This study aimed to determine the selectivity of herbicides applied in pre- and post-emergence for alfalfa crops. Three separate experiments were carried out under greenhouse conditions. The first experiment was arranged in a completely randomized design with three replications in a 4 x 11 + 1 factorial scheme , with eleven herbicides (bentazon, chlorimuron-ethyl, fomesafen, fluazifop-p-butyl, saflufenacil, imazethapyr, clethodim, nicosulfuron, imazaquin, haloxyfop-methyl and MSMA), four doses of each herbicide (0.5 D, 0.75 D, 1.0 D and 1.25 D, where D = recommended dose), plus an untreated control. The products were applied to alfalfa plants at the stage of 4 to 5 leaf pairs. In the second experiment, the effect of pre-emergent herbicides on early alfalfa development was observed through a completely randomized design with five replications in a 3 x 4 x 2 factorial scheme, with three herbicides (hexazinone, atrazine + simazine, S-metolachlor), four doses (0.5 D, 0.75 D, 1.0 D and 1.25 D), and two types of soil texture (loamy and clay soil), plus an untreated control. The third experiment evaluated the action of atrazine, 2,550 g ha-1; clomazone - 600 g ha-1; diclosulam - 25 g ha-1; diuron+hexazinone - 936 + 264 g ha-1 and diuron+hexazinone +sulfometuron - 1,386 + 391 + 33.35 g ha-1 on alfalfa sown at different times after herbicide application. The effects of the treatments on alfalfa were evaluated according to visual phytotoxicity symptoms, plant height, and biomass of roots and shoots. Among the herbicides applied at post-emergence, imazethapyr, clethodim, haloxyfop-p-methyl and MSMA were selective for alfalfa, while among those applied at pre-emergence, none were selective, regardless of soil texture. The results of the third experiment showed that the herbicides diclosulam, hexazinone + diuron and atrazine caused less toxicity in alfalfa plants.

A sinúsia herbácea terrícola ainda é pouco estudada em florestas tropicais e subtropicais, principalmente no que se refere a sua relação a fatores ambientais. O objetivo deste trabalho foi descrever a estrutura desta sinúsia em floresta estacional, no Parque Estadual do Turvo em duas estações do ano e relacioná-la ao grau de abertura do dossel, medido pelo uso de fotografias hemisféricas. O levantamento de 30 unidades amostrais de 2 x 2 m resultou na amostragem de 29 espécies herbáceas, sendo 13 pteridófitas e 16 magnoliófitas. Lastreopsis effusa (Sw.) Tindale e Thelypteris scabra (C. Presl) Lellinger foram as espécies com maiores freqüências e coberturas. Didymochlena truncatula (Sw.) J. Sm. obteve elevado valor de cobertura e Olyra humilis Nees e Pharus lappulaceus Aubl. alta freqüência. A média das alturas foi de 37 cm (s ± 27). A diversidade (H') estimada no verão foi de 2,771 (nats) e a equabilidade (J') de 0,823, não diferindo significativamente do inverno (P = 0,509), assim como a composição e a cobertura herbácea não diferiram entre as duas estações (P = 0,728). Houve correlação entre a cobertura absoluta por parcela e o grau de abertura do dossel (P = 0,005). As maiores riqueza e diversidade e o porte e a cobertura elevados, comparados com outros estudos no sul do Brasil, são conseqüências da descontinuidade do dossel desta floresta, que permite maior incidência de luz no sub-bosque. A variação não significativa na estrutura da sinúsia herbácea é um indício do baixo grau de deciduidade da floresta.

