Preparative high-speed counter-current chromatography (HSCCC) was successfully applied for separation and purification of sesquiterpenoids from an extract of Tussilago farfara L. with a two-phase solvent system composed of n-hexane-ethyl acetate- methanol-water (1:0.5:1.1:0.3, v/v/v/v). The separation produced a total of 32 mg of tussilagone, 18 mg of 14-acetoxy-7β-(3'-ethyl cis-crotonoyloxy)-lα-(2'-methyl butyryloxy)-notonipetranone and 21 mg of 7β-(3'-ethyl cis-crotonoyloxy)-lα-(2'- methyl butyryloxy)-3,14-dehydro-Z-notonipetranone from 500 mg of the crude extract in one step separation with the purity of 99.5, 99.4 and 99.1%, respectively, as determined by HPLC. The structures of these compounds were identified by ESI-MS, ¹H-NMR and 13C-NMR.

Supercritical fluid extraction was used to extract active compounds from the Chinese traditional medicinal D. dasycarpus under the pressure of 30 MPa and temperature of 45 ºC. Further separation and purification was established by high-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (1:0.8:1.3:0.9, volume ratio). The separation yielded a total of 47 mg of dictamnine, 24 mg of obacunone and 83 mg of fraxinellone from 1.0 g of the crude extract in one step separation with the purity of 99.2, 98.4 and 99.0%, respectively, as determined by HPLC. The chemical structures of these compounds were identified by ESI-MS, IR, ¹H-NMR and 13C-NMR.

Electron paramagnetic resonance (EPR) spectroscopy was used to study the main structural accommodations of spin labels in bilayers of saturated phosphatidylcholines with acyl chain lengths ranging from 16 to 22 carbon atoms. EPR spectra allowed the identification of two distinct spectral components in thermodynamic equilibrium at temperatures below and above the main phase transition. An accurate analysis of EPR spectra, using two fitting programs, enabled determination of the thermodynamic profile for these major probe accommodations. Focusing the analysis on two-component EPR spectra of a spin-labeled lipid, the influence of 40 mol % cholesterol in DPPC was studied.

The technique of pH-zone-refining counter-current chromatography was successfully applied to preparatively separate three C19-diterpenoid alkaloids from the crude extracts of Aconitum carmichaelii for the first time using a two-phase solvent system of petroleum ether-ethyl acetate-methanol-water (5:5:1:9, v/v/v/v). Mesaconitine (I), hypaconitine (II), and deoxyaconitine (III) were obtained from 2.5 g of the crude alkaloids in a one-step separation; the yields were 4.16%, 16.96%, and 5.05%, respectively. The purities of compounds I, II, and III were 93.0%, 95%, and 96%, respectively, as determined by HPLC. The chemical structures of the three compounds were identified by electrospray ionization mass spectrometry (ESI-MS) and NMR.

An effective method for the rapid separation and purification of three stilbenes from the radix of Polygonum cillinerve (Nakai) Ohwl by macroporous resin column chromatography combined with high-speed counter-current chromatography (HSCCC) was successfully established. In the present study, a two-phase solvent system composed of chloroform-n-butanol-methanol-water (4:1:4:2, v/v/v/v) was used for HSCCC separation. A one-step separation in 4 h from 150 mg of crude extract produced 26.3 mg of trans-resveratrol-3-O-glucoside, 42.0 mg of pieceid-2"-O-gallate, and 17.9 mg of trans-resveratrol with purities of 99.1%, 97.8%, and 99.4%, respectively, as determined by high-performance liquid chromatography (HPLC). The chemical structures of these compounds were identified by nuclear magnetic resonance (NMR) spectroscopy.

