This study evaluated whether pentoxifylline (PTX) present in the flushing extender influenced the function of equine epididymal spermatozoa after recovery and after thawing. For this experiment, 58 testicles from 29 Brazilian Jumping Horses were used. Cauda epididymides of each stallion were separated and flushed with a skim milk extender, with or without 7.18 mM PTX and then subjected to the freezing process. Samples flushed with the extender containing PTX showed a significant increase in total motility, progressive motility, straight line velocity, curvilinear velocity, and percentage of rapid sperm immediately after the recovery of epididymal sperm and after 15 minutes of incubation at 37°C (P < .05). However, the presence of PTX in the flushing extender did not affect the post-thaw motility parameters or plasma membrane integrity (P > .05). The results of this study showed that the PTX present in the flushing extender improved motility parameters of recently recovered epididymal sperm and had no deleterious effects on plasma membrane integrity and freezability of equine epididymal sperm. © 2013 Elsevier Inc. Summary: The objective of this work was to evaluate the sperm motility of 13 Steindachneridion parahybae males using open-source software (ImageJ/CASA plugin). The sperm activation procedure and image capture were initiated after semen collection. Four experimental phases were defined from the videos captured of each male as follows: (i) standardization of a dialogue box generated by the CASA plugin within ImageJ; (ii) frame numbers used to perform the analysis; (iii) post-activation motility between 10 and 20 s with analysis at each 1 s; and (iv) post-activation motility between 10 and 50 s with analysis at each 10 s. The settings used in the CASA dialogue box were satisfactory, and the results were consistent. These analyses should be performed using 50 frames immediately after sperm activation because spermatozoa quickly lose their vigor. At 10 s post-activation, 89.1% motile sperm was observed with 107.2 μm s-1 curvilinear velocity, 83.6 μm s-1 average path velocity, 77.1 μm s-1 straight line velocity; 91.6% were of straightness and 77.1% of wobble. The CASA plugin within ImageJ can be applied in sperm analysis of the study species by using the established settings. © 2013 Blackwell Verlag GmbH.

Valuable genetic material can be preserved by the cryopreservation of epididymal sperm. This study evaluated the viability of pre-freezing and post-thawed sperm samples recovered from the epididymal cauda of buffaloes. Epididymides from eight Murrah buffaloes with 18 months of age were used. Semen samples were diluted in two different freezing extenders: Botu-bov (BB) and Tris (TRIS). Immediately after slaughter, both testicles from each animal were collected and transported at 4 degrees C for six hours interval. In laboratory, the removed epididymides were flushed to obtain sperm and the fractions were diluted in both freezing extenders (BB and TRIS). Semen doses were analyzed before and after frozen at -196 degrees C. BB and TRIS pre-freezing results were 38.54 +/- 22.33%(b) and 14.17 +/- 12.78%(a) for total motility (TM), 25.00 +/- 16.12(a) and 9.44 +/- 9.11(a) for progressive motility (PM), 7.21 +/- 0.98(a) and 5.09 +/- 2.65(a) for percentage of rapid cells (RAP), 91.08 +/- 12.53(b) and 63.33 +/- 31.47(a) for velocity of trajectory (VAP), 73.54 +/- 20.17(b) and 49.50 +/- 9.11(a) for linear progressive velocity (VSL), 172.21 +/- 24.55(a) and 116.94 +/- 59.48(a) for curvilinear velocity (VCL), respectively (P < 0.05). BB and TRIS post-thawing results were 42.25 +/- 21.50(b) and 17.62 +/- 19.46(a) for TM, 27.25 +/- 24.86(a) and 18.00 +/- 13.68(a) for PM, 7.35 +/- 0.98(a) and 6.26 +/- 1.13(a) for RAP, 91.42 +/- 16.86(a) and 75.96 +/- 13.17(a) for VAP, 67.96 +/- 12.13(a) and 60.04 +/- 10.42(a) for VSL, 177.54 +/- 23.53(b) and 141.29 +/- 24.97a for VCL, respectively (P < 0.05). The sperm recovered from the epididymal cauda, after 6 h storage of epididymides at 5 degrees C ensures sperm preservation demonstrating that the diluent Botu-bov had higher total motility both pre-and post-freezing when compared with TRIS. Additionally, the sperm frozen with the diluent Botu-bov showed higher values of VSL at post-thawing. These findings may reflect in improvement of conception rates.

