Background: Plant-derived condensed tannins (CT) show promise as a complementary option to treat gastrointestinal helminth infections, thus reducing reliance on synthetic anthelmintic drugs. Most studies on the anthelmintic effects of CT have been conducted on parasites of ruminant livestock. Oesophagostomum dentatum is an economically important parasite of pigs, as well as serving as a useful laboratory model of helminth parasites due to the ability to culture it in vitro for long periods through several life-cycle stages. Here, we investigated the anthelmintic effects of CT on multiple life-cycles stages of O. dentatum. Methods: Extracts and purified fractions were prepared from five plants containing CT and analysed by HPLC-MS. Anthelmintic activity was assessed at five different stages of the O. dentatum life cycle; the development of eggs to infective third-stage larvae (L3), the parasitic L3 stage, the moult from L3 to fourth-stage larvae (L4), the L4 stage and the adult stage. Results: Free-living larvae of O. dentatum were highly susceptible to all five plant extracts. In contrast, only two of the five extracts had activity against L3, as evidenced by migration inhibition assays, whilst three of the five extracts inhibited the moulting of L3 to L4. All five extracts reduced the motility of L4, and the motility of adult worms exposed to a CT-rich extract derived from hazelnut skins was strongly inhibited, with electron microscopy demonstrating direct damage to the worm cuticle and hypodermis. Purified CT fractions retained anthelmintic activity, and depletion of CT from extracts by pre-incubation in polyvinylpolypyrrolidone removed anthelmintic effects, strongly suggesting CT as the active molecules. Conclusions: These results suggest that CT may have promise as an alternative parasite control option for O. dentatum in pigs, particularly against adult stages. Moreover, our results demonstrate a varied susceptibility of different life-cycle stages of the same parasite to CT, which may offer an insight into the anthelmintic mechanisms of these commonly found plant compounds.

Key points The physiological metabolite, lactate and the two-pore domain leak potassium channel, TREK1 are known neuroprotectants against cerebral ischaemia. However, it is not known whether lactate interacts with TREK1 channel to provide neuroprotection. In this study we show that lactate increases TREK1 channel activity and hyperpolarizes CA1 stratum radiatum astrocytes in hippocampal slices. Lactate increases open probability and decreases longer close time of the human (h)TREK1 channel in a concentration dependent manner. Lactate interacts with histidine 328 (H328) in the carboxy terminal domain of hTREK1 channel to decrease its dwell time in the longer closed state. This interaction was dependent on the charge on H328. Lactate-insensitive mutant H328A hTREK1 showed pH sensitivity similar to wild-type hTREK1, indicating that the effect of lactate on hTREK1 is independent of pH change. AbstractA rise in lactate concentration and the leak potassium channel TREK1 have been independently associated with cerebral ischaemia. Recent literature suggests lactate to be neuroprotective and TREK1 knockout mice show an increased sensitivity to brain and spinal cord ischaemia; however, the connecting link between the two is missing. Therefore we hypothesized that lactate might interact with TREK1 channels. In the present study, we show that lactate at ischaemic concentrations (15-30mm) at pH7.4 increases TREK1 current in CA1 stratum radiatum astrocytes and causes membrane hyperpolarization. We confirm the intracellular action of lactate on TREK1 in hippocampal slices using monocarboxylate transporter blockers and at single channel level in cell-free inside-out membrane patches. The intracellular effect of lactate on TREK1 is specific since other monocarboxylates such as pyruvate and acetate at pH7.4 failed to increase TREK1 current. Deletion and point mutation experiments suggest that lactate decreases the longer close dwell time incrementally with increase in lactate concentration by interacting with the histidine residue at position 328 (H328) in the carboxy terminal domain of the TREK1 channel. The interaction of lactate with H328 is dependent on the charge on the histidine residue since isosteric mutation of H328 to glutamine did not show an increase in TREK1 channel activity with lactate. This is the first demonstration of a direct effect of lactate on ion channel activity. The action of lactate on the TREK1 channel signifies a separate neuroprotective mechanism in ischaemia since it was found to be independent of the effect of acidic pH on channel activity. Con el objetivo de conocer los principales helmintos gastrointestinales que afectan a bovinos menores de un año en la zona del municipio de Matagalpa, se realizó este estudio en diez (10) fincas representativas de la zona que se encuentran en distintos puntos geográficos del municipio. Investigación que se llevó a efecto entre noviembre del 2005 - abril del 2006. Los parámetros climáticos del municipio en esta época oscila entre 26 - 28 °C de temperatura y 80 % de humedad relativa, ± un 5 %. Se realizaron cuatro muestreos en serie a 91 terneros menores de un año, correspondiente al 20 % del total de una población de 455 animales, la que se les extrajo del recto aproximadamente de 80 a l 00 gramos de heces fecales, de los cuales se utilizó 5 gramos para realizar el método cualitativo de presencia de huevos de helmintos por medio del método de flotación de Sheather. Posterior al análisis cualitativo, se procedió a realizar el cálculo de la carga parasitaria, utilizando para esto la metodología de Gordon & Whitllock (Macmaster modificado). Para la identificación de especies, se realizaron cultivos de larva de las que se obtuvieron muy pocas formas larvarias, se observó baja eclosión de larvas, posiblemente a la no viabilidad de los huevos. Se encontraron 82 animales positivos, de los cuales los géneros Strongyloides s y Cooperia presentaron 33 % y 16.5 % respectivamente siendo los más predominantes, seguido por Trichuris 6.6 %, Toxocara; 2.2 %, Oesophagostomum; 1.1%, Bunostomum; l.l %. Además se presentaron asociaciones de helmintos con una prevalencia de 29.6 % de los casos positivos.

