The environmental impact of agro-chemicals for fish production was extensively reviewed. The positive contribution of agro- chemicals and the devastating effect on aquaculture was x-rayed to alert users to this obvious environmental problem. Lime and fertilizers are commonly used in fish farming to increase pH of pond soil and water and to increase alkalinity and hardness, reduce humic acid content and to initiate primary and secondary productivity. Devastating effect of lime on environment is likely to be minimal. In the case of fertilizers, over utilization of this agro-chemical could impair water quality as phytoplankton bloom become excessive which consequently raises BOD. The use of Therapeutants in aquaculture was discovered to be more popular in Europe and North America than in the tropics (Africa). Commonly used therapeutants include antibiotics and antimicrobials. For fish pathology chemicals like formalin, potassium permanganate, Dipterex and malachite green are widely in use. Effluent from farms where these chemicals are commonly in use can distort the aquatic ecosystem. The changes in water quality, aquatic community structure and productivity caused by intensive aquaculture are typical of the impacts of pollution from a wide variety of sources like sewage, agricultural run-off and effluent discharges from industry

Oxygen consumption in Oreochromis mossambicus, Peters (3-60g in weight) was measured under different stress conditions at a constant temperature of 20±1°C. The rate of oxygen consumption was significantly higher (0.170 ml gˉ¹hˉ¹)at a salinity of 30x10ˉ³ compared with that (0.132ml gˉ¹hˉ¹) in freshwater. The oxygen consumption was also found to be affected by changes in pH. Weight specific rate decreased significantly from 0.113 to 0.045 ml gˉ¹hˉ¹ with increasing body weight. A positive correlation was recorded between availability of dissolved oxygen and the rate of oxygen consumption by the fish. While copper sulphate and malachite green inhibited the respiratory metabolism, formaldehyde treatment raised it from 0.088 to 0.118ml gˉ¹hˉ¹.

During a two years research hydrogen peroxide efficacy evaluated for Persian sturgeon, Chinese carps and common carp eggs. These series of the experiments conducted in various conditions different concentration of hydrogen peroxide include 250, 500, 750, 1,000 1,500 2,000 3,000 and 9,000 PPM used as ten and fifteen minutes baths, compared with Malachite green and natural control . In the next phase effect of Levaemisole hydrochloride as an immunostimulator which applied as 5 mg/I in twenty minutes baths from day sixth after hatch evaluated by daily mortality rate and leukocytes counts. The results shown that according fertilization percent and temperature condition hydrogen peroxide at 1,000 and 1,500 PPM concentrations is a effective antifungal agent during incubation periods of Persian sturgeon and even sometimes increasing hatching rates significantly comparing with natural controls and Malachite green. In Chinese carps although hydrogen peroxide controls water molds but it is not recommended in high temperatures because it make shortened incubation time and mold infections will decrease. Also the results shown 750 PPM concentration of hydrogen peroxide in common carp eggs controls water moulds infections and increase hatching rate significantly comparing with Malachite green and natural control. Daily mortality rates accessing of Persian sturgeon fries show that 20 minutes baths of 5mg/1 levamisole hydrochloride decreases daily mortality rate during yolk sac absorption. Nitrogenous compounds: nitrate and ammonium differ significantly between treated tanks with control. Blood leucocytes concentrations as an immune index was different significantly in treated fishes by levamisole hydrochloride comparing with controls. In Chinese carps because yolks sac absorption time is short there is not necessary to use the levamisole hydrochloride. Although treated larvae were more active than controls. As a result our suggestions is to use hydrogen peroxide in Persian sturgeon and common carp artificial propagation and also suggest the use levamisole hydrochloride for Persian sturgeon beside management method in stress and pollution condition

