Mature berries of Pinot Noir grapevines were sampled across a latitudinal gradient in Europe, from southern Spain to central Germany. Our aim was to study the influence of latitude-dependent environmental factors on the metabolite composition (mainly phenolic compounds) of berry skins. Solar radiation variables were positively correlated with flavonols and flavanonols and, to a lesser extent, with stilbenes and cinnamic acids. The daily means of global and erythematic UV solar radiation over long periods (bud break-veraison, bud break-harvest, and veraison-harvest), and the doses and daily means in shorter development periods (5–10 days before veraison and harvest) were the variables best correlated with the phenolic profile. The ratio between trihydroxylated and monohydroxylated flavonols, which was positively correlated with antioxidant capacity, was the berry skin variable best correlated with those radiation variables. Total flavanols and total anthocyanins did not show any correlation with radiation variables. Air temperature, degree days, rainfall, and aridity indices showed fewer correlations with metabolite contents than radiation. Moreover, the latter correlations were restricted to the period veraison-harvest, where radiation, temperature, and water availability variables were correlated, making it difficult to separate the possible individual effects of each type of variable. The data show that managing environmental factors, in particular global and UV radiation, through cultural practices during specific development periods, can be useful to promote the synthesis of valuable nutraceuticals and metabolites that influence wine quality.

Berry size and crop yield are widely recognized as important factors that contribute to wine quality. The final berry size indirectly affects the phenolic concentration of the wine due to skin surface-to-berry volume ratio. The effects of different irrigation levels, soil management and plant crop level on growth of ‘Trincadeira’ berries were studied. In order to test the influence of different irrigation levels (rainfed, pre-veraison and post-veraison), different soil management (tillage and natural cover crops) and different plant crop levels (8 and 16 clusters per vine), leaf water potential, skin anthocyanin, polyphenols, berry skin and seed fresh weight were measured in fruits. The segregation of berries into three different berry classes: small, medium and large, allowed to identify different levels of contribution of soil management and irrigation level into berry, skin and seeds ratios. As expected, higher water availability due to irrigation and soil tillage management during berry development induced an increase in berry flesh weight and this was more evident in larger berries; however, berry skin and seed fresh weight remained unchanged. Also, anthocyanins did not show significant differences.

A vitivinicultura no Rio Grande do Sul tem reconhecimento no cenário enológico nacional e a Campanha Gaúcha tem sua participação neste processo. Com vinhedos implantados na década de 70, no município de Santana do Livramento- RS, para produção de uvas finas, a região tem apresentado crescente desenvolvimento com instalação de novas vinícolas. Apesar dos benefícios climáticos, a Campanha apresenta invernos irregulares, tanto na distribuição como na qualidade do frio, provocando brotações irregulares, baixa produtividade e desuniformidade na produção dos vinhedos. Buscando novas tecnologias para melhorar a qualidade de brotação e consequente qualidade produtiva, o presente trabalho objetivou avaliar o comportamento de brotação de diferentes cultivares de Vitis vinifera em relação às diferentes épocas de poda com a interação de indutores químicos de brotação.

A videira é uma planta caducifólia, perde suas folhas e inicia a endodormência com as primeiras horas de frio (HF, horas em temperatura <7,2°C) no outono. Portanto, suprindo a exigência de HF, que pode ser distinta entre genótipos, a planta tende a uma brotação plena e uniforme e pode ser um parâmetro de seleção para as condições locais de cultivo. O trabalho tem por objetivo caracterizar a evolução do potencial de brotação de clones de Moscato Branco ao longo do período da endodormência.

A Campanha Gaúcha vem sendo classificada entre as regiões vitivinícolas mais promissoras no Rio Grande do Sul, especialmente pelas suas condições edafoclimáticas com maior restrição hídrica e drenagem do solo. Apesar do início da vitivinicultura nesta região datar da década de 70, a maior intensificação dos plantios e produção ocorreu nos últimos dez anos, porém sem muitos subsídios técnicos. Os objetivos deste trabalho foram avaliar os efeitos da antecipação da poda hibernal e concentrações de cianamida hidrogenada (CH) sobre o potencial produtivo da uva ?Merlot?/SO4 conduzida nos sistemas de poda Guyot Duplo (DG) e Cordão Esporonado (CE).

