Este artigo apresenta os resultados de análise de composição florística, abundância e aspectos fitossociológicos da flora pteridofítica em três ambientes da Área de Pesquisa Ecológica do Rio Guamá, localizada em Belém, PA. Os ambientes estudados foram floresta terra firme (Reserva Mocambo), Floresta de Igapó (Reserva Catu) e a transição entre estes dois ambientes. Em cada sítio foram sorteadas seis parcelas de 5m x 10 m. Dentro de cada parcela foram registradas as espécies ocorrentes, contados os indivíduos e anotadas as formas de vida. A flora de pteridófitas inventariada foi de 12 espécies distribuídas em 11 gêneros e nove famílias botânicas. A maior riqueza específica medida foi na área de igapó, seguida da área de transição e de terra firme, embora a de terra firme tenha apresentado maior densidade de indivíduos. Algumas espécies apresentam elevada freqüência e abundância em mais de um hábitat, enquanto outras podem ser localmente raras e restritas a um só ambiente. A floresta de igapó apresentou menor similaridade tanto internamente como quando comparada com a de terra firme e a de transição. As epífitas verdadeiras ou holoepífitas dominaram nos três ambientes estudados, sendo mais representativas no igapó, onde a inundação do solo deve inibir as formas terrestres. Estratégias de proteção para este grupo de plantas devem priorizar unidades de conservação que incluam variação ambiental, uma vez que mesmo ambientes próximos e interligados podem incluir considerável número de espécies exclusivas ou restritas.

Os íons de metais pesados são conhecidos tanto pela sua importância fisiológica e industrial, bem como pelo risco ambiental e à saúde humana. Para elucidar o comportamento dessas espécies nos corpos hídricos, os quais recebem grande parte da descarga de metais, seja de origem antrópica ou por fontes naturais, é necessário entender as interações que elas apresentam com o meio, principalmente a especiação química. Um dos mais importantes processos pelos quais passam as espécies metálicas em corpos aquáticos naturais é a interação com a matéria orgânica dissolvida (MOD), que pode ser por adsorção, reações de troca iônica ou por complexação. Neste trabalho foram realizados vários experimentos com o objetivo de descrever o comportamento da complexação de três importantes cátions Cu(II), Cd(II) e Pb(II) com a matéria orgânica (ácido húmico comercial), sob condições diversas de força iônica em meio tamponado. Os resultados foram avaliados de acordo com o modelo de van den Berg e Kramer para a complexação de metais. O modelo foi aplicado na determinação da capacidade de complexação dos íons em amostras reais, oriundas de três rios maranhenses que integram a Amazônia legal: Itapecuru, Bacanga e Pericumã. Nas águas dos rios utilizou-se o parâmetro carbono orgânico dissolvido (COD) para expressar a MOD. Os resultados confirmaram forte interação entre a MOD e íons de metais pesados e que o modelo de van den Berg e Kramer é satisfatório para se estimar a constante de complexação (K) e a concentração de sítios de complexação (Lt). Nas amostras simuladas em laboratório a ordem de complexação dos metais foi Cu(II) > Cd(II) > Pb(II) e a capacidade de complexação mostrou ser linear em função da concentração de ácido húmico comercial. Acredita-se que por ter menor raio iônico, o íon Cu(II) possui maior afinidade com os sítios de complexação. Nas amostras retiradas dos corpos aquáticos, observou-se que o rio com maior concentração de COD (rio Bacanga) apresentou maior capacidade de complexação; entretanto, a ordem foi Pb(II) > Cu(II) > Cd(II) provavelmente devido à presença de íons Cu(II) em maior quantidade nas águas dos rios.

