In this work evaluate the technical characteristics of the fibers grown in settlements Guamaré, colored cotton seeds were donated existing in the Germplasm Bank of Embrapa Cotton. We sought through the breeding program, raising the resistance, fineness, length and uniformity of cotton fibers, as well as stabilize the staining of fibers in the BRS Topaz, BRS Brown and BRS Green shades and raise their productivity in the field. First, the individual selections to test progeny seeds, and thereafter the hybridization method followed by family selection to obtain variations in the color tones were performed. The BRS Topaz, BRS Brown and BRS Green varieties were produced, analyzed and compared with existing cottons in the region which is the White cotton. The properties amount of impurities and neps, length, length uniformity, short fiber content, fineness and tensile strength of the fibers were sized in Classifiber, NATI, Pressley and Micronaire devices. 10 trials each with 10 tests for all four fiber types were carried out. The White and Topaz fibers showed greater length (32-34mm) and greater resistance (7.94 lb/mg and 7.97 lb/mg respectively) and showed finesse with lower micronaire index 3,71μg/inch and 3, 73μg/inch and a low rate of short fibers. The results were very promising for the use of genetically improved cotton in the manufacturing of fabric and yarn in the textile industry. O adensamento da cultura do algodão aumenta a competição entre plantas por recursos, como luz, nutrientes e água; logo, o período de florescimento é reduzido e, consequentemente, a marcha de acúmulo de matéria seca é alterada. O objetivo deste trabalho foi determinar a marcha de absorção de N, P e K pelo algodoeiro, identificando a época de maior absorção e a quantidade absorvida e exportada em espaçamentos adensado (48 cm - 20,58 plantas m-2), intermediário (75 cm - 13,30 plantas m-2) e convencional (96 cm - 10,39 plantas m-2). Foram amostradas três plantas úteis por parcela aos 46, 69, 99, 139, 148 e 166 dias após a emergência (DAE), em que foram determinados o acúmulo de matéria seca, N, P e K. O acúmulo de matéria seca nas plantas cultivadas no espaçamento de 48 cm foi maior no início do desenvolvimento, igualando-se às demais ao final do ciclo, o que resultou em maior acúmulo de N entre os 69 e 99 DAE, no menor espaçamento. O adensamento antecipa o pico de absorção de nutrientes, sugerindo a antecipação das adubações de cobertura com N e K no algodoeiro cultivado nesse sistema. Para condições de fertilidade média/alta, a dose de nutrientes a ser aplicada não precisaria ser alterada em função do aumento da densidade de plantas, pois não há variação nas quantidades de N, P e K exportados por kg de algodão produzido, em fibra ou em caroço.

It is known that the transmission of hospital infections, whether environmental or cross infection, is facilitated by the enhanced survival of microorganisms on dry surfaces that is caused by the presence of biological fluids. To demonstrate the need for care with bodily substances in the routine of cleaning, this study evaluated the influence of some body fluids (blood, urine and artificial saliva), deposited in the same way on various surfaces and allowed to dry, on the survival of Staphylococcus aureus (ATCC 25923). Blood was able to preserve bacterial viability for up to 72 days when deposited on ceramic flooring. Fabric of cotton fiber allowed longer survival than synthetic fabric. These results show that the composition of biological fluid and type of support influence bacterial survival in normal conditions.

The aim of this study was to evaluate the in vitro growth of Cattleya loddigesii in alternative agents to agar with starch and physical matrix with acclimatization of regenerated plants. Protocorms with 90 days after sowing (0.5 cm of length) were subcultured in 1/2 MS culture medium among the treatments consisting of: agar 7 g L-1 (T1, which corresponds the control), agar 3,5 g L-1 with cassava starch 30 g L-1 (T2), cassava starch 60 g L-1 (T3), cotton fiber (T4) and chopped polyurethane foam (T5). Plantlets were retained in these treatments for over 150 days, and at the end of in vitro culture, were analyzed by their biometric data and acclimatized in a greenhouse during 120 days and evaluated the survival and relative growth rate (RGR). The substrate comprising of chopped polyurethane foam (T5) showed greater efficiency for growth in vitro and also increased survival rate, while substrate cassava starch (T3) provided delay for plantlet growth. Therefore, chopped polyurethane foam is recommended because of low cost and suitable characteristics for the propagation of Cattleya loddigesii.

