In this article, we describe our ongoing efforts in addressing the environment and energy challenges facing the world today. Tapping solar thermal energy seems to be the right choice for a country like India. We look at three solar-thermal technologies in the laboratory — water purification/distillation, Stirling engine, and air-conditioning/refrigeration.

A solvent-free synthesis of alpha-aminonitriles and beta-nitroamines by oxidative cross-dehydrogenative coupling under aerobic condition is reported. A catalytic amount of molybdenum(VI) acetylacetonoate was found to catalyze cyanation of tertiary amines to form alpha-aminonitriles, whereas vanadium pentoxide was found to promote aza-Henry reaction to furnish beta-nitroamines. Both of these environmentally benign reactions are performed in the absence of solvents using molecular oxygen as an oxidant.

Serine hydroxymethyltransferase (SHMT), a pyridoxal-5V-phosphate (PLP)-dependent enzyme catalyzes thetetrahydrofolate (H4-folate)- dependent retro-aldol cleavage of serine to form 5,10-methylene H4-folate and glycine. The structure–function relationship of SHMT wasstudied in our laboratory initially by mutation of residues that are conserved in all SHMTs and later by structure-based mutagenesis of residues located in the active site. The analysis of mutants showed that K71, Y72, R80, D89, W110, S202, C203, H304, H306 and H356 residues are involved in maintenance of the oligomeric structure. The mutation of D227, a residue involved in charge relay system, led to the formation of inactive dimers, indicating that this residue has a role in maintaining the tetrameric structure and catalysis. E74, a residue appropriately positioned in the structure of the enzyme to carry out proton abstraction, was shown by characterization of E74Q and E74K mutants to be involved in conversion of the enzyme from an ‘open’ to ‘closed’ conformation rather than proton abstraction from the hydroxylgroup of serine. K256, the residue involved in the formation of Schiffs base with PLP, also plays a crucial role in the maintenance of the tetrameric structure. Mutation of R262 residue established the importance of distal interactions in facilitating catalysis and Y82 is not involved in the formaldehyde transfer via the postulated hemiacetal intermediate but plays a role in stabilizing the quinonoid intermediate.The mutational analysis of scSHMT along with the structure of recombinant Bacillus stearothermophilus SHMT and its substrate(s)complexes was used to provide evidence for a direct transfer mechanism rather than retro-aldol cleavage for the reaction catalyzed by SHMT.
India's biodiversity hotspot under anthropogenic pressure: A case study of Nilgiri Biosphere Reserve

This paper presents data on the impact of biotic pressure in terms of grazing by livestock and wood cutting by humans on the plant community in the Nilgiri Biosphere Reserve of India. Grass, and herbaceous plant biomass, number of cattle dung piles, number of woody stems available and damaged by human activities and weed biomass were assessed at different proximity along transects radiating from village-forest boundary to forest interior to measure the ecological impact of livestock grazing and fire wood collection. The grass biomass was positively correlated to overgrazing indicating the adverse effect on natural vegetation by cattle. Woodcutting was intense along the forest boundary and significantly declined as distance increased. Similarly, weed biomass and number of thorny species declined positively with proximity from village-forest boundary and the weed biomass was significantly higher in the pastoral sites compared to residential sites. The results suggest that human impact adversely affects natural vegetation and promotes weed proliferation in forest areas adjoining human settlements in the ecologically important Nilgiri Biosphere Reserve. Continued anthropogenic pressure could cause reduction in fodder availability to large herbivores like elephants, which in turn leads to an increase in human-elephant conflict. The van der Waals and Platteuw (vdVVP) theory has been successfully used to model the thermodynamics of gas hydrates. However, earlier studies have shown that this could be due to the presence of a large number of adjustable parameters whose values are obtained through regression with experimental data. To test this assertion, we carry out a systematic and rigorous study of the performance of various models of vdWP theory that have been proposed over the years. The hydrate phase equilibrium data used for this study is obtained from Monte Carlo molecular simulations of methane hydrates. The parameters of the vdWP theory are regressed from this equilibrium data and compared with their true values obtained directly from simulations. This comparison reveals that (i) methane-water interactions beyond the first cage and methane-methane interactions make a significant contribution to the partition function and thus cannot be neglected, (ii) the rigorous Monte Carlo integration should be used to evaluate the Langmuir constant instead of the spherical smoothed cell approximation, (iii) the parameter values describing the methane-water interactions cannot be correctly regressed from the equilibrium data using the vdVVP theory in its present form, (iv) the regressed empty hydrate property values closely match their true values irrespective of the level of rigor in the theory, and (v) the flexibility of the water lattice forming the hydrate phase needs to be incorporated in the vdWP theory. Since methane is among the simplest of hydrate forming molecules, the conclusions from this study should also hold true for more complicated hydrate guest molecules.