INTRODUCTION: EORTC trial 22991 was designed to evaluate the addition of concomitant and adjuvant short-term hormonal treatments to curative radiotherapy in terms of disease-free survival for patients with intermediate risk localized prostate cancer. In order to assess the compliance to the 3D conformal radiotherapy protocol guidelines, all participating centres were requested to participate in a dummy run procedure. An individual case review was performed for the largest recruiting centres as well. MATERIALS AND METHODS: CT-data of an eligible prostate cancer patient were sent to 30 centres including a description of the clinical case. The investigator was requested to delineate the volumes of interest and to perform treatment planning according to the protocol. Thereafter, the investigators of the 12 most actively recruiting centres were requested to provide data on five randomly selected patients for an individual case review. RESULTS: Volume delineation varied significantly between investigators. Dose constraints for organs at risk (rectum, bladder, hips) were difficult to meet. In the individual case review, no major protocol deviations were observed, but a number of dose reporting problems were documented for centres using IMRT. CONCLUSIONS: Overall, results of this quality assurance program were satisfactory. The efficacy of the combination of a dummy run procedure with an individual case review is confirmed in this study, as none of the evaluated patient files harboured a major protocol deviation. Quality assurance remains a very important tool in radiotherapy to increase the reliability of the trial results. Special attention should be given when designing quality assurance programs for more complex irradiation techniques.

In a recent paper Iyama and Yoshino consider two interesting examples of isolated singularities over which it is possible to classify the indecomposable maximal Cohen-Macaulay modules in terms of linear algebra data. In this paper we present two new approaches to these examples. In the first approach we give a relation with cluster categories. In the second approach we use Orlov's result on the graded singularity category. We obtain some new results on the singularity category of isolated singularities which may be interesting in their own right.

INTRODUCTION: Solid tumors are known to have an abnormal vasculature that limits the distribution of chemotherapy. We have recently shown that tumor vessel modulation by low-dose photodynamic therapy (L-PDT) could improve the uptake of macromolecular chemotherapeutic agents such as liposomal doxorubicin (Liporubicin) administered subsequently. However, how this occurs is unknown. Convection, the main mechanism for drug transport between the intravascular and extravascular spaces, is mostly related to interstitial fluid pressure (IFP) and tumor blood flow (TBF). Here, we determined the changes of tumor and surrounding lung IFP and TBF before, during, and after vascular L-PDT. We also evaluated the effect of these changes on the distribution of Liporubicin administered intravenously (IV) in a lung sarcoma metastasis model. MATERIALS AND METHODS: A syngeneic methylcholanthrene-induced sarcoma cell line was implanted subpleurally in the lung of Fischer rats. Tumor/surrounding lung IFP and TBF changes induced by L-PDT were determined using the wick-in-needle technique and laser Doppler flowmetry, respectively. The spatial distribution of Liporubicin in tumor and lung tissues following IV drug administration was then assessed in L-PDT-pretreated animals and controls (no L-PDT) by epifluorescence microscopy. RESULTS: L-PDT significantly decreased tumor but not lung IFP compared to controls (no L-PDT) without affecting TBF. These conditions were associated with a significant improvement in Liporubicin distribution in tumor tissues compared to controls (P < .05). DISCUSSION: L-PDT specifically enhanced convection in blood vessels of tumor but not of normal lung tissue, which was associated with a significant improvement of Liporubicin distribution in tumors compared to controls.

BACKGROUND: Photodynamic therapy (PDT) at low drug-light conditions can enhance the transport of intravenously injected macromolecular therapeutics through the tumor vasculature. Here we determined the impact of PDT on the distribution of liposomal doxorubicin (Liporubicin™) administered by isolated lung perfusion (ILP) in sarcomas grown on rodent lungs. METHODS: A syngeneic methylcholanthrene-induced sarcoma cell line was implanted subpleurally in the left lung of Fischer rats. Treatment schemes consisted in ILP alone (400 μg of Liporubicin), low-dose (0.0625 mg/kg Visudyne®, 10 J/cm(2) and 35 mW/cm(2)) and high-dose left lung PDT (0.125 mg/kg Visudyne, 10 J/cm(2) and 35 mW/cm(2)) followed by ILP (400 μg of Liporubicin). The uptake and distribution of Liporubicin in tumor and lung tissues were determined by high-performance liquid chromatography and fluorescence microscopy in each group. RESULTS: Low-dose PDT significantly improved the distribution of Liporubicin in tumors compared to high-dose PDT (p < 0.05) and ILP alone (p < 0.05). However, both PDT pretreatments did not result in a higher overall drug uptake in tumors or a higher tumor-to-lung drug ratio compared to ILP alone. CONCLUSIONS: Intraoperative low-dose Visudyne-mediated PDT enhances liposomal doxorubicin distribution administered by ILP in sarcomas grown on rodent lungs which is predicted to improve tumor control by ILP.

