Owing to the demand for genuine mozzarella, some 330 water buffaloes are being slaughtered every year in Switzerland albeit a stunning procedure meeting animal welfare and occupational safety requirements remains to be established. To provide a basis for improvements, we sized anatomical specifics in water buffaloes and cattle and we assessed brain lesions after stunning with captive bolts or handguns by diagnostic imaging. In water buffaloes and cattle, the median distance from the frontal skin surface to the inner bone table was 74.0 mm (56.0–100.0 mm) vs 36.6 mm (29.3–44.3 mm) and from skin to the thalamus 144.8 mm (117.1–172.0 mm) vs 102.0 (101.0–121.0 mm), respectively. Consequently, customary captive bolt stunners may be inadequate. Free bullets are potentially suitable for stunning buffaloes but involve occupational safety hazards. The results of the present study shall be used to develop a device allowing effective and safe stunning of water buffaloes.

The prevalence of and possible risk factors for tuberculosis were studied in water buffalo from Para, Brazil. In this study, 3,917 pregnant and nonpregnant female Murrah and Mediterranean buffaloes were studied; 2,089 originated from Marajo Island, and 1,108 were from the mainland. The comparative cervical tuberculin test was used as a diagnostic test for tuberculosis in these animals. The prevalence of positive buffaloes was 3.5 % (100/2,809) on Marajo Island and 7.2% (80/1,108) on the mainland. The municipalities with the highest tuberculosis prevalence rates in animals were Ipixuna do Para (10.1 %), Marapanim (9.8 %), Chaves (9.4 %), Paragominas (8.6 %), and Cachoeira do Arari (6.7 %). The tuberculosis prevalence was not significantly different between the Murrah (4.3 %) and Mediterranean (4.8 %) breeds or between pregnant (5%) and nonpregnant (4.3%) buffaloes. Tuberculosis was detected in water buffaloes from Para, Brazil; the mainland buffalo exhibited the highest tuberculosis prevalence. These results indicate that this disease is dangerous to public health and buffalo farming in Para.

Water buffaloes (Bubalus bubalis) are quite well adapted to climatic conditions in the Amazon, and in this biome, they are noted for the considerable amount of meat and milk they produce and how hard they are able to work. Because of a lack of research dedicated to improving the rearing of buffaloes in the Amazon, the objective of this study was to genetically characterize the Murrah and Mediterranean breeds, as well as a mixed-breed population, based on polymorphisms in the diacylglycerol O-acyltransferase 1 gene (DGAT1), and associate the genotypes with milk production. By using the polymerase chain reaction-single-strand conformation polymorphism technique, the alleles A (0.79), B (0.20), and D (0.01) were found in the Murrah breed. In the Mediterranean and mixed-breed buffaloes, we found alleles A (0.69) and (0.77) and B (0.31) and (0.23), respectively. The Murrah breed had the genotypes AA (0.63), AB (0.29), BB (0.05), and AD (0.03), and the Mediterranean and mixed-breed buffaloes had the genotypes AA (0.44) and (0.61), AB (0.50) and (0.31), and BB (0.06) and (0.08), respectively. For the Murrah, Mediterranean, and mixedbreed buffaloes, respectively, the expected heterozygosity values were 0.34, 0.43, and 0.35, the inbreeding coefficients were 0.78, -0.15, and 0.17, and the Hardy-Weinberg probabilities were 0.70, 0.67, and 0.52. The genotypes evaluated did not have an effect on milk production; however, the single nucleotide polymorphisms can be used in studies on genetic variability.

During 12 years feces samples from cows, water buffaloes, sheeps and goats were examined by sequencial tamis filtration to show the occurence of Fasciola hepatica eggs. The material came from 129 municipalities of Santa Catarina State, and 5 g of feces per animal were examined. The occurrence of F. hepatica was confirmed in 64.82% of the municipalities. Considering the host, F. hepatica was confirmed in goats from Florianópolis, São José, São João Batista and Guaramirim municipalities; in sheeps from Brusque, Pomerode, Palhoça and São José; in water buffaloes from 9 and in cows from 86 municipalities. For this study, 13,762 feces samples were examined and in 3,814 the presence of eggs of F. hepatica was demonstrated. The percentage of occurence for host species was 27.86 in cows, 24.72 in water buffaloes, 16.92 in sheeps and 15.66 in goats. By the results it was demonstrated that Itajaí Valley at Southest Hidrographic Basin, in Santa Catarina State is an endemic area of F. hepatica, even though Uruguai Hidrographic Basin was not referred as a geographical record for this parasite.