The envelope protein (E1-E2) of Hepatitis C virus (HCV) is a major component of the viral structure. The glycosylated envelope protein is considered to be important for initiation of infection by binding to cellular receptor(s) and also known as one of the major antigenic targets to host immune response. The present study was aimed at identifying mouse monoclonal antibodies which inhibit binding of virus like particles of HCV to target cells. The first step in this direction was to generate recombinant HCV-like particles (HCV-LPs) specific for genotypes 3a of HCV (prevalent in India) using the genes encoding core, E1 and E2 envelop proteins in a baculovirus expression system. The purified HCV-LPs were characterized by ELISA and electron microscopy and were used to generate monoclonal antibodies (mAbs) in mice. Two monoclonal antibodies (E8G9 and H1H10) specific for the E2 region of envelope protein of HCV genotype 3a, were found to reduce the virus binding to Huh7 cells. However, the mAbs generated against HCV genotype 1b (D2H3, G2C7, E1B11) were not so effective. More importantly, mAb E8G9 showed significant inhibition of the virus entry in HCV JFH1 cell culture system. Finally, the epitopic regions on E2 protein which bind to the mAbs have also been identified. Results suggest a new therapeutic strategy and provide the proof of concept that mAb against HCV-LP could be effective in preventing virus entry into liver cells to block HCV replication.

A molecular dynamics (MD) investigation of LiCl in water, methanol, and ethylene glycol (EG) at 298 K is reported. Several; structural and dynamical properties of the ions as well as the solvent such as self-diffusivity, radial distribution functions, void and neck distributions, velocity autocorrelation functions, and mean residence times of solvent in the first solvation shell have been computed. The results show that the reciprocal relationship between the self-diffusivity of the ions and the viscosity is valid in almost all solvents with the exception of water. From an analysis of radial distribution functions and coordination numbers the nature of hydrogen bonding within the solvent and its influence on the void and neck distribution becomes evident. It is seen that the solvent solvent interaction is important in EG while solute solvent interactions dominate in water and methanol. From Voronoi tessellation, it is seen that the voids and necks within methanol are larger as compared to those within water or EG. On the basis of the void and neck distributions obtained from MD simulations and literature experimental data of limiting ion conductivity for various ions of different sizes we show that there is a relation between the void and neck radius on e one hand and dependence of conductivity on the ionic radius on the other. It is shown that the presence of large diameter voids and necks in methanol is responsible for maximum in limiting ion conductivity (lambda(0)) of TMA(+), while in water in EG, the maximum is seen for Rb+. In the case of monovalent anions, maximum in lambda(0) as a function ionic radius is seen for Br- in water EG but for the larger ClO4- ion in methanol. The relation between the void and neck distribution and the variation in lambda(0) with ionic radius arises via the Levitation effect which is discussed. These studies show the importance of the solvent structure and the associated void structure.

The electrocaloric effect (ECE) of 0.85PbMg(1/3)Nb(2/3)O(3-)0.15PbTiO(3) (0.85PMN-0.15PT) thin films deposited on (111) Pt/TiO2/SiO2/Si substrate by pulsed laser deposition (PLD) has been calculated. The reversible adiabatic temperature was calculated indirectly using the Maxwell's relation Delta T = -T/C rho integral(E2)(E1) (partial derivative P/partial derivative T)(sigma,E)dE. Permittivity and P-E measurements show an anomaly at 11 degrees C on heating only. This anomaly previously reported are claimed to arise due to the PNR depolarization upon heating. The absence of this anomaly during cooling suggests that no structural phase transition takes place. A negative electrocaloric effect is observed which is explained by the increase in the entropy term.

