using several materials and different surfaces.

Canned tuna is one of the most widespread and recognizable fish commodities in the world. Over all oceans 80% of the total tuna catches are caught by purse seine fishery and in tropical waters their target species are: yellowfin (Thunnus albacares), bigeye (Thunnus obesus) and skipjack (Katsuwonus pelamis). Even if this fishing gear is claimed to be very selective, there are high levels of by-catch especially when operating under Fish Aggregating Devices (FADs). The main problem is underestimation of by-catch data. In order to solve this problem the scientific community has developed many specific programs (e.g. Observe Program) to collect data about both target species and by-catch with observers onboard. The purposes of this study are to estimate the quantity and composition of target species and by-catch by tuna purse seiner fishery operating in tropical waters and to underline a possible seasonal variability in the by-catch ratio (tunas versus by-catch). Data were collected with the French scientific program ”Observe” on board of the French tuna purse seiner “Via Avenir” during a fishing trip in the Gulf of Guinea (C-E Atlantic) from August to September 2012. Furthermore some by-catch specimens have been sampled to obtain more information about size class composition. In order to achieve those purposes we have shared our data with the French Institute of Research for the Development (IRD), which has data collected by observers onboard in the same study area. Yellowfin tuna results to be the main specie caught in all trips considered (around 71% of the total catches) especially on free swimming schools (FSC) sets. Instead skipjack tuna is the main specie caught under FADs. Different percentages of by-catch with the two fishing modes are observed: the by-catch incidence is higher on FADs sets (96.5% of total by-catch) than on FSC sets (3.5%) and the main category of by-catch is little-tuna (73%). When pooling data for both fishing sets used in purse seine fishery the overall by-catch/catch ratio is 5%, a lower level than in other fishing gears like long-lining and trawling.

In dieser Arbeit wird eine Einteilung der degenerativen strukturellen Veränderungen der Synovialmembran vorgestellt. Anhand der Kriterien Fibrosierung des Stromas, Rückgang des Gefäßnetzes, Auftreten von Hyalinose und chondroider Metaplasie mit und ohne Nachweis von CPPD Kristallen wurden Präparate der Synovialmembran von 59 Patienten mit Nachweis degenerativer strukturellen Veränderungen in 4 Stadien eingeteilt. rnHyalinose (Stadium 3) konnte in den untersuchten Schnitten nur relativ selten beobachtet werden, so dass am ehesten von einem Vorstadium zur chondroiden Metaplasie auszugehen ist. rnDie Verteilung der Erkrankungsdauer und des Alters in den verschiedenen Stadien lassen darauf schließen, dass höhere Stadien mit höherem Alter und längerer Erkrankungsdauer korrelieren. rnAus der vorhandenen Literatur ergeben sich Hinweise, welche Faktoren zu der Entstehung der strukturellen Veränderungen beitragen können: rnAus dem Netzwerk der Zytokine scheinen TGF-beta und die BMP’s an der Zunahme der Fibrose und an der Entstehung chondroider Metaplasie beteiligt zu sein. Makrophagen scheinen dabei eine wichtige Rolle zu spielen. Dies weist darauf hin, dass entzündliche und strukturelle Veränderungen miteinander vernetzt sind. rnBei der Entstehung der chondroiden Metaplasie kommen zusätzlich mechanische Einflüsse in Form von zyklischen Kompressionen als Einflussfaktor in Frage. rnDie Regulierung der Angiogenese ist noch zu wenig verstanden, um den Gefäßrückgang bei fortgeschrittenen strukturellen Veränderungen zu erklären. Erklärungsansätze sind zum einen zunehmende mechanische Schädigung bei zunehmender Inkongruenz der Gelenkflächen. Zum anderen könnte eine beginnende chondroide Metaplasie mit Expression von Chondromodulin I eine entscheidende Rolle spielen. rnInsgesamt muss man davon ausgehen, dass die zunehmenden strukturellen Veränderungen die Ernährung des Knorpels erschweren. Dabei ist an erster Stelle der Rückgang des Gefäßnetzes zu nennen. Dies erschwert nicht nur die Versorgung mit Nährstoffen, sondern auch den Abtransport von Stoffwechselprodukten. Ab einem gewissen Punkt ist aber auch davon auszugehen, dass die Funktion der Deckzellschicht beeinträchtigt wird. Wenn die Konzentration der Hyaluronsäure in der Synovia dadurch sinkt, kann dies durch eine vermehrte Permeabilität der Synovialmembran zum verstärkten Ausstrom von Wasser aus der Gelenkhöhle führen. Durch ein Ödem des umliegenden Gewebes kann dadurch der Blutfluss im Bereich des Gelenks zusätzlich vermindert werden. rnAuch die zunehmende Fibrosierung der Synovialmembran kann einen Einfluss auf die Permeabilität der Synovialmembran haben. Ob und in welchen Stadien der Veränderungen das einen relevanten Einfluss für die Ernährung der Chondrozyten hat, ist noch unklar.rn