Cellobiohydrolases hydrolyze cellulose releasing cellobiose units. They are very important for a number of biotechnological applications, such as, for example, production of cellulosic ethanol and cotton fiber processing. The Trichoderma cellobiohydrolase I (CBH1 or Cel7A) is an industrially important exocellulase. It exhibits a typical two domain architecture, with a small C-terminal cellulose-binding domain and a large N-terminal catalytic core domain, connected by an O-glycosylated linker peptide. The mechanism by which the linker mediates the concerted action of the two domains remains a conundrum. Here, we probe the protein shape and domain organization of the CBH1 of Trichoderma harzianum (ThCel7A) by small angle X-ray scattering (SAXS) and structural modeling. Our SAXS data shows that ThCel7A linker is partially-extended in solution. Structural modeling suggests that this linker conformation is stabilized by inter- and intra-molecular interactions involving the linker peptide and its O-glycosylations. © 2013 Springer Science+Business Media Dordrecht.

Fruit purees, combined or not with polysaccharides, have been used in some studies to elaborate edible films. The present study was conducted to evaluate tensile properties and water vapor barrier of alginate-acerola puree films plasticized with corn syrup, and to study the influence of cellulose whiskers from different origins (cotton fiber or coconut husk fiber, the latter submitted to one- or multi-stage bleaching) on the film properties. The whiskers improved the overall tensile properties (except by elongation) and the water vapor barrier of the films. The films with coconut whiskers, even those submitted only to a one-stage bleaching, presented similar properties to those of films with cotton whiskers, despite the low compatibility between the matrix and the remaining lignin in coconut whiskers. This was probably ascribed to a counterbalancing effect of the higher aspect ratios of the coconut whiskers. (C) 2011 Elsevier Ltd. All rights reserved.

Diversos biomateriais podem ser aplicados como suportes na imobilização de células totais de fungos filamentosos ou enzimas isoladas, visando a manutenção e o prolongamento da atividade enzimática em processos biocatalíticos. Exemplos promissores de biomateriais são a fibroína da seda e o alginato de sódio. A fibroína é um material protéico com alta estabilidade térmica, elasticidade, resistência à tensão, não sofre ataque microbiano, baixo custo de purificação e alta tenacidade, o alginato é um biopolímero versátil, devido a suas propriedades gelificantes em soluções aquosas. Assim, neste trabalho empregou-se micélios do fungo derivado de ambiente marinho, Penicillium citrinum CBMAI 1186, livres e imobilizados em biopolímeros (fibra de algodão, fibra de fibroína da seda e fibra de paina) na biorredução quimiosseletiva, regiosseletiva e enantiosseletiva da ligação α,β-C=C de enonas α,β-, α,β,γ,δ- e di-α,β-insaturadas previamente sintetizados pela a reação de condensação aldólica. Foi possível a utilização do fungo P. citrinum CBMAI 1186 na redução quimiosseletiva, regiosseletiva e enantiosseletiva da ligação dupla carbono-carbono de sistemas α,β-insaturados. A imobilização do fungo P. citrinum CBMAI 1186 em biopolímeros (algodão, fibroína da seda, paina e quitosana) permitiu a prolongamento da atividade celular do fungo. O protocolo desenvolvido foi capaz de obter compostos até então descritos apenas por síntese clássica. Também foi realizado reações de resolução enzimática de derivados de haloidrinas por diferentes lipases microbianas de: Pseudomonas fluorescens, Candida cylindracea, Rhizopus niveus e Aspergillus niger. A lipase de P. fluorescens foi imobilizada em esferas de fibroína do bicho da seda (método 1, via adsorção) e em blenda com alginato de cálcio (método 2, via encapsulação) em diferentes condições, tais como, variação de solvente, variação da quantidade de enzima imobilizada e tempo de reação. As condições otimizadas foram empregadas em diferentes haloidrinas, rendendo elevados excessos enantioméricos (ee > 99%) e alta razão enanantiomérica (E > 200) para os produtos acetilados. Foi possível desenvolver um protocolo simples, barato e prático para a síntese enantiosseletiva de haloidrina reforçando a versatilidade da fibroína e do alginato como suportes de imobilização para catalisadores heterogêneos. Também foi possível utilizar a lipase imobilizada (método 2) na reação de transesterificação para obtenção do biodiesel etílico. As melhores condições para o bom funcionamento do biocatalisador foram: 30% do biocatalisador, 20% de n-hexano, relação óleo e etanol de 1:4 a 32 ºC por 48 h em agitação magnética (400 rpm). Essas condições permitiram a formação de 42% de rendimento do biodiesel etílico. O biocatalisador apresentou algumas limitações reacionais, tais como, fragilidade frente a elevadas temperaturas (> 32 ºC) e prolongado tempo de agitação magnética. Porém, permaneceu apto no meio por 4 ciclos consecutivas. Conclui-se que os biomateriais (fibroína, alginato e quitosana) podem ser utilizados como alternativas versáteis na imobilização de micélios de fungos filamentoso e de enzimas isoladas para aplicações em biocatalíticas.