Enterolobium schomburgkii Benth. (orelha-de-macaco) é uma árvore de interesse madeireiro que possui potencial para plantios florestais em áreas degradadas, por ser heliófila e nodulífera. Os objetivos deste trabalho foram caracterizar morfologicamente os frutos, sementes e plântulas e definir o estádio de desenvolvimento da "plântula normal" de E. schomburgkii, visando fornecer informações valiosas para a padronização dos estudos de propagação da espécie na área de tecnologia de sementes. O fruto é um legume nucóide, sublenhoso, polispérmico, auriculiforme e circular, com as dimensões de 21,2 × 3,2 cm para o legume estendido; e 6,2 ×5,9 ×0,5 cm para o fruto na forma original. O peso fresco de um fruto foi de 6,7 g, com 16,1 sementes sadias por fruto, sendo necessários 62 frutos para obter 1.000 sementes sadias. O número de sementes por quilograma foi de 18.749 e o peso de mil sementes foi de 53,6 g. A semente é elipsóide, de superfície lisa, brilhosa, marrom-amarelo escuro, com as dimensões de 8,0 ×3,9 ×2,6 mm e peso de 0,054 g. O eixo embrionário é reto e visível, com plúmula bem desenvolvida. Observou-se que a germinação é do tipo epígea fanerocotiledonar, com cotilédones foliáceos. A protrusão da raiz primária foi de 95,7%, e para a formação de plântulas normais foi de 80,2%. A "plântula normal" foi formada entre 7 e 15 dias após a semeadura, possuindo raiz primária bem desenvolvida, raízes secundárias curtas, hipocótilo alongado, cotilédones semi-abertos e primeiros eofilos visíveis, com o mesmo comprimento dos cotilédones.

Plants were regenerated from leaf-derived callus culture of Stylosanthes scabra, a polyploid legume tolerant to drought and adapted to acid soils. A total of 168 regenerants were planted out in Leonard jars in a complete randomized design. Nitrogen fixation and vegetative growth were indirectly evaluated by shoot dry weight, root dry weight, shoot N content and acetylene reduction activity. The results showed higher variation in the regenerants than in controls not submitted to tissue culture. Significant differences were found for all nitrogen fixation related-traits

The presence of phaseolin (a vicilin-like 7S storage globulin) peptides in the seed coat of the legume Phaseolus lunatus L. (lima bean) was demonstrated by N-terminal amino acid sequencing. Utilizing an artificial seed system assay we showed that phaseolin, isolated from both cotyledon and testa tissues of P. lunatus, is detrimental to the nonhost bruchid Callosobruchus maculatus (F) (cowpea weevil) with ED50 of 1.7 and 3.5%, respectively. The level of phaseolin in the seed coat (16.7%) was found to be sufficient to deter larval development of this bruchid. The expression of a C. maculatus-detrimental protein in the testa of nonhost seeds suggests that the protein may have played a significant role in the evolutionary adaptation of bruchids to legume seeds.

The soluble and insoluble cotyledon (SPF-Co and IPF-Co) and tegument (SPF-Te and IPF-Te) cell wall polymer fractions of common beans (Phaseolus vulgaris) were isolated using a chemical-enzymatic method. The sugar composition showed that SPF-Co was constituted of 38.6% arabinose, 23.4% uronic acids, 12.7% galactose, 11.2% xylose, 6.4% mannose and 6.1% glucose, probably derived from slightly branched and weakly bound polymers. The IPF-Co was fractionated with chelating agent (CDTA) and with increasing concentrations of NaOH. The bulk of the cell wall polymers (29.4%) were extracted with 4.0M NaOH and this fraction contained mainly arabinose (55.0%), uronic acid (18.9%), glucose (10.7%), xylose (10.3%) and galactose (3.4%). About 8.7% and 10.6% of the polymers were solubilised with CDTA and 0.01M NaOH respectively and were constituted of arabinose (52.0 and 45.9%), uronic acids (25.8 and 29.8%), xylose (9.6 and 10.2%), galactose (6.1 and 3.9%) and glucose (6.5 and 3.8%). The cell wall polymers were also constituted of small amounts (5.6 and 7.2%) of cellulose (CEL) and of non-extractable cell wall polymers (NECW). About 16.8% and 17.2% of the polymers were solubilised with 0.5 and 1.0M NaOH and contained, respectively, 92.1 and 90.7% of glucose derived from starch (IST). The neutral sugar and polymers solubilization profiles showed that weakly bound pectins are present mainly in SPF-Co (water-soluble), CDTA and 0.01-0.1M NaOH soluble fractions. Less soluble, highly cross-linked pectins were solubilised with 4.0M NaOH. This pectin is arabinose-rich, probably highly branched and has a higher molecular weight than the pectin present in SPF-Co, CDTA and 0.01-0.1M NaOH fractions.

