Le besoin pour des biocapteurs à haute sensibilité mais simples à préparer et à utiliser est en constante augmentation, notamment dans le domaine biomédical. Les cristaux colloïdaux formés par des microsphères de polymère ont déjà prouvé leur fort potentiel en tant que biocapteurs grâce à l’association des propriétés des polymères et à la diffraction de la lumière visible de la structure périodique. Toutefois, une meilleure compréhension du comportement de ces structures est primordiale avant de pouvoir développer des capteurs efficaces et polyvalents. Ce travail propose d’étudier la formation et les propriétés des cristaux colloïdaux résultant de l’auto-assemblage de microsphères de polymère en milieu aqueux. Dans ce but, des particules avec différentes caractéristiques ont été synthétisées et caractérisées afin de corréler les propriétés des particules et le comportement de la structure cristalline. Dans un premier temps, des microsphères réticulées de polystyrène anioniques et cationiques ont été préparées par polymérisation en émulsion sans tensioactif. En variant la quantité de comonomère chargé, le chlorure de vinylbenzyltriméthylammonium ou le sulfonate styrène de sodium, des particules de différentes tailles, formes, polydispersités et charges surfaciques ont été obtenues. En effet, une augmentation de la quantité du comonomère ionique permet de stabiliser de façon électrostatique une plus grande surface et de diminuer ainsi la taille des particules. Cependant, au-dessus d’une certaine concentration, la polymérisation du comonomère en solution devient non négligeable, provoquant un élargissement de la distribution de taille. Quand la polydispersité est faible, ces microsphères chargées, même celles non parfaitement sphériques, peuvent s’auto-assembler et former des cristaux colloïdaux diffractant la lumière visible. Il semble que les répulsions électrostatiques créées par les charges surfaciques favorisent la formation de la structure périodique sur un grand domaine de concentrations et améliorent leur stabilité en présence de sel. Dans un deuxième temps, le besoin d’un constituant stimulable nous a orientés vers les structures cœur-écorce. Ces microsphères, synthétisées en deux étapes par polymérisation en émulsion sans tensioactif, sont formées d’un cœur de polystyrène et d’une écorce d’hydrogel. Différents hydrogels ont été utilisés afin d’obtenir des propriétés différentes : le poly(acide acrylique) pour sa sensibilité au pH, le poly(N-isopropylacrylamide) pour sa thermosensibilité, et, enfin, le copolymère poly(N-isopropylacrylamide-co-acide acrylique) donnant une double sensibilité. Ces microsphères forment des cristaux colloïdaux diffractant la lumière visible à partir d’une certaine concentration critique et pour un large domaine de concentrations. D’après les changements observés dans les spectres de diffraction, les stimuli ont un impact sur la structure cristalline mais l’amplitude de cet effet varie avec la concentration. Ce comportement semble être le résultat des changements induits par la transition de phase volumique sur les interactions entre particules plutôt qu’une conséquence du changement de taille. Les interactions attractives de van der Waals et les répulsions stériques sont clairement affectées par la transition de phase volumique de l’écorce de poly(N-isopropylacrylamide). Dans le cas des microsphères sensibles au pH, les interactions électrostatiques sont aussi à considérer. L’effet de la concentration peut alors être mis en relation avec la portée de ces interactions. Finalement, dans l’objectif futur de développer des biocapteurs de glucose, les microsphères cœur-écorce ont été fonctionnalisées avec l’acide 3-aminophénylboronique afin de les rendre sensibles au glucose. Les effets de la fonctionnalisation et de la complexation avec le glucose sur les particules et leur empilement périodique ont été examinés. La structure cristalline est visiblement affectée par la présence de glucose, même si le mécanisme impliqué reste à élucider.

Small interfering RNA (siRNA), antisense oligonucleotides (ODNs), ribozymes and DNAzymes have emerged as sequence-specific inhibitors of gene expression that may have therapeutic potential in the treatment of a wide range of diseases. Due to their rapid degradation in vivo, the efficacy of naked gene silencing nucleic acids is relatively short lived. The entrapment of these nucleic acids within biodegradable sustained-release delivery systems may improve their stability and reduce the doses required for efficacy. In this study, we have evaluated the potential in vitro and in vivo use of biodegradable poly (d,l-lactide-co-glycolide) copolymer (PLGA) microspheres as sustained delivery devices for ODNs, ribozyme, siRNA and DNA enzymes. In addition, we investigated the release of ODN conjugates bearing 5′-end lipophilic groups. The in vitro sustained release profiles of microsphere-entrapped nucleic acids were dependent on variables such as the type of nucleic acid used, the nature of the lipophilic group, and whether the nucleic acid used was single or double stranded. For in vivo studies, whole body autoradiography was used to monitor the bio-distribution of either free tritium-labelled ODN or that entrapped within PLGA microspheres following subcutaneous administration in Balb-c mice. The majority of the radioactivity associated with free ODN was eliminated within 24 h whereas polymer-released ODN persisted in organs and at the site of administration even after seven days post-administration. Polymer microsphere released ODN exhibited a similar tissue and cellular tropism to the free ODN. Micro-autoradiography analyses of the liver and kidneys showed similar bio-distribution for polymer-released and free ODNs with the majority of radioactivity being concentrated in the proximal convoluted tubules of the kidney and in the Kupffer cells of the liver. These findings suggest that biodegradable PLGA microspheres offer a method for improving the in vivo sustained delivery of gene silencing nucleic acids, and hence are worthy of further investigation as delivery systems for these macromolecules.

