AbstractFilms obtained by blends between starch and other polymers and films developed with the addition of an oil can show higher water vapor barriers and improved mechanical properties. Films with starch/PVOH/alginate were obtained by adding copaiba and lemongrass essential oils (EOs). Films without oil served as the control. The microstructure, water vapor permeability (PVA), mechanical properties, and antifungal activity were determined for the films. The effects of the addition of the EOs on the properties of the films were dependent of the concentration and type of oil. The films with 0.5% lemongrass EO were similar to the control films. These films showed a 2.02 × 10-12 g s-1Pa m-1 PVA, 11.43 MPa tensile stress, 13.23% elongation, and 247.95 MPa/mm resistance at perforation. The addition of 1% of copaiba EO increased the PVA from 0.5 × 10-12 to 12.1 × 10-12 g s-1 Pa m-1 and the diffusion coefficient from 0.17 × 10-8 to 7.15 × 10-8m2/day. Films with quantities of EOs displayed fissures and micropores; the control films developed micropores with smaller diameters than films with EOs. The addition of EOs did not change the resulting infrared spectrum of the films. The films with oil displayed a diminished development of the Fusarium sp. culture, and the film without EOs did not display notable differences in the development of the culture. The starch/PVOH/alginate films with 0.5% lemongrass EO were the most suited for the development of a packaging active system.

Suurlujuusteräskonstruktioiden suunnittelu eroaa perinteisten rakenneterästen vastaavasta lukuisten lisähuomiota vaativien ilmiöiden ja seikkojen osalta, jotka kytkeytyvät toisiinsa rakenteen valmistettavuuden ja toiminnallisuuden kautta. Lujuuden sallima vapaampi suunnittelu esimerkiksi ohuempine materiaalivahvuuksineen ja näennäisesti vapaampine elementtigeometrioineen kumuloituu suurlujuusteräsmateriaalin kautta vaativampana työstettävyytenä. Tämä heijastuu suunnittelijalle materiaalin pienen murtovenymän ja käytettävissä olevat laitteistoresurssien kautta. Suurlujuusterästen muovaaminen vaatii perinteisiin rakenneteräksiin nähden suurempaa voimankäyttöä, mutta pieni murtovenymä rajoittaa saavutettavissa olevia muotoja. Materiaalivahvuuksien muuttuessa vaatii hitsausliitosten suunnittelu esimerkiksi railomuotojen uudelleensuunnittelua, sekä lämmöntuonnin aiheuttamien muutosten tarkempaa huomioimista. Näiden yhteensovittaminen ohjaa suunnittelijan poikkeuksetta hakemaan ratkaisuja erilaisista hitsausaineista ja -prosesseista. Suuren lujuuden ja pienen murtovenymän suurlujuusteräselementtien mekaanisten ominaisuuksien ja erilaisen elementtigeometrian yhdistelmänä kumuloituva rakennetekninen käyttäytyminen eroaa selvästi perinteisistä teräslaaduista. Yksittäisten elementtien suhteen korostuvat esimerkiksi stabiliteettiomiinaisuudet, sekä kokonaisten rakenteiden suhteen liitosratkaisujen vaikutus koko konstruktion toimintaan. Onnistuneella suurlujuusteräsuunnittelulla saavutettuja etuja ovat käyttökohteesta riippuen esimerkiksi kymmenien prosenttien massa- ja kustannussäästöjen kautta kumuloituvat konstruktion toiminnalliset edut. Suuren lujuuden terästen materiaaliteknisten erityispiirteiden suhteen ei kuitenkaan ole tarjolla perinteisiä rakenneteräksiä vastaavaa määrää yhteisesti sovittuja standardoituja suunnitteluohjeita. Tämän johdosta korostuu suunnittelijan kyky hahmottaa teräsmateriaalin lujuusluokan taakse piiloutuvat ilmiöt, sekä materiaalitekniset erityispiirteet ja niiden kytkeytyminen toisiinsa valmistettavuuden ja toiminnallisuuden kautta.