Baru (Dipteryx alata Vog.), a species of legume found in the Brazilian savannas, was investigated in this study for the composition of its flesh and seed. Thermal analyses, Thermogravimetry (TG), and Differential Scanning Calorimetry (DSC) were used to investigate the proteins in defatted meal, concentrate, and protein isolate. The protein concentrate was extracted at pH 10, followed by a precipitation at the isoelectric point to obtain the isolate that was spray dried. The thermogravimetric curves were obtained under a nitrogen atmosphere with a 100 mL/minutes flow. The initial, final and peak temperatures and mass loss were analyzed. Within the performed temperature ranges studied, the defatted meal and concentrate presented four steps of mass loss, while the isolate showed only two steps. The protein content of defatted meal from Baru nuts was higher than that of the isolate. On the other hand, there was a reduction in enthalpy, which suggests that the process applied to obtain the baru concentrate and isolate led to protein denaturation.

Baru (Dipteryx alata Vog.) is an abundant legume in the Brazilian Savanna. Its nuts can be exploited sustainably using its protein and lipid fractions. This study aimed to analyze the proteins of the nuts present in the defatted flour and protein concentrate in terms of their functional properties, the profile of their fractions, and the in vitro digestibility. The flour was defatted with hexane and extracted at the pH of higher protein solubility to obtain the protein concentrate. The electrophoretic profile of the protein fractions was evaluated in SDS-PAGE gel. The functional properties of the proteins indicate the possibility of their use in various foods, like soybeans providing water absorption capacity, oil absorption capacity, emulsifying properties, and foamability. Globulins, followed by the albumins, are the major fractions of the flour and protein concentrate, respectively. Digestibility was greater for the concentrate than for the defatted flour.

The Brazilian Savannah, known as "Cerrado," has an extensive biodiversity, but it is under explored. Among the native vegetables is the jatobá-do-cerrado (Hymenaea stigonocarpa Mart.), a legume with great potential for exploration for its content of dietary fiber. Legumes are an important source of nutrient compounds, such as phenolic compounds and vitamins that have antioxidant properties. This study aimed at determining the chemical composition and antioxidant activity of the jatobá flour. The jatobá flour showed high fiber content (insoluble and soluble fiber 47.8 and 12.8 g.100 g- 1, respectively), significant amounts of carotenoids such as beta-carotene and lutein, and some minerals such as calcium: 145 mg.100 g- 1, magnesium: 125 mg.100 g- 1, and potassium: 1352 mg.100 g- 1. The jatobá flour extracted with different solvents (water, methanol, and acetone) exhibited antioxidant activity by the DPPH, FRAP, and ORAC methods. The solvent used in the extraction affected the total phenolic content and antioxidant activity. Acetone extraction produced the best results. Therefore, the jatobá flour is an ingredient that can be used to develop new products with properties that promote health.

Abstract Germinated grains have been known as sources of Gamma-aminobutyric acid (GABA) that provide beneficial effects for human health. This study was aimed to investigate GABA production, dietary fiber, antioxidant activity, and the effect of cooking on GABA loss in germinated legumes and sesame. The highest GABA content was found in germinated mung bean, (0.8068 g kg-1, 24 h incubation) followed by germinated soybean, germinated black bean and soaked sesame. Beside GABA, dietary fiber content also increased in all grains during germination where the insoluble dietary fiber fractions were always found in higher proportions to soluble dietary fiber fractions. Our results also confirmed that germinated mung bean is a rich source of GABA and dietary fibers. Microwave cooking resulted in the smallest loss of GABA in mung bean and sesame, while steaming led to the least GABA content loss in soybean and black bean. Therefore microwave cooking and steaming are the most recommended cooking processes to preserve GABA in germinated legumes and sesame.