El objeto de esta investigación fue el de determinar la prevalencia e incidencia de Nemathelmintos parásitos en la heces de bovinos jóvenes del Departamento de Masaya. Se inicio en el mes de Agosto de 1973 y termino noviembre del mismo año. Para realizar esta investigación se utilizo el diseño de muestreo bietapico. Se seleccionaron al azar 10 fincas y 2 animales por fincas en cada uno de los nueve municipios del Departamento, sin considerar el sexo. Las muestras se analizaron de acuerdo a la técnica de dilucion de Stoll: colentadose por medio de masajes rectales para identificar y cuantificar a los parásitos en el laboratorio se utilizo la parte media de las deposiciones. Se realizo un análisis de varianza y una prueba de diferencia mínima significativa para cada uno de los géneros encontrados. La identificación de los parásitos se hizo con un microscopio de luz y por comparación con microfotografia (3). El orden de prevalencia de todos los generos de Nemathelmintos en el Departamento fue el siguiente: Haemonchus sp., Trichostrongylus sp., Toxocara sp., Oesophagostomum sp., Strongyloides sp., Buxtonella sp., Neoascaris sp., Arcaris sp., Trichuris sp. El orden de los municipios, relacionados de mayor a menor infestacion para todos los nematodos fue el siguiente: Catarina, Masaya, Masatepe, Niquinohomo, Tisma, La Concepcion, Nandasmo, San Juan de Oriente, Nindiri. La mas alta infestacion detectada fue de 26.220 huevos y correspondio al genero Haemonchus sp., en el municipio de Masaya y la menos fue de 230 huevos correspondiendo al genero Trichuris sp., en el municipio de Nandasmo. El 63% de infestaciones se calificaron como moderadas e intensas siendo de 43% y 20% respectivamente. La infestacion mas frecuente para cada genero encontrado fue la Moderada. El 37% de las infestaciones restantes, se distribuyo entre infestaciones Ligeras, muy ligeras, muy intensas e inexistentes. En algunos municipios no se encontraron los géneros Trichuris sp., Neoascaris sp., Buxtonella sp., y AScaris sp., que fueron los menos prevalecientes. (Ver cuadro 1).

