This study was designed to investigate the effect of calcium and fluoride intake, and parity and lactation on the risk of spinal osteoporosis. Height loss was used as a surrogate measure for spinal fractures by taking advantage of documented changes in height found during the 25-year follow-up of the Charleston Heart Study cohort. Women who had lost 2-4" in height or who had no change in height during the follow-up period were defined as case and comparison subjects respectively. Calcium intake when the subjects were "about 25" and in the recent past, average intake of fluoride over 25 years, and parity and history of breastfeeding were ascertained by questionnaire from 54 case and 77 comparison subjects. Low calcium intake in the past decreased the risk of height loss (age-adjusted OR = 0.3, 95%CI: 0.1-0.96) although several potentially important confounding variables could not be adjusted for. There was no association between risk of height loss and present calcium intake (OR = 0.8, 95%CI: 0.3-2.6 for low versus high intake) after adjustment for past calcium intake. High fluoride intake decreased the risk of height loss (adjusted OR = 0.4, 95%CI: 0.1-1.2). The effect of fluoride or calcium intake in the present was modified by the level of the other nutrient. Compared to a low intake of both calcium and fluoride, a high intake of one increased the risk of height loss (crude OR = 3.3 for high fluoride/low calcium, crude OR = 6.0 for high calcium/low fluoride) although a high intake of both was slightly protective (crude OR = 0.7). It is estimated that a "high" nutrient intake in this population was greater than 850mg/day for calcium and 2mg/day for fluoride. After adjustment for age, increasing parity decreased the risk of height loss in women who had never breastfed (OR = 0.2, 95%CI: 0.01-1.7 for 4 or more children). Women who had breastfed were also at lower risk of height loss than nulliparous women (OR = 0.3, 95%CI: 0.1-1.2 for 4 or more children) although at any level of parity, breastfeeding women had a greater risk of height loss than did non-breastfeeding women. ^

Over the last several decades, human activities have resulted in environmental changes that have increased the number of stressors that can act on a single environment. In Canadian Shield lakes, two recent stressors, the invasion of Bythotrephes longimanus and calcium decline, have been documented. Widespread acidification of hundreds of North American lakes has resulted in the precipitous decline of lake water calcium concentration. Crustacean zooplankton with high calcium demands are likely to be vulnerable to calcium decline, especially <1.5 mg Ca/L, where survival and reproduction rates are reduced. These taxa are also vulnerable to predation by Bythotrephes that has been implicated in the loss of pelagic biodiversity in soft water lakes. Despite laboratory and field studies aimed at understanding the independent impact of these stressors, it is unclear how their co-occurrence will influence community response. Using a combination of data from a large regional lake survey and field experiments, I examined the individual and joint effects of Bythotrephes and calcium decline on native zooplankton community structure. Results demonstrated that much is known about Bythotrephes and our findings of reduced total zooplankton and species richness, due to the loss of Cladocera, are consistent with field surveys and other experimental studies. While we did not detect strong evidence for an effect of calcium on zooplankton using the lowest calcium concentration among invaded lakes (1.2 mg Ca/L), there is evidence that, as lake water calcium concentrations fall <1 mg Ca/L, per capita growth rates of a broad variety of taxa are expected to decline. At the regional scale, negative effects of Bythotrephes and calcium on abundances of small cladocerans and Daphnia pulicaria, respectively, were in agreement with my experimental observations. We also observed significant interactions between Bythotrephes and calcium for a broad variety of taxa. As Bythotrephes continues to spread and invade lakes that are also declining in aqueous calcium, both stressors are likely to amplify negative effects on Cladocera that appear the most vulnerable. Loss of these important zooplankton in response to both Bythotrephes and calcium decline, is likely to lower zooplankton productivity, with potential effects on phytoplankton and higher trophic levels.

The effects of serum and brain calcium concentration on rat behavior were tested by maintaining animals on either distilled water (N = 60) or water containing 1% calcium gluconate (N = 60) for 3 days. Animals that were maintained on high calcium drinking water presented increased serum calcium levels (control = 10.12 ± 0.46 vs calcium treated = 11.62 ± 0.51 µg/dl). Increase of brain calcium levels was not statistically significant. In the behavioral experiments each rat was used for only one test. Rats that were maintained on high calcium drinking water showed increased open-field behavior of ambulation (20.68%) and rearing (64.57%). on the hole-board, calcium-supplemented animals showed increased head-dip (67%) and head-dipping (126%), suggesting increased ambulatory and exploratory behavior. The time of social interaction was normal in animals maintained on drinking water containing added calcium. Rats supplemented with calcium and submitted to elevated plus-maze tests showed a normal status of anxiety and elevated locomotor activity. We conclude that elevated levels of calcium enhance motor and exploratory behavior of rats without inducing other behavioral alterations. These data suggest the need for a more detailed analysis of several current proposals for the use of calcium therapy in humans, for example in altered blood pressure states, bone mineral metabolism disorders in the elderly, hypocalcemic states, and athletic activities.

