A new series of donor acceptor copolymers were synthesized via the Witting route and applied as an active layer in organic thin-films solar cells. These copolymers are composed of fluorene thiophene and phenylene thiophene units. The ratio between those was systematically varied, and copolymers containing 0%, 50%, and 75% of phenylene thiophene were characterized and evaluated when used in photovoltaic devices. The copolymers' composition, photophysical, electrical, and morphological properties are addressed and correlated with device performance. The 50% copolymer ratio was found to be the best copolymer of the series, yielding a power conversion efficiency (PCE) under air mass (AM) 1.5 conditions of 2.4% in the bilayer heterojunction with the C-60 molecule. Aiming at flexible electronics applications, solutions based on the heterojunction of this copolymer with PCBM (6,6-phenyl-C-61-butyric acid methyl ester) were also successfully deposited using an inkjet printing method and used as an active layer in solar cells.

The oil industry uses gas separators in production wells as the free gas present in the suction of the pump reduces the pumping efficiency and pump lifetime. Therefore, free gas is one of the most important variables in the design of pumping systems. However, in the literature there is little information on these separators. It is the case of the inverted-shroud gravitational gas separator. It has an annular geometry due to the installation of a cylindrical container in between the well casing and pioduction pipe (tubing). The purpose of the present study is to understand the phenomenology and behavior of inverted-shroud separator. Experimental tests were performed in a 10.5-m-length inclinable glass tube with air and water as working fluids. The water flow rate was in the range of 8.265-26.117 l/min and the average inlet air mass flow rate was 1.1041 kg/h, with inclination angles of 15 degrees, 30 degrees, 45 degrees, 60 degrees, 75 degrees, 80 degrees and 85 degrees. One of the findings is that the length between the inner annular level and production pipe inlet is one of the most important design parameters and based on that a new criterion for total gas separation is proposed. We also found that the phenomenology of the studied separator is not directly dependent on the gas flow rate, but on the average velocity of the free surface flow generated inside the separator. Maps of efficiency of gas separation were plotted and showed that liquid flow rate, inclination angle and pressure difference between casing and production pipe outlet are the main variables related to the gas separation phenomenon. The new data can be used for the development of design tools aiming to the optimized project of the pumping system for oil production in directional wells. (C) 2012 Elsevier Inc. Abstract: Background: The testis-specific isoform of angiotensin-converting enzyme (tACE) is exclusively expressed in germ cells during spermatogenesis. Although the exact role of tACE in male fertility is unknown, it clearly plays a critical function in spermatogenesis. The dipeptidase domain of tACE is identical to the C-terminal catalytic domain of somatic ACE (sACE). Bradykinin potentiating peptides (BPPs) from snake venoms are the first natural sACE inhibitors described and their structure–activity relationship studies were the basis for the development of antihypertensive drugs such as captopril. In recent years, it has been showed that a number of BPPs – including BPP-10c – are able to distinguish between the N- and C-active sites of sACE, what is not applicable to captopril. Considering the similarity between tACE and sACE (and since BPPs are able to distinguish between the two active sites of sACE), the effects of the BPP-10c and captopril on the structure and function of the seminiferous epithelium were characterized in the present study. BPP-10c and captopril were administered in male Swiss mice by intraperitoneal injection (4.7 μmol/kg for 15 days) and histological sections of testes were analyzed. Classification of seminiferous tubules and stage analysis were carried out for quantitative evaluation of germ cells of the seminiferous epithelium. The blood-testis barrier (BTB) permeability and distribution of claudin-1 in the seminiferous epithelium were analyzed by hypertonic fixative method and immunohistochemical analyses of testes, respectively. Results: The morphology of seminiferous tubules from animals treated with BPP-10c showed an intense disruption of the epithelium, presence of atypical multinucleated cells in the lumen and degenerated germ cells in the adluminal compartment. BPP-10c led to an increase in the number of round spermatids and total support capacity of Sertoli cell in stages I, V, VII/VIII of the seminiferous epithelium cycle, without affecting BTB permeability and the distribution of claudin-1 in the seminiferous epithelium. Interestingly, no morphological or morphometric alterations were observed in animals treated with captopril. Conclusions: The major finding of the present study was that BPP-10c, and not captopril, modifies spermatogenesis by causing hyperplasia of round spermatids in stages I, V, and VII/VIII of the spermatogenic cycle.