Nonsteroid anti-inflammatory drugs (NSAIDs) represent standard therapy for the alleviation of pain and inflammation. At present various classes of compounds have been reported as selective inhibitors of cyclooxygenase-2 (COX-2). However, they are associated with adverse side effects. To address these issues, we report here a new class of compounds that exhibit potent analgesic and anti-inflammatory response. Substituted bromo-benzothiophene carboxamides (4-11) were examined for their analgesic and anti-inflammatory properties. Our findings demonstrate that newly synthesized bromo-benzothiophene carboxamide derivatives 4, 6, and 8 attenuate nociception and inflammation at lower concentration than classical NSAIDs, such as ibuprofen. These compounds act by selectively inhibiting COX-2 and by disrupting the prostaglandin-E2-dependent positive feedback of COX-2 regulation, which was further substantiated by reduction in the levels of cytokines, chemokines, neutrophil accumulation, synthesis of prostaglandin-E2, expression of COX-2, and neutrophil activation at lower concentration than the classic NSAID ibuprofen. Toxicological study reveals that these compounds are well tolerated and metabolized to avoid any toxicity. Thus, these molecules represent a new class of analgesic and anti-inflammatory agents. Aims: Administration of estradiol or compounds with estrogenic activity to newborn female rats results in irreversible masculinization as well as defeminization in the brain and the animals exhibit altered reproductive behavior as adults. The cellular and molecular mechanism involved in inducing the irreversible changes is largely unknown. In the present study, we have monitored the changes in the expression of selected synaptogenesis related genes in the sexually dimorphic brain regions such as POA, hypothalamus and pituitary following 17 beta-estradiol administration to neonatal female rats. Main methods: Female Wistar rats which were administered 17 beta-estradiol on day 2 and 3 after birth were sacrificed 120 days later and the expression levels of genes implicated in synaptogenesis were monitored by semi-quantitative reverse transcription PCR. Since estradiol induced up-regulation of COX-2 in POA is a marker for estradiol induced masculinization as well as defeminization, in the present study only animals in which the increase in expression of COX-2 gene was observed in POA were included in the study. Key findings: Down-regulation of genes such as NMDA-2B, NETRIN-1, BDNF, MT-5 MMP and TNF-alpha was observed in the pre-optic area of neonatally E2 treated female rat brain but not in hypothalamus and pituitary compared to the vehicle- treated controls as assessed by RT-PCR and Western blot analysis. Significance: Our results suggest a possibility that down-regulation of genes associated with synaptogenesis in POA, may be resulting in disruption of the cyclical regulation of hormone secretion by pituitary the consequence of which could be infertility and altered reproductive behavior. El presente trabajo se realizó en el periodo comprendido entre Noviembre y Diciembre de 1989, con el objetivo de investigar la infestación de parásitos gastrointestinales y pulmonares de mayor importancia económica en los bovinos del complejo 1 y 2 de la empresa genética Agenor Gómez. Boaco, Nicaragua. Se muestrearon 242 animales, entre bovinos de la raza Aberdeen Angus y Brahman, distribuidos en las categorías siguientes: C1 (0-1 año), C2 (1-3 años) y C3 (>3 años). Las muestras fueron extraídas directamente del recto animal, luego refrigeradas a 5°C y fueron analizadas en laboratorio de parasitología del MIDINRA, localizado en municipio de Camoapa V región. La cantidad de huevos y larvas se registró para cada género empleando las técnicas de sedimentación en agua, flotación con solución salina saturada y el método de larvoscopia e identificación con fotografías. Al realizar el ANDEVA por género de parásitos reportados en el análisis coprológico para la variable N° de huevos y larvas/gr de heces, se encontró que existen diferencias significativas con P>0.05 entre los promedios observados de los diferentes hatos. Los niveles de infestación promedio en los distintos hatos oscilaron entre 0u y 2lu; donde u= número de huevos y larvas/gramos de heces. De estos 48 promedios (8 hatos por 6 géneros de parásitos), 6 alcanzaron valores >9; 1 hato C1 de la upe Las Brisas con media de 2lu del género Haemonchus, 1 hato e de la upe San Miguel con media de 20.87u del género Strongyloides, 1 hato C2 de la upe San Jorge con media de 17.69u del género Strongyloides, 2 hatos C1 de las upe las Brisas y San Miguel con medias de 10.67u y medias de 10.5u respectivamente, ambos del género Coccidia y 1 hato e2 de la upe San Jorge con media de 9.6lu del género Coccidia. En los promedios restantes se obtuvieron valores <6u perteneciendo a la calificación leve. Posteriormente se realizó un análisis de correlación para determinar el grado de asociación existente entre cada uno de los géneros de parásitos económicamente importantes, encontrándose que el género Coccidia presentó el mayor grado de correlación con respecto a los demás géneros encontrados, se encontró que las correlaciones >= 0.25 corresponden a los géneros Coccidia, Moniezia, Neoascaris, Strongyloides y Haemonchus con r de 0.25, 0.25, 0.48, 0.30 respectivamente, similar para Haemonchus y Moniezia con r de 0.289.