The temporoammonic (TA) pathway is the direct, monosynaptic projection from layer III of entorhinal cortex to the distal dendritic region of area CA1 of the hippo­ campus. Although this pathway has been implicated in various functions, such as memory encoding and retrieval, spatial navigation, generation of oscillatory activity, and control of hippocampal excitability, the details of its physiology are not well understood. In this thesis, I examine the contribution of the TA pathway to hippocampal processing. I find that, as has been previously reported, the TA pathway includes both excitatory, glutamatergic components and inhibitory, GABAergic components. Several new discoveries are reported in this thesis. I show that the TA pathway is subject to forms of short-term activity-dependent regulation, including paired-pulse and frequency­ dependent plasticity, similar to other hippocampal pathways such as the Schaffer collateral (SC) input from CA3 to CA1. The TA pathway provides a strongly excitatory input to stratum radiatum giant cells of CA1. The excitatory component of the TA pathway undergoes a long-lasting decrease in synaptic strength following low-frequency stimulation in a manner partially dependent on the activation of NMDA receptors. High­ frequency activation of the TA pathway recruits a feedforward inhibition that can prevent CA1 pyramidal cells from spiking in response to SC input; this spike-blocking effect shows that the TA pathway can act to regulate information flow through the hippocampal trisynaptic pathway.

While RNA interference (RNAi) has been deployed to facilitate gene function studies in diverse helminths, parasitic nematodes appear variably susceptible. To test if this is due to inter-species differences in RNAi effector complements, we performed a primary sequence similarity survey for orthologs of 77 Caenorhabditis elegans RNAi pathway proteins in 13 nematode species for which genomic or transcriptomic datasets were available, with all outputs subjected to domain-structure verification. Our dataset spanned transcriptomes of Ancylostoma caninum and Oesophagostomum dentatum, and genomes of Trichinella spiralis, Ascaris suum, Brugia malayi, Haemonchus contortus, Meloidogyne hapla, Meloidogyne incognita and Pristionchus pacificus, as well as the Caenorhabditis species C. brenneri, C. briggsae, C. japonica and C. remanei, and revealed that: (i) Most of the C. elegans proteins responsible for uptake and spread of exogenously applied double stranded (ds)RNA are absent from parasitic species, including RNAi-competent plant-nematodes; (ii) The Argonautes (AGOs) responsible for gene expression regulation in C. elegans are broadly conserved, unlike those recruited during the induction of RNAi by exogenous dsRNA; (iii) Secondary Argonautes (SAGOs) are poorly conserved, and the nuclear AGO NRDE-3 was not identified in any parasite; (iv) All five Caenorhabditis spp. possess an expanded RNAi effector repertoire relative to the parasitic nematodes, consistent with the propensity for gene loss in nematode parasites; (v) In spite of the quantitative differences in RNAi effector complements across nematode species, all displayed qualitatively similar coverage of functional protein groups. In summary, we could not identify RNAi effector deficiencies that associate with reduced susceptibility in parasitic nematodes. Indeed, similarities in the RNAi effector complements of RNAi refractory and competent nematode parasites support the broad applicability of this research genetic tool in nematodes.