Paracoccidoides brasiliensis adhesion to lung epithelial cells is considered an essential event for the establishment of infection and different proteins participate in this process. One of these proteins is a 30 kDa adhesin, pI 4.9 that was described as a laminin ligand in previous studies, and it was more highly expressed in more virulent P. brasiliensis isolates. This protein may contribute to the virulence of this important fungal pathogen. Using Edman degradation and mass spectrometry analysis, this 30 kDa adhesin was identified as a 14-3-3 protein. These proteins are a conserved group of small acidic proteins involved in a variety of processes in eukaryotic organisms. However, the exact function of these proteins in some processes remains unknown. Thus, the goal of the present study was to characterize the role of this protein during the interaction between the fungus and its host. To achieve this goal, we cloned, expressed the 14-3-3 protein in a heterologous system and determined its subcellular localization in in vitro and in vivo infection models. Immunocytochemical analysis revealed the ubiquitous distribution of this protein in the yeast form of P. brasiliensis, with some concentration in the cytoplasm. Additionally, this 14-3-3 protein was also present in P. brasiliensis cells at the sites of infection in C57BL/6 mice intratracheally infected with P. brasiliensis yeast cells for 72 h (acute infections) and 30 days (chronic infection). An apparent increase in the levels of the 14-3-3 protein in the cell wall of the fungus was also noted during the interaction between P. brasiliensis and A549 cells, suggesting that this protein may be involved in host-parasite interactions, since inhibition assays with the protein and this antibody decreased P. brasiliensis adhesion to A549 epithelial cells. Our data may lead to a better understanding of P. brasiliensis interactions with host tissues and paracoccidioidomycosis pathogenesis.

Insect storage proteins accumulate at high levels during larval development of holometabolous insects. During metamorphosis they are degraded, supplying energy and amino acids for the completion of adult development. The genome of Culex quinquefasciatus contains eleven storage protein-coding genes. Their transcripts are more abundant in larvae than in pupae and in adults. In fact, only four of these genes are transcribed in adults, two of which in blood-fed adult females but not in adult males. Transcripts corresponding to all Cx. quinquefasciatus storage proteins were detected by RT-PCR, while mass spectrometric analysis of larval and pupal proteins identified all storage proteins with the exception of one encoded by Cq LSP1.8. Our results indicate that the identified Cx. quinquefasciatus storage protein-coding genes are candidates for identifying regulatory sequences for the development of molecular tools for vector control

[EN]Low cost real-time depth cameras offer new sensors for a wide field of applications apart from the gaming world. Other active research scenarios as for example surveillance, can take ad- vantage of the capabilities offered by this kind of sensors that integrate depth and visual information. In this paper, we present a system that operates in a novel application context for these devices, in troublesome scenarios where illumination conditions can suffer sudden changes. We focus on the people counting problem with re-identification and trajectory analysis.

[EN]African dust pulses have important effects on the climate and marine biogeochemistry of the Canary Region. Common factors influencing the air mass concentration, elemental and composition of aerosol particles and atmospheric deposition fluxes of soluble Fe, Al, Mn, Ti, Cu and Co were examined. Aerosol samples have been collected at three stations in Gran Canaria Island (Taliarte at sea level, Tafira at 269 m altitude and Pico de la Gorra 1930 m altitude) since 2002. Air-mass back trajectories (HySPLIT, NOAA) suggested that the Sahara desert is the major sources of dust (dominant during 32-50% of days) while the Sahel desert was the major source only 2-10% of the time (maximum in summerautumn).