La investigación se realizó en la finca San Antonio del municipio de Santo Domingo. Los objetivos fueron; Evaluar la efectividad de tres protocolos de sincronización en vaquillas. El trabajo consistió en la comparación de tres protocolos (DIB, Prostaglandina + Prostaglandina y una sola dosis de Prostaglandina) para la sincronización de celo en vaquillas. Para el experimento se conformaron tres grupos de 20 vaquillas cada uno, con una edad promedio de 2.5 años, las cuales se examinaron por el método de palpación para definir el estado ginecológico de ellas, se definió el estado corporal por medio de la observación de la condición corporal (CC) y las variables evaluadas fueron: (1) Porcentaje de Preñez a Primer Servicio (PPPS) (2) Porcentaje de Preñez a Segundo Servicio (PPSS) (3) Porcentaje de Preñez Acumulada Sincronización más Toro (PAST) Los resultados derivados del análisis para tratamiento 1 (DIB) elporcentaje de preñez al primer servicio fue del 80% y a segundo servicio fue de 50% para una preñez acumulada del 90%, con el tratamiento 2 (PGF2α + PGF2α) el resultado de preñez a primer servicio fue 50% y a segundo servicio fue de 33.33% con una preñez acumulada de 60%, con el tratamiento 3 (una sola aplicación de PGF2 α) el resultado de preñez a primer servicio fue 40 % y a segundo servicio fue de 7.70% para una preñez acumulada del 40 %, el grupo testigo presento una efectividad de 30 %. Se concluye que bajo las condiciones en que se realizó el experimento las vaquillas responden a los tratamientos hormonales exógenos, y se puede obtener un 90 % de éxito en la preñez. Asimismo se puede disminuir el tiempo de la incorporación de las vaquillas a primer servicio lo que se traduce en una mayor vida útil del animal y una mayor cantidad de partos, además, este tratamiento es una alternativa efectiva para enfrentar el problema de detección de celo en los rebaños, además, las vaquillas presentaron ciclicidad post tratamiento. Se recomienda la utilización del DIB pues permite obtener buenos resultados en la taza de preñez también recomendamos el uso del protocolo prostaglandina más prostaglandina en fincas con difícil acceso a la inseminación artificial, pues, es un método de bajo costo y fácil utilización para el productor y se puede implementar el uso de toros, se debe realizar estudios con la categoría de vacas paridas para determinar el comportamiento en la reproducción, se debe tomar en cuenta el estado nutricional y la condición corporal de los animales ya que es determinante para obtener buenos resultados en el programa y realizar chequeo ginecológico periódico en las vaquillas que tienen peso y edad adecuados con el fin de realizar tratamientos y evitar así preñeces tardías.

Through LSC funding, Thurrock Adult Community College (TACC) purchased a single-deck bus, equipped with all the essential equipment that has allowed them to deliver fundamental IT skills within local communities. The bus travels to various local towns and villages which in turn has brought learning to the learners. This has proved to be very useful for those who perhaps have difficulty travelling to the main campuses or do not have the confidence to enrol on a course.

South Essex College has developed a college intranet called C_Space. This has grown from a handful of HTML pages to an intranet hosting 70 in-house developed applications and two million staff and student-generated pages. In the past three years, the College has developed a range of social networking tools within the site to engage students. This has provided a 360-degree appraisal system for the College, which has informed the content and design of the intranet, as well as influenced the overall development of the College.
