Eighteen isolates of the red algae Chondrus crispus were collected from Northern Atlantic sites, together with C. ocellatus, C. yendoi and C. pinnulatus from the North Pacific. The nuclear rDNA internal transcribed spacer (ITS) was sequenced and compared, spanning both the ITS regions and the 5.8S rRNA gene. Percentage of nucleotide variation for C. crispus ranged from 0.3% to 4.0%. Phylogenetic analyses were performed using maximum parsimony (MP), neighbor-joining (NJ) and minimum evolution methods. They showed that two main clades existed within the C. crispus samples examined and that suggested C. crispus had a single Atlantic origin. The clustering however did not follow the geographic origin. We hypothesized that the current distribution of C. crispus populations might be a result of three main factors: temperature boundaries, paleoclimate and paleoceanography. ITS data exhibited abundant molecular information not only for phylogeographical investigation but also for systematics studies.

An electrochemical technique for the real-time detection of hydrogen peroxide (H2O2) was employed to describe respiratory burst activity (RBA) of phagocytes in plasma which can be used to evaluate the ability of immune system and disease resistance. The method is based upon the electric current changes, by redox reaction on platinum electrode of extracellular hydrogen peroxide (H2O2) released from phagocytes stimulated by the zymosan at 680 mV direct current (d.c.). Compared with the control, activation of respiratory burst by zymosan particles results in a high amperometric response, and a current peak was obtained during the whole monitoring process. The peak current was proved by addition Of Cu2+ and other controls, to be the result of intense release of H2O2 from phagocytes. The peak area was calculated and used to evaluate the quantity of effective H2O2, which represents the quantity of H2O2 beyond the clearance of related enzymes in plasma. According to Faraday's law, the phagocytes' ability of prawns to generate effective H2O2 was evaluated from 1.253 x 10(-14) mol/cell to 6.146 x 10(-14) mol/cell, and carp from 1.689 x 10(-15) Mol/Cell to 7.873 x 10(-1)5 mol/cell. This method is an acute and quick detection of extracellular effective H2O2 in plasma and reflects the capacity of phagocytes under natural conditions, which could be applied for selecting species and parents with high immunity for breeding in aquaculture. Resting cysts of the marine phytoplanktonic dinoflagellate Scrippsiella spp. are encountered in coastal habitats and shallow seas all over the world. Identification of Scrippsiella species requires information on cyst morphology because the plate pattern of the flagellated cell is conserved. Cysts from sediments of the East China Sea were identified based on traits from both the cysts and the thecal patterns of germinated cells. Calcareous cysts belonged predominantly to S. trochoidea (F. Stein) A. R. Loebl., S. rotunda J. Lewis, and S. precaria Montresor et Zingone. The former two species also produced smooth and noncalcified cysts in the field. A new species, S. donghaienis H. Gu sp. nov, was obtained from six noncalcified cysts with organic spines. These cysts are spherical, full of pale white and greenish granules with a mesoepicystal archeopyle. The vegetative cells consist of a conical epitheca and a round hypotheca with a plate formula of po, x, 4', 3a, 7 '', 6c (5c + t), 6 s, 5''', 2'''' and are morphologically indistinguishable from S. trochoidea. Results of internal transcribed spacer (ITS) sequence comparisons revealed that S. donghaienis was distinct from the S. trochoidea complex and appeared nested within the Calciodinellum/Calcigonellum clade. Culture experiments showed that the presence of a red body in the cyst and the shape of the archeopyle were constant within cell lines from one generation to the next, while the morphological features of the cyst wall, such as calcification and spine shape, appeared to be phenotypically plastic.

