Oil is a hydrocarbon mixture of various sizes, including saturated and aromatic compounds. Natural gas is a mixture of gaseous hydrocarbons and its main component is methane. In our society, the great demand for these fuels requires fast extraction, transportation and refining, increasing the number of accidents that compromise the environment. Oil is a finite resource and it is necessary to reduce the problems related to the question concerning environmental pollution which has encouraged the search for alternative fuel sources in our country. So today we have two major biofuels: ethanol and biodiesel. Concurrently, many studies have been done directed toward the isolation of microorganisms capable of degrading petrochemical industrial wastes, most of them using as a source of isolation soil and water collected in a contaminated environment. Isolation from alternative substrates has emerged as a new strategy that has provided satisfactory results. In this work, we present the leaf-cutter ants of the Attini tribe as a source for the isolation of micro-fungi with the potential for hydrocarbon degradation. These insects have a social way of life and a highly specialized system of intra and interspecific communication, which is based on the recognition of individuals through volatile chemical compounds, the majority hydrocarbons, stored in their exoskeleton. The micro-environment exoskeleton of Attini ants (genus Atta) used in this work proved to be a rich source of microbial biodiversity, as other studies have found. The flotation isolation technique applied here allowed the achievement of 214 micro-fungi, 118 representatives of the dematiaceous fungi group and 96 hyaline filamentous fungi. In this work we investigate the dissociation of heteronuclear diatomic molecules subjected to laser pulses. This phenomenon can be modeled by the classical forced Morse oscillator. This system presents a chaotic dynamics associated with the anharmonicity of the internuclear potential and with the coupling of permanent dipole of molecule with the electric field of laser. We want to verify how the dissociation probability evolves while we change the intensity and frequency of laser. We study the phase space of molecules to have a better understanding of system dynamics. We make the calculations changing two parameters of laser (intensity and frequency) and checking how this parameters influences on molecule dissociation. We compare the results of HF molecule (Fluoride acid) and CO molecule (Carbon monoxide) to check how the dipole moment of each molecule can influence on laser interaction

Atualmente são conhecidas aproximadamente 10.000 substâncias do metabolismo secundário de invertebrados e microorganismos marinhos. Dentre os organismos marinhos estudados do ponto de vista químico e farmacológico, destacam-se, as esponjas, as ascídias, os briozoários e os octocorais. No entanto, pouco se sabe quanto ao potencial imunomodulador de compostos isolados de microrganismos marinhos, em especial daqueles isolados da costa brasileira. A propriedade anti-inflamatória de diferentes extratos brutos foi inicialmente avaliada pelo potencial destes compostos quanto à inibição da produção de óxido nítrico (NO) em linhagem de macrófagos, RAW 264.7. Posteriormente as amostras que se mostraram promissoras foram também avaliadas em relação ao potencial imunomodulador quanto a expressão de moléculas de superfície relacionadas migração (Mac-1 ou CD11b) e ativação celular (CD80 e CD86) em linhagens de macrófagos estimulados com LPS. Nossos resultados mostram que dentre as 289 amostras testadas apenas o extrato DLM33 e as substâncias Ma(M3%)J-MeOH e Dr(M3%)6-MeOH/H2O foram inicialmente considerados promissores quanto capacidade de inibir a síntese de NO por macrófagos. O extrato DLM33 foi capaz de de modular apenas a porcentagem de macrófagos positivos para CD80 na presença de LPS. No entanto, a substância Dr(M3%)6-MeOH/H2O não se mostrou eficiente quanto a modulação da expressão de moléculas de superfície Mac-1, CD80 e CD86. Surpreendentemente, a substância Ma(M3%)J-MeOH apresentou um potencial imunoestimulador quanto a expressão de Mac-1+/CD80+, mas não de CD86, em macrófagos sugerindo um possível efeito adjuvante desta substância. In materials science, the search for technological improvements have become one of the main subject of study of researchers. This is especially true in the case of materials with reduced sizes, in the nanometer scale. Important phenomena to be studied in these cases are the desorption and adsorption on two-dimensional materials, such as graphene. These phenomena are