Adsorbents functionalized with chelating agents are effective in removal of heavy metals from aqueous solutions. Important properties of such adsorbents are high binding affinity as well as regenerability. In this study, aminopolycarboxylic acid, EDTA and DTPA, were immobilized on the surface of silica gel, chitosan, and their hybrid materials to achieve chelating adsorbents for heavy metals such as Co(II), Ni(II), Cd(II), and Pb(II). New knowledge about the adsorption properties of EDTA- and DTPA-functionalizedadsorbents was obtained. Experimental work showed the effectiveness, regenerability, and stability of the studied adsorbents. Both advantages and disadvantages of the adsorbents were evaluated. For example, the EDTA-functionalized chitosan-silica hybrid materials combined the benefits of the silica gel and chitosan while at the same time diminishing their observed drawbacks. Modeling of adsorption kinetics and isotherms is an important step in design process. Therefore, several kinetic and isotherm models were introduced and applied in this work. Important aspects such as effect of error function, data range, initial guess values, and linearization were discussed and investigated. The selection of the most suitable model was conducted by comparing the experimental and simulated data as well as evaluating the correspondence between the theory behind the model and properties of the adsorbent. In addition, modeling of two-component data was conducted using various extended isotherms. Modeling results for both one- and twocomponent systems supported each other. Finally, application testing of EDTA- and DTPA-functionalized adsorbents was conducted. The most important result was the applicability of DTPA-functionalized silica gel and chitosan in the capturing of Co(II) from its aqueous EDTA-chelate. Moreover, these adsorbents were efficient in various solution matrices. In addition, separation of Ni(II) from Co(II) and Ni(II) and Pb(II) from Co(II) and Cd(II) was observed in two- and multimetal systems. Lastly, prior to their analysis, EDTA- and DTPA-functionalized silica gels were successfully used to preconcentrate metal ions from both pure and salty waters

This thesis is about development broadband feed for two-mirror antenna system that match following requirements: beamwidth from 45 to 90 degrees at -3 dB level, circular polarization, absence of radiation to the lower hemisphere area. Literature review was done in the areas of the UWB antennas creation. During the work attempts were made to create a feed in a form of the quad ridged horn and "eleven" antennas. The latter is introduced as the most effective feed among all antennas discussed in thesis. Radiation patterns and other results for "eleven" antenna were obtained. Results were saved as far field sources and placed slightly below focal point into the two-mirror antenna system, because phase center of the “eleven” antenna is predominantly shifted upwards. Directivity patterns for the two-mirror system were obtained and the conclusions about the work results have been made

In this work, Sr2FeMoO6 (SFMO) thin films were studied with the main focus on their magnetic and magneto-transport properties. The fabrication process of pulsed laser deposited SFMO films was first optimized. Then the effects of strain, film thickness and substrate were thoroughly investigated. In addition to these external factors, the effect of intrinsic defects on the magnetic properties of SFMO were also clarified. Secondly, the magnetoresistivity mechanims of SFMO films were studied and a semiempirical model of the temperature dependence of resistivity was introduced. The films were grown on single crystal substrates using a ceramic target made with sol-gel method. The structural characterization of the films were carried out with X-ray diffraction, atomic force microscopy, transmission electron microscopy and high kinetic energy photoelectron spectroscopy. The magnetic properties were measured with SQUID magnetometer and the magneto-transport properties by magnetometer with a resistivity option. SFMO films with the best combination of structural and magnetic properties were grown in Ar atmosphere at 1050 °C . Their magnetic properties could not be improved by the ex situ post-annealing treatments aside from the treatments in ultra-high vacuum conditions. The optimal film thickness was found to be around 150 nm and only small improvement in the magnetic properties with decreasing strain was observed. Instead, the magnetic properties were observed to be highly dependent on the choice of the substrate due to the lattice mismatch induced defects, which are best avoided by using the SrTiO3 substrate. The large difference in the Curie temperature and the saturation magnetization between the SFMO thin film and polycrystalline bulk samples was connected to the antisite disorder and oxygen vacancies. Thus, the Curie temperature of SFMO thin films could be improved by increasing the amount of oxygen vacancies for example with ultra-high vacuum treatments or improving the B-site ordering by further optimization of the deposition parameters. The magneto-transport properties of SFMO thin films do not follow any conventional models, but the temperature dependence of resistivity was succesfully described with a model of two spin channel system. Also, evidences that the resistivity-temperature behaviour of SFMO thin films is dominated by the structural defects, which reduce the band gap in the majority spin band were found. Moreover, the magnetic field response of the resistivity in SFMO thin films were found to be superposition of different mechanisms that seems to be related to the structural changes in the film.