The motivation for the work presented in this thesis is to retrieve profile information for the atmospheric trace constituents nitrogen dioxide (NO2) and ozone (O3) in the lower troposphere from remote sensing measurements. The remote sensing technique used, referred to as Multiple AXis Differential Optical Absorption Spectroscopy (MAX-DOAS), is a recent technique that represents a significant advance on the well-established DOAS, especially for what it concerns the study of tropospheric trace consituents. NO2 is an important trace gas in the lower troposphere due to the fact that it is involved in the production of tropospheric ozone; ozone and nitrogen dioxide are key factors in determining the quality of air with consequences, for example, on human health and the growth of vegetation. To understand the NO2 and ozone chemistry in more detail not only the concentrations at ground but also the acquisition of the vertical distribution is necessary. In fact, the budget of nitrogen oxides and ozone in the atmosphere is determined both by local emissions and non-local chemical and dynamical processes (i.e. diffusion and transport at various scales) that greatly impact on their vertical and temporal distribution: thus a tool to resolve the vertical profile information is really important. Useful measurement techniques for atmospheric trace species should fulfill at least two main requirements. First, they must be sufficiently sensitive to detect the species under consideration at their ambient concentration levels. Second, they must be specific, which means that the results of the measurement of a particular species must be neither positively nor negatively influenced by any other trace species simultaneously present in the probed volume of air. Air monitoring by spectroscopic techniques has proven to be a very useful tool to fulfill these desirable requirements as well as a number of other important properties. During the last decades, many such instruments have been developed which are based on the absorption properties of the constituents in various regions of the electromagnetic spectrum, ranging from the far infrared to the ultraviolet. Among them, Differential Optical Absorption Spectroscopy (DOAS) has played an important role. DOAS is an established remote sensing technique for atmospheric trace gases probing, which identifies and quantifies the trace gases in the atmosphere taking advantage of their molecular absorption structures in the near UV and visible wavelengths of the electromagnetic spectrum (from 0.25 μm to 0.75 μm). Passive DOAS, in particular, can detect the presence of a trace gas in terms of its integrated concentration over the atmospheric path from the sun to the receiver (the so called slant column density). The receiver can be located at ground, as well as on board an aircraft or a satellite platform. Passive DOAS has, therefore, a flexible measurement configuration that allows multiple applications. The ability to properly interpret passive DOAS measurements of atmospheric constituents depends crucially on how well the optical path of light collected by the system is understood. This is because the final product of DOAS is the concentration of a particular species integrated along the path that radiation covers in the atmosphere. This path is not known a priori and can only be evaluated by Radiative Transfer Models (RTMs). These models are used to calculate the so called vertical column density of a given trace gas, which is obtained by dividing the measured slant column density to the so called air mass factor, which is used to quantify the enhancement of the light path length within the absorber layers. In the case of the standard DOAS set-up, in which radiation is collected along the vertical direction (zenith-sky DOAS), calculations of the air mass factor have been made using “simple” single scattering radiative transfer models. This configuration has its highest sensitivity in the stratosphere, in particular during twilight. This is the result of the large enhancement in stratospheric light path at dawn and dusk combined with a relatively short tropospheric path. In order to increase the sensitivity of the instrument towards tropospheric signals, measurements