Objetivou-se comparar as perdas, a dinâmica microbiológica e as alterações fermentativas e nutritivas de silagens de cana-de-açúcar in natura ou queimada e tratadas ou não com Lactobacillus buchneri durante o período de armazenamento. Os tratamentos foram cana-de-açúcar in natura ou tratada com L. buchneri e cana-de-açúcar queimada ou tratada com L. buchneri, avaliadas com 0, 1, 4, 7, 14, 28 e 56 dias de fermentação, em três repetições por tratamento. Apenas antes da ensilagem foi constatada diferença significativa na população de leveduras entre cana-de-açúcar in natura e queimada, cujos valores foram de 4,85 e 5,43 log ufc/g de forragem, respectivamente. No decorrer do tempo de fermentação, ocorreu redução da recuperação de matéria seca e as perdas se prolongaram até o dia 56. A concentração de etanol foi alta até o dia 28 e estável até o dia 56, sem diferença entre tratamentos. A digestibilidade verdadeira in vitro da matéria seca sofreu severa redução já no primeiro dia de armazenamento, chegando a reduzir cerca de 20 unidades percentuais aos 56 dias de armazenamento. Silagens de cana-de-açúcar queimada apresentam maiores perdas e alterações fermentativas e nutricionais que silagens de cana-de-açúcar in natura. O L. buchneri deve ser utilizado em silagens de cana-de-açúcar queimada, mas nas de cana-de-açúcar in natura seu uso é dispensável.

Vegetated riparian buffer strips have been established in Southern Quebec (Canada) in order to intercept nutrients such as nitrate (NO(3)(-)) and protect water quality near agricultural fields. Buffer strips may also favour denitrification through a combination of high soil moisture, NO(3)(-) and carbon supply, which could lead to the production of nitrous oxide (N(2)O), a greenhouse gas. Denitrification could be further amplified by the presence of earthworms, or by plant species that promote earthworm and bacterial activity in soils. Soils from four farms, comprising maize fields and adjacent buffer strips, were sampled in the fall of 2008. A total of six earthworm species were found, but average earthworm biomass did not differ between buffer strips and maize agroecoecosystems. Nitrate concentrations and net nitrification rates were higher in the maize fields than in the buffer strips: there was no difference in N(2)O production in soils collected from the two sampling locations. Potential denitrification, measured by acetylene inhibition, varied by two orders of magnitude, depending on experimental conditions: when amended with H(2)O or with H(2)O + NO3-, potential denitrification was higher (P < 0.05) in soils from buffer strips than from maize fields. Potential denitrification was highest in soils amended with H(2)O+glucose, or with H(2)O+ NO(3)(-) + glucose. Using microcosms, we tested the effect of litter-soil mixtures on earthworm growth, and the effect of earthworm-litter-soil mixtures on potential denitrification. Based on four categories of chemical assays, litters of woody species (oak, apple, Rhododendron) were generally of lower nutritional quality than litter from agronomic species (alfalfa, switchgrass, corn stover). Alfalfa litter had the most positive effect, whereas apple litter had the most negative effect, on earthworm growth. Potential denitrification was 2-4 times higher in earthworm-litter-soil mixtures than in plain soil. Litter treatments that included corn stover had lower potential denitrification than those that included alfalfa or switchgrass, whereas litter treatments that included oak had lower potential denitrification than those that included apple or Rhododendron. Results suggest that potential N(2)O emissions may be higher in riparian buffer strips than in adjacent maize fields, that N(2)O emissions in buffer strips may be amplified by comminuting earthworms, and that plant litters that reduce earthworm growth may not be best at mitigating N(2)O emissions. The organic production systems based on natural processes do not allow the use of synthetic fertilizers with a high solubility, so it is required the use of technologies in accordance with the organic production legislation in order to make the technical, economical and environmental sustainable activities viable. To evaluate the effects of the usage of mineral and organic natural fertilizers, associated with liquid biofertilizers (BLE= enriched liquid biofertilizer and BF= leaf biofertilizers) on the production of the sugar apple fruit crop Annona squamosa L., replacing totally the chemical fertilizers, used by conventional farming systems, a nine year old orchard was driven in the irrigated plot n degrees 1295, irrigated project Nilo Coelho, district of Petrolina - PE. The experiment was carried out from September 2005 to February 2006. The experimental design was done through randomized blocks with eight treatments and four replications in plots of three plants each, with 96 plants in total. The treatments were the following: T(1) - plants with no fertilizer; T(2) - 60 g de N + 32 g de P(2)O(5) + 48 g de K(2)O; T(3) - 90g de N + 32 g de P(2)O(5) + 48 g de K(2)O; T(4) - 30 g de N + 32 g de P(2)O(5) + 48 g de K(2)O + 15 L de BLE + BF a 5 %; T(5) - 60g de N + 32 g de P(2)O(5) + 48 g de K(2)O + 30 L de BLE + BF a 5 %; T(6)- 90g de N + 32 g de P(2)O(5) + 48 g de K(2)O + 45 L de BLE + BF a 5 %; T(7) - 120g de N + 32 g de P(2)O(5) + 48 g de K(2)O + 60 L de BLE + BF a 5 %; T(8) - 150 g de N + 32 g de P(2)O(5) + 48 g de K(2)O + 75 L de BLE + BF a 5 % The fruit growth analyses (diameter and average length), total yield, productivity, average fruit weight, average number of fruits, fresh weight of the skin and pulp and dry weight of the fruits were used to evaluate the experiment. The results were relevant to the total production, fresh weight of the skin and dry weight of the fruits. The use of organominerals associated with liquid fertilizers increased significantly the productivity in values ranging from 10.44% (T(5) = 1.98 t ha(-1)) to 24.52 % (T(7) = 12.34 %). The treatment T(3) stood out (90g de N + 32g de P(2)O(5) + 48g de K(2)O) with a higher production than the control. The fruit weight increased in the fertilized treatments, with values increasing from treatment (T(2) = 5.26 %) to the treatment (T(7) = 12.34 %) in relation to the control. The fruit development was characterized by an average growth increase of 82 % in length and 84 % in diameter until the 56(th) day, with a sigmoidal growth pattern.