En 1997, dans un souci de justice sociale et de solidarité entre tous les québécois, le gouvernement a instauré un Régime général d’assurance médicaments (RGAM) devant permettre de fournir à moindre coût et de manière équitable, des médicaments prescrits assurant une meilleure prise en charge de la maladie des individus. Depuis sa mise en place, le rythme soutenu d’évolution des coûts des médicaments remboursés par le Régime est le sujet d’un nombre croissant de débats. Le Québec ne fait pas figure d’exception car au cours des dernières décennies, la croissance des dépenses de médicaments des régimes d’assurance médicaments des pays industrialisés a connu un rythme de progression élevé, cela malgré l’instauration successive de mécanismes de régulation ciblant une utilisation optimale des médicaments et un meilleur contrôle de la trajectoire de leurs dépenses. La recherche propose une réflexion sur les cadres communs d’action dans lesquels ces outils de régulation sont implantés et évoluent. Elle s’intéresse à l’ensemble des facteurs relationnels et contextuels qui les constituent. Le devis de recherche s’appuie sur une étude de cas unique qu’est le processus menant à l’utilisation du médicament prescrit couvert par le RGAM. Il se compose de trois unités d’analyse identifiées comme étant des cibles importantes de régulation : l’inscription du médicament à la liste du RGAM, la prescription et l’accès au médicament couvert par le Régime. La perspective conceptuelle retenue est celle de l’analyse stratégique de Crozier et Friedberg (1987). La démarche analytique s’appuie sur des entrevues réalisées auprès des régulateurs, de fabricants de médicaments brevetés et de génériques, de médecins, de pharmaciens et de patients impliqués dans l’utilisation des médicaments remboursés par le RGAM et sur une revue du cadre réglementaire concernant ces acteurs, le médicament d’ordonnance et le RGAM. La recherche décrit et analyse la structuration du processus menant à l’utilisation du médicament couvert par le RGAM. Elle conduit au développement d’un construit organisationnel- le système d’action concret- fondé sur les interdépendances des cinq groupes d’acteurs et les relations de pouvoir qu’ils entretiennent entre eux. Ceux-ci évoluent dans des contextes organisationnels singuliers constitués d’enjeux, de stratégies et de ressources. Ils développent diverses stratégies pour accroître leurs possibilités d’action (capacités stratégiques) face aux problèmes qu’ils ont à résoudre. Les capacités stratégiques, inégales entre les groupes d’acteurs, découlent des relations de pouvoir qu’ils exercent entre eux et les amènent à occuper des positions différentes dans le système d’action concret. La recherche démontre qu’en fonction des capacités stratégiques dont ils disposent, les acteurs sont portés à s’approprier certaines règles en tentant de les influencer, de les contourner ou de ne pas les respecter. La connaissance empirique des cadres communs d’action permet d’établir les bases d’un système d’action concret dans un contexte de « phénomène organisation » (Friedberg, 1997). Cette connaissance d’une part, établit que les retombées de certaines stratégies d’acteurs peuvent s’avérer peu compatibles avec les objectifs du RGAM et, d’autre part, pose les limites de certains mécanismes de régulation en vigueur tout en questionnant les façons de concevoir la régulation dans un système complexe.