with the telescope pointing the horizon (offaxis DOAS) have to be performed. In this circumstances, the light path in the lower layers can become very long and necessitate the use of radiative transfer models including multiple scattering, the full treatment of atmospheric sphericity and refraction. In this thesis, a recent development in the well-established DOAS technique is described, referred to as Multiple AXis Differential Optical Absorption Spectroscopy (MAX-DOAS). The MAX-DOAS consists in the simultaneous use of several off-axis directions near the horizon: using this configuration, not only the sensitivity to tropospheric trace gases is greatly improved, but vertical profile information can also be retrieved by combining the simultaneous off-axis measurements with sophisticated RTM calculations and inversion techniques. In particular there is a need for a RTM which is capable of dealing with all the processes intervening along the light path, supporting all DOAS geometries used, and treating multiple scattering events with varying phase functions involved. To achieve these multiple goals a statistical approach based on the Monte Carlo technique should be used. A Monte Carlo RTM generates an ensemble of random photon paths between the light source and the detector, and uses these paths to reconstruct a remote sensing measurement. Within the present study, the Monte Carlo radiative transfer model PROMSAR (PROcessing of Multi-Scattered Atmospheric Radiation) has been developed and used to correctly interpret the slant column densities obtained from MAX-DOAS measurements. In order to derive the vertical concentration profile of a trace gas from its slant column measurement, the AMF is only one part in the quantitative retrieval process. One indispensable requirement is a robust approach to invert the measurements and obtain the unknown concentrations, the air mass factors being known. For this purpose, in the present thesis, we have used the Chahine relaxation method. Ground-based Multiple AXis DOAS, combined with appropriate radiative transfer models and inversion techniques, is a promising tool for atmospheric studies in the lower troposphere and boundary layer, including the retrieval of profile information with a good degree of vertical resolution. This thesis has presented an application of this powerful comprehensive tool for the study of a preserved natural Mediterranean area (the Castel Porziano Estate, located 20 km South-West of Rome) where pollution is transported from remote sources. Application of this tool in densely populated or industrial areas is beginning to look particularly fruitful and represents an important subject for future studies.

Über cDNA-Banken und RT-PCR wurden erstmals 15 Intermediärfilament-Proteine (IF-Proteine) des Flussneunauges Lampetra fluviatilis (Agnatha, kieferlose Wirbeltiere) kloniert und sequenziert: drei Typ I-Keratine, vier Typ II-Keratine, fünf keratinartige IF-Proteine (drei Kγ, zwei Kα), die Typ III-Proteine Vimentin und Desmin sowie ein Typ IV-Neurofilament-Protein (NF).Die IF-Proteine wurden aus verschiedenen Organen isoliert und durch zweidimensionale Polyacrylamid-Gelelektrophorese (2D-PAGE) aufgetrennt. Biochemische sowie massenspektrometrische Analysen anhand der 2D-PAGE ermöglichten in Kombination mit den Sequenzdaten die Identifizierung von Vimentin, Desmin sowie aller sequenzierten Keratine bis auf zwei der fünf Kα/Kγ-Proteine. Die meisten Keratine ließen sich darüber hinaus in die Kategorien „E“ (von „epidermal“) und „S“ (von „simple epithelial“) einteilen.Von den sequenzierten Keratinen ist das IIS-Keratin K8 wahrscheinlich ortholog zu den bekannten K8-Sequenzen höherer Vertebraten. Die Bezeichnung K18 für das einzige IS-Keratin des Neunauges in Anlehnung an das IS-Keratin K18 des Menschen basiert auf der stets beobachteten Koexpression mit K8 in einfachen Epithelien.Die Sequenz des Neunaugen-Vimentins zeigt große Übereinstimmungen mit den bekannten Desminsequenzen der Vertebraten. Die keratinartigen Proteine Kα und Kγ sind bis jetzt nur von Agnathen (Neunaugen und Schleimaale) bekannt.In molekularen Stammbäumen können K8, K18, Vimentin, Desmin und das NF_L des Neunauges gut als Außengruppe definiert werden.