Increased consumption of low-fat milk products is inversely associated with the risk of hypertension. The beneficial effect of milk on blood pressure is attributed to high calcium and potassium content but also to specific peptide sequences, which are cleaved from milk protein during gastrointestinal digestion, fermentation of milk with proteolytic starter cultures or enzymatic hydrolysis. Milk products fermented with Lactobacillus helveticus contain casein-derived tripeptides isoleucine-proline-proline (Ile-Pro-Pro) and valine-proline-proline (Val-Pro-Pro), which have been shown to possess antihypertensive effects in humans and in experimental animals. The aim of the present series of studies was to investigate the effects of tripeptides Ile-Pro-Pro and Val-Pro-Pro or fermented milk products containing them on vascular function and blood pressure and to elucidate the mechanisms behind them by using different experimental models of hypertension. Another aim was to characterize the acute effects of tripeptides on blood pressure and arterial stiffness in mildly hypertensive humans. Ile-Pro-Pro and Val-Pro-Pro or fermented milk products containing them attenuated the development of hypertension in two experimental models of hypertension, spontaneously hypertensive rat (SHR) and type 2 diabetic Goto-Kakizaki (GK) rat fed with high-salt diet. Significant differences in systolic blood pressure (SBP) were seen after 8 weeks treatment with tripeptide-containing products compared to control product. Plant sterols did not enhance this effect. Two differently produced tripeptide powders produced a similar attenuating effect on SBP in SHR. In mildly hypertensive subjects, a single administration of tripeptide- and plant sterol-containing fermented milk product decreased both SBP and diastolic blood pressure (DBP) over a period of 8 hours. Protective effect of tripeptides Ile-Pro-Pro and Val-Pro-Pro and fermented milk products containing them on vascular function was demonstrated in in vitro studies and long-term experimental studies. The effect was shown to be endothelium-dependent and possibly involving endothelium-derived hyperpolarizing factor (EDHF). In the clinical study, single administration of tripeptide-containing fermented milk product did not affect measures of arterial stiffness. Long-term treatment with fermented milk product containing Ile-Pro-Pro and Val-Pro-Pro inhibited angiotensin-converting enzyme (ACE) and decreased aldosterone levels thus showing beneficial effects on the renin-angiotensin system (RAS) in SHR and GK. No changes in the components of RAS were observed by the single administration of the same product in mildly hypertensive subjects. Increased levels of cGMP, NOx and citrulline suggest increased nitric oxide (NO) production by the tripeptides. Taken together, Ile-Pro-Pro and Val-Pro-Pro -containing products attenuate the development of hypertension after long-term treatment in experimental models of hypertension and possess an acute antihypertensive effect in mildly hypertensive subjects. In addition, these tripeptides show endothelium-mediated beneficial effects on vascular function. Attenuation of blood pressure increase by the tripeptides in experimental animals involves RAS, but its role in the antihypertensive effect in humans remains to be elucidated.

S100 family of calcium-binding proteins is commonly upregulated in a variety of tumor types and is often associated with tumor progression. Among several S100 members, altered expression of S100A2 is a potential diagnostic and prognostic marker in cancer. Several reports suggest a role for S100A2 in metastasis. Earlier, our studies established regulation of S100A2 by transforming growth factor- (TGF-) and its involvement in TGF--mediated cancer cell invasion and migration. However, the molecular mechanisms of S100A2 protumorigenic actions remain unexplored. In the present study, we demonstrate that overexpression of S100A2 in A549 lung cancer cells induced epithelialmesenchymal transition (EMT) followed by increased invasion, loose colony morphology in soft agar and enhanced Akt phosphorylation (Ser-473). Furthermore, overexpression of S100A2 led to increased tumor growth in immunocompromised mice. In agreement, immunohistochemical examination of resected xenograft tumors established inverse correlation between S100A2 and E-cadherin expression together with activated Akt signaling. Interestingly, our study demonstrates a strong dependence of S100A2 and Smad3 in TGF--induced Hep3B cell EMT and invasion. Most importantly, we demonstrate that these effects of S100A2 are manifested through functional interaction with Smad3, which is enhanced in the presence of high calcium and TGF-. S100A2 stabilizes Smad3 and binds to its C-terminal MH2 domain. Additionally, loss of S100A2 attenuates the transcription of TGF-/Smad3 target genes involved in tumor promotion, such as PA1-1 and vimentin. Collectively, our findings present the first mechanistic details of S100A2 protumorigenic actions and its involvement in TGF--mediated cancer cell invasion and EMT.