Il sito archeologico di Suasa è stato oggetto di una lunga campagna di scavi iniziata negli anni sessanta, che ha portato alla pubblicazione di diversi saggi e ha stimolato l’attenzione di numerosi studiosi nel corso degli ultimi anni. Questo interesse, però, rimane un fenomeno ristretto agli addetti al settore e non vede una vera riflessione sul piano turistico. Suasa, infatti, rimane un gioiello culturale scarsamente conosciuto nel territorio ed escluso dai principali percorsi turistici. Il primo obiettivo del nostro intervento è quello di porre l’attenzione su un’area di così grande interesse e avvicinare le persone, esperti di storia e non, all’antica città, in modo che questa possa acquisire una nuova vita e ritrovare una sua valenza nel territorio. Il primo passo in questa direzione è stato quello di mettere in relazione il parco archeologico con gli altri siti archeologici della zona inserendolo in un sistema di offerta culturale volto all’esplorazione e all’approfondimento del territorio marchigiano e della sua storia. La posizione di Suasa sul fiume Cesano, inoltre, risulta particolarmente favorevole ad una integrazione dell’area con i percorsi cicloturistici, in quanto fornisce la possibilità di creare un parco fluviale di supporto a queste nuove tratte di collegamento, così come alle esistenti, andando ad aggiungere valore all’area. Questo elemento risulterà un fattore chiave per la collocazione e la definizione del visitor center del parco archeologico, il quale fa del rapporto tra il sito archeologico e il territorio il suo punto fondante. L’organizzazione del parco richiama quella che era l’organizzazione della città romana, cercando di restituire al visitatore non tanto l’immagine precisa di una ricostruzione, ma la logica e la successione degli spazi che l’impianto urbano poteva assumere. Gli interventi sulle emergenze, quindi, sono realizzati seguendo una coerenza progettuale e materica volta a minimizzare l’impatto sull’archeologia e nel contesto.

Il percorso progettuale intrapreso ci ha permesso di confrontarci con un’analisi del territorio e della storia romana, che ne ha influenzato l’evoluzione. Considerando tutti i frammenti come parte di un sistema più grande dove non è possibile comprenderne uno se non attraverso gli altri, si è deciso di valorizzare con una strategia diversa con criteri ben precisi. Così viene ristabilita la rete di connessioni e relazioni che secondo noi più aiutano nella comprensione del sito e della sua storia, cercando di integrare nel contesto la varietà di sistemi archeologici, storici e culturali che caratterizzano il sito. I resti della città romana vengono trattati con approcci diversi, a seconda del grado di conoscenza e di esperienza che possiamo avere di ogni reperto. Per le tracce meno evidenti sono stati usati interventi più leggeri, reversibili, mentre per le rovine di cui abbiamo maggiori informazioni, sono stati proposti interventi più strutturati, che accompagnino il visitatore nella loro scoperta e comprensione. Le tracce di cui non si hanno molte informazioni, sono state trattate con segni molto più leggeri e reversibili, usando una strategia di lining out che comporti l’impiego di essenze, scelte in base ad associazioni con quello che vanno a simboleggiare. Per le emergenze archeologiche evidenti, è stato pensato un percorso interno che possa renderle fruibili, lasciando le rovine più intatte possibile, mentre l’intervento di musealizzazione più importante è destinato alla domus, che contiene i resti più rilevanti e quelli che necessitano una maggiore protezione.