A Sesbania virgataé uma espécie arbórea, pioneira e de ocorrência natural no Brasil. Pertence à família Leguminosae-Faboideae sendo recomendada para recuperação de áreas degradadas devido a sua rusticidade e capacidade de estabelecer simbiose com rizóbio. Mas, apesar de sua ampla distribuição, até o momento não havia uma descrição morfológica das estruturas de propagação e da planta na fase inicial de desenvolvimento. O objetivo deste trabalho foi descrever os caracteres morfológicos dos frutos, sementes e plântulas, bem como caracterizar o processo de germinação de Sesbania virgata. Para o estudo do fruto, os seguintes aspectos foram observados: tipo; cor; dimensões; textura e consistência do pericarpo; deiscência; e número de sementes por fruto. Os aspectos observados para as sementes foram: cor; dimensões; peso de 1000 sementes; textura e consistência dos tegumentos; forma; bordo, posição do hilo e de outras estruturas presentes e características do embrião. As plântulas foram caracterizadas em dois estádios. O estádio de plântula foi considerado quando os protófilos já estavam totalmente formados e de planta jovem, a partir do surgimento do 2° protófilo. Os elementos vegetativos descritos e ilustrados foram radícula, coleto, hipocótilo, cotilédones, epicótilo, protófilos e caule. O fruto de Sesbania virgata é um legume indeiscente medindo 5,8 cm e contendo de 2 a 6 sementes. A semente de Sesbania virgata é reniforme, endospérmica e desprovida do tegma, sendo que o hilo e o estrófíolo são facilmente distinguíveis. O eixo-embrionário encontra-se inserido aos cotilédones, sendo apical e invaginado papilonáceo. A germinação da semente é do tipo epígea. A plântula jovem apresenta protófilos compostos com 4 a 9 pares de folíolos pequenos opostos e glabros com peciolo e pulvino, e no ápice há uma pequena expansão laminar glabra. A radícula é sublenhosa, de cor branca ou marrom castanho.

O presente trabalho teve por objetivo descrever a morfologia dos frutos, das sementes e o desenvolvimento pós-seminal de faveira (Clitoria fairchildiana R. A. Howard. - Fabaceae). Os frutos e as sementes foram caracterizados quanto à forma e dimensões. Periodicamente, unidades representativas de cada fase de germinação foram retiradas para as descrições morfológicas. Os frutos são do tipo legume, deiscentes e de coloração marrom. As sementes são exalbuminosas, de forma orbicular e achatada, com tegumento de coloração castanho-esverdeada. O hilo tem forma elíptica, homocromo e de tamanho pequeno em relação à semente. Os cotilédones são livres, de coloração verde, maciços e plano-convexos e o embrião é invaginado. O início do desenvolvimento pós-seminal é marcado pelo rompimento do tegumento e emissão da raiz primária, glabra, de coloração amarelo-esverdeada e de forma cilíndrica. Posteriormente, observa-se o desenvolvimento das raízes secundárias, levemente esbranquiçadas, curtas e filiformes. Em seguida, o crescimento do hipocótilo proporciona a emergência dos cotilédones e da plúmula acima do substrato. O epicótilo alonga-se e, em seguida, observa-se a expansão dos eófilos, simples, opostos, com tricomas simples e esparsos, pecíolos curtos com estípulas em sua base. Posteriormente ao desenvolvimento dos eófilos, ocorre a formação do segundo par de folhas, alternas, trifolioladas, estipuladas e com estipelas na base dos peciólulos.