The prevalence of inflammatory based diseases has increased in industrialized countries over the last decades. For allergic diseases, two primary hypotheses have been proposed to explain this phenomenon, namely the hygiene and dietary evolution based hypothesis. Particularly, the reduced early exposure to microbes and an increase in the amount of polyunsaturated fatty acids (especially n-6 PUFA) in the diet have been discussed. Often, these two factors have been studied independently, even though both factors have been shown to possess potential health benefits and their mode of action to share similar mechanisms. The hypothesis of the present study was that demonstrate that PUFA and probiotics are not separate entities as such but do interact with each other. In the present study, we investigated whether maternal diet and atopic status influence the PUFA composition of breast milk and serum fatty acids of infants, and whether the fatty acid absorption and utilization of infant formula fatty acids is affected by supplementation of infant formula with probiotic bacteria (Lactobacillus GG and Bifidobacterium lactis Bb-12). Moreover, we investigated the mechanisms by which different PUFA influence the physicochemical and functional properties of probiotics as well as functionality of epithelial cells in vitro. We demonstrated a carry-over effect of dietary fatty acids from maternal diet via breast milk into infants’ serum lipid fatty acids. Our data confirmed the previously shown allergy –related PUFA level imbalances, though it did not fully support the impaired desaturation and elongation capacity hypothesis. We also showed that PUFA incorporation into phospholipids of infants was influenced by probiotics in infant formula in a strain dependent manner. Especially,Bifidobacterium lactis Bb-12 in infant formula promoted the utilization of n-3 PUFA. Mechanistically, we demonstrated that probiotics (Lactobacillus GG, Lactobacillus casei Shirota and Lactobacillus bulgaricus) did incorporate and interconvert exogenous free PUFA in the growth medium into bacterial fatty acids strain and PUFA dependently. In general, high concentrations of free PUFA inhibited the growth and mucus adhesion of probiotics, whereas low concentrations of specific long chain PUFA were found to promote the growth and mucus adhesion of Lactobacillus casei Shirota. These effects were paralleled with only minor alterations in hydrophobicity and electron donor – electron acceptor properties of lactobacilli. Furthermore, free PUFA were also demonstrated to alter the adhesion capacity of the intestinal epithelial cells; n-6 PUFA tended to inhibit the Caco-2 adhesion of probiotics, whereas n-3 PUFA had either no or minor effects or even promote the bacterial adhesion (especially Lactobacillus casei Shirota) to PUFA treated Caco-2 cells. The results of this study demonstrate the close and bilateral interactions between dietary PUFA and probiotics. Probiotics were shown to influence the absorption and utilization of dietary PUFA, whereas PUFA were shown to alter the functional properties of both probiotics and mucosal epithelia. These findings suggest that a more thorough understanding of interactions between PUFA and intestinal microbiota is a prerequisite, when the beneficial effects of new functional foods containing probiotics are designed and planned for human intervention studies.

Työssä tutkitaan PI-säätimen käyttöä dynaamisessa kireydensäädössä ilman varsinaista kireyden takaisinkytkentää. Kireyttä säädetään epäsuorasti käyttämällä takaisinkytkentätietona kahden telan välistä paikkaeroa. Kireyssäädin toteutetaan nopeussäätimen rinnalle. Rinnakkaisrakenteella pyritään kireyden muutoksiin nopeasti reagoivaan säätöratkaisuun. Rakenne toteutetaan osaksi taajuusmuuttajan säätöketjua. Työssä esitetään telasysteemin simulointimalli, jonka toimivuus varmistetaan käytännön mittauksin. Lisäksi työssä arvioidaan kireyssäädön toimintaa dynaamisessa kireydensäädössä simulointien ja testilaitteistolla suoritettavien mittausten perusteella.

The objective of this study was to evaluate the effect of the ethanolic extract of Serjania lethalis leaves and stems on the diaspore germination and seedling growth of wild poinsettia (Euphorbia heterophylla) and barnyardgrass (Echinochloa crus-galli). The crude ethanolic extract was prepared from 100 g of dry plant material dissolved in 500 ml of ethanol. The extracts were solubilized in a buffer solution containing dimethyl sulfoxide (DMSO) at concentrations of 10.0, 7.5, 5.0 and 2.5 mg mL-1. The effect of these extracts was compared with herbicide oxyfluorfen in bioassays. The ethanolic extracts of S. lethalis leaves and stems inhibited the germination and seedling growth of barnyardgrass and wild poinsettia in a concentration-dependent manner. The reduction in the root length of E. heterophylla seedlings might be attributed to the reduced elongation of metaxylem cells. The phytotoxicity of the extracts ranged according to the receptor species, and for some variables, the inhibitory effect was similar, and even superior, to that of the commercial herbicide. Thus, S. lethalis extracts might be a promising alternative for sustainable weed management.