L’esigenza di contestualizzare le rovine di Suasa Senonum all’interno di un Parco Archeologico che racconti la composizione della città e le relazioni che questa ha intessuto con il territorio, ha condotto alla formulazione di un progetto territoriale; il Parco Archeologico è così inteso come un’occasione per rilanciare la conoscenza di un sistema che connette aspetti culturali, storici e paesaggistici. La città è intersecata da uno dei percorsi più antichi e importanti della Regione: la Salaria Gallica che attraversa città, borghi, vallate, fiumi e parchi archeologici tra cui quello di Suasa Senonum e offre la possibilità di leggere le dinamiche che hanno modellato il territorio sin dall’antichità. Il punto di forza di questa via consiste nell’offrire la possibilità di leggere come la natura e l’uomo abbiano interagito nel tempo e di come esso eserciti la propria opera modellatrice sul risultato delle loro relazioni. Queste considerazioni hanno portato ad affrontare temi che pongono l’accento su questioni che vanno dalle connessioni territoriali all’aggiunta di stratificazione sullo spessore del tempo culminando in un risultato che riconsegna la città alla collettività. Il progetto traduce queste riflessioni affrontando la costruzione di un ponte sul fiume Cesano quale punto di ricucitura della via Salaria Gallica, dell’inserimento di un visitor centre e della rivitalizzazione dell’anfiteatro quale luogo di spettacolo. Il filo conduttore del tutto, dal lining out all’opera land art, sta nella volontà di intaccare il meno possibile quell’equilibrio con cui la natura ha preso il sopravvento sulle attività umane di un tempo celandone le tracce; la sfida è stata quella di rendere l’organismo urbano nuovamente leggibile attraverso il progetto di architettura. Abbiamo così cercato di reinserire in un circuito ritrovato ciò che il tempo ha in parte cancellato.

AIM: To describe a method of carrying out apical surgery of a maxillary molar using ultrasonics to create a lateral sinus window into the maxillary sinus and an endoscope to enhance visibility during surgery. SUMMARY: A 37-year-old female patient presented with tenderness to percussion of the maxillary second right molar. Root canal treatment had been undertaken, and the tooth restored with a metal-ceramic crown. Radiological examination revealed an apical radiolucency in close proximity to the maxillary sinus. Apical surgery of the molar was performed through the maxillary sinus, using ultrasonics for the osteotomy, creating a window in the lateral wall of the maxillary sinus. During surgery, the lining of the sinus was exposed and elevated without perforation. The root-end was resected using a round tungsten carbide drill, and the root-end cavity was prepared with ultrasonic retrotips. Root-end filling was accomplished with MTA(®) . An endoscope was used to examine the cut root face, the prepared cavity and the root-end filling. No intraoperative or postoperative complications were observed. At the 12-month follow-up, the tooth had no clinical signs or symptoms, and the radiograph demonstrated progressing resolution of the radiolucency. KEY LEARNING POINTS: When conventional root canal retreatment cannot be performed or has failed, apical surgery may be considered, even in maxillary molars with roots in close proximity to the maxillary sinus. Ultrasonic sinus window preparation allows more control and can minimize perforation of the sinus membrane when compared with conventional rotary drilling techniques. The endoscope enhances visibility during endodontic surgery, thus improving the quality of the case.

Inheritance of a mutant allele of the von Hippel-Lindau tumor suppressor gene predisposes affected individuals to develop renal cysts and clear cell renal cell carcinoma. Von Hippel-Lindau gene inactivation in single renal tubular cells has indirectly been showed by immunohistochemical staining for the hypoxia-inducible factor alpha target gene product carbonic anhydrase IX. In this study we were able to show von Hippel-Lindau gene deletion in carbonic anhydrase IX positive nonneoplastic renal tubular cells, in epithelial cells lining renal cysts and in a clear cell renal cell carcinoma of a von Hippel-Lindau patient. This was carried out by means of laser confocal microscopy and immunohistochemistry in combination with fluorescence in situ hybridization. Carbonic anhydrase IX negative normal renal tubular cells carried no von Hippel-Lindau gene deletion. Furthermore, recent studies have indicated that the von Hippel-Lindau gene product is necessary for the maintenance of primary cilia stability in renal epithelial cells and that disruption of the cilia structure by von Hippel-Lindau gene inactivation induces renal cyst formation. In our study, we show a significant shortening of primary cilia in epithelial cells lining renal cysts, whereas, single tubular cells with a von Hippel-Lindau gene deletion display to a far lesser extent signs of cilia shortening. Our in vivo results support a model in which renal cysts represent precursor lesions for clear cell renal cell carcinoma and arise from single renal tubular epithelial cells owing to von Hippel-Lindau gene deletion.