The objectives of this study were to determine (a) the influence of fiber on the sensory characteristics of dry dog foods; (b) differences of coated and uncoated kibbles for aroma and flavor characteristics; (c) palatability of these dry dog foods; and (d) potential associations between palatability and sensory attributes. A total of eight fiber treatments were manufactured: a control (no fiber addition), guava fiber (3%, 6%, and 12%), sugar cane fiber (9%; large and small particle size), and wheat bran fiber (32%; large and small particle size). The results indicated significant effects of fibers on both flavor and texture properties of the samples. Bitter taste and iron and stale aftertaste were examples of flavor attributes that differed with treatment, with highest intensity observed for 12% guava fiber and small particle size sugar cane fiber treatments. Fracturability and initial crispness attributes were lowest for the sugar cane fiber treatments. Flavor of all treatments changed after coating with a palatant, increasing in toasted, brothy, and grainy attributes. The coating also had a masking effect on aroma attributes such as stale, flavor attributes such as iron and bitter taste, and appearance attributes such as porosity. Palatability testing results indicated that the control treatment was preferred over the sugar cane or the wheat bran treatment. The treatment with large sugarcane fiber particles was preferred over the treatment with small particles, while both of the wheat bran treatments were eaten at a similar level. Descriptive sensory analysis data, especially textural attributes, were useful in pinpointing the underlying characteristics and were considered to be reasons that may influence palatability of dog foods manufactured with inclusion of different fibers.

Self-assembly relies on the association of pre-programmed building blocks through non-covalent interactions to give complex supramolecular architectures. Previous studies provided evidence for the unique self-assembly properties of semi-synthetic lipophilic guanosine derivatives which can sequestrate ions from an aqueous phase, carry them into an organic phase where they promote the generation of well-defined supramolecular assemblies. In the presence of cations lipophilic guanosines form columnar aggregates while in their absence they generate supramolecular ribbons. The aim of this thesis has been the synthesis of guanine derivatives, in particular N9-alkylated guanines and a guanosine functionalized as a perchlorotriphenylmetil moiety (Gace-a-HPTM) in order to observe their supramolecular behaviour in the absence of sugar (ribose or deoxyribose) and in the presence of a bulky and chiral substituent respectively. By using guanine instead of guanosine, while maintaining all the hydrogen bond acceptor and donor groups required for supramolecular aggregation, the steric hindrance to supramolecular aggregation is notably reduced because (i.e. guanines with groups in N9 different from sugar are expected to have a greatest conformational freedom even in presence of bulky groups in C8). Supramolecular self-assembly of these derivatives has been accomplished in solutions by NMR and CD spectroscopy and on surface by STM technique. In analogy with other guanosine derivatives, also N9-substituted guanines and GAceHPTM form either ribbon-like aggregates or cation-templated G-quartet based columnar structures.