Several recent studies have further clarified the role of chemotherapy in newly diagnosed anaplastic glioma. For newly diagnosed glioblastoma, combined daily radiotherapy with daily temozolomide followed by six cycles of adjuvant temozolomide improves overall survival. This benefit is especially observed in patients with a methylated promotor of the MGMT gene which encodes an alkyltransferase; this observation however, needs confirmation. Although oligodendroglial tumours are sensitive to chemotherapy, classical adjuvant nitrosourea-based chemotherapy does not improve overall survival in newly diagnosed anaplastic oligodendroglioma, even in the subset of 1p/19q loss tumours. It may increase progression-free survival however, and further studies must show if combined modality treatment with daily chemotherapy during radiotherapy increases survival. Trials exploring the role of chemotherapy in low-grade glioma are ongoing. No standard chemotherapy is currently available for highly anaplastic glioma failing first-line temozolomide-based therapy.

Fluorescence imaging for detection of non-muscle-invasive bladder cancer is based on the selective production and accumulation of fluorescing porphyrins-mainly, protoporphyrin IX-in cancerous tissues after the instillation of Hexvix®. Although the sensitivity of this procedure is very good, its specificity is somewhat limited due to fluorescence false-positive sites. Consequently, magnification cystoscopy has been investigated in order to discriminate false from true fluorescence positive findings. Both white-light and fluorescence modes are possible with the magnification cystoscope, allowing observation of the bladder wall with magnification ranging between 30× for standard observation and 650×. The optical zooming setup allows adjusting the magnification continuously in situ. In the high-magnification (HM) regime, the smallest diameter of the field of view is 600 microns and the resolution is 2.5 microns when in contact with the bladder wall. With this cystoscope, we characterized the superficial vascularization of the fluorescing sites in order to discriminate cancerous from noncancerous tissues. This procedure allowed us to establish a classification based on observed vascular patterns. Seventy-two patients subject to Hexvix® fluorescence cystoscopy were included in the study. Comparison of HM cystoscopy classification with histopathology results confirmed 32?33 (97%) cancerous biopsies and rejected 17?20 (85%) noncancerous lesions.

Abstract: Background: Cancer/testis (CT) genes are expressed only in the germ line and certain tumors and are most frequently located on the X-chromosome (the CT-X genes). Amongst the best studied CT-X genes are those encoding several MAGE protein families. The function of MAGE proteins is not well understood, but several have been shown to potentially influence the tumorigenic phenotype. Methodology/Principal Findings: We undertook a mutational analysis of coding regions of four CT-X MAGE genes, MAGEA1, MAGEA4, MAGEC1, MAGEC2 and the ubiquitously expressed MAGEE1 in human melanoma samples. We first examined cell lines established from tumors and matching blood samples from 27 melanoma patients. We found that melanoma cell lines from 37% of patients contained at least one mutated MAGE gene. The frequency of mutations in the coding regions of individual MAGE genes varied from 3.7% for MAGEA1 and MAGEA4 to 14.8% for MAGEC2. We also examined 111 fresh melanoma samples collected from 86 patients. In this case, samples from 32% of the patients exhibited mutations in one or more MAGE genes with the frequency of mutations in individual MAGE genes ranging from 6% in MAGEA1 to 16% in MAGEC1. Significance: These results demonstrate for the first time that the MAGE gene family is frequently mutated in melanoma.
