Synaptonemal complex (SC) component Zip1 plays a role in meiotic recombination independent of SC polymerization along the chromosomes.

Zip1 is a yeast synaptonemal complex (SC) central region component and is required for normal meiotic recombination and crossover interference. Physical analysis of meiotic recombination in a zip1 mutant reveals the following: Crossovers appear later than normal and at a reduced level. Noncrossover recombinants, in contrast, seem to appear in two phases: (i) a normal number appear with normal timing and (ii) then additional products appear late, at the same time as crossovers. Also, Holliday junctions are present at unusually late times, presumably as precursors to late-appearing products. Red1 is an axial structure component required for formation of cytologically discernible axial elements and SC and maximal levels of recombination. In a red1 mutant, crossovers and noncrossovers occur at coordinately reduced levels but with normal timing. If Zip1 affected recombination exclusively via SC polymerization, a zip1 mutation should confer no recombination defect in a red1 strain background. But a red1 zip1 double mutant exhibits the sum of the two single mutant phenotypes, including the specific deficit of crossovers seen in a zip1 strain. We infer that Zip1 plays at least one role in recombination that does not involve SC polymerization along the chromosomes. Perhaps some Zip1 molecules act first in or around the sites of recombinational interactions to influence the recombination process and thence nucleate SC formation. We propose that a Zip1-dependent, pre-SC transition early in the recombination reaction is an essential component of meiotic crossover control. A molecular basis for crossover/noncrossover differentiation is also suggested.

Processing of the leader mRNA plays a major role in the induction of thrS expression following threonine starvation in Bacillus subtilis.

The threonyl-tRNA synthetase gene, thrS, is a member of a family of Gram-positive genes that are induced following starvation for the corresponding amino acid by a transcriptional antitermination mechanism involving the cognate uncharged tRNA. Here we show that an additional level of complexity exists in the control of the thrS gene with the mapping of an mRNA processing site just upstream of the transcription terminator in the thrS leader region. The processed RNA is significantly more stable than the full-length transcript. Under nonstarvation conditions, or following starvation for an amino acid other than threonine, the full-length thrS mRNA is more abundant than the processed transcript. However, following starvation for threonine, the thrS mRNA exists primarily in its cleaved form. This can partly be attributed to an increased processing efficiency following threonine starvation, and partly to a further, nonspecific increase in the stability of the processed transcript under starvation conditions. The increased stability of the processed RNA contributes significantly to the levels of functional RNA observed under threonine starvation conditions, previously attributed solely to antitermination. Finally, we show that processing is likely to occur upstream of the terminator in the leader regions of at least four other genes of this family, suggesting a widespread conservation of this phenomenon in their control.

Human immunodeficiency virus type 1 viral background plays a major role in development of resistance to protease inhibitors.

The observed in vitro and in vivo benefit of combination treatment with anti-human immunodeficiency virus (HIV) agents prompted us to examine the potential of resistance development when two protease inhibitors are used concurrently. Recombinant HIV-1 (NL4-3) proteases containing combined resistance mutations associated with BMS-186318 and A-77003 (or saquinavir) were either inactive or had impaired enzyme activity. Subsequent construction of HIV-1 (NL4-3) proviral clones containing the same mutations yielded viruses that were severely impaired in growth or nonviable, confirming that combination therapy may be advantageous. However, passage of BMS-186318-resistant HIV-1 (RF) in the presence of either saquinavir or SC52151, which represented sequential drug treatment, produced viable viruses resistant to both BMS-186318 and the second compound. The predominant breakthrough virus contained the G48V/A71T/V82A protease mutations. The clone-purified RF (G48V/A71T/V82A) virus, unlike the corresponding defective NL4-3 triple mutant, grew well and displayed cross-resistance to four distinct protease inhibitors. Chimeric virus and in vitro mutagenesis studies indicated that the RF-specific protease sequence, specifically the Ile at residue 10, enabled the NL4-3 strain with the triple mutant to grow. Our results clearly indicate that viral genetic background will play a key role in determining whether cross-resistance variants will arise.