Im Rahmen der Projekte CARIBIC ('Civil Aircraft for the Regular Investigation of the atmosphere Based on an Instrument Container') und INDOEX ('Indian Ocean Experi-ment') wurde ein Gaschromatograph mit Massenspektrometer zur Analyse von Luftproben auf Nicht-Methan-Kohlenwasserstoffe (NMKW) im ppb- und ppt-Bereich entwickelt. Während INDOEX erfolgte die Probennahme auf dem Forschungsschiff Ronald Brown, während CARIBIC mit einem automatischen Probensammler an Bord eines Passagierflugzeuges (Boeing 767-ER, LTU). Die NMKW-Meßergebnisse wurden zusammen mit Ergebnissen von Mes-sungen von Kohlenmonoxid (CO, einschließlich Isotopenzusam-mensetzung), Ozon (O3), Methan (CH4), Kohlendioxid (CO2), Distickstoffmonoxid (N2O), Schwefelhexafluorid (SF6) und Aerosoleigenschaften sowie meteorologischen Daten inter-pretiert. Während INDOEX (Februar / März 1999) wurde in der maritimen Grenzschicht (MBL) des Indischen Ozeans (IO) eine starke Variabilität diverser Spurengase beobachtet, die teilweise durch regionale Emissionen hervorgerufen wurde, die stärkste Variabilität war jedoch durch Langstrecken-transport aus mittleren Breiten der Nordhemisphäre bedingt. Aufgrund der Abweichungen vom klimatologischen Mittel, sollten regionale Quellen die MBL des IO im allgemeinen stärker beeinflussen. Die Einteilung des IO in meteorologi-sche Luftmassenregime wurde bestätigt. Starke Spurengasgra-dienten an der innertropischen Konvergenzzone (ITCZ) zeigen, daß die ITCZ in erster Linie den Austausch von Luftmassen zwischen den Hemisphären behindert. Bei CARIBIC werden Messungen von Spurengasen (ein-(schließlich NMKW) und Aerosoleigenschaften auf Flügen in der oberen Troposphäre / unteren Stratosphäre durchgeführt. Während eines Flug über Afrika wurden der Einfluß von durch Konvektion in die obere Troposphäre eingebrachten, frischen Abgasen aus Biomassenverbrennung nachgewiesen. Andere Luft-massen wurden durch Emissionen von Erdgas bzw. durch die Stratosphäre beeinflußt.

Zusammenfassung:In dieser Studie werden Deformationsprozesse im mesozoischen Torlesse Akkretionskeil (Neuseeland) quantifiziert, um Aufschluß über die Dynamik in Akkretionskeilen zu erhalten. Absolute und relative Verformungsmessungen zeigen sowohl im lokalen als auch regionalen Maßstab eine stark heterogene Deformation des Torlesse Keils. Die regionale Deformation wurde mit Hilfe einer Tensordurchschnittsberechnung, unter Benutzung einzelner lokaler Verformungsdaten, als uniaxiale Verkürzung entlang einer subvertikalen, maximalen Verkürzungsachse charakterisiert. Absolute Verformungsmessungen an niedriggradigen Metasandsteinen belegen darüber hinaus durchschnittliche Volumenverluste von ca. 20% SiO2. Volumenveränderungen in tieferkrustalen Aufschlüssen wurden mittels einer geochemischen Massenbilanzanalyse abgeschätzt. Chemische Zusammensetzungen höhergradiger Zonen weichen je nach Grad der Volumenverformung von der Protolitzusammensetzung ab und zeigen somit Verluste von 15% SiO2 an. Da Speicherorte für das gelöste Material nicht bekannt sind, muss angenommen werden, dass das Material aus dem Keil abtransportiert wurde. Die Verformungsergebnisse geben weiterhin Aufschluß über den Grad der Kopplung zwischen Akkretionskeil und subduzierter Platte. Die ermittelten Scherwerte in den Gesteinen liegen deutlich unter den zu erwartenden Scherwerten, die mittels eines einfachen Modells berechnet wurden, das sowohl verschiedene Konvergenzgeschwindigkeiten als auch Exhumierungsraten berücksichtigt. Dies belegt, dass der Torlesse Keil stark von der subduzierten pazifischen Platte entkoppelt war und die Deformation hauptsächlich durch den Fluß der Sedimente in und aus dem Keil bestimmt wurde.