Samples of ten penises of Mediterranean buffaloes and ten penises of Red Sindhi cattle were used. The thickness of the tunica albuginea (TA), distribution of smooth muscle cells (SMC) and volume density (Vv) of elastic system fibers in TA, corpus cavernosum (CC) and corpus spongiosum (CS) were evaluated. The Vv of elastic system fibers in buffalo and bovine penis was respectively 4.07% ±0.88% and 3.36% ±1.21% in TA; 17.32% ±2.21% and 13.14% ±1.27% (CC), 26.58% ±4.31% and 31.36% ±3.67% (CS). The CC of buffalo presented higher Vv of elastic fibers than bovine, while in the CS the Vv of elastic fibers in buffaloes was smaller than in cattle. The TA thickness showed a significant difference among the species studied. The arrangement of SMC in the bovine penises and in the water buffalo suggests that this pattern is common to animals that have fibroelastic penises.

Os objetivos neste trabalho foram estimar as correlações entre medidas obtidas por ultrassonografia e na carcaça e acompanhar o desenvolvimento da área de olho-de-lombo e a deposição de gordura de búfalos em diferentes períodos de confinamento. Foram utilizados 20 bubalinos da raça Murrah, castrados, abatidos aos 75, 100, 125 ou 150 dias de confinamento. As medidas realizadas foram área de olho-de-lombo, espessura de gordura subcutânea do dorso e espessura de gordura da garupa, mensuradas por ultrassonografia quando os búfalos atingiram o tempo de abate e na carcaça, após o abate. A espessura de gordura subcutânea da carcaça foi avaliada utilizando-se paquímetro e a área de olho-de-lombo da carcaça, régua do quadrante de pontos. O período de confinamento apresentou efeito linear crescente na área de olho-de-lombo e nas espessuras de gordura do dorso e da garupa avaliadas por ultrassom e na carcaça. O efeito linear positivo entre 75 e 150 dias de confinamento, tanto para a área de olho-de-lombo como para espessura de gordura medidas no dorso e na garupa, comprovou que os animais ainda não haviam atingido a maturidade. As correlações entre as medidas obtidas por ultrassom e na carcaça foram 0,82 para área de olho-de-lombo e 0,85 para espessura de gordura subcutânea no dorso. As características de carcaça medidas por ultrassom em bubalinos apresentam boas correlações com as medidas realizadas na carcaça no momento do abate, o que permite, em bubalinos da raça Murrah castrados e oriundos de rebanho leiteiro, acompanhar as alterações dessas características durante o desenvolvimento.

Toxocara vitulorum, a nematode parasite in the small intestine of cattle and water buffaloes, causes high morbidity and mortality of 1-3 months old buffalo calves. This research evaluated the specific perieneteric antigens (Pe) reactivity of anti-T. vitulorum-Pe antibody (Tv-Pe-Ab) in both immune sera and colostrum from buffalo cows immediately post-partum from buffalo cows. The presence of Tv-Pe-Ab in sera of buffalo newborn calves was also examined at 1 day before and after suckling the colostrum as well as in sera from naturally infected calves at the beginning and peak of the maximum infection and then again during the period of rejection and post-rejection of the parasite. Pe antigens were characterized for Tv-Pe-Ab by SDS-PAGE and Western blot (WB). The SDS-PAGE showed that Pe contained nine protein bands (11, 14, 31, 38, 58, 76, 88,112 and 165 kDa). All Pe bands were recognized by Tv-Pe-Ab in sera and colostrum of buffalo cows. Only the serum antibodies of buffalo calves at 1 day of age after suckling the colostrum and during the beginning of T. vitulorum infection recognized Pe antigen's nine bands. In contrast, serum antibodies from 1-day-old buffalo calves, taken before suckling colostrum, did not react with any protein band. In suckling calves, which reached peak egg output, rejection and post-rejection stages of the infection, serum Tv-Pe-Ab reactivity with lower molecular weight protein bands (11-76 kDa) was lost and only reactivity with the Pe protein bands of higher molecular weight (88, 112 and 165 kDa) remained. O objetivo deste trabalho foi avaliar a estrutura populacional e os progressos genéticos, fenotípicos e ambientais de características de desenvolvimento ponderal, em bubalinos da raça Mediterrâneo. Foram utilizadas informações dos pesos ajustados aos 205, 365 e 550 dias de idade, de 6.243 bubalinos nascidos no período de 1974 a 2003, provenientes de quatro fazendas. As estimativas dos coeficientes de herdabilidade foram altas para os pesos ajustados nas três idades. Os rebanhos apresentaram ganho genético positivo quanto a essas três características, porém, aquém do possível, provavelmente em razão da seleção baseada somente no fenótipo, realizada pelos produtores. Para aumento nos ganhos genéticos, são necessários a redução do intervalo de geração, o aumento do tamanho efetivo, o uso de reprodutores avaliados com base nos valores genéticos preditos pelo BLUP e o controle dos acasalamentos de animais aparentados.