In organic and biodynamic vineyards, canopy management practices should be carefully and timely modulated, particularly in a context of climate change, for successfully achieving balanced plants, ventilated and exposed berries, elevated grape and wine quality. In 2013 and 2014, characterized by contrasting climatic conditions, the implications of post-veraison (late) or pea-size trimming, post-veraison or pre-harvest late defoliations and shoot-positioning (post-veraison) were assessed against long-shoots non treated controls, under field conditions on organically-cultivated cv. Sangiovese. The key agronomic and enological relevance of late trimming and defoliations clearly emerged in both seasons. Berry skin phenolics (e.g. anthocyanins, flavonols) increased markedly, without changes in technological parameters. In case of early trimming, such positive effects were observed only in 2013. Maintaining long shoots for shading decreased anthocyanins, flavonols and total phenolics concentrations and promoted the production of compact bunches. Experimental data strongly designated late trimming, a practice proved to contain yield and bunch compactness, as a valuable alternative to cluster thinning. Late trimming, defoliations and shoot positioning reduced the severity of Botrytis cluster rot. The highest levels of berry skins phenolic compounds in late trimmed and defoliated plants could have contributed control the severity of this pathogen. The enological benefits induced by late trimming and defoliations and shoot positioning emerged in both young and aged wines. For the first time, cell cultures from cv. Sangiovese berry tissues were obtained and enabled to investigate, in controlled conditions, the relations between mechanisms regulating secondary metabolism in grapevine cells and changes induced by environmental and agronomic factors. The Doctoral Dissertation strongly highlights the need to consider, for a proper interpretation of the multiple modifications induced by canopy management strategies, physiological mechanisms other than the canonic source-sink relationships, in particular their impact on the vine hormonal status.

Il massiccio utilizzo di fungicidi in Viticoltura e i loro effetti negativi sulla salute dell’uomo e dell’ambiente sollecitano lo sviluppo di strategie agronomiche e di sistemi agrari innovativi. Il Lavoro di Tesi ha investigato l’influenza della cv Isabella su cultivars di Vitis vinifera in campo, in Fari Agroecologici, sistemi agrari frutto di una gestione partecipativa. L'esperimento ha dimostrato, per la prima volta, che la cultivar Isabella, nota per la sua tolleranza ai principali patogeni fungini, può contenere l’Incidenza della peronospora in cultivars di Vitis vinifera consociate e contigue. Studi precedenti dimostrano che i VOCs emessi dalla cv Isabella sono in grado di ridurre la presenza di patogeni fungini. La ricerca pioneristica ha evidenziato il ruolo chiave della biodiversità nella gestione dell’ecosistema vigneto, nella prospettiva della progettazione di sistemi viticoli agroecologici.

È necessario intraprendere una via maggiormente ecosostenibile e vantaggiosa per gestire le problematiche presenti in vivaio, e in generale in vigneto, adottando tecniche ammesse in agricoltura biologica o, meglio ancora, con una visione agroecologica della gestione. Le osservazioni condotte presso i Vivai Maiorana hanno indagato la diversa suscettibilità di genotipi di vite ai principali patogeni fungini e gli effetti della consociazione sui sintomi causati da tali patogeni. In primo luogo si è osservata la differenza di suscettibilità tra le cultivars di Merlot e la varietà portinnesto Kober 5BB. I genotipi di Vitis vinifera analizzati si sono dimostrati suscettibili, a differenza del portinnesto Kober 5BB che è risultato completamente esente da sintomi. In seguito, le osservazioni si sono concentrate nel lotto del barbatellaio, in cui tre filari di Calabrese e tre filari di Nerello Mascalese sono separati da un filare di Isabella. Le osservazioni svolte hanno permesso di indagare gli effetti della consociazione, sfruttando l’attuale impostazione del vivaio, in cui diversi genotipi di vite vengono coltivate a stretto contatto. Si è osservata una variazione dei sintomi di peronospora su Vitis vinifera in relazione alla distanza dalla varietà Isabella. L’effetto è presumibilmente mediato da VOCs specifici. Questi benefici, se opportunamente sfruttati, consentirebbero di ridurre sensibilmente la necessità di intervenire con prodotti esogeni nella gestione del barbatellaio, aumentando la possibilità di impostare una produzione biologica che soddisfi le necessità della viticoltura moderna, con vantaggi agronomici ed ecologici.

A considerable portion of Brazil's commercial eucalypt plantations is located in areas Subjected to periods of water deficit and grown in soils with low natural fertility, particularly poor In potassium. Potassium is influential in controlling water relations of plants. The objective of this study was to verify the influence of potassium fertilization and soil water potential (psi(w)) oil the dry matter production and oil water relations Of eucalypt seedlings grown under greenhouse conditions. the experimental units were arranged in 4x4x2 randomized blocks factorial design, as follow: four species of Eucalyptus (Eucalyptus grandis, Eucalyptus urophylla, Eucalyptus camaldulensis and hybrid Eucalyptus grandis x Eucalyptus urophylla), four dosages of K (0, 50, 100 and 200 mg dm(-3)) and two soil water potentials (-0.01 M Pa and -0.1 M Pa). Plastic containers with 15 cm diameter and 18 cm height, with Styrofoam base, containing 3.0 dm(3) of soil and two plants per container were used. Soil water potential was kept at -0.01 MPa for 40 days after seeding. Afterward, the experimental units were divided into two groups: in one group the potential was kept at 0.01 MPa, and in the other one, at -0.10 MPa. Sol I water potential was control led gravimetrically twice a day with water replacement until the desired potential was reestablished. A week before harvesting, the leaf water potential (psi), the photosynthetic rate (A), the stomatal conductance (gs) and the transpiration rate were evaluated. The last week before harvesting, the mass of the containers was recorded daily before watering to determine the consumption of water by the plants. After harvesting, total dry matter and leaf area were evaluated. the data were Submitted to analysis of variance, to Tukey's tests and regression analyses. The application of K influenced A, gs and the transpiration rate. Plants deficient in K showed lower A and higher Us and transpiration rates. There were no statistical differences in A, gs and transpiration rates ill plants with and Without water deficit. The addition of K reduced the consumption of water per unit of leaf area and, in general, plants submitted to water deficit presented a lower consumption of water.