Työn tavoitteena oli tutkia lujan nuorrutetun painelaiteteräksen P500QL2 hitsattavuutta ja koehitsausten avulla löytää optimaaliset hitsausparametrit ja lämmöntuonti teräksen hitsaukseen. Työn tavoitteena oli myös selvittää ja käsitellä kaikkien painelaiteterässtandardissa esitettyjen lujien painelaiteterästen hitsauksessa huomioon otettavia asioita. Työn teoriaosuudessa käsitellään lujien painelaiteterästen hitsauksessa huomioitavia erityispiirteitä, kuten lämmöntuontia, jäähtymisaikaa, esilämmitystä sekä hitsausaineiden valintaa. Lisäksi teoriaosuudessa käsitellään painelaitteiden valmistusta, painelaiteterässtandardiin kuuluvia lujia painelaiteteräksiä sekä keinoja lujien terästen hitsattavuuden arviointiin. Työn kokeellisessa osassa tutkittiin aineenvahvuudeltaan 50 mm paksun P500QL2-teräksen päittäisliitoksen mekaanisia ominaisuuksia eri lämmöntuonneilla hitsattuna. Kokeellisessa osassa tutkittiin myös myöstön poisjättämisen vaikutuksia liitoksen mekaanisiin ominaisuuksiin. Mekaanisia ominaisuuksia tutkittiin toteuttamalla koekappaleiden aineenkoetus menetelmäkoestandardin vaatimuksia soveltaen. Tutkimuksessa käytettyjä testausmenetelmiä olivat silmämääräinen tarkastus, magneettijauhetarkastus, ultraäänitarkastus, mikro- ja makrorakennetarkastelu, kovuuskokeet, vetokokeeet ja iskukokeet. Testauksessa saatujen tulosten avulla lujan painelaiteteräksen P500QL2 hitsaukseen laadittiin alustava hitsausohje. Hitsausliitosten testauksessa saatujen tulosten perusteella havaittiin hitsien lujuuden ja kovuuden laskevan lämmöntuonnin kasvaessa. Hitsausliitosten iskusitkeysominaisuudet olivat erinomaiset vielä suurellakin lämmöntuonnilla, mutta liitosten murtovenymäarvot laskivat lämmöntuonnin kasvaessa. Myöstön havaittiin parantavan hitsin mekaanisia ominaisuuksia huomattavasti. Tutkimuksen tulosten perusteella painelaiteteräs P500QL2 on hitsattavissa suurella lämmöntuonnilla ja suurella tuottavuudella liitoksen täyttäessä painelaitevalmistuksen edellyttämät vaatimukset.

The allelopathic effect studied in many cultures has currently generated great expectations that displayed a natural and environmentally friendly tool for weed management using bioherbicides. The objective of this work was to assess allelopathic influence of residues of S. trilobata on the germination and growth of weeds, as well as their relation with some crops and effects on soil properties. Results show that residues from S. trilobata have inhibited the germination of weeds (31.6 - 72%), increasingly with the applied dose. All residue doses of this specie have inhibited dicotyledonous germination, but only maximum concentration has affected monocotyledons. The residues did not affect onion germination, but stimulated it in radish and tomato, while the dose applied at 50% produced tomato stimulation and inhibition of cabbage. The effects of residues on hypocotyl growth in different crops showed changes in species response. For onion, the three doses had negative effects on the growth of hypocotyl, while tomato was stimulated. For radish, the growth was hindered by any dose applied, and were only different (50 and 100%) compared to control. For cabbage, only hypocotyl length was stimulated, when maximum dose (100%) was applied. For the radicle growth, in onion and radish no differences were found compared to control. While the tomato radicle growth was inhibited, in cabbage, all doses encouraged the elongation of the radicle. The dry mass of weed was affected by increased dose of residue (0.49 - 8.8 g m-2), however the soil microflora was stimulated, while the population of Azotobacter spp. was not affect. Some soil properties were affected, the level of organic material, Na+ and electrical conductivity were increased, while pH (H2O) decreased a bit, however it remained basic.