of great importance in the study of interactions between organic films, synthesis or catalysis of reactions on surfaces and even in the creation of nanoscale devices [1, 2, 3, 4]. Between the most important topics related to these phenomena are the storage of gases in low-dimensional systems and the study of nanostructured fuel cells or batteries. In this context we used two different parametrizations for the reactive force field ReaxFF to study the potential barriers and reaction barriers of our system. First we made a study about the Reaction Barriers and Energy Barriers for bonds between graphene and the following atoms: sulfur, fluorine, hydrogen, nitrogen and oxygen. It is important to have this information in order to make it possible to understand how these atoms react with the graphene sheet. Subsequently, we calculate reaction barriers for mixed structures where fluorine is a fixed element bonded to graphene and other element is simultaneously bonded to graphene. This other element (N, O, H or S) is varied in its possible relative positions (ortho, meta and para in relation to fluorine in either: the same side and in the opposite side of the graphene membrane)

Estudos têm demonstrado que o reconhecimento inicial de microrganismos é mediado por receptores celulares expressos em células da imunidade inata denominados receptores de reconhecimento de padrões (PRRs). Assim, a interação entre moléculas de superfície dos patógenos e receptores homólogos presentes na membrana celular de monócitos, modula a fagocitose, a ativação da célula e conseqüentemente a produção de citocinas. Trabalhos têm demonstrado a importância da estimulação de receptores toll-like 2 e 4 (TLR2 e TLR4), receptores de manose (MR) e dectina-1 de monócitos, tanto em infecções bacterianas como fúngicas, culminando com indução de produção de várias citocinas. A IL-18 é uma citocina indutora de IFN-g e possui uma ação extremamente importante, por ser capaz de promover tanto uma resposta do tipo Th1 ou Th2, dependendo do contexto de estimulação e do microambiente de citocinas. Em trabalho recente demonstramos que a IL-18 possui uma ação importante sobre o aumento da expressão de MR em monócitos humanos, e o reconhecimento do Paracoccidioides brasiliensis via esse receptor causaria aumento do crescimento fúngico no interior da célula. Dessa forma, os objetivos do presente projeto foram: 1) Avaliar a ação da IL- 18 sobre a expressão do receptor dectina-1 por monócitos humanos desafiados in vitro com diferentes cepas do P. brasiliensis; b) Avaliar a participação da dectina-1 na indução da produção de IL-18, TNF-a e IL-10 por monócitos desafiados in vitro com diferentes cepas do P. brasiliensis. Assim, monócitos de indivíduos normais tratados in vitro com IL-18 foram desafiados com diferentes de cepas do P. brasiliensis, e a expressão do receptor dectina-1 foi avaliada pela técnica de citometria de fluxo. Cutaneous melanoma is a malignancy caused by the uncontrolled proliferation of melanocytes. Has been increasing in incidence and mortality over the years. This type of tumor may arise in the skin or any other place where there was migration of neural crest cells. Melanocytes are located in the basal layer of the epidermis forming the epidermal melanin unit, involving a melanocyte to 36 keratinocytes. The proliferation of melanocytes depends on this relationship with the keratinocytes, gap junctions, E-cadherin and desmoglein. The basal membrane is responsible for promoting the integrity of the skin, their adhesion structures shown to be altered in several skin diseases. Some molecules that play the role of adherence in this membrane are the laminins, type IV collagen and type XVII and perlecan. Analysis of protein expression of cell adhesion molecules in melanoma by immunohistochemistry on tissue microarray (TMA). We selected paraffin blocks of patients who were diagnosed with cutaneous melanoma in the period 1995 to 2010, reaching a sample of 124 cases. All these have gone through review of preparation for subsequent histological analysis of tissue microarray (TMA). The protein expression will be analyzed by immunohistochemistry, using markers anti-laminin, anti-collagen type IV and type XVII and anti-perlecan. There was a predominance of female patients, Caucasians and southeastern Brazil. There were frequent in type of extensive superficial ulcers with Breslow depth greater than 4 mm, Clark level IV and mitotic index greater than 6 mitoses per cga. The patients, most of them were female with mean age of 57.13 years and Caucasian. The most frequent histological type was extensive superficial