Ein neu konstruierter Kondensationskernzähler COPAS (COndensation PArticle counting System) für in-situ-Messungen der Konzentration von Aitken-Teilchen und ultrafeinen Aerosolpartikeln wurde im Rahmen dieser Arbeit erstmals erfolgreich bei Flugzeugmessungen eingesetzt. COPAS ist ein für flugzeuggestützte Messungen an Bord des Forschungsflugzeuges „Geophysica“ in der oberen Troposphäre und unteren Stratosphäre angepaßtes und voll automatisiertes System. Die Verfahrensweise, die Aerosolpartikel des Größenbereichs mit Durchmessern d < 100 nm zum Anwachsen zu bringen, um sie mittels optischer Detektion zu erfassen, ist im COPAS durch das Prinzip der thermischen Diffusion realisiert, wodurch eine kontinuierliche Messung der Aerosolkonzentration mit der untersten Nachweisgrenze für Partikeldurchmesser von d = 6 nm gewährleistet ist. Durch die Verwendung einer Aerosolheizung ist die Unterscheidung von volatilem und nichtvolatilem Anteil des Aerosols mit COPAS möglich. In umfassenden Laborversuchen wurde das COPAS-System hinsichtlich der unteren Nachweisgrenze in Abhängigkeit von der Betriebstemperatur und bei verschiedenen Druckbedingungen charakterisiert sowie die Effizienz der Aerosolheizung bestimmt. Flugzeuggestützte Messungen fanden in mittleren und polaren Breiten im Rahmen des EUPLEX-/ENVISAT-Validierungs–Projektes und in den Tropen während der TROCCINOX/ENVISAT-Kampagne statt. Die Messungen der vertikalen Konzentrationsverteilung des Aerosols ergaben in polaren Breiten eine Zunahme der Konzentration oberhalb von 17 km innerhalb des polaren Vortex mit hohem Anteil nichtvolatiler Partikel von bis zu 70 %. Als Ursache hierfür wird der Eintrag von meteoritischen Rauchpartikeln aus der Mesosphäre in die obere und mittlere Stratosphäre des Vortex angesehen. Ferner konnte in der unteren Stratosphäre des polaren Vortex der Einfluß troposphärischer Luft aus niedrigen Breiten festgestellt werden, die sich in einer hohen Variabilität der Aerosolpartikelkonzentration manifestiert. In tropischen Breiten wurde die Tropopausenregion untersucht. Dabei wurden Konzentrationen von bis zu 104 ultrafeiner Aerosolpartikel mit 6 nm < d < 14 nm pro cm-3 Luft gemessen, deren hoher volatiler Anteil einen sicheren Hinweis darauf gibt, daß die Partikel durch den Prozeß der homogenen Nukleation gebildet wurden. Damit konnte erstmals die Schlußfolgerungen von Brock et al. (1995) durch direkte Messungen der ultrafeinen Partikelkonzentration weitergehend belegt werden, daß in der tropischen Tropopausenregion die Neubildung von Aerosolpartikeln durch homogene Nukleation stattfindet. Die vertikalen Verteilungen der stratosphärischen Aerosolpartikelkonzentration mittlerer Breiten verdeutlichen die Ausbildung einer über 6 Jahre hinweg nahezu konstanten Hintergrundkonzentration des stratosphärischen Aerosols unter vulkanisch unbeeinflußten Bedingungen. Ferner gibt die vergleichende Untersuchung der stratosphärischen Aerosolpartikelkonzentration aus polaren, mittleren und tropischen Breiten Aufschluß über den Transport und die Prozessierung des stratosphärischen Aerosols und insbesondere über den Austausch von Luftmassen zwischen der Stratosphäre und der Troposphäre.