Para este estudo, quinze papilas de fêmeas de búfalos e uma de fêmea bovina foram obtidas no matadouro. Estudos macro e microscópico foram realizados por meio de diferentes técnicas: morfológica, histológica e análises, biométrica, histométrica e mesoscópica. As médias foram: papila cranial direita (PCD) 20,95g; papila caudal direita (PCaD) 21,49g; papila cranial esquerda (PCE) 23,52g; papila caudal esquerda (PCaE) 21,35g. O comprimento médio do ducto papilar, do número de pregas e do comprimento das cisternas papilares foi respectivamente: 0,67/5,5 e 2,17cm para a PCD; 0,62/6,0 e 2,26cm para a PCaD; e para o antímero esquerdo, PCD: 0,71/6,0 e 2,26cm e para PCaD: 0,74/5,7 e 2,57cm. A análise de Pearson revelou uma correlação negativa entre o peso do corpo e o peso da glândula mamária. Foi feita a amostragem das três áreas proximal, média e distal e estas foram fixadas em formaldeído a 10%, seguindo-se a investigação mesoscópica em amostras de espessuras de 10, 20 e 40µm coradas pelas técnicas Azan, F3BA Picrosirus e Weigert - Van Gieson. As análises histológicas e morfométricas foram feitas em amostras de 5µm de espessura o que mostrou que a camada muscular estava disposta em dois arranjos, interna circular e externa longitudinal com epitélio cubóide bi-estratificado. A camada muscular bem desenvolvida estava relacionada com a porção média revelando uma espessura de 22,90mm com uma grande quantidade de tecido conectivo (7,10mm).

The aim was to study the seroprevalence of Toxoplasma gondii in water buffaloes (Bubalus bubalis) from State of Pará, Brazil. Three hundred and nineteen buffaloes were randomly selected into seven municipalities of Marajó Island. For comparative purposes, 128 buffaloes of five municipalities in the state of Pará were also evaluated. The seroprevalence of T. gondii was evaluated by Indirect Enzyme Linked Immunosorbent Assay (iELISA). The samples diagnosed as positive in iELISA were subjected to Immunofluorescence Antibody Test (IFAT). We evaluated risk factors: location, breed, pregnancy and co-infection with Brucella abortus or Mycobacterium bovis. The frequency of animals positive for T. gondii in iELISA were compared by chi-square (x2) with 95% confidence. Variables with p <0.2 were subjected to logistic regression analysis; the model was built based on the odds ratios test. The prevalence of T. gondii in iELISA was 41,6% (186/447). In IFAT, 86,5% (161/186) had their positivity for T. gondii confirmed. The average prevalence in the municipalities of the Marajó Island and of the mainland was 32% (103/319) and 55% (70/128), respectively. The municipalities with the highest prevalence were Soure (53%) and Salvaterra (49%) in Marajó Island, and Castanhal (55%) and Thailândia (50%) in the Continent. The breed and co-infection with Brucella abortus or Mycobacterium bovis presented no influence on the prevalence of T. gondii. Additionally, pregnant animals were 57% more positive for T. gondii than nonpregnant animals. The presence of antibodies is an indicative of T. gondii in buffaloes in the state of Pará, and these findings represent a risk not only for farm animals, but to public health as a source of infection.

The aim of the study was to estimate the prevalence of Babesia bovis and Babesia bigemina in water buffaloes of the Marajó Island, State of Pará, Brazil. We used an indirectenzyme- linked immunosorbent assay (iELISA), with total antigen containing proteins outer surface, and polymerase chain reaction (qPCR), involving the use of SYBR Green based on amplification of a small fragment of the cytochrome b gene. The prevalence of positive animals in iELISA to B. bovis B. bigemina and mixed infection was 24.87% (199/800), 20.75% (166/800) and 18.75% (150/800), respectively. Using the PCR, the presence of B. bovis wasdetected in 15% (18/199) and B. bigemina in 16% (19/199) of animals, and of these, 58% (11/19) presented co-infected by the two agents. The results show a low prevalence of antibodies anti-B. bovis and anti-B. bigemina in water buffaloes from Marajó Island. However, it was observed that the agents of bovine babesiosis circulate in buffaloes, and these may act as reservoirs.