The general method for determining organomercurials in environmental and biological samples is gas chromatography with electron capture detection (GC-ECD). However, tedious sample work up protocols and poor chromatographic response show the need for the development of new methods. Here, Atomic Fluorescence-based methods are described, free from these deficiencies. The organomercurials in soil, sediment and tissue samples are first released from the matrices with acidic KBr and cupric ions and extracted into dichloromethane. The initial extracts are subjected to thiosulfate clean up and the organomercury species are isolated as their chloride derivatives by cupric chloride and subsequent extraction into a small volume of dichloromethane. In water samples the organomercurials are pre-concentrated using a sulfhydryl cotton fiber adsorbent, followed by elution with acidic KBr and CuSO 4 and extraction into dichloromethane. Analysis of the organomercurials is accomplished by capillary column chromatography with atomic fluorescence detection.

This thesis reports the synthesis and/or applications of three types of block copolymers that each bear a low-surface-energy block. First, poly(dimethylsiloxane)-block-poly(2-cinnamoyloxyethyl acrylate) (PDMS-b-PCEA) was synthesized and characterized. Cotton coating using a micellar solution of this block copolymer yielded superhydrophobic cotton fabrics. X-ray photoelectron spectroscopy (XPS) and surface property analyses indicated that the PDMS block topped the polymer coating. Photocuring the cotton swatches crosslinked the underlying PCEA layer and yielded permanent coatings. More interestingly, hydrophilically patterned superhydrophobic cotton fabrics were produced using photolithography that allowed the crosslinking of the coating around irradiated fibers but the removal, by solvent extraction, of the coating on fibers that were not irradiated. Since water-based ink only permeated the uncoated regions, such patterned fabric was further used to print ink patterns onto substrates such as fabrics, cardboard, paper, wood, and aluminum foil. Then, another PDMS-based diblock copolymer poly(dimethylsiloxane)-block-poly(glycidyl methacrylate) (PDMS-b-PGMA) was prepared. Different from PCEA that photocrosslinked around cotton fibers, PGMA reacted with hydroxyl groups on cotton fiber surfaces to get covalently attached. Further, different PGMA chains crosslinked with each other. PDMS-b-PGMA-coated cotton fabrics have been used for oil-water separations. In addition, polymeric nanoparticles were grafted onto cotton fiber surface before PDMS-b-PGMA was used to cover the surfaces of the grafted spheres and the residual surfaces of the cotton fibers. These two types of fabrics, coated by the block copolymer alone or by the polymer nanospheres and then the copolymer, were characterized by scanning electron microscope (SEM), Fourier transform infrared spectroscopy (FT-IR), and water repellency analyses. A comprehensive comparative study was made of their performances in oil-water separation. Finally, a fluorinated ABC triblock copolymer poly(acrylic acid)-block-poly(2-cinnamoyloxyethyl methacrylate)-block-poly(2-perfluorooctylethyl methacrylate) (PAA-b-PCEMA-b-PFOEMA) was used to iii encapsulate air nanobubbles. The produced air nanobubbles were thermodynamically stable in water and were some 100 times more stable than commercially available perfluorocarbon-filled microbubbles under ultrasound. These nanobubbles, due to their small sizes and thus ability to permeate the capillary networks of organs and to reach tumors, may expand the applications of microbubbles in diagnostic ultrasonography and find new applications in ultrasound-regulated drug delivery.

No presente trabalho pretende-se caraterizar a cinética de secagem de papel obtido partir de fibra de algodão. A componente experimental é executada com recurso a uma estação de secagem, munida com equipamentos de controlo e de leitura. O objetivo é realizar a análise da cinética de secagem, com base nas condições impostas através dos parâmetros de entrada, temperatura a humidade relativa e velocidade do ar de secagem. Os resultados permitem determinar a influência dos parâmetros temperatura, humidade relativa, velocidade do escoamento e espessura do material sobre a cinética de secagem. Foi determinada a difusividade em cada ensaio e desenvolvido e validado um modelo matemático de secagem, com recurso aos valores obtidos experimentalmente. A modelação da secagem é realizada através de um modelo que recorre à segunda lei de Fick. São apresentados os resultados da modelação e respetivos desvio padrão relativamente aos valores experimentais.

Raman spectroscopy has been applied to characterize fiber dyes and determine the discriminating ability of the method. Black, blue, and red acrylic, cotton, and wool samples were analyzed. Four excitation sources were used to obtain complementary responses in the case of fluorescent samples. Fibers that did not provide informative spectra using a given laser were usually detected using another wavelength. For any colored acrylic, the 633-nm laser did not provide Raman information. The 514-nm laser provided the highest discrimination for blue and black cotton, but half of the blue cottons produced noninformative spectra. The 830-nm laser exhibited the highest discrimination for red cotton. Both visible lasers provided the highest discrimination for black and blue wool, and NIR lasers produced remarkable separation for red and black wool. This study shows that the discriminating ability of Raman spectroscopy depends on the fiber type, color, and the laser wavelength.