followed by acral lentiginous. Predominated cases very deep, with Breslow thickness greater than 4 mm, Clark IV, ulcerated and on average by 9 cga mitosis, leading to worse prognosis in this sample. In many movies of scientific fiction, machines were capable of speaking with humans. However mankind is still far away of getting those types of machines, like the famous character C3PO of Star Wars. During the last six decades the automatic speech recognition systems have been the target of many studies. Throughout these years many technics were developed to be used in applications of both software and hardware. There are many types of automatic speech recognition system, among which the one used in this work were the isolated word and independent of the speaker system, using Hidden Markov Models as the recognition system. The goals of this work is to project and synthesize the first two steps of the speech recognition system, the steps are: the speech signal acquisition and the pre-processing of the signal. Both steps were developed in a reprogrammable component named FPGA, using the VHDL hardware description language, owing to the high performance of this component and the flexibility of the language. In this work it is presented all the theory of digital signal processing, as Fast Fourier Transforms and digital filters and also all the theory of speech recognition using Hidden Markov Models and LPC processor. This work aims to give greater visibility to the issue of software security, due to people talk a lot in security conferences, that much of both IT (Information Technology) staff and, more specifically, IS (Information Security) staff does not know this, and, thanks to the spread of the mobile computing and of the cloud computing, this lack of deeper knowledge on this subject is increasingly becoming worrisome. It aims too, make applications to be developed in a security manner, priorizing the security of the information processed. It attempts to demonstrate the secure coding techniques, the principles of software security, the means to identify software vulnerabilities, the cutting-edge software exploitation techniques and the mechanisms of mitigation. Nowadays, the security guys are in charge of the most of the security tests in applications, audits and pentests, and it is undeniable that the so-called security experts, most often come from computer network field, having few experience in software development and programming. Therefore, the development process does not consider the security issue, thanks to the lack of knowledge on the subject by the developer, and the security tests could be improved whether security experts had a greater know-how on application development. Propomos avaliar o potencial de uma abordagem imunoterápica que tem o fator de crescimento do endotélio vascular (VEGF) como alvo. A angiogênese é necessária para nutrir o tumor e propicia sua metastatização. Sabe-se que VEGF é produzido pela maioria dos tumores sólidos metastatizantes. A eficiência do anticorpo anti-VEGF, BEVACIZUMAB, já foi demonstrada. Entretanto, efeitos adversos limitam sua utilização em alguns casos e, com base na teoria da rede idiotípica e na experiência do grupo com anticorpos anti-idiotípicos (anti-Id ou AB2), anticorpos AB2 capazes de mimetizar VEGF foram obtidos. Assim, com o objetivo de explorar o rationale, dois anticorpos monoclonais (MAb) anti-Id de Bevacizumab, 10.D7 e 3.E3, foram produzidos em nosso laboratório. O controle da purificação de anticorpos foi feito através da avaliação do material purificado em géis de poliacrilamida. Immunoblots comprovaram a especificidade do reconhecimento do idiotipo de Bevacizumab pelos MAbs anti-Id. Foi mostrado que os AB2 obtidos se ligam a moléculas da superfície de células endoteliais, que expressam receptores de VEGF, sugerindo que esses anticorpos mimetizam VEGF. Entretanto, não foi possível verificar inibição do crescimento dessas células quando cultivadas em presença desses MAbs. Será necessária a otimização dos protocolos utilizados para uma avaliação mais definitiva. Uma vez demonstrada a atividade anti-idiotípica dos AB2 in vitro, 10.D7 e 3.E3 foram utilizados como imunógenos em camundongos Balb/c, com o objetivo de avaliar a resposta AB3 ou anti-anti-Id. Por meio de ensaio imunoenzimático (ELISA) indireto foi verificada a presença de anticorpos anti-anti-Id ligantes de VEGF nos soros dos camundongos imunizados. Os animais imunizados com o MAb 10.D7 responderam melhor do que os imunizados com 3.E3. Anticorpos IgG purificados dos soros desses animais reconheceram VEGF em ensaios imunoenzimáticos.