The structure of the fruit and seed in development of Chorisia speciosa are described with the main purpose of clarifying the origin and nature of the hairs that cover the seeds and aiding future taxonomical and ecological studies of the group. The fruit is an ellipsoid loculicide capsule and presents the exocarp formed by 7-10 cells layers, with very thick walls and evident simple pits. A great number of mucilage secretory cavities and ramified vascular bundles, accompanied by fibers, occur in the parenchymatic mesocarp. The endocarp derives from the ventral epidermis of the ovary wall, whose cells undergo a gradual elongation, become lignified, and constitute the trichomes which cover the mature seeds. The fruit aperture occurs by means of a suture evident in the ovarian wall in the middle region of the carpel leaf. Anatropous and bitegmic ovules, provided by a hypostase, give rise to campilotropous and bitegmic seeds. The testa is uniseriate, the exotegmen is completely formed by macrosclereids, and mucilage secretory cavities occur in the mesotegmen. The endotegmen, which is differentiated in the endothelium, is crushed in the mature seed. The plicate embryo, which occupies practically the entire seminal cavity, is found between endosperm layers, both being rich in lipids.

Changes in the polyamine content were analyzed in different embryo developmental stages and tissues during seed development in the conifer Araucaria angustifolia (Bert.) O. Ktze. Free polyamine contents varied according to the tissue and stage of embryo development, the highest levels occurring in the embryonic axis at the early stages, when putrescine and spermidine were most abundant. The levels of spermidine were higher from the stage where cotyledons arise, whereas putrescine decreases. The putrescine/spermine+spermidine ratio was higher during the initial phases of seed development, corresponding to cell multiplication and elongation, with a decrease in the final stages, corresponding to stabilization of the dry matter content.

In Velloziaceae, the number of subsidiary cells has been used to characterize species and support groups. Nevertheless, the homology of the stomatal types have not been scrutinized. Stomatal ontogenesis of Vellozia epidendroides and V. plicata, assigned to have tetracytic stomata, and of V. glauca and Barbacenia riparia, assigned to have paracytic stomata, were investigated. In the four species studied, stomata followed perigenic development. Subsidiary cells arise from oblique divisions of neighbouring cells of the guard mother cell (GMC). These cells are elongated and parallel to the longer axis of the stoma. Polar cells show wide variation, following the shape and size of the epidermal cells in the vicinity. Hence, these cells cannot be called subsidiary cells. This wide variation is due to a much higher density of stomata in some regions of the leaf blade. This distribution of stomata forces the development of short polar cells, leading to an apparently tetracytic stomata. In regions of low concentration of stomata, higher spatial availability between the GMCs allows the elongation of polar cells, leading to evident paracytic stomata. Therefore, the four studied species are considered braquiparacytic, questioning the classification of stomata into tetracytic and paracytic in Velloziaceae.

In this work we attempted to characterize the diaspores and the germination process of Piper aduncum L., as well as to verify the influence of the interaction between presence and absence of light (photoperiod of 12 hours and dark) and temperature (25 °C, 30 °C and 20-30 °C) and also of gibberellin (0, 50, 100, 200 and 400 mg L-1) on the root protrusion and normal seedlings formation. The diaspores are very small with a thousand seed weight of 0.3645 g, 13% moisture and protein reserve. Diaspores are strict positively photoblastic in the tested temperature range and the optimum temperature for root protrusion was 30 °C, while for normal seedlings was 25 °C. The previous permanence in the dark led to an increase in the speed of root protrusion and percentage and speed of seedling formation. The application of gibberellic acid negatively interfered with the protrusion and growth of the radicle while favoring the elongation of hypocotyls.

Axon growth and guidance represent complex biological processes in which probably intervene diverse sets of molecular cues that allow for the appropriate wiring of the central nervous system (CNS). The extracellular matrix (ECM) represents a major contributor of molecular signals either diffusible or membrane-bound that may regulate different stages of neural development. Some of the brain ECM molecules form tridimensional structures (tunnels and boundaries) that appear during time- and space-regulated events, possibly playing relevant roles in the control of axon elongation and pathfinding. This short review focuses mainly on the recognized roles played by proteoglycans, laminin, fibronectin and tenascin in axonal development during ontogenesis.