The aims of this work were to investigate the role of nuclear Phospholipase C beta 1 (PI-PLCβ1) in human and mouse cell lines and to identify new binding partners of nuclear PI-PLCβ1 to further understand the functional network in which the enzyme acts. The intracellular distribution of PI-PLCβ1 was further investigated in human leukaemia cell lines (NB4, HL60, THP1, CEM, Jurkat, K562). With the exception of HL60, a high endogenous level of PI-PLCβ1 was detected in purified nuclei in each of the cell lines. We found that also in Ba/F3 pro-B cells overexpressing PI-PLCβ1b the protein localize within the nucleus. Although our data demonstrated that PI-PLCβ1b was not involved in cell proliferation and IGF-1 response as shown in other cell lines (FELC and Swiss 3T3), there was an effect on apoptosis. Activation of early apoptotic markers caspase-3 and PARP was delayed in PI-PLCβ1b overexpressing Ba/F3 cells treated with 5 gr/ml mitomycin C for 24h. We performed an antibody-specific immunoprecipitation on nuclear lysates from FELC-PLCβ1b cells. Mass spectrometry analysis (nano-ESI-Q-TOF) of co-immunoprecipitated proteins allowed for identification of 92 potential nuclear PI-PLCβ1b interactors. Among these, several already documented PI-PLCβ1b interacting partners (Srp20, LaminB, EF1α2) were identified, further validating our data. All the identified proteins were nuclear, mostly localized within the nuclear speckles. This evidence is particularly relevant as PI-PLCβ1 is known to localize in the same domains. Many of the identified proteins are involved in cell cycle, proliferation and transcriptional control. In particular, many of the proteins are components of the spliceosome multi-complex, strengthening the idea that PI-PLCβ1b is involved in mRNA processing and maturation. Future work will aim to better characterize the regulatory role of PI-PLCβ1b in mRNA splicing.

Im Rahmen dieser Arbeit wurden zwei verschiedene Systeme untersucht, deren verbindende Gemeinsamkeit in den verwendeten ortsauflösenden, spektroskopischen Messmethoden der Oberflächenanalytik, wie z.B. abbildendes XPS, Röntgennahkanten-Photoemissionsmikroskopie (XANES-PEEM) und Augerspektroskopie (AES) liegt. Im ersten Teil der Arbeit wurden Diamant-Nukleationsdomänen auf Ir/SrTiO3 untersucht und mit vorherrschenden Modellen aus der Literatur verglichen. Die Nukleationsdomänen, wie sie im Mikrowellen-induzierten CVD Prozess unter Verwendung der BEN Prozedur (bias-enhanced nucleation) entstehen, bilden die „Startschicht“ für ein heteroepitaktisches Wachstum einer hoch orientierten Diamantschicht. Sie entwickeln sich aber unter Bedingungen, unter denen 3D-Diamant abgetragen und weggeätzt wird. Mittels XANES-PEEM Messungen konnte erstmals die lokale Bindungsumgebung des Kohlenstoffs in den Nukleationsdomänen ortsaufgelöst aufgezeigt werden und aus AES Messungen ließ sich die Schichtdicke der Nukleationsdomänen abschätzen. Es zeigte sich, dass die Nukleationsdomänen Bereiche mit etwa 1 nm Dicke darstellen, in denen der Übergang von eine sp2-koordinierte amorphen Kohlenstoff- zu einer Diamantschicht mit hohem sp3 Anteil abläuft. Zur Erklärung des Nukleationsprozesses wurde auf das „Clustermodell“ von Lifshitz et al. zurückgegriffen, welches um einen wesentlichen Aspekt erweitert wurde. Die Stabilität der Nukleationsdomänen gegen die Ätzwirkung des Nukleationsprozesses auf Volumendiamant wird durch eine starke Wechselwirkung zwischen dem Diamant und dem Iridiumsubstrat erklärt, wobei die Dicke von etwa 1 nm als Maß für die Ausdehnung dieses Wechselwirkungsbereichs angesehen wird. Der zweite Teil der Arbeit beschäftigt sich mit der Charakterisierung präsolarer SiC-Körner und darin eingeschlossener Spurenelemente. Neben den Hauptelementen Si und C wurden auch Spinelle wie Chromit (FeCr2O4), Korund (Al2O3) und auch verschiedene Spurenelemente (z. B. Al, Ba und Y) nachgewiesen. Ferner wurden XPS-Linien bei Energien nachgewiesen, welche sich den Seltenen Erden Erbium, Thulium und Dysprosium zuordnen lassen. Aufgrund von Abweichungen zur Literatur bzgl. der ausgeprägten Intensität der XPS-Linien, wurde als alternative Erklärungsmöglichkeit für verschiedene Signale der Nachweis von stark schwefelhaltigen Körnern (z.B. so genannte „Fremdlinge“) mit Aufladungen von mehreren Volt diskutiert. Es zeigt sich, dass abbildendes XPS und XANES-PEEM Methoden zur leistungsfähigen chemischen Charakterisierung von SiC-Körnern und anderer solarer und präsolarer Materie im Größenbereich bis herab zu 100 – 200 nm Durchmesser (z.B. als Grundlage für eine spätere massenspektrometrische Isotopenanalyse)darstellen.