Background: High-throughput SNP genotyping has become an essential requirement for molecular breeding and population genomics studies in plant species. Large scale SNP developments have been reported for several mainstream crops. A growing interest now exists to expand the speed and resolution of genetic analysis to outbred species with highly heterozygous genomes. When nucleotide diversity is high, a refined diagnosis of the target SNP sequence context is needed to convert queried SNPs into high-quality genotypes using the Golden Gate Genotyping Technology (GGGT). This issue becomes exacerbated when attempting to transfer SNPs across species, a scarcely explored topic in plants, and likely to become significant for population genomics and inter specific breeding applications in less domesticated and less funded plant genera. Results: We have successfully developed the first set of 768 SNPs assayed by the GGGT for the highly heterozygous genome of Eucalyptus from a mixed Sanger/454 database with 1,164,695 ESTs and the preliminary 4.5X draft genome sequence for E. grandis. A systematic assessment of in silico SNP filtering requirements showed that stringent constraints on the SNP surrounding sequences have a significant impact on SNP genotyping performance and polymorphism. SNP assay success was high for the 288 SNPs selected with more rigorous in silico constraints; 93% of them provided high quality genotype calls and 71% of them were polymorphic in a diverse panel of 96 individuals of five different species. SNP reliability was high across nine Eucalyptus species belonging to three sections within subgenus Symphomyrtus and still satisfactory across species of two additional subgenera, although polymorphism declined as phylogenetic distance increased. Conclusions: This study indicates that the GGGT performs well both within and across species of Eucalyptus notwithstanding its nucleotide diversity >= 2%. The development of a much larger array of informative SNPs across multiple Eucalyptus species is feasible, although strongly dependent on having a representative and sufficiently deep collection of sequences from many individuals of each target species. A higher density SNP platform will be instrumental to undertake genome-wide phylogenetic and population genomics studies and to implement molecular breeding by Genomic Selection in Eucalyptus.

High yields of hemicellulosic and cellulosic sugars are critical in obtaining economical conversion of agricultural residues to ethanol. To optimize pretreatment conditions, we evaluated oxalic acid loading rates, treatment temperatures and times in a 2(3) full factorial design. Response-surface analysis revealed an optimal oxalic acid pretreatment condition to release sugar from the cob of Zea mays L ssp. and for Pichia stipitis CBS 6054. To ferment the residual cellulosic sugars to ethanol following enzymatic hydrolysis, highest saccharification and fermentation yields were obtained following pretreatment at 180 degrees C for 50 min with 0.024 g oxalic acid/g substrate. Under these conditions, only 7.5% hemicellulose remained in the pretreated substrate. The rate of cellulose degradation was significantly less than that of hemicellulose and its hydrolysis was not as extensive. Subsequent enzymatic saccharification of the residual cellulose was strongly affected by the pretreatment condition with cellulose hydrolysis ranging between 26.0% and 76.2%. The residual xylan/lignin ratio ranged from 0.31 to 1.85 depending on the pretreatment condition. Fermentable sugar and ethanol were maximal at the lowest ratio of xylan/lignin and at high glucan contents. The model predicts optimal condition of oxalic acid pretreatment at 168 degrees C, 74 min and 0.027 g/g of oxalic acid. From these findings, we surmised that low residual xylan was critical in obtaining maximal glucose yields from saccharification. Published by Elsevier Ltd.

Two new Tetranychidae (Prostigmata) mites are described from Brazil Monoceronychus tchecensis n. sp., a bryobiine collected from weeping fingergrass, Eustachys distichophylla (Lag.) Nees (Poaceae), in the State of Rio Grande do Sul; and Oligonychus fileno n. sp., a tetranychine collected from grape, Vitis vinifera L. (Vitaceae), in the State of Minas Gerais. Monoceronychus tchecensis n. sp. is the second species in this genus described from South America. In addition to the description of these new species, the tetranychine Eotetranychus smithi Pritchard and Baker, 1955 was recorded for the first time for South America.