The early life-history of Chinese rock carp Procypris rabaudi was investigated during a 56-day rearing period: 318 artificially propagated P. rabaudi larvae were reared throughout metamorphosis in a small-scale recirculation system (345 L water volume, 10 x 18 L rearing tanks, 150 L storage and filter compartment with bioballs, 20-30 larvae L-1) at the Institute of Hydrobiology, Wuhan, Hubei Province, China. The newly hatched larvae had an initial total length of 8.93 +/- 0.35 mm SD (n = 10) at 3 days post-hatch and reached an average total length of 33.29 mm (+/- 1.88 mm SD, n = 10) 56 days after hatching. Length increment averaged 0.45 mm day(-1), resulting in a mean growth of 24.4 mm within the 56-day period. High mortality rates of up to 92% derived from an introduced fungus infection and subsequent treatment stress with malachite green. Our results indicate that Chinese rock carp can be raised successfully from artificially fertilized eggs. We therefore assume this species to be a candidate for commercial aquaculture.

Malachite Green (MG), Crystal Violet (CV) and Brilliant Green (BC) are antibacterial, antifungal and antiparasitic agents that have been used for treatment and prevention of diseases in fish. These dyes are metabolized into reduced leuco forms (LMG, LCV, LBG) that can be present in fish muscles for a long period. Due to the carcinogenic properties they are banned for use in fish for human consumption in many countries including the European Union and the United States. HPLC and LC-MS techniques are generally used for the detection of these compounds and their metabolites in fish. This study presents the development of a fast enzyme-linked immunosorbent assay (ELISA) method as an alternative for screening purposes. A first monoclonal cell line producing antibodies to MG was generated using a hybridoma technique. The antibody had good cross-reactivates with related chromatic forms of triphenylmethane dyes such as CV, BC, Methyl Green, Methyl Violet and Victoria Blue R. The monoclonal antibody (mAb) was used to develop a fast (20 min) disequilibrium ELISA screening method for the detection of triphenylmethanes in fish. By introducing an oxidation step with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) during sample extraction the assay was also used to detect the presence of the reduced metabolites of triphenylmethanes. The detection capability of the assay was 1 ng g(-1) for MG, LMG, CV, LCV and BC which was below the minimum required performance limit (MRPL) for the detection method of total MG (sum of MG and LMG) set by the Commission Decision 2004/25/EC (2 ng g(-1)). The mean recoveries for fish samples spiked at 0.5 MRPL and MRPL levels with MG and LMG were between 74.9 and 117.0% and inter- and intra-assay coefficients of variation between 4.7 and 25.7%. The validated method allows the analysis of a batch of 20 samples in two to three hours. Additionally, this procedure is substantially faster than other ELISA methods developed for MG/LMG thus far. The stable and efficient monoclonal cell line obtained is an unlimited source of sensitive and specific antibody to MG and other triphenylmethanes. (C) 2011 Elsevier B.V. All rights reserved.

La saprolégniose est une maladie fongique causée par le champignon aquatique Saprolegnia sp. qui affecte les poissons sauvages et ceux provenant des piscicultures. L’apparition de touffes cotonneuses semblables à de la ouate de couleur blanche à grise est souvent la première indication de l’infection. Ce saprophyte ubiquitaire se nourrit habituellement des œufs de poissons morts, mais peut se propager rapidement aux œufs sains causant la mort de ces derniers. La saprolégniose est souvent une infection secondaire, mais des souches virulentes peuvent facilement se développer sur les salmonidés ayant subi un stress ou une mauvaise manipulation. De grandes pertes économiques associées à la saprolégniose sont rapportées chaque année à travers le monde surtout dans l’industrie de la pisciculture. Jusqu’en 2002, le contrôle de la saprolégniose pouvait se faire par l’utilisation du vert de malachite, un colorant organique ayant une grande activité antifongique. Malheureusement, cette molécule a été bannie à cause de ses propriétés cancérigènes. Aucun composé aussi efficace n’est actuellement disponible pour traiter les infections de la saprolégniose. Des molécules ou extraits naturels ayant un potentiel antifongique ont donc été testés à l’aide de deux techniques (par graines de chanvre et par cylindre d’agar). Les molécules d’un extrait de propolis (cire de ruches d’abeilles) démontrant de l’activité anti-Saprolegnia ont été identifiées. De plus, une bactérie, Pseudomonas aeruginosa, pouvant être retrouvée dans le même environnement que Saprolegnia sp. a démontré un effet antagoniste au champignon. Une molécule de signalisation intercellulaire produite par P. aeruginosa, 4-hydroxy-2-heptylquinoline (HHQ), a été identifiée comme responsable de l’effet antagoniste contre Saprolegnia sp.