Variations of cellular total lipid, total carbohydrate and total protein content of two dominant bloom-forming species (Skeletonema costatum and Prorocentrum donghaiense) isolated from the Yangtze River Estuary were examined under six different nutrient conditions in batch cultures. Daily samples were collected to estimate the cell growth, nutrient concentration and three biochemical compositions content during 7 days for S. costatum and the same sampling procedure was done every other day during 10 days for P. donghaiense. Results showed that for S. costatum, cellular total lipid content increased under phosphorus (P) limitation, but not for nitrogen (N) limitation; cellular carbohydrate were accumulated under both N and P limitation: cellular total protein content of low nutrient concentration treatments were significantly lower than that of high nutrient concentration treatments. For P. donghaiense, both cellular total lipid content and total carbohydrate content were greatly elevated as a result of N and P exhaustion, but cellular total protein content had no significant changes under nutrient limitation. In addition, the capability of accumulation of three biochemical constituents of P. donghaiense was much stronger than that of S. costatum. Pearson correlation showed that for both species, the biochemical composition of three constituents (lipid, carbohydrate and protein) had no significant relationship with extracellular N concentration, but had positive correlation with extracellular and intracellular P concentration. The capability of two species to accumulate cellular total lipid and carbohydrate under nutrient limitation may help them accommodate the fluctuating nutrient condition of the Yangtze River Estuary. The different responses of two species of cellular biochemical compositions content under different nutrient conditions may provide some evidence to explain the temporal characteristic of blooms Caused by two species in the Yangtze River Estuary. In this paper, the detailed morphology of Prorocentrum donghaiense Lu from both field samples and cultures was examined, and a taxonomic comparison was made between P donghaiense and some related Prorocentrum spp. using morphological and molecular data and other published information. There were distinct differences among these species in morphological characteristics that historically have been presented as conservative features. The discrepancies extended beyond that of individual variations within the same species due to environmental factors. Therefore, these morphological features may not be conservative but, rather, polymorphic depending on environmental conditions. Based on this analysis, we suggest that the high-biomass bloom-forming species in the East China Sea, previously reported as Prorocentrum dentatum Stein, is P donghaiense Lu. The species reported from the East China Sea and Japanese and Korean waters appear to be the same species. Molecular data also suggest that P. dentatum (CCMP1517) and P. donghaiense are genetically identical. Therefore, the geographic distribution of P. donghaiense may be much wider than expected. To investigate harmful effects of the dinoflagellate Alexandrium species on microzooplankton, the rotifer Brachionus plicatilis was chosen as an assay species, and tested with 10 strains of Alexandrium including one known non-PSP-producer (Alexandrium tamarense, AT-6). HPLC analysis confirmed the PSP-content of the various strains: Alexandrium lusitanicum, Alexandrium minutum and Alexandrium tamarense (ATHK, AT5-1, AT5-3, ATC102, ATC103) used in the experiment were PSP-producers. No PSP toxins were detected in the strains Alexandrium sp1, Alexandrium sp2. Exposing rotifer populations to the densities of 2000 cells ml(-1) of each of these 10 Alexandrium strains revealed that the (non-PSP) A. tarnarense (AT-6) and two other PSP-producing algae: A. lusitanicum, A. minutum, did not appear to adversely impact rotifer populations. Rotifers exposed to these three strains were able to maintain their population numbers, and in some cases, increase them. Although some increases in rotifer population growth following exposures to these three algal species were noted, the rate was less than for the non-exposed control rotifer groups. In contrast, the remaining seven algal strains (A. tamarense ATHK, AT5-1, AT5-3, ATC102, ATC103; also Alexandrium sp1 and Alexandrium sp2) all have adverse effects on the rotifers. Dosing rotifers with respective algal cell densities of 2000 cells ml-1 each, for Alexandrium spl, Alexandrium sp2, and A. tamarense strains ATHK and ATC103 showed mean lethal time (LT50) on rotifer populations of 21, 28, 29, and 36h, respectively. The remaining three species (A. tamarense strains AT5-1, AT5-3, ATC102) caused respective mean rotifer LT50S of 56, 56, and 71 h, compared to 160 h for the unexposed "starved control" rotifers. Experiments to determine ingestion rates for the rotifers, based on changes in their Chlorophyll a content, showed that the rotifers could feed on A. lusitanicum, A. minutum and A. tamarense strain AT-6, but could graze to little or no extent upon algal cells of the other seven strains. The effects on rotifers exposed to different cell densities, fractions, and growth phases of A. tamarense algal culture were respectively compared. It was found that only the whole algal cells had lethal effects, with strongest impact being shown by the early exponential growth phase of A. tamarense. The results indicate that some toxic mechanism(s), other than PSP and present in whole algal cells, might be responsible for the adverse effects on the exposed rotifers. (C) 2004 Elsevier B.V. The recovery and fate of three species of dinoflagellates, Alexandrium tamarense, Cochlodinium polykrikoides and Scrippsiella trochoidea, after having been sedimented by yellow clay, were investigated in the laboratory. The effect of burying period in yellow clay pellet and mixing on the recovery of settled algal cells were studied. The morphological changes of algal cells in yellow clay pellet were also tracked. Results showed that there was almost no recovery for A. tamarense and C. polykrikoides, and the cells decomposed after 2-3 days after visible changes in morphology and chloroplasts. There was some recovery for S. trochoidea. Moreover, S. trochoidea cysts were formed in clay pellet during the period of about 14 days, with the highest abundance of 87 000 cysts g(-1) clay and the incidence of cyst formation of 6.5%, which was considered as a potential threat for the further occurrence of algal blooms. S. trochoidea cysts were isolated from yellow clay and incubated to test their viability, and a germination ratio of more than 30% was obtained after incubation for 1 month. These results showed the species specificity of the mitigation effect of yellow clay. It is suggested that cautions be taken for some harmful species and thorough risk assessments be conducted before using this mitigation strategy in the field.

