Methods of semen cryopreservation allow changes in spermatic cells, such as damage in plasma and acrossomal membrane and modifications in mitochondrial function due to a disorder in the lipidic bilayer. For effective oocyte fertilization, spermatozoa require functional competent membranes, and intact organelles, acrosome and DNA. However, most laboratory methods used to evaluate semen quality are not highly correlated with fertilizing capacity. The discovery of a variety of fluorochromes and compounds conjugated to fluorescent probes has enabled an accurate assessment of the viability, integrity and function of spermatozoa. Among the most used probes that label the various compartments of the sperm cell there are the membrane impermeable fluorescent dyes to test the membrane integrity, as well as acylated dyes that pass the intact membrane. For the acrossomal integrity the most commonly used method is lectins labeled by a fluorescent probe. The acrosome reaction and spermatic capacitation is detected by the evaluation of membrane architecture and disorder of lipids in plasma membrane. Mitochondrial function can be determined using markers for their aerobic activity. The DNA status of spermatozoa has been determined using the metachromatic properties of Acridine Orange, and the DNA fragmentation can also be assessed by TUNEL assay. Finally, DNA condensation is analyzed using a single cell DNA gel electrophoresis assay that indicates DNA compactation. This monograph aims to compile the various tests used to detect damaged spermatozoa under cryopreservation methods, searching for improve the predictive value of semen analysis with the intention of a successful conception

As formações vegetais são altamente dinâmicas e dependem principalmente da taxa de crescimento das espécies vegetais, entretanto, esta está intimamente relacionada e suscetível a estresses abióticos, como elevada ou baixa radiação solar, temperaturas extremas, e bióticos, como taque de patógenos e de herbívoros, o que faz com que as plantas necessitem de um sistema de defesa altamente eficiente o qual consiste na atuação de compostos secundários, e de enzimas como por exemplo peroxidases e polifenoloxidases. A atividade das peroxidases constitui a mais importante via do sistema de defesa vegetal a patógenos no que se refere a cicatrização de ferimentos e à herbivoria, propiciando um aumento da lignificação nos tecidos lesados, o que aumenta a resistência dos mesmos, dificultando assim a ocorrência de novas lesões. Quanto às polifenoloxidases, também pertencentes ao grupo das oxidorredutases, são enzimas que também atuam diretamente na via de defesa das plantas ao ataque de patógenos e na cicatrização de ferimentos e processos de senescência. Por meio da extração e análise da atividade de Peroxidases e Polifenoloxidases em porção de raiz, caule, ápice e em folhas de indivíduos de Erythrina speciosa Andrews, Eugenia uniflora L., Hevea brasiliensis M. Arg., Hymenaea courbaril L. var. stilbocarpa (Hayne) Lee et Lang, Joannesia princeps Vell., Licania tomentosa (Benth.) Fritsch, Pachira aquatica Aubl. e Psidium guajava L., obteve-se comportamento pioneiro para as espécies Eugenia uniflora L., Hymenaea courbaril L. var. stilbocarpa (Hayne) Lee et Lang, Joannesia princeps Vell. e Psidium guajava L., e comportamento não-pioneiro para as espécies Erythrina speciosa Andrews, Hevea brasiliensis M. Arg., Licania tomentosa (Benth.) Fritsch e Pachira aquatica Aubl.. Dessa maneira foi possível verificar que a análise da atividade peroxidásica... (Resumo completo, clicar acesso eletrônico abaixo)

The main objectives of this work were to evaluate the antioxidant activity and to determine of an ethanolic ginger extract total phenolic compounds concentration, as well as verifying its behavior by means of the oxidative stability, when added to refined soybean oil in concentrations of 0, 500, 1000, 1500, 2000 and 2500 mg/kg. The maximum antioxidant activity and EC50, concentration of the extract to achieve 50% of the antioxidant activity, value determined by free radical DPPH method were 79.1% and 42.6 g/mL, respectively. The total phenolic compounds concentration, determined to Folin-Ciocalteu method, was 251 mg/g. The induction period of the samples increased according to the augment of the extract concentration in the oil when evaluated using oxidative stability through the Rancimat equipament. It was possible to conclude that the ginger extracts possess effective action against the lipid oxidation and can be applied in foods as natural antioxidant.

Lipids have important biological functions, suchas membrane constituents, hormone precursors, and are efficient energy reserves, due to thers high caloric content. In fish, but not in mammals, lipid storage patterns are quite diverse. The aim of this study was to determine lipid distribution in somatic and reproductive tissues of matrinxã, Brycon cephalus, both male and female, highlighting somatic indices related to lipid dynamics. From Oct 98 to Jan 99, a total of 174 fish (8-12 each month) were sampled. After fish anesthesia, heparinized blood was collected for plasma triacylglycerol determination. Fish were individually weighed and measured, and liver, gonads and visceral fat were collected and weighed for HSI (hepatosomatic index), GSI (gonad somatic index) and MFI (mesenteric fat index) calculation. The highest values of total lipids were found in red muscle (about 18%), liver and gonads (about 16.5%). The white muscle had a lower concentration (2.5%). Analyses of variance of HSI and MFI showed fluctuations along the experimental period with lowest concentrations in the periods of highest temperatures. Matrinxã store lipids in several body tissues, including mesenteric fat, liver, muscles and gonads.