Clade V nematodes comprise several parasitic species that include the cyathostomins, primary helminth pathogens of horses. Next generation transcriptome datasets are available for eight parasitic clade V nematodes, although no equine parasites are included in this group. Here, we report next generation transcriptome sequencing analysis for the common cyathostomin species, Cylicostephanus goldi. A cDNA library was generated from RNA extracted from 17 C. goldi male and female adult parasites. Following sequencing using a 454 GS FLX pyrosequencer, a total of 475,215 sequencing reads were generated, which were assembled into 26,910 contigs. Using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases, 27% of the transcriptome was annotated. Further in-depth analysis was carried out by comparing the C. goldi dataset with the next generation transcriptomes and genomes of other clade V nematodes, with the Oesophagostomum dentatum transcriptome and the Haemonchus contortus genome showing the highest levels of sequence identity with the cyathostomin dataset (45%). The C. goldi transcriptome was mined for genes associated with anthelmintic mode of action and/or resistance. Sequences encoding proteins previously associated with the three major anthelmintic classes used in horses were identified, with the exception of the P-glycoprotein group. Targeted resequencing of the glutamate gated chloride channel α4 subunit (glc-3), one of the primary targets of the macrocyclic lactone anthelmintics, was performed for several cyathostomin species. We believe this study reports the first transcriptome dataset for an equine helminth parasite, providing the opportunity for in-depth analysis of these important parasites at the molecular level. Sequences encoding enzymes involved in key processes and genes associated with levamisole/pyrantel and macrocyclic lactone resistance, in particular the glutamate gated chloride channels, were identified. This novel data will inform cyathostomin biology and anthelmintic resistance studies in future.

Les patients atteints d'épilepsie du lobe temporal (TLE) ainsi que les rats injectés à l'acide kaïnique (KA) exhibent des patrons pathophysiologiques similaires de crises, de sclérose de l'hippocampe et de perte de certains types neuronaux. Parmi les cellules atteintes dans le modèle KA du TLE on retrouve certains interneurones inhibiteurs du CA1. En effet, certains interneurones des couches oriens et alveus (O/A-IN) meurent suite à une injection de KA chez le rat, contrairement aux interneurones à la bordure des couches radiatum et lacunosum/moleculare (R/LM-IN) de la même région. Bien que cette perte soit empêchée par des antagonistes des récepteurs glutamatergiques métabotropes de groupe I (mGluR1/5), la cause de cette perte sélective des O/A-INs reste à être précisée. Au cours des travaux de cette thèse, nous avons effectué des enregistrements de patch-clamp en configuration cellule-entière en modes courant- et voltage-imposé couplés à l'imagerie calcique pour étudier les causes de la vulnérabilité sélective des O/A-INs dans ce modèle. Dans un premier temps, nous avons évalué les effets d'une application aiguë de KA sur les propriétés membranaires et calciques pour voir s'il y avait des différences entre les O/A-INs et R/LM-INs qui pourraient expliquer la vulnérabilité. Nos résultats montrent que les dépolarisations et variations de résistance d'entrée ainsi que les augmentations de calcium intracellulaire, dépendantes principalement des récepteurs -amino-3-hydroxy-5-methyl-4-isoxasole propionic acid (AMPA), sont similaires entre les deux types d'interneurones suite à des applications aigües de KA. Ceci indique que l'effet aigu du KA sur les interneurones ne serait pas la cause de la vulnérabilité des O/A-INs. Dans un second temps nous avons comparé l'implication des sous-types de récepteurs mGluR1 et 5 dans l'activité épileptiforme des deux types d'interneurones évoquée dans un modèle de tranche désinhibée. Dans ce cas, nos données montrent un rôle important des mGluR1 et 5 activés synaptiquement lors des décharges épileptiformes et ce, de manière spécifique aux O/A-INs. Les courants synaptiques sous-tendant ces décharges impliquent des récepteurs ionotropes et métabotropes du glutamate. En présence d'antagonistes des récepteurs ionotropes glutamatergiques, les courants synaptiques sont biphasiques et formés de composantes rapide et lente. Les récepteurs mGluR1 et 5 sont différemment impliqués dans ces composantes: les mGluR5 étant impliqués dans les composantes rapide et lente, et les mGluR1 que dans la composante lente. Ces résultats indiquent que les mGluR1 et 5 contribuent différemment à l'activité épileptiforme, et spécifiquement dans les O/A-INs, et pourraient donc être impliqués dans la vulnérabilité sélective de ces interneurones dans le modèle KA.