This paper provides basic information on the general ecology and life history cycles of various flatfish species in the Bohai Sea, China. The species studied are Paralichthys olivaceus (Temminck & Schlegel), Cleisthenes herzensteini (Schmidt), Eopsetta grigorjewi (Herzenstein), Verasper variegatus (Temminck & Schlegel), Pleuronichthys cornutus (Temminck & Schlegel), Pseudopleuronectes yokohamae (Gunther), Pseudopleuronectes herzensteini (Jordan & Snyder), Kareius bicoloratus (Basilewsky), Zebrias zebra (Bloch), Cynoglossus semilaevis Gunther, Cynoglossus abbreviatus (Gray) and Cynoglossus joyneri Gunther. Information on reproduction, eggs and larval distribution, growth and adult abundance is presented. Based on the biology and ecology of these flatfish, artificial enhancement of the commercial species in the Bohai Sea is discussed.

The effects of fresh thalli and culture medium filtrates from two species of marine macroalgae, Ulva pertusa Kjellm (Chlorophyta) and Gracilaria lemaneiformis (Bory) Dawson (Rhodophyta), on growth of marine microalgae were investigated in co-culture under controlled laboratory conditions. A selection of microalgal species were used, all, being identified as bloom-forming dinoflagellates: Prorocentrum donghaiense Lu sp., Alexandrium tamarense (Lebour) Balech, Amphidinium carterae Hulburt and Scrippsiella trochoide (Stein) Loeblich III. Results showed that the fresh thalli of either U. pertusa or G. lemaneiformis significantly inhibited the microalgal growth, or caused mortality at the end of the experiment. However, the overall effects of the macroalgal culture filtrates on the growth of the dinoflagellates were species-specific (inhibitory, stimulatory or none) for different microalgal species. Results indicated an allelopathic effect of macroalga on the co-cultured dinoflagellate. We then took P. donghaiense as an example to further assess this hypothesis. The present study was carried out under controlled conditions, thereby excluded the fluctuation in light and temperature. Nutrient assays showed that nitrate and phosphate were almost exhausted in G. lemaneiformis co-culture. but remained at enough high levels in U pertusa co-culture, which were well above the nutrient limitation for the microalgal growth, when all cells of P. donghaiense were killed in the co-culture. Daily f/2 medium enrichment greatly alleviated the growth inhibition on P. donghaiense in G. lemaneiformis co-culture, but could not eliminate it. Other environmental factors, such as carbonate limitation, bacterial presence and the change of pH were also not necessary for the results. We thus concluded that allelopathy was the most possible reason leading to the negative effect of U. pertusa on P. donghaiense, and the combined roles of allelopathy and nutrient competition were essential for the effect of G. lemaneiformis on P. donghaiense. The effects of Heterosigma akashiwo on the early development of Argopecten irradians Lamarck: eggs, D-shaped larvae, eye-spot larvae and juveniles, were investigated under laboratory conditions. Exposing fertilized eggs to various densities of H. akashiwo algal culture revealed that the development of the embryos to the gastrula was significantly slowed at densities of more than 1 X 10(4) cells/ml algal cells, and mostly was arrested when the embryos reached the trochophore larvae stage. At this stage, several trochophore larvae were adhered together by the algal cells, resulting in the inhibition of their swimming activity. Larvae had still not developed into D-shaped larvae after 30 h, and therefore did not finish the hatching process. The attachment and adherence of the algal cells to the larvae might be an important process in the mechanism of the impact on egg hatching success. The activity of the D-shaped larvae was significantly inhibited after 48 h exposure to H. akashiwo at a density of 15 X 10(4) cells/ml and after 96 h at 10 X 10(4) cells/ml. The survival rate of the eye-spot larvae was decreased significantly after 48 h exposure to the algal culture at densities of more than 1 X 10(4) cells/ml. However, all the juveniles could survive and their climbing and attachment activity were not affected after 1 and 5 h exposure to the algal culture at all the various algal cell densities tested from 5 to 20 X 10(4) cells/ml. The results indicated that susceptibility of embryos or larvae to the alga H. akashiwo differs depending on the developmental stage. The embryos and the eye-spot larvae of A. irradians are more sensitive stages to the toxicity of H. akashiwo. Observed effects of H. akashiwo exposure on early development of A. irradians serve to point out to the potential danger of this alga for scallop populations. The possible toxicological mechanisms of H. akashiwo on the scallop embryos and larvae are discussed. (c) 2005 Elsevier B.V All rights reserved.