Objetivou-se nesse estudo avaliar o papel atenuador da espermidina exógena sobre a germinação, vigor de sementes e crescimento de plântulas de cultivares das forrageiras alfafa, guandu e labe-labe submetidas ao estresse salino. A semeadura foi realizada em caixas de plástico tipo "gerbox" forradas com papel de filtro umedecidos com soluções salinas nas concentrações de 0, 20, 40, 60, 80 e 120 mM de NaCl contendo 0 ou 0,5 mM de espermidina. O delineamento experimental foi inteiramente casualizado em esquema fatorial 3 x 6 x 2 (forrageiras x salinidade x espermidina) com cinco repetições de 25 sementes. As avaliações da germinação foram realizadas no quarto e décimo dias, juntamente com o índice de velocidade de germinação (IVG), avaliado até o nono dia após a semeadura. Determinou-se a concentração salina que reduz em 50% a germinação e a massa seca da parte aérea e raiz no décimo dia após a semeadura. A aplicação de espermidina exógena proporcionou maior acúmulo de massa seca das raízes de labe-labe cv. Rongai, além de ter possibilitado a germinação de 50% das sementes de guandu cv. Caqui até a concentração salina de 72,09 mM de NaCl, assim como maior IVG nas sementes das leguminosas alfafa cv. Crioula e labe-labe cv. Rongai.

ABSTRACT Fescues consist of wild and cultivated grasses that have adapted to a wide range of environmental conditions. They are an excellent model species for evolutionary ecology studies that investigate symbiosis and polyploidization and their effects on plant performance. First, they are frequently infected with symbiotic endophytic fungi known to affect a plant’s ability to cope with biotic and abiotic environmental factors. Second, fescue species have been reported to have substantial intraspecific variation in their ploidy level and morphology. In my thesis, I examined large-scale generalizations for frequency of polyploidy and endophyte infections and their effects on plant morphology. As a model species, I selected red (Festuca rubra) and viviparous sheep’s (F. vivipara) fescues. They are closely related, but they differ in terms of distribution and endophyte infection frequency. I investigated the biogeographic pattern and population biology of 29 red and 12 viviparous sheep’s fescue populations across ≈300 latitudes in Europe (400-690 N). To examine plant ploidy levels, I implemented time- and cost-efficient plate-based high throughput flow cytometric analysis. This efficient procedure enabled me to analyze over 1000 red fescue individuals. I found three ploidy levels among them: overall 84 %, 9 % and 7 % of the red fescue plants were hexaploid, tetraploid and octoploid, respectively. However, all viviparous sheep’s fescue plants were tetraploid. Ploidy level of red fescue appeared to some extent follow gradients in latitude and primary production as suggested by previous studies, but these results could be explained better by taking the sampling design and local adaptation into account. Three Spanish populations were mostly tetraploids and one high elevation population in northernmost Finland (Halti) was octoploid, while most other populations (25 sites) were dominated by hexaploids. Endophyte infection frequencies of wild fescue populations varied from 0 to 81 % in red fescue populations and from 0 to 30 % in viviparous sheep’s fescue populations. No gradients with latitude or primary production of the sites were detected. As taxonomy of red fescues is somewhat unclear, I also studied morphology, ploidy variation and endophyte status of proposed subspecies of European red fescues. Contrary to previous literature, different ploidy levels occurred in the same subspecies. In addition to wild fescues, I also used two agronomically important cultivars of meadow and tall fescue (Schedonorus phoenix and S. pratensis). As grass-legume mixtures have an agronomic advantage over monocultures in meadows, I carried out a mixture/competition experiment with fescues and red clover to find that species composition, nutrient availability and endophyte status together determined the total biomass yield that was higher in mixtures compared to monocultures. The results of this thesis demonstrate the importance of local biotic and abiotic factors such as grazing gradients and habitat types, rather than suggested general global geographical or environmental factors on grass polyploidization or its association with symbiotic endophytic fungi. I conclude that variation in endophyte infection frequencies and ploidy levels of wild fescues support the geographic mosaic theory of coevolution. Historical incidents, e.g., glaciation and present local factors, rather than ploidy or endophyte status, determine fescue morphology.