Glass microspheres containing radionuclides are used to treat liver cancer. A promising alternative therapy is being developed based on the magnetic hyperthermia which is related to the heat supplied by a magnetic material under an alternating current magnetic field. The advantage of this option is that most of killed cells are cancer cells which are more susceptible to the temperature raise. In the present work aluminum iron silicate glasses containing minor glass modifiers and nucleating agents were synthesized as irregular shape particles which were further transformed in microspheres by using a petrol liquefied gas-oxygen torch. The optimized processing parameters which lead to microspheres that give a response to the magnetic field were determined. The dissolution rate in water at 90 degrees C was determined to be 3 x 10(-8) g cm(-2) min(-1). The microsphere size distribution was determined by laser scattering. The crystalline phase responsible for the ferromagnetic response was identified as magnetite. Since this phase has a high saturation magnetization and high Curie temperature, it is potentially useful for biomedical applications. The hysteresis magnetic loop was measured for materials produced in different conditions, and some of them showed to be appropriated for thermotherapy. The ratio Fe(3+)/Fe(total) was determined by Mossbauer spectroscopy. (C) 2010 Elsevier B.V. All rights reserved.

Basic oxygen furnace (BOF) slag media were studied as a potential treatment material in on-site sanitation systems. Batch and column studies were conducted to evaluate attenuation of the bacteriophage PR772 and 0.190 mu m diameter microspheres by BOF media, and to delineate the relative contributions of two principle processes of virus attenuation: inactivation and attachment. In the batch studies, conducted at 4 degrees C, substantial inactivation of PR772 did not occur in the pH 7.6 and 9.5 suspensions. At pH 11.4, bimodal inactivation of PR772 was observed, at an initial rate of 2.1 log C/C(0) day(-1) for the first two days, followed by a much slower rate of 0.124 log C/C(0) day(-1) over the following 10 days. Two column studies were conducted at 4 degrees C at a flow rate of 1 pore volume day(-1) using two slag sources (Stelco, Ontario; Tubarao, Brazil) combined with sand and pea gravel. In both column experiments, the effluent microsphere concentration approached input concentrations over time (reductions of 0.1-0.2 log C/C(0)), suggesting attachment processes for microspheres were negligible. Removal of PR772 virus was more pronounced both during the early stages of the experiments, but also after longer transport times (0.5-1.0 log C/C(0)). PR772 reduction appeared to be primarily as a result of virus inactivation in response to the elevated pH conditions generated by the BOF mixture (10.6-11.4). On-site sanitation systems using BOF media should be designed to maintain sufficient contact time between the BOF media and the wastewater to allow sufficient residence time of pathogens at elevated pH conditions. Drug delivery systems involving the use of polymers are widely studied and discovery of biocompatible polymers has become the focus of research in this area. Psoralen loaded poly(DL-lactide-co-glycolide) (PLGA) microspheres to be used in PUVA therapy (psoralen and UVA irradiation (ultraviolet A, 320-400 nm) of psoriasis were identified in paraffin sections by histological analysis. The psoralen loaded PLGA microspheres were prepared using the solvent evaporation technique. They were spherical and possessed an external smooth surface as observed by scanning electron microscopy (SEM) analysis. This study describes a modification in the routine preparation of microsphere samples for examination by light microscopy. The changes involved fixative agents and/or stains allowing the identification of microspheres containing a non-fluorescent material. The preservation and identification of microspheres in tissues for histological processing in paraffin was greatly improved by these modifications as proven by our results. (c) 2007 Elsevicr Ltd. Heat-transfer studies were carried out in a packed bed of glass beads, cooled by the wall, through which air percolated. Tube-to-particle diameter ratios (D/dp) ranged from 1.8 to 55, while the air mass flux ranged from 0.204 to 2.422 kg/m2·s. The outlet bed temperature (TL) was measured by a brass ring-shaped sensor and by aligned thermocouples. The resulting radial temperature profiles differed statistically. Angular temperature fluctuations were observed through measurements made at 72 angular positions. These fluctuations do not follow a normal distribution around the mean for low ratios D/dp. The presence of a restraining screen, as well as the increasing distance between the temperature measuring device and the bed surface, distorts TL. The radial temperature profile at the bed entrance (T0) was measured by a ring-shaped sensor, and T 0 showed to be a function of the radial position, the particle diameter, and the fluid flow rate.