The present paper reports six genera nine species of Palaemonidae collected from Clipperton Island, eastern tropical Pacific, including three new species of the pontoniine genus Chacella: C. bicornuta, C. quadricornuta, and C. sexicornuta. We provide a key to identify all the six known species of Chacella.

Four new species, Eudesme huanghaiensis Ding et Lu, E. qingdaoensis Ding et Lu, E. shandongensis Ding et Lu and Sphaerotrichia huanghaiensis Ding et Lu, from the western Yellow Sea coast of China are described. Eudesme huanghaiensis is mainly characterized by its spherical or sub-spherical sub-cortical cells, its rhizoidal filaments developing from the basal cells of sub-cortex and its broad sub-cortical and medullary layers. E. qingdaoensis is mainly characterized by its long medullary cells, generally hollow center of the medulla, short sub-cortex with only 3-4 cylindrical cells and long, slender and clavate terminal cells of the rhizoidal filaments. E. shandongensis is mainly characterized by its hollow frond, thick cell walls of both medulla and inner sub-cortical layers and the spherical terminal cells of the rhizoid filaments. Sphaerotrichia huanghaiensis is mainly characterized by its cylindrical, sparsely branched frond with acute angle, and its thick 5-6 layered sub-cortex with long assimilating filaments of 6-10 cells.

In this paper, four new species of malacocarpic Sargassum are described: S. fuliginosoides Tseng et Lu sp. nov. is characterized by its discoid holdfast, the presence of the bulbs on the basal parts of the primary branches and very thick, lanceolate leaves. S. gemmiphorum Tseng et Lu sp. nov. is characterized by its conical holdfast, usually branched axis and very long, narrow, thin, denticulate leaves. S. shandongense Tseng, Zhang et Lu sp. nov. is characterized by its discoid holdfast, flattened primary branches, its leaves mostly entire or wavy at the margins and its racemose to paniculate receptacles. S. qingdaoense Tseng et Lu sp. nov. is characterized by the presence of the glandular dots on the ultimate branches and elongated lanceolate, acute, mostly wavy leaves.

A detailed study of materials from our herbarium shows the presence of two species of Gelidiella and three species of Pterocladiella, including two new records and a new species. Previously recorded species include G. acerosa and P. capillacea. G. bornetii and P. caerulescens are new records for China and P. yinggehaiensis is believed to be a new species. This new species is characterized by its small thalli, 1.5-2 cm high, that are more densely branched on the middle to upper parts for one half their lengths. These characteristics are so distinctive that they can be used to separate this species readily from other species now reported in this genus.