Les maladies constituent présentement la cause la plus importante de perte économique en aquaculture moderne. Chez certaines espèces, notamment les salmonidés (Oncorhynchus sp. et Salmo sp.), on rapporte des pertes annuelles atteignant cinquante pour cent de la production. À l’heure actuelle, les infections fongiques occupent le second rang derrière les maladies bactériennes en fonction de leur importance économique. Ces poissons sont particulièrement vulnérables à une infection fongique causée par Saprolegnia sp. qui infecte habituellement les oeufs morts. Le saprophyte ubiquitaire se propage ensuite aux oeufs sains et aux individus matures. Malheureusement, le traitement efficace de cette infection, souvent primaire et parfois secondaire, est de plus en plus difficile en raison de nouvelles réglementations restrictives entourant le vert de malachite. Jadis, ce colorant constituait le fongicide le plus efficace dans la lutte contre la saprolégniose, mais son potentiel cancérigène en limite maintenant l’utilisation. Jusqu'à présent, aucun traitement disponible n’est aussi efficace que le vert de malachite pour le contrôle de la saprolégniose. Récemment, nous sommes parvenus à isoler trois bactéries capables d’inhiber la croissance de Saprolegnia sp. in vitro. Ces trois Pseudomonas fluorescens proviennent d’une pisciculture dans laquelle survenaient des cas d’infections à Saprolegnia parasitica. En poussant la caractérisation de l’activité grâce à des analyses de chromatographie liquide haute performance et de spectrométrie de masse, nous avons réussi à isoler et à identifier la molécule responsable. L’acide phénazine-1-carboxylique (PCA), sécrété par deux de nos trois souches, cause l’inhibition de la croissance de Saprolegnia.

Thermal diffusivity (TD) measurements were performed on some industrially important dyes – auramine O (AO), malachite green and methylene blue (MB) – adsorbed K-10 montmorillonites using photoacoustic method. The TD value for the dye-adsorbed clay mineral was observed to change with a variation in dye concentration. The contribution of the dye towards TD was also determined. The repeatedly adsorbed samples with MB and AO exhibited a lower TD than the single-adsorbed samples. TD values of sintered MB samples were also obtained experimentally. These sintered samples exhibit a higher TD, although they show a trend similar to that of non-sintered pellets. A variation in dye concentration and sintering temperature can be used for tuning the TD value of the clay mineral to the desired level.

The purpose of this study was to verify the occurrence of gastrointestinal parasites in fecal samples from cats of the Andradina city, SP. This work was carried out from March to November of 2007, and used 51 cats delivered to the Center of Zoonoses Control of that city. Techniques of Willis and Faust were used in the fecal examination and resulted in detection of Ancylostoma spp. in 96.1% of the animals; Toxocara spp. in 43.1%; Cystoisospora spp. in 43.1%; Dipylidium caninum in 21.6% and Giardia spp. in 5.9% samples. Cryptosporidium spp. oocysts were detected in 3.9% fecal samples by the use of malachite green negative stain. There was no significant association between the occurrence of endoparasites and consistency of fecal samples. The results confirm that these cats represent important hosts of parasites, some of those with high zoonotic potential.