In order to provide information that may help researchers to understand the main cause(s) of differences in bull fertility frequently observed in field trials, this study aimed to investigate conception rates as well as several in vitro sperm characteristics of different sires of unknown fertility utilized in a Timed-AI (TAI) program. Suckled Nelore cows submitted to the same TAI protocol were allocated into eight breeding groups of approximately 120 animals each. Frozen semen doses from three Angus bulls and three different batches from each bull were utilized. Approximately 100 doses from each batch were used in TAI. Sires, batches and AI technicians were equally distributed across breeding groups. Cows were examined for pregnancy diagnosis 40 d after TAI. For in vitro sperm analyses, the same thawing procedure was repeated in the laboratory to mimic field conditions. The following in vitro sperm characteristics were assessed: computerized motility, thermal resistance, plasma and acrosomal membrane integrity, lipid peroxidation, morphology, morphometry and chromatin structure. No effect of breeding group, body condition score, AI technician and sire was observed. However, some significant differences among bulls were detected in laboratory analyses. Semen from sire presenting numerically lower (P > 0.05) pregnancy/AI also presented lower (P < 0.05) values in all sperm characteristics analyzed in thermal resistance test at 4 h (Total Motility, Progressive Motility, Average Path Velocity, Straight-Line Velocity, Curvilinear Velocity, Amplitude of Lateral Head Displacement, Beat Cross Frequency, Straightness, Linearity, and Percentage of Rapidly Moving Cells), higher (P < 0.05) Major and Total Defects in sperm morphological test, lower (P < 0.05) Length, Ellipticity and Fourier parameter (Fourier 0) in sperm morphometric analysis as well as higher (P < 0.05) chromatin heterogeneity. It was concluded that, although no bull effect was observed in the field experiment, the sire that presented numerically lower pregnancy/AI also presented lower semen quality according to the laboratory analyses performed. Mounting an immune response against pathogens incurs costs to organisms by its effects on important life-history traits, such as reproductive investment and survival. As shown recently, immune activation produces large amounts of reactive species and is suggested to induce oxidative stress. Sperm are highly susceptible to oxidative stress, which can negatively impact sperm function and ultimately male fertilizing efficiency. Here we address the question as to whether mounting an immune response affects sperm quality through the damaging effects of oxidative stress. It has been demonstrated recently in birds that carotenoid-based ornaments can be reliable signals of a male's ability to protect sperm from oxidative damage. In a full-factorial design, we immune-challenged great tit males while simultaneously increasing their vitamin E availability, and assessed the effect on sperm quality and oxidative damage. We conducted this experiment in a natural population and tested the males' response to the experimental treatment in relation to their carotenoid-based breast coloration, a condition-dependent trait. Immune activation induced a steeper decline in sperm swimming velocity, thus highlighting the potential costs of an induced immune response on sperm competitive ability and fertilizing efficiency. We found sperm oxidative damage to be negatively correlated with sperm swimming velocity. However, blood resistance to a free-radical attack (a measure of somatic antioxidant capacity) as well as plasma and sperm levels of oxidative damage (lipid peroxidation) remained unaffected, thus suggesting that the observed effect did not arise through oxidative stress. Towards the end of their breeding cycle, swimming velocity of sperm of more intensely colored males was higher, which has important implications for the evolution of mate choice and multiple mating in females because females may accrue both direct and indirect benefits by mating with males having better quality sperm.

Two-stroke outboard boat engines using total loss lubrication deposit a significant proportion of their lubricant and fuel directly into the water. The purpose of this work is to document the velocity and concentration field characteristics of a submerged swirling water jet emanating from a propeller in order to provide information on its fundamental characteristics. Measurements of the velocity and concentration field were performed in a turbulent jet generated by a model boat propeller (0.02 m diameter) operating at 1500 rpm and 3000 rpm. The measurements were carried out in the Zone of Established Flow up to 50 propeller diameters downstream of the propeller. Both the mean axial velocity profile and the mean concentration profile showed self-similarity. Further, the stand deviation growth curve was linear. The effects of propeller speed and dye release location were also investigated.

Aging in humans is associated with a loss in neuromuscular function and performance. This is related, in part, to the reduction in muscular strength and power caused by a loss of skeletal muscle mass (sarcopenia) and changes in muscle architecture. Due to these changes, the force-velocity (f-v) relationship of human muscles alters with age. This change has functional implications such as slower walking speeds. Different methods to reverse these changes have been investigated, including traditional resistance training, power training and eccentric (or eccentrically-biased) resistance training. This review will summarise the changes of the f-v relationship with age, the functional implications of these changes and the various methods to reverse or at least partly ameliorate these changes.