372 osteochondrodysplasias and genetically determined dysostoses were reported in 2007 [Superti-Furga and Unger, 2007]. For 215 of these conditions, an association with one or more genes can be stated, while the molecular changes for the remaining syndromes remain illusive to date. Thus, the present dissertation aims at the identification of novel genes involved in processes regarding cartilage/ bone formation, growth, differentiation and homeostasis, which may serve as candidate genes for the above mentioned conditions. Two different approaches were undertaken. Firstly, a high throughput EST sequencing project from a human fetal cartilage library was performed to identify novel genes in early skeletal development (20th week of gestation until 2nd year of life) that could be investigated as potential candidate genes. 5000 EST sequences were generated and analyzed representing 1573 individual transcripts, corresponding to known (1400) and to novel, yet uncharacterized genes (173). About 7% of the proteins were already described in cartilage/ bone development or homeostasis, showing that the generated library is tissue specific. The remaining profile of this library was compared to previously published libraries from different time points (8th–12th, 18th–20th week and adult human cartilage) that also showed a similar distribution, reflecting the quality of the presented library analyzed. Furthermore, three potential candidate genes (LRRC59, CRELD2, ZNF577) were further investigated and their potential involvement in skeletogenesis was discussed. Secondly, a disease-orientated approach was undertaken to identify downstream targets of LMX1B, the gene causing Nail-Patella syndrome (NPS), and to investigate similar conditions. Like NPS, Genitopatellar syndrome (GPS) is characterized by aplasia or hypoplasia of the patella and renal anomalies. Therefore, six GPS patients were enrolled in a study to investigate the molecular changes responsible for this relatively rare disease. A 3.07 Mb deletion including LMX1B and NR5A1 (SF1) was found in one female patient that showed features of both NPS and GPS and investigations revealed a 46,XY karyotype and ovotestes indicating true hermaphroditism. The microdeletion was not seen in any of the five other patients with GPS features only, but a potential regulatory element between the two genes cannot be ruled out yet. Since Lmx1b is expressed in the dorsal limb bud and in podocytes, proteomic approaches and expression profiling were performed with murine material of the limbs and the kidneys to identify its downstream targets. After 2D-gel electrophoresis with protein extracts from E13.5 fore limb buds and newborn kidneys of Lmx1b wild type and knock-out mice and mass spectrometry analysis, only two proteins, agrin and carbonic anhydrase 2, remained of interest, but further analysis of the two genes did not show a transcriptional down regulation by Lmx1b. The focus was switched to expression profiles and RNA from newborn Lmx1b wild type and knock-out kidneys was compared by microarray analysis. Potential Lmx1b targets were almost impossible to study, because of the early death of Lmx1b deficient mice, when the glomeruli, containing podocytes, are still immature. Because Lmx1b is also expressed during limb development, RNA from wild type and knock-out Lmx1b E11.5 fore limb buds was investigated by microarray, revealing four potential Lmx1b downstream targets: neuropilin 2, single-stranded DNA binding protein 2, peroxisome proliferative activated receptor, gamma, co-activator 1 alpha, and short stature homeobox 2. Whole mount in situ hybridization strengthened a potential down regulation of neuropilin 2 by Lmx1b, but further investigations including in situ hybridization and protein-protein interaction studies will be needed.