BACKGROUND Lactococcus garvieae is a bacterial pathogen that affects different animal species in addition to humans. Despite the widespread distribution and emerging clinical significance of L. garvieae in both veterinary and human medicine, there is almost a complete lack of knowledge about the genetic content of this microorganism. In the present study, the genomic content of L. garvieae CECT 4531 was analysed using bioinformatics tools and microarray-based comparative genomic hybridization (CGH) experiments. Lactococcus lactis subsp. lactis IL1403 and Streptococcus pneumoniae TIGR4 were used as reference microorganisms. RESULTS The combination and integration of in silico analyses and in vitro CGH experiments, performed in comparison with the reference microorganisms, allowed establishment of an inter-species hybridization framework with a detection threshold based on a sequence similarity of >or= 70%. With this threshold value, 267 genes were identified as having an analogue in L. garvieae, most of which (n = 258) have been documented for the first time in this pathogen. Most of the genes are related to ribosomal, sugar metabolism or energy conversion systems. Some of the identified genes, such as als and mycA, could be involved in the pathogenesis of L. garvieae infections. CONCLUSIONS In this study, we identified 267 genes that were potentially present in L. garvieae CECT 4531. Some of the identified genes could be involved in the pathogenesis of L. garvieae infections. These results provide the first insight into the genome content of L. garvieae.

Résumé : Contexte: Les maladies cardiovasculaires (MCV) sont un enjeu contemporain de santé publique. Or, des recherches cliniques démontrent que certaines interventions sont efficaces dans leur traitement et prévention. Il s’agit d’interventions nutritionnelles éducatives priorisant des aliments végétaux minimalement transformés (VMT). Ces interventions promeuvent l’adoption de postures alimentaires se caractérisant par la consommation à volonté d’une grande variété d’aliments d’origine végétale (e.g. légumineuses, céréales entières, fruits, légumes) et par une diminution de la consommation d’aliments d’origine animale (e.g. viandes, œufs et produits laitiers) et ultra-transformés (e.g. riches en sucres, sel ou gras, et faibles en fibres). Objectifs: À l’aide d’un devis mixte concomitant imbriqué, nous avons évalué les effets d’un programme d’interventions éducatives visant à augmenter la consommation de VMT chez des adultes à risque de MCV et exploré les déterminants des modifications comportementales observées. Méthodologie : Divers paramètres physiologiques et anthropométriques ont été mesurés pré-post programme (n = 72) puis analysés avec un test t pour échantillons appariés ou un test signé des rangs de Wilcoxon. D’autre part, 10 entretiens semi-dirigés ont été réalisés post-programme et soutenus par un guide d’entretien basé sur le Food Choice Process Model. Les verbatims intégraux ont été codés selon la méthode d’analyse thématique. Résultats : Après 12 semaines, le poids (-10,5 lb, IC 95 %: 9,0-12,0), le tour de taille (-7,4 cm, IC 95 %:6,5-8,4), la tension artérielle diastolique (-3,2 mmHg, IC 95 %: 0,1-6,3), le cholestérol total (-0,87 mmol/ L, IC 95 %:0,57-1,17), le cholestérol LDL (-0,84 mmol/ L, IC 95 %: 0,55-1,13) et l’hémoglobine glyquée (-1,32 %, IC 95 %:-0,17-2,80) se sont significativement améliorés. L’analyse thématique des données qualitatives révèle que le programme, par la stimulation de valeurs de santé, d’éthique et d’intégrité, favorise la transformation des choix alimentaires vers une posture davantage axée sur les VMT durant une période clé du parcours de vie (i.e. pré-retraite). D’autres déterminants pouvant favoriser l’adoption d’une alimentation VMT ont été identifiés, dont les bénéfices importants observables à court terme, l’absence de restriction à l’égard de la quantité d’aliments VMT et le développement de compétences de planification dans l’acquisition et la préparation des aliments. Conclusion : Une intervention priorisant les VMT permet d’améliorer le profil cardiométabolique d’individus pré-retraités en raison de ses caractéristiques intrinsèques, mais aussi parce qu’elle modifie les valeurs impliquées dans les choix alimentaires.