Examining the Relationship Between Passion and Perceptions of Cohesion in Athletes

The purpose of the present study was to examine the relationship of harmonious and obsessive passion to perceptions of task and social cohesion in team sport athletes. Participants were 370 competitive (N=252) and recreational (N=118) athletes ranging from 18- to 28-years-old (Mage=20.20, SD=1.52) from a wide variety of team sports. Participants completed the Passion Scale (Vallerand et al., 2003) and the Group Environment Questionnaire (Carron et al., 1985). A MANOVA revealed that competitive athletes were more passionate and had higher perceptions of cohesion than did recreational athletes. Multiple regression analyses revealed a positive relationship between both harmonious and obsessive passion and both task (ATG-T, GI-T) and social (ATG-S, GI-S) cohesion. Theoretical and practical implications are discussed pertaining to the importance of harmonious and obsessive passion in athletes and perceptions of cohesion in competitive and recreational sport.

Just Need Some Satisfaction: Examining the Relationship between Passion for Exercise and the Basic Psychological Needs

Individuals often pursue activities for which they are passionate about, and this passion is operationalized as being harmonious (an autonomous desireto engage in the activity) or obsessive (a controlled desire to engage in the activity) in nature (Vallerand et al., 2003). With regard to harmonious passion, Vallerand et al. (2003) suggests that it is fostered in environments that nurture innate needs for autonomy, competence, and relatedness. The purpose of the present study was to explore the nature of the passion-basic psychological needs (competence, autonomy, relatedness) relationship. Kinesiology students (N = 917; Mage = 18.54 SD = 1.66) completed the Passion Scale (Vallerand et al., 2003) and the Psychological Need Satisfaction in Exercise Scale (Wilson et al., 2006). Results from the SEM path analysis indicated that harmonious passion was positively related to competence (SPE = .43) and relatedness (SPE = .43) and obsessive passion was negatively related to autonomy (SPE = -.18)(CFI = .90, RMSEA = .07, SRMR = .07). Implications for exercise participation/enjoyment are discussed.

Russia is currently the world’s second most popular destination country for international migrants (second only to the United States). In recent years, Russia’s relatively high economic growth has attracted foreign workers from poorer neighbouring republics in Central Asia, as well as from Ukraine and Belarus. In the absence of a consistent immigration policy, the largescale influx of immigrants has become a major issue affecting social relations in Russia. The majority of Russians oppose the arrival of both foreign workers and internal migrants from Russia’s North Caucasus republics, claiming that their presence in Russia contributes to the escalation of ethnic and religious tensions, fuels organised crime and corruption, and increases competition on the labour market. As many as 70% of Russians are in favour of restricting the number of immigrants allowed into the country, calling on the government for a more stringent policy on immigration. Since the end of July the authorities have responded to these calls by carrying out a series of raids on markets and construction sites across Moscow, where most immigrants tend to find employment. The raids have led to arrests and deportations. However, these measures should not be seen as a serious attempt to deal with the problem of economic migrants in the capital, mainly because of the highly selective and staged nature of the crackdown. This, coupled with the timing of the initiative, might indicate that the raids are a part of an ongoing election campaign, particularly in the run-up to the Moscow mayoral elections scheduled for 8 September. By adopting anti-immigration rhetoric, the Kremlin is seeking both to garner support among Russian voters, who tend to be easily swayed by nationalist sentiments, and to steal the anti-immigration card from the opposition and its leader Alexei Navalny. The opposition has been calling for a clearer policy on this issue and has blamed the government for the current lack of control over migrant numbers, accusing the authorities of benefiting from the widespread corruption linked to immigration. In a broader context, the actions taken by the government are a response to the declining legitimacy of the current ruling elite. By attempting to address the immigration issue, the Kremlin is trying to restore its image as a government attentive to social problems and capable of solving them effectively.