The Ca2+-modulated, dimeric proteins of the EF-hand (helix-loop-helix) type, S100A1 and S100B, that have been shown to inhibit microtubule (MT) protein assembly and to promote MT disassembly, interact with the type III intermediate filament (IF) subunits, desmin and glial fibrillary acidic protein (GFAP), with a stoichiometry of 2 mol of IF subunit/mol of S100A1 or S100B dimer and an affinity of 0.5-1.0 µM in the presence of a few micromolar concentrations of Ca2+. Binding of S100A1 and S100B results in inhibition of desmin and GFAP assemblies into IFs and stimulation of the disassembly of preformed desmin and GFAP IFs. S100A1 and S100B interact with a stretch of residues in the N-terminal (head) domain of desmin and GFAP, thereby blocking the head-to-tail process of IF elongation. The C-terminal extension of S100A1 (and, likely, S100B) represents a critical part of the site that recognizes desmin and GFAP. S100B is localized to IFs within cells, suggesting that it might have a role in remodeling IFs upon elevation of cytosolic Ca2+ concentration by avoiding excess IF assembly and/or promoting IF disassembly in vivo. S100A1, that is not localized to IFs, might also play a role in the regulation of IF dynamics by binding to and sequestering unassembled IF subunits. Together, these observations suggest that S100A1 and S100B may be regarded as Ca2+-dependent regulators of the state of assembly of two important elements of the cytoskeleton, IFs and MTs, and, potentially, of MT- and IF-based activities.

Additive manufacturing is a fast growing manufacturing technology capable of producing complex objects without the need for conventional manufacturing process planning. During the process the work piece is built by adding material one layer at a time according to a digital 3D CAD model. At first additive manufacturing was mainly used to make prototypes but the development of the technology has made it possible to also make final products. Welding is the most common joining method for metallic materials. As the maximum part size of additive manufacturing is often limited, it may sometimes be required to join two or more additively manufactured parts together. However there has been almost no research on the welding of additively manufactured parts so far, which means that there has been very little information available on the possible differences compared to the welding of sheet metal parts. The aim of this study was to compare the weld joint properties of additively manufactured parts to those of sheet metal parts. The welding process that was used was TIG welding and the test material was 316L austenitic stainless steel. Weld joint properties were studied by making tensile, bend and hardness tests and by studying the weld microstructures with a microscope. Results show that there are certain characteristics in the welds of additively manufactured parts. The building direction of the test pieces has some impact on the mechanical properties of the weld. Nevertheless all the welds exhibited higher yield strength than the sheet metal welds but at the same time elongation at break was lower. It was concluded that TIG welding is a feasible process for welding additively manufactured parts.

Greenhouse gases emitted from energy production and transportation are dramatically changing the climate of Planet Earth. As a consequence, global warming is affecting the living conditions of numerous plant and animal species, including ours. Thus the development of sustainable and renewable liquid fuels is an essential global challenge in order to combat the climate change. In the past decades many technologies have been developed as alternatives to currently used petroleum fuels, such as bioethanol and biodiesel. However, even with gradually increasing production, the market penetration of these first generation biofuels is still relatively small compared to fossil fuels. Researchers have long ago realized that there is a need for advanced biofuels with improved physical and chemical properties compared to bioethanol and with biomass raw materials not competing with food production. Several target molecules have been identified as potential fuel candidates, such as alkanes, fatty acids, long carbon‐chain alcohols and isoprenoids. The current study focuses on the biosynthesis of butanol and propane as possible biofuels. The scope of this research was to investigate novel heterologous metabolic pathways and to identify bottlenecks for alcohol and alkane generation using Escherichia coli as a model host microorganism. The first theme of the work studied the pathways generating butyraldehyde, the common denominator for butanol and propane biosynthesis. Two ways of generating butyraldehyde were described, one via the bacterial fatty acid elongation machinery and the other via partial overexpression of the acetone‐butanol‐ethanol fermentation pathway found in Clostridium acetobutylicum. The second theme of the experimental work studied the reduction of butyraldehyde to butanol catalysed by various bacterial aldehyde‐reductase enzymes, whereas the final part of the work investigated the in vivo kinetics of the cyanobacterial aldehyde deformylating oxygenase (ADO) for the generation of hydrocarbons. The results showed that the novel butanol pathway, based on fatty acid biosynthesis consisting of an acyl‐ACP thioesterase and a carboxylic acid reductase, is tolerant to oxygen, thus being an efficient alternative to the previous Clostridial pathways. It was also shown that butanol can be produced from acetyl‐CoA using acetoacetyl CoA synthase (NphT7) or acetyl‐CoA acetyltransferase (AtoB) enzymes. The study also demonstrated, for the first time, that bacterial biosynthesis of propane is possible. The efficiency of the system is clearly limited by the poor kinetic properties of the ADO enzyme, and for proper function in vivo, the catalytic machinery requires a coupled electron relay system.