A total of 145 capybara (Hydrochoerus hydrochaeris) fecal samples from the state of Sdo Paulo, Brazil, were screened for Cryptosporidium spp. oocysts using the malachite green method. Eight samples (5.52%) showed positive results and were further submitted to nested PCR reaction for amplification of fragments of 18S rRNA gene and 60-kDa glycoprotein gene for determination of species, alleles and subtypes of Cryptosporidium. Sequencing of the PCR products of the 18S rRNA gene fragments and 60-kDa glycoprotein gene fragments showed that for both genes all Cryptosporidium isolates from capybara were respectively 100% genetically similar to a bovine isolate of C. parvum and to C parvum subtype IIaA15G2R1. To the best of our knowledge this is the first report of Cryptosporidium infection in this rodent. The finding of zoonotic C parvum infection in a semi-aquatic mammal that inhabits anthroponotic habitats raises the concern that human water supplies may be contaminated with zoonotic Cryptosporidium oocysts from wildlife. In this work the influence of two different iron sources, Fe(NO3)(3) and complexed ferrioxalate (FeOx), on the degradation efficiency of 4-chlorophenol (4CP), malachite green, formaldehyde, dichloroacetic acid (DCA) and the commercial products of the herbicides diuron and tebuthiuron was studied. The oxidation of 4CP, DCA, diuron and tebuthiuron shows a strong dependence on the iron source. While the 4CP degradation is favored by the use of Fe(NO3)(3), the degradation of DCA and the herbicides diuron and tebuthiuron is most efficient when ferrioxalate is used. on the other hand, the degradation of malachite green and formaldehyde is not very influenced by the iron source showing only a slight improvement when ferrioxalate is used. In the case of formaldehyde, DCA, diuron and tebuthiuron, despite of the additional carbon introduced by the use of ferrioxalate, higher mineralization percentages were observed, confirming the beneficial effect of ferrioxalate on the degradation of these compounds. The degradation of tebuthiuron was studied in detail using a shallow pond type solar flow reactor of 4.5 L capacity and 4.5 cm solution depth. Solar irradiation of tebuthiuron at a flow rate of 9 L h(-1), in the presence of 10.0 mmol L-1 H2O2 and 1.0 mmol L-1 ferrioxalate resulted in complete conversion of this herbicide and 70% total organic carbon removal. A new strategy for minimization of Cu2+ and Pb2+ interferences on the spectrophotometric determination of Cd2+ by the Malachite green (MG)-iodide reaction using electrolytic deposition of interfering species and solid phase extraction of Cd2+ in flow system is proposed. The electrolytic cell comprises two coiled Pt electrodes concentrically assembled. When the sample solution is electrolyzed in a mixed solution containing 5% (v/v) HNO3, 0.1% (v/v) H2SO4 and 0.5 M NaCl, Cu2+ is deposited as Cu on the cathode, Pb2+ is deposited as PbO2 on the anode while Cd2+ is kept in solution. After electrolysis, the remaining solution passes through an AG1-X8 resin (chloride form) packed minicolumn in which Cd2+ is extracted as CdCl4/2-. Electrolyte compositions, flow rates, timing, applied current, and electrolysis time was investigated. With 60 s electrolysis time, 0.25 A applied current, Pb2+ and Cu2+ levels up to 50 and 250 mg 1-1, respectively, can be tolerated without interference. For 90 s resin loading time, a linear relationship between absorbance and analyte concentration in the 5.00-50.0 μg Cd 1-1 range (r2 = 0.9996) is obtained. A throughput of 20 samples per h is achieved, corresponding to about 0.7 mg MG and 500 mg KI and 5 ml sample consumed per determination. The detection limit is 0.23 μg Cd 1-1. The accuracy was checked for cadmium determination in standard reference materials, vegetables and tap water. Results were in agreement with certified values of standard reference materials and with those obtained by graphite furnace atomic absorption spectrometry at 95% confidence level. The R.S.D. for plant digests and water containing 13.0 μg Cd 1-1 was 3.85% (n = 12). The recoveries of analyte spikes added to the water and vegetable samples ranged from 94 to 104%.