Cinnamon (Cinnamomum verum) has been shown to have anti-inflammatory and antimicrobial properties, but effects on parasitic worms of the intestine have not been investigated. Here, extracts of cinnamon bark were shown to have potent in vitro anthelmintic properties against the swine nematode Ascaris suum. Analysis of the extract revealed high concentrations of proanthocyanidins (PAC) and trans-cinnamaldehyde (CA). The PAC were subjected to thiolysis and HPLC-MS analysis which demonstrated that they were exclusively procyanidins, had a mean degree of polymerization of 5.2 and 21% of their inter-flavan-3-ol links were A-type linkages. Purification of the PAC revealed that whilst they had activity against A. suum, most of the potency of the extract derived from CA. Trichuris suis and Oesophagostomum dentatum larvae were similarly susceptible to CA. To test whether CA could reduce A. suum infection in pigs in vivo, CA was administered daily in the diet or as a targeted, encapsulated dose. However, infection was not significantly reduced. It is proposed that the rapid absorption or metabolism of CA in vivo may prevent it from being present in sufficient concentrations in situ to exert efficacy. Therefore, further work should focus on whether formulation of CA can enhance its activity against internal parasites.

The aim of this study was to identify molecular pathways involved in audiogenic seizures in the epilepsy-prone Wistar Audiogenic Rat (WAR). For this, we used a suppression-subtractive hybridization (SSH) library from the hippocampus of WARs coupled to microarray comparative gene expression analysis, followed by Northern blot validation of individual genes. We discovered that the levels of the non-protein coding (npc) RNA BC1 were significantly reduced in the hippocampus of WARs submitted to repeated audiogenic seizures (audiogenic kindling) when compared to Wistar resistant rats and to both naive WARs and Wistars. By quantitative in situ hybridization, we verified lower levels of BC1 RNA in the GD-hilus and significant signal ratio reduction in the stratum radiatum and stratum pyramidale of hippocampal CA3 subfield of audiogenic kindled animals. Functional results recently obtained in a BC1-/- mouse model and our current data are supportive of a potential disruption in signaling pathways, upstream of BC1, associated with the seizure susceptibility of WARs. Descrevem-se os achados clínico, cirúrgico e anatomopatológico de intussuscepção em quatro bovinos fêmeas. Observaram-se intussuscepção do jejuno, com obstrução total, em três animais, e intussuscepção do cólon, com obstrução parcial, em um animal. A presença de parasitismo intenso por Oesophagostomum sp. em dois animais sugere a participação desse parasita na gênese da intussuscepção.

O estudo foi desenvolvido no período de outubro de 2001 a maio de 2002, com 51 javalis (Sus scrofa scrofa) proveniente de duas propriedades situadas nos municípios de Mirassol e Fernandópolis, Estado de São Paulo, sendo 18 animais variando de 150 a 360 dias (Grupo I) e 33 animais entre 30 a 120 dias de vida (Grupo II). O objetivo da investigação foi identificar a helmintofauna e obter subsídio para melhor conhecimento da instalação das infecções helmínticas. Foram identificadas nos dois grupos, respectivamente, oito espécies de nematódeos com as seguintes prevalências: Trichuris suis (16,7% e 30,3%); Metastrongylus salmi (50,0% e 15,2%), Metastrongylus pudendotectus (5,6% e 3,0%); Strongyloides ransomi (27,8% e 12,1%); Ascaris suum (0,0% e 3,0%), Ascarops strongylina (27,8% e 0,0%); Physocephalus sexalatus (5,6% e 0,0%); Oesophagostomum dentatum (22,2% e 0,0%). O total de nematódeos colhidos e identificados nos dois grupos de animais foi de 7958, assim distribuídos: 6573 no intestino grosso (82,6%), 1246 no pulmão (15,7%), 89 no intestino delgado (1,1%), e 50 no estômago (0,6%). As maiores variações de intensidade foram obtidas por T. suis de 1 a 1764 e por M. salmi 1 a 248 exemplares.
