A colorectal cancer genome-wide association study in a Spanish cohort identifies two variants associated with colorectal cancer risk at 1p33 and 8p12

Background: Colorectal cancer (CRC) is a disease of complex aetiology, with much of the expected inherited risk being due to several common low risk variants. Genome-Wide Association Studies (GWAS) have identified 20 CRC risk variants. Nevertheless, these have only been able to explain part of the missing heritability. Moreover, these signals have only been inspected in populations of Northern European origin. Results: Thus, we followed the same approach in a Spanish cohort of 881 cases and 667 controls. Sixty-four variants at 24 loci were found to be associated with CRC at p-values <10-5. We therefore evaluated the 24 loci in another Spanish replication cohort (1481 cases and 1850 controls). Two of these SNPs, rs12080929 at 1p33 (P-replication=0.042; P-pooled=5.523x10(-03); OR (CI95%)=0.866(0.782-0.959)) and rs11987193 at 8p12 (P-replication=0.039; P-pooled=6.985x10(-5); OR (CI95%)=0.786(0.705-0.878)) were replicated in the second Phase, although they did not reach genome-wide statistical significance. Conclusions: We have performed the first CRC GWAS in a Southern European population and by these means we were able to identify two new susceptibility variants at 1p33 and 8p12 loci. These two SNPs are located near the SLC5A9 and DUSP4 loci, respectively, which could be good functional candidates for the association signals. We therefore believe that these two markers constitute good candidates for CRC susceptibility loci and should be further evaluated in other larger datasets. Moreover, we highlight that were these two SNPs true susceptibility variants, they would constitute a decrease in the CRC missing heritability fraction.

A Constituinte começa a discutir a Reforma Tributária. Lideranças preparam a regulamentação das eleições municipais. A aprovação desse projeto afasta a prorrogação dos mandatos de prefeitos e vereadores. Com votações rápidas o Plenário decide a defesa do Estado. Numa primeira etapa, foram criados o Estado de Sítio e o Estado de Defesa, figura que substitui o Estado de Emergência. Depois, foi a vez do papel das Forças Armadas, que continuam com a mesma função de defesa do território e da ordem constitucional. O Plenário derrubou uma emenda que tornava o serviço militar facultativo. O serviço militar continua obrigatório, ressalvados os casos imperativos de consciência, como por exemplo, motivos religiosos. No capítulo da segurança pública, foram criadas as guardas municipais e as patrulhas rodoviárias federais foram vinculadas à Polícia Federal.

Quando a Constituinte terminar esta fase do trabalho, começa a votação em segundo turno de tudo o que já foi aprovado. No segundo turno, nada pode ser acrescentado ao texto, mas pode haver emenda supressiva, ou seja, muita coisa já aprovada pode ser retirada. Alguns constituintes lembram que houve avanços, principalmente na área dos direitos sociais, que devem permanecer no texto. Todavia, muitos constituintes acham que alguns direitos dos trabalhadores não devem permanecer no texto constitucional, assim como algumas das matérias que dizem respeito às restrições ao capital estrangeiro. Constituintes temem a interferência do Governo na votação, no segundo turno, suprimindo matérias aprovadas no primeiro turno.

Asociación Argentina de Limnología (AAL)

Este año se cumplieron veinticinco años de la creación de la Asociación Argentina de Limnología. En 1984, en la vieja aula magna de la Facultad de Ciencias Naturales y Museo (en esa época esta unidad académica funcionaba en las instalaciones del Museo de La Plata) un conjunto de mujeres y hombres relacionados con esta disciplina concretaban ese ansiado anhelo aprobando los fundamentos de su formación y eligiendo su primer comisión directiva. Los años siguientes fueron de gran impulso concretando la realización de talleres reuniones, ediciones de boletines y una gran comunicación entre sus asociados. En 1991 se realiza la primer reunión internacional en la ciudad de La Plata con gran repercusión y convocatoria, posteriormente se hacen los congresos de 1994 y 1998 en la ciudades de Tucumán y Buenos Aires respectivamente. A partir de esta fecha comienza a diluirse la actividad de la asociación para desaparecer en los primeros años del presente siglo. No obstante, ya sin el marco de la asociación, se realizan congresos en 2004 y 2008, en las ciudades de Chascomús y San Carlos de Bariloche respectivamente, con singular éxito. Este archivo es una pequeña muestra de algo que unió a un grupo de profesionales con un objetivo común: el desarrollo y proyección regional de esta disciplina. A simple vista lo podríamos calificar de un nuevo fracaso, ya que tuvo el mismo fin de la ALOA (Asociación Limnológica y Oceanográfica Argentina), pero no sería totalmente cierto, ya que la magnitud de las últimas reuniones nos indica que el crecimiento de esta ciencia es importante. Por ello, me parece adecuado que dejemos a un sociólogo y un historiador de la ciencia la interpretación de este fragmento de la rica historia del “limnobios” en nuestro país.