Le sommeil est un besoin vital et le bon fonctionnement de l’organisme dépend de la quantité et de la qualité du sommeil. Le sommeil est régulé par deux processus : un processus circadien qui dépend de l’activité des noyaux suprachiasmatiques de l’hypothalamus et qui régule le moment durant lequel nous allons dormir, et un processus homéostatique qui dépend de l’activité neuronale et se reflète dans l’intensité du sommeil. En effet, le sommeil dépend de l’éveil qui le précède et plus l’éveil dure longtemps, plus le sommeil est profond tel que mesuré par des marqueurs électroencéphalographiques (EEG). Des études ont montré que le bon fonctionnement de ces deux processus régulateurs du sommeil dépend de la plasticité synaptique. Ainsi, les éléments synaptiques régulant la communication et la force synaptique sont d’importants candidats pour agir sur la physiologie de la régulation du sommeil. Les molécules d’adhésion cellulaire sont des acteurs clés dans les mécanismes de plasticité synaptique. Elles régulent l’activité et la maturation des synapses. Des études ont montré que leur absence engendre des conséquences similaires au manque de sommeil. Le but de ce projet de thèse est d’explorer l’effet de l’absence de deux familles de molécule d’adhésion cellulaire, les neuroligines et la famille des récepteur Eph et leur ligand les éphrines dans les processus régulateurs du sommeil. Notre hypothèse est que l’absence d’un des membres de ces deux familles de molécule affecte les mécanismes impliqués dans le processus homéostatique de régulation du sommeil. Afin de répondre à notre hypothèse, nous avons étudié d’une part l’activité EEG chez des souris mutantes n’exprimant pas Neuroligine‐1 (Nlgn1) ou le récepteur EphA4 en condition normale et après une privation de sommeil. D’autre part, nous avons mesuré les changements moléculaires ayant lieu dans ces deux modèles après privation de sommeil. Au niveau de l’activité EEG, nos résultats montrent que l’absence de Nlgn1 augmente la densité des ondes lentes en condition normale et augment l’amplitude et la pente des ondes lentes après privation de sommeil. Nlgn1 est nécessaire au fonctionnement normal de la synchronie corticale, notamment après une privation de sommeil, lui attribuant ainsi un rôle clé dans l’homéostasie du sommeil. Concernant le récepteur EphA4, son absence affecte la durée du sommeil paradoxal ainsi que l’activité sigma qui dépendent du processus circadien. Nos résultats suggèrent donc que ce récepteur est un élément important dans la régulation circadienne du sommeil. Les changements transcriptionnels en réponse à la privation de sommeil des souris n’exprimant pas Nlgn1 et EphA4 ne sont pas différents des souris sauvages. Toutefois, nous avons montré que la privation de sommeil affectait la distribution des marques épigénétiques sur le génome, tels que la méthylation et l’hydroxyméthylation, et que l’expression des molécules régulant ces changements est modifiée chez les souris mutantes pour le récepteur EphA4. Nos observations mettent en évidence que les molécules d’adhésion cellulaire, Nlgn1 et le récepteur EphA4, possèdent un rôle important dans les processus homéostatique et circadien du sommeil et contribuent de manière différente à la régulation du sommeil.

It is observed that reclamation of natural rubber latex based rubber using 2,2'-dibenzamidodiphenvldisulphide as reclaiming agent is an optional methodology for recycling of waste latex rubber (WLR). For progressive replacement of virgin natural rubber by the reclaim, two alternatives curing system were investigated: adjustment or reduction of the curing system with increasing reclaim content, to compensate for the extra amount of curatives brought along by the reclaim. For fixed curing system, as if the reclaim were equivalent to virgin NR. The cure behavior, final crosslink density and distribution, mechanical properties, and dynamic viscoelastic properties of the blends with reclaimed WLR are measured and compared with the virgin compound. The morphology of the blends, sulfur migration, and final distribution are analyzed.The mechanical and dynamic viscoelastic properties deteriorate for both curing systems, but to a lesser extent for fixed curing system compared to adjusted curing system. With the fixed cure system, many properties like tensile strength and compression set do still deteriorate, but tan 6 and Mrrr„/Murxr, representative for the rolling resistance of tires are improved. On the other hand, with the adjusted cure system both mechanical and dynamic properties still deteriorate.

This thesis has focused on the synthesis and analysis of some important phosphors (nano, bulk and thin film) for display applications. ACTFEL device with SrS:Cu as active layer was also fabricated.Three bulk phosphors: SrS:Cu,CI; SrS:Dy,Cl; and SrS:Dy,Cu,Cl were synthesized and their structural, optical and electrical properties were investigated. Special emphasis was given to, the analysis of the role of defects and charge compensating centers, on the structural changes of the host and hence the luminance. A new model describing the sensitizing behaviour of Cu in SrS:Dy,Cu,Cl two component phosphor was introduced. It was also found that addition of NH4CI as flux in SrS:Cu caused tremendous improvement in the structural and luminescence properties.A novel technique for ACTFEL phosphor deposition at low temperature was introduced. Polycrystalline films of SrS:Cu,F were synthesized at low temperature by concomitant evaporation of host and dopant by electron beam evaporation and thermal evaporatin methods.Copper doped strontium sulphide nanophosphor was synthesized for the first time. Improvement in the luminescence properties was observed in the nanophosphor with respect to it' s bulk counterpart.

