The main objective of the present thesis consists on the development of an analytical preconcentration technology for the concomitant extraction and concentration of human pollution tracers from wastewater streams. Due to the outstanding tunable properties of ionic liquids (ILs), aqueous biphasic systems (ABS) composed of ILs can provide higher and more selective extraction efficiencies for a wide range of compounds, being thus a promising alternative to the volatile and hazardous organic solvents (VOCs) typically used. For that purpose, IL-based ABS were employed and adequately characterized as an one-step extraction and concentration technique. The applicability of IL-based ABS was verified by their potential to completely extract and concentrate two representative pharmaceutical pollution tracers, namely caffeine (CAF) and carbamazepine (CBZ), from wastewaters. The low concentration of these persistent pollutants (usually found in μg·dm-3 and ng·dm-3 levels, respectively) by conventional analytical equipment does not permit a proper detection and quantification without a previous concentration step. Preconcentration methods commonly applied are costly, timeconsuming, with irregular recoveries and make use of VOCs. In this work, the ABS composed of the IL tetrabutylammonium chloride ([N4444]Cl) and the salt potassium citrate (K3[C6H5O7]) was investigated while demonstrating to be able to completely extract and concentrate CAF and CBZ, in a single-step, overcoming thus the detection limit of the applied analytical equipment. Finally, the hydrotropic effect responsible for the ability of IL-based ABS to extract and concentrate a wide variety of compounds was also investigated. It was shown that the IL rules the hydrotropic mechanism in the solubility of CAF in aqueous solutions, with an increase in solubility up to 4-fold. Moreover, the proper selection of the IL enables the design of the system that leads to a more enhanced solubility of a given solute in the IL-rich phase, while allowing a better extraction and concentration. IL-based ABS are a promising and more versatile technique, and are straightforwardly envisaged as selective extraction and concentration routes of target micropollutants from wastewater matrices.

Presence and concentrations of radionuclides could be as a result of natural and human activities. This study examined the associations and differences among soil, sediment and water specific activities of long-lived radioactive element (LLRE). Gamma spectroscopy was used to measure the concentration of the LLRE along the Mini Okoro/Oginigba Creek, Port Harcourt. Specific activities of three selected LLRE were derived. Correlation analysis was carried out to examine associations among the specific activities across different substrates. A strong and a significant negative correlation exists between the specific activities of Water 40K and Soil 232Th (r =-0.721, p<0.05); Water 238U and Soil 238U (r = -0.717, p<0.05) and Water 40K and Sediment 238U (r=-0.69, p<0.05). Comparison using Mann-Whitney U test shows that, soil and sediment are similar in their specific activities with Z values of -0.408, -1.209 and -1.021 (p > 0.05) for 40K, 232Th and 238U respectively. The concentration of solid samples (soil and sediment) is different from the liquid (water) samples. These associations can be attributed to some specific underlying factors. And in other to understand them there is need for more studies.

The first days of radioactivity, the discoveries of X-rays, radioactivity, of alpha- and beta- particles and gamma- radiation, of new radioactive elements, of artificial radioactivity, the neutron and positron and nuclear fission are reviewed as well as several adverse historical marks, such as the Manhattan project and some nuclear and radiological accidents. Nuclear energy generation in Brazil and the world, as an alternative to minimize environmental problems, is discussed, as are the medicinal, industrial and food applications of ionizing radiation. The text leads the reader to reflect on the subject and to consider its various aspects with scientific and technological maturity.

cDNA arrays are a powerful tool for discovering gene expression patterns. Nylon arrays have the advantage that they can be re-used several times. A key issue in high throughput gene expression analysis is sensitivity. In the case of nylon arrays, signal detection can be affected by the plastic bags used to keep membranes humid. In this study, we evaluated the effect of five types of plastics on the radioactive transmittance, number of genes with a signal above the background, and data variability. A polyethylene plastic bag 69 μm thick had a strong shielding effect that blocked 68.7% of the radioactive signal. The shielding effect on transmittance decreased the number of detected genes and increased the data variability. Other plastics which were thinner gave better results. Although plastics made from polyvinylidene chloride, polyvinyl chloride (both 13 μm thick) and polyethylene (29 and 7 μm thick) showed different levels of transmittance, they all gave similarly good performances. Polyvinylidene chloride and polyethylene 29 mm thick were the plastics of choice because of their easy handling. For other types of plastics, it is advisable to run a simple check on their performance in order to obtain the maximum information from nylon cDNA arrays.

Absolute dating methods have been used in chronological studies of geological processes and sedimentary units of Quaternary age in Central Amazonia, Brazil. Although radiocarbon dating has been very useful in archaeological research and soil studies, the temporal interval of this method is inefficient in evaluating the sedimentation aspects and geological events from the beginning of the Quaternary in the Amazon basin. The use of crystal luminescence dating has been one of the most promising tool for determining the absolute dating of Quaternary deposits in the Amazonian region. Optically stimulated luminescence (OSL) dating, following the MAR and SAR protocols, in a tectonic-sedimentary study of Quaternary fluvial deposits in the confluence area of the Negro and Solimões rivers, indicated ages from 1.3 (Holocene) to about 67.4 kyears (Late Pleistocene) for these sediments. Low radioactive isotope concentrations were found about 2ppm for 235U and 238U; 5ppm for 232Th; and the 40K concentrations were almost zero. A comparison was made between MAR and SAR protocols taking into account the fluvial depositional process.
