We previously reported nonaqueous silicone elastomer gels (SEGs) for sustained vaginal administration of the CCR5-targeted entry inhibitor maraviroc (MVC). Here, we describe chemically modified SEGs (h-SEGs) in which the hydrophobic cyclomethicone component was partially replaced with relatively hydrophilic silanol-terminated polydimethylsiloxanes (st-PDMS). MVC and emtricitabine (a nucleoside reverse transcriptase inhibitor), both currently under evaluation as topical microbicides to counter sexual transmission of human immunodeficiency virus type 1 (HIV-1), were used as model antiretroviral (ARV) drugs. Gel viscosity and in vitro ARV release were significantly influenced by st-PDMS molecular weight and concentration in the h-SEGs. Unexpectedly, gels prepared with lower molecular weight grades of st-PDMS showed higher viscosities. h-SEGs provided enhanced release over 24 h compared with aqueous hydroxyethylcellulose (HEC) gels, did not modify the pH of simulated vaginal fluid (SVF), and were shown to less cytotoxic than standard HEC vaginal gel. ARV solubility increased as st-PDMS molecular weight decreased (i.e., as percentage hydroxyl content increased), helping to explain the in vitro release trends. Dye ingression and SVF dilution studies confirmed the increased hydrophilicity of the h-SEGs. h-SEGs have potential for use in vaginal drug delivery, particularly for ARV-based HIV-1 microbicides.

Several dermocosmetic bases even without active substances, can increase the cutaneous hydration, resulting in a beneficial effect to the skin. The evidence and interpretation of possible hydration effect of formulations in the skin can be carried through by means of histopathological and histomorphometrical evaluation, a time that allows the analysis of the epithelial tissue, of dermis and also of the cellular characteristics. The objective of this research was to evaluate the skin hydration after exposition to the aqueous and hydroalcoholic bases and silicone emulsion. Swines had areas submitted to treatments during 15 days with three different formulations (F1 - aqueous gel, F2 - hydroalcoholic gel and F3 - silicone emulsion). By means of histometric and histopathological techniques were gotten the thickness of the epidermis and stratum corneum. Comparison of means was done using ANOVA followed by the Tukey test. The F1 provoked significant increase in the thickness of the epidermis. The formulaton F2 provoked significant reduction in the thickness of the epidermis and stratum corneum. F3 not presented significant difference in this structures. According to the study, the type of base chosen intervenes with the skin hydration.

The aim of this study is to demonstrate through a case report, a proposed treatment for discolored teeth, with and without pulp vitality, by the technique of external and internal tooth bleaching with hydrogen peroxide to 35% Lase Peroxide Sensy (DMC) using Whitening Lase II Device (DMC), and a silicone guide (3M ESPE) in the palatine portion of the upper teeth. In this clinical case, the patient had darkened dental elements 11 and 22, and dissatisfaction with the coloring of other elements. It was observed that the techniques used and the materials chosen allowed for an excellent aesthetic result, with technical simplicity and low cost, and minimal occurrence of signs and symptoms

Esse trabalho teve como objetivo avaliar, clinicamente, a efetividade de dois agentes clareadores aplicados sobre a superfície dentária durante 45 minutos, sem troca do gel. Foram selecionados 10 pacientes de acordo com os critérios de inclusão e exclusão, e a arcada superior dos pacientes foi dividida em duas hemiarcadas (n = 10): na hemiarcada direita (HD), foi aplicado o peróxido de hidrogênio a 35% (PH) Clàriant Office (Angelus); e, na hemiarcada esquerda (HE), o PH 38% Opalescence Boost (Ultradent). Foram realizadas duas sessões clínicas, de 45 minutos cada, com intervalo de uma semana entre as sessões. Inicialmente, ao tratamento clareador, os pacientes receberam profilaxia, moldagem para guia de mensuração de cor com silicone de condensação e avaliação de cor por meio de um aparelho espectrofotômetro Vita Easyshade (Vita Zhanfabrik, Alemanha). Após 14 dias do término do tratamento clareador, foi realizada a mensuração final da cor dos dentes. Os dados foram submetidos ao teste t e de Tukey (p < 0,05). Os resultados demonstraram não haver diferença estatisticamente significativa entre as hemiarcadas quanto à efetividade do clareamento (p = 0,146) e nem quanto à sensibilidade. Pode-se concluir que a aplicação dos materiais avaliados sem troca do gel, por 45 minutos, foi eficaz em promover o clareamento de dentes vitais e com baixa sensibilidade dentária.

A simple and scalable procedure was used to obtain thin, stable, homogeneous, and easy-to-handle films composed of silicone derived from dimethicones containing dispersed hydrotalcite-type materials previously organo-modified with amino acids. The absence of the typical X-ray pattern of the bioinorganic LDH filler suggested an exfoliation process that was further indirectly evidenced by a drastic change in the rheological behavior, which turned from a quasi-Newtonian behavior for the silicone free of LDH filler to an extensive developed gel-like structure for the nanocomposite derivatives. Visualized by the shear-thinning exponent of the complex viscosity in the low-frequency range, the percolation threshold was evident for filler loading as low as <5 w/W%, suggesting the presence of a largely developed interface between the filler and the polymer. The increase of more than one order of magnitude in viscosity was explained by the rather strong attrition phenomenon between the tethered amino acid anions and the silicone chains. UVB radiation absorption profiles make such bioinorganic polymer nanocomposites potentially applicable in skin protection. Thermo-gravimetric analysis revealed significant improvement in the thermal stability, especially in the final step of the polymer combustion, thus underlining the role of the hybrid material as a thermal retardant agent. Objectives: The antimicrobial efficacy of a chlorhexidine gluconate (CHG) intravascular catheter gel dressing was evaluated against methicillin-resistant Staphylococcus aureus (MRSA) and an extended-spectrum β-lactamase (ESBL)-producing Escherichia coli. Chlorhexidine deposition on the skin surface and release from the gel were determined. Methods: The antimicrobial efficacy was evaluated in in vitro studies following microbial inoculation of the dressing and application of the dressing on the inoculated surface of a silicone membrane and donor skin [with and without a catheter segment and/or 10% (v/v) serum] on diffusion cells. Antimicrobial activity was evaluated for up to 7 days. Chlorhexidine skin surface deposition and release were also determined. Results: MRSA and E. coli were not detectable within 5 min following direct inoculation onto the CHG gel dressing. On the silicone membrane, 3 log and 6 log inocula of MRSA were eradicated within 5 min and 1 h, respectively. Time to kill was prolonged in the presence of serum and a catheter segment. Following inoculation of donor skin with 6 log cfu of MRSA, none was detected after 24 h. Chlorhexidine was released from the gel after a lag time of 30 min and increasing amounts were detected on the donor skin surface over the 48 h test period. The CHG gel dressing retained its antimicrobial activity on the artificial skin for 7 days. Conclusions: The CHG intravascular catheter site gel dressing had detectable antimicrobial activity for up to 7 days, which should suppress bacterial growth on the skin at the catheter insertion site, thereby reducing the risk of infection. © The Author 2011.