The main objective of the present study is to understand different mechanisms involved in the production and evolution of plasma by the pulsed laser ablation and radio frequency magnetron sputtering. These two methods are of particular interest, as these are well accomplished methods used for surface coatings, nanostructure fabrications and other thin film devices fabrications. Material science researchers all over the world are involved in the development of devices based on transparent conducting oxide (TCO) thin films. Our laboratory has been involved in the development of TCO devices like thin film diodes using zinc oxide (ZnO) and zinc magnesium oxide (ZnMgO), thin film transistors (TFT's) using zinc indium oxide and zinc indium tin oxide, and some electroluminescent (EL) devices by pulsed laser ablation and RF magnetron sputtering.In contrast to the extensive literature relating to pure ZnO and other thin films produced by various deposition techniques, there appears to have been relatively little effort directed towards the characterization of plasmas from which such films are produced. The knowledge of plasma dynamics corresponding to the variations in the input parameters of ablation and sputtering, with the kind of laser/magnetron used for the generation of plasma, is limited. To improve the quality of the deposited films for desired application, a sound understanding of the plume dynamics, physical and chemical properties of the species in the plume is required. Generally, there is a correlation between the plume dynamics and the structural properties of the films deposited. Thus the study of the characteristics of the plume contributes to a better understanding and control of the deposition process itself. The hydrodynamic expansion of the plume, the composition, and SIze distribution of clusters depend not only on initial conditions of plasma production but also on the ambient gas composition and pressure. The growth and deposition of the films are detennined by the thermodynamic parameters of the target material and initial conditions such as electron temperature and density of the plasma.For optimizing the deposition parameters of various films (stoichiometric or otherwise), in-situ or ex-situ monitoring of plasma plume dynamics become necessary for the purpose of repeatability and reliability. With this in mind, the plume dynamics and compositions of laser ablated and RF magnetron sputtered zinc oxide plasmas have been investigated. The plasmas studied were produced at conditions employed typically for the deposition of ZnO films by both methods. Apart from this two component ZnO plasma, a multi-component material (lead zirconium titanate) was ablated and plasma was characterized.

This thesis entitled Reliability Modelling and Analysis in Discrete time Some Concepts and Models Useful in the Analysis of discrete life time data.The present study consists of five chapters. In Chapter II we take up the derivation of some general results useful in reliability modelling that involves two component mixtures. Expression for the failure rate, mean residual life and second moment of residual life of the mixture distributions in terms of the corresponding quantities in the component distributions are investigated. Some applications of these results are also pointed out. The role of the geometric,Waring and negative hypergeometric distributions as models of life lengths in the discrete time domain has been discussed already. While describing various reliability characteristics, it was found that they can be often considered as a class. The applicability of these models in single populations naturally extends to the case of populations composed of sub-populations making mixtures of these distributions worth investigating. Accordingly the general properties, various reliability characteristics and characterizations of these models are discussed in chapter III. Inference of parameters in mixture distribution is usually a difficult problem because the mass function of the mixture is a linear function of the component masses that makes manipulation of the likelihood equations, leastsquare function etc and the resulting computations.very difficult. We show that one of our characterizations help in inferring the parameters of the geometric mixture without involving computational hazards. As mentioned in the review of results in the previous sections, partial moments were not studied extensively in literature especially in the case of discrete distributions. Chapters IV and V deal with descending and ascending partial factorial moments. Apart from studying their properties, we prove characterizations of distributions by functional forms of partial moments and establish recurrence relations between successive moments for some well known families. It is further demonstrated that partial moments are equally efficient and convenient compared to many of the conventional tools to resolve practical problems in reliability modelling and analysis. The study concludes by indicating some new problems that surfaced during the course of the present investigation which could be the subject for a future work in this area.