The objective of the present study was the development and characterization of ethylcellulose microspheres containing diclofenac and the determination of the in vitro drug release profile. Microspheres were prepared by emulsification/solvent evaporation method using ethyl acetate as solvent for the polymer and water as non solvent. The microspheres were characterized by morphologic and granulometric analyses. The amount of encapsulated drug as well as its release profile in vitro were also determined. The product obtained was microparticles with smooth surface and narrow size distribution, about 50% of the particles being smaller than 5 μm. The methodology used allowed drug encapsulation with a good yield and the system provided a controlled release of diclofenac.

Purpose: The present study aimed to evaluate an injectable extended-release formulation of prednisolone acetate (PA) for orbital administration. Methods: Microspheres (MEs) of poly-ε-caprolactone (PCL) containing PA were developed by the method of solvent evaporation. The MEs obtained were characterized by scanning electron microscopy (SEM), differential scanning calorimetry (DSC), encapsulation efficiency and in vitro release profile. The in vivo release profile was evaluated in rabbits after periocular injection of an aqueous suspension of MEs. The local biocompatibility of the system was verified by histopathologic analysis of the deployment region. Results: After MEs preparation, morphological analysis by SEM showed the feasibility of the employed method. The content of PA encapsulated was 43 ± 7% and can be considered as satisfactory. The system characterization by DSC technique, in addition to confirm the system stability, did not indicate the existence of interaction between the drug and the polymer. The in vitro release study showed the prolonged-release features of the developed system. Preliminary in vivo study showed the absence of local toxicity and confirmed the prolonged release profile of PA from MEs, suggesting the viability of the developed system for the treatment of orbital inflammatory diseases. Conclusion: The results obtained in this work are relevant and accredit the system developed as a possible alternative to the treatment of inflammatory orbitopathy.

Polymers blends represent an important approach to obtain materials with modulated properties to reach different and desired properties in designing drug delivery systems in order to fulfill therapeutic needs. The aim of this work was to evaluate the influence of drug loading and polymer ratio on the physicochemical properties of microparticles of cross-linked high amylose starch-pectin blends loaded with diclofenac for further application in controlled drug delivery systems. Thermal analysis and X-ray diffractograms evidenced the occurrence of drug-polymer interactions and the former pointed also to an increase in thermal stability due to drug loading. The rheological properties demonstrated that drug loading resulted in formation of weaker gels while the increase of pectin ratio contributes to origin stronger structures. © 2012 Elsevier Ltd.

The results of the histopathological analyses after the implantation of highly crystalline PVA microspheres in subcutaneous tissues of Wistar rats are here in reported. Three different groups of PVA microparticles were systematically studied: highly crystalline, amorphous, and commercial ones. In addition to these experiments, complementary analyses of architectural complexity were performed using fractal dimension (FD), and Shannon's entropy (SE) concepts. The highly crystalline microspheres induced inflammatory reactions similar to the ones observed for the commercial ones, while the inflammatory reactions caused by the amorphous ones were less intense. Statistical analyses of the subcutaneous tissues of Wistar rats implanted with the highly crystalline microspheres resulted in FD and SE values significantly higher than the statistical parameters observed for the amorphous ones. The FD and SE parameters obtained for the subcutaneous tissues of Wistar rats implanted with crystalline and commercial microparticles were statistically similar. Briefly, the results indicated that the new highly crystalline microspheres had biocompatible behavior comparable to the commercial ones. In addition, statistical tools such as FD and SE analyses when combined with histopathological analyses can be useful tools to investigate the architectural complexity tissues caused by complex inflammatory reactions. © 2012 WILEY PERIODICALS, INC.

Polyvinyl alcohol (PVA) microspheres with different degree of crystallinity were used as solid supports for Rhizomucor miehei lipase immobilization, and the enzyme-PVA complexes were used as biocatalysts for the transesterification of soybean oil to fatty acid ethyl esters (FAEE). The amounts of immobilized enzyme on the polymeric supports were similar for both the amorphous microspheres (PVA4) and the high crystalline microspheres (PVA25). However, the enzymatic activity of the immobilized enzymes was depended on the crystallinity degree of the PVA microspheres: enzymes immobilized on the PVA4 microspheres have shown low enzymatic activity (6.13 U mg-1), in comparison with enzymes immobilized on the high crystalline PVA25 microspheres (149.15 U mg-1). A synergistic effect was observed for the enzyme-PVA25 complex during the transesterification reaction of soybean oil to FAEE: transesterification reactions with free enzyme with the equivalent amount of enzyme that were immobilized onto the PVA25 microspheres (5.4 U) have yielded only 20% of FAEE, reactions with the pure highly crystalline microsphere PVA25 have not yielded FAEE, however reactions with the enzyme-PVA25 complexes have yielded 66.3% of FAEE. This synergistic effect of an immobilized enzyme on a polymeric support has not been observed before for transesterification reaction of triacylglycerides into FAEE. Based on ATR-FTIR, 23Na- and 13C-NMR-MAS spectroscopic data and the interaction of the polymeric network intermolecular hydrogen bonds with the lipases residual amino acids a possible explanation for this synergistic effect is provided. © 2013 Elsevier Ltd.