Lung macrophages, that is, the intravascular, interstitial, pleural, and surface macrophages, are part of the mononuclear phagocyte system. They are derived from the hematopoietic stem cell in the bone marrow with the monocytes as their putative precursors. Macrophages residing on the inner surfaces of the lungs and immersed within the lung lining layer, that is, the alveolar and the airway macrophages, are constantly exposed to the environment; it is those cells that are recognized as first line of cellular host defense.

Because the recommendation to use flowables for posterior restorations is still a matter of debate, the objective of this study was to determine in a nationwide survey in Germany how frequently, for what indications, and for what reasons, German dentists use flowable composites in posterior teeth. In addition, the acceptance of a simplified filling technique for posterior restorations using a low stress flowable composite was evaluated. Completed questionnaires from all over Germany were returned by 1,449 dentists resulting in a response rate of 48.5%; 78.6% of whom regularly used flowable composites for posterior restorations. The most frequent indications were cavity lining (80.1%) and small Class I fillings (74.2%). Flowables were less frequently used for small Class II fillings (22.7%) or other indications (13.6%). Most frequent reasons given for the use of flowables in posterior teeth were the prevention of voids (71.7%) and superior adaptation to cavity walls (72.9%), whereas saving time was considered less important (13.8%). Based on the subjective opinion of the dentists the simplified filling technique seemed to deliver advantages compared to the methods used to date particularly with regard to good cavity adaptation and ease of use. In conclusion, resin composites are the standard material type used for posterior restorations by general dental practitioners in Germany and most dentists use flowable composites as liners.

Mechanical ventilation is not only a life saving treatment but can also cause negative side effects. One of the main complications is inflammation caused by overstretching of the alveolar tissue. Previously, studies investigated either global strains or looked into which states lead to inflammatory reactions in cell cultures. However, the connection between the global deformation, of a tissue strip or the whole organ, and the strains reaching the single cells lining the alveolar walls is unknown and respective studies are still missing. The main reason for this is most likely the complex, sponge-like alveolar geometry, whose three-dimensional details have been unknown until recently. Utilizing synchrotron-based X-ray tomographic microscopy, we were able to generate real and detailed three-dimensional alveolar geometries on which we have performed finite-element simulations. This allowed us to determine, for the first time, a three-dimensional strain state within the alveolar wall. Briefly, precision-cut lung slices, prepared from isolated rat lungs, were scanned and segmented to provide a three-dimensional geometry. This was then discretized using newly developed tetrahedral elements. The main conclusions of this study are that the local strain in the alveolar wall can reach a multiple of the value of the global strain, for our simulations up to four times as high and that thin structures obviously cause hotspots that are especially at risk of overstretching.

With the increasing advances in hip joint preservation surgery, accurate diagnosis and assessment of femoral head and acetabular cartilage status is becoming increasingly important. Magnetic resonance imaging (MRI) of the hip does present technical difficulties. The fairly thin cartilage lining necessitates high image resolution and high contrast-to-noise ratio (CNR). With MR arthrography (MRA) using intraarticular injected gadolinium, labral tears and cartilage clefts may be better identified through the contrast medium filling into the clefts. However, the ability of MRA to detect varying grades of cartilage damage is fairly limited and early histological and biochemical changes in the beginning of osteoarthritis (OA) cannot be accurately delineated. Traditional MRI thus lacks the ability to analyze the biological status of cartilage degeneration. The technique of delayed gadolinium-enhanced MRI of cartilage (dGEMRIC) is sensitive to the charge density of cartilage contributed by glycosaminoglycans (GAGs), which are lost early in the process of OA. Therefore, the dGEMRIC technique has a potential to detect early cartilage damage that is obviously critical for decision-making regarding time and extent of intervention for joint-preservation. In the last decade, cartilage imaging with dGEMRIC has been established as an accurate and reliable tool for assessment of cartilage status in the knee and hip joint.This review outlines the current status of dGEMRIC for assessment of hip joint cartilage. Practical modifications of the standard technique including three-dimensional (3D) dGEMRIC and dGEMRIC after intra-articular gadolinium instead of iv-dGEMRIC will also be addressed.