A associação inversa da ingestão de cálcio dietético com adiposidade corporal e pressão arterial está documentada em estudos epidemiológicos. Achados experimentais sugerem que este fenômeno pode ser mediado por alterações na concentração intracelular de cálcio ([Ca]i). Existem poucos estudos relacionando o cálcio dietético com a [Ca]i. O objetivo do presente estudo foi avaliar a relação da ingestão habitual de cálcio dietético com a [Ca]i, adiposidade corporal, perfil metabólico, biomarcadores inflamatórios, pressão arterial e função endotelial em mulheres. Para tanto, foi desenvolvido estudo transversal, com 76 mulheres na pré-menopausa submetidas à avaliação: dietética (questionário de frequência alimentar validado); da [Ca]i em eritrócitos (espectrometria de absorbância atômica); da gordura corporal (GC) total [índice de massa corporal (IMC) e % GC por bioimpedância elétrica] e central [perímetro da cintura (PC), e razão cintura quadril (RCQ)]; do perfil metabólico (glicose, colesterol e frações, insulina e HOMA-IR); dos biomarcadores inflamatórios [adiponectina e proteína C-reativa (PCR)]; dos biomarcadores da função endotelial [molécula de adesão intracelular-1 (ICAM-1), molécula de adesão celular vascular-1 (VCAM-1) e E-Selectina]; da função endotelial avaliada pelo equipamento Endo-PAT2000; e da pressão arterial. Calcitriol, paratormônio, cálcio sérico e cálcio urinário completaram o metabolismo do cálcio. As participantes foram estratificadas em 2 grupos de acordo com a ingestão habitual de cálcio: Grupo com baixa ingestão de cálcio ou BIC (n=32; ingestão de cálcio <600mg/d) e Grupo com elevada ingestão de cálcio ou AIC (n=44; ingestão de cálcio ≥600mg/d). A média da idade foi semelhante entre os grupos (Grupo BIC: 31,41,4 vs Grupo AIC: 31,41,4anos; p=0,99). Após ajustes para fatores de confundimento (idade, ingestão de energia, bebida alcoólica, proteína, carboidratos e lipídios), o Grupo AIC, em comparação com o BIC, apresentou valores significativamente mais baixos de IMC (25,65,3 vs 26,9 6,0 kg/m; p=0,02), PC (84,413,6 vs 87,815,3cm; p=0,04), % GC (31,15,9 vs 33,35,6 %; p=0,003), pressão arterial diastólica (68,210,8 vs 72,411,2 mm Hg; p=0,04) e pressão arterial média (80,1310,94 vs 83,8611,70 mmHg; p=0,04); e significativamente mais altos de HDL-colesterol (58,612,2 vs 52,912,2 mg/dL; p=0,004) e adiponectina (34572,1 19472,8 vs 31910,319385,1 ng/mL; p=0,05). A [Ca]i e as outras variáveis avaliadas não diferiram entre os grupos, mesmo após ajustes. Neste estudo realizado com mulheres, o maior consumo de cálcio se associou com valores mais baixos de adiposidade corporal total e central, pressão arterial diastólica e média; além de valores mais elevados de HDL-colesterol e adiponectina

Stimulation operations have with main objective restore or improve the productivity or injectivity rate in wells. Acidizing is one of the most important operations of well stimulation, consist in inject acid solutions in the formation under fracture formation pressure. Acidizing have like main purpose remove near wellbore damage, caused by drilling or workover operations, can be use in sandstones and in carbonate formations. A critical step in acidizing operation is the control of acid-formation reaction. The high kinetic rate of this reaction, promotes the consumed of the acid in region near well, causing that the acid treatment not achive the desired distance. In this way, the damage zone can not be bypassed. The main objective of this work was obtain stable systems resistant to the different conditions found in field application, evaluate the kinetic of calcite dissolution in microemulsion systems and simulate the injection of this systems by performing experiments in plugs. The systems were obtained from two non ionic surfactants, Unitol L90 and Renex 110, with sec-butanol and n-butanol like cosurfactants. The oily component of the microemlsion was xilene and kerosene. The acqueous component was a solution of HCl 15-26,1%. The results shown that the microemulsion systems obtained were stable to temperature until 100ºC, high calcium concentrations, salinity until 35000 ppm and HCl concentrations until 25%. The time for calcite dissolution in microemulsion media was 14 times slower than in aqueous HCl 15%. The simulation in plugs showed that microemulsion systems promote a distributed flux and promoted longer channels. The permeability enhancement was between 177 - 890%. The results showed that the microemulsion systems obtained have potential to be applied in matrix acidizing

Active tufas in the form of waterfalls and dams occur along drainage channels in the Serra do Andre Lopes region (State of Sao Paulo, southeastern Brazil) and are associated with the karst system that developed on a dolomitic plateau with a superhumid subtropical climate. The predominance of autogenic waters enables the groundwater to become enriched in calcium carbonate, with low terrigenous sediment content. The tufas that were studied are composed of calcite and have high calcium contents and low magnesium contents. Eroded tufa beds that originate from changes in the position of fluvial channels or river flow rates also occur in this region. In the Sapatu deposit, phytohermal tufas with complex morphologies are arranged in levels constituting various temporally repeated sequences that were deposited between 10,570 and 4,972 cal years BP. In the Frias deposit, distal fluvial deposits of tufa are massive with a relatively greater quantity of terrigenous material and show evidence of dissolution and reprecipitation. The base of this deposit is composed of a cemented breccia dated at 25,390 years BP, which is younger than the overlying tufas ([42,000 years BP). In the two deposits, the levels of terrigenous sediments (quartz sand and lithic pebbles) and terrestrial gastropod shells are interpreted as phases of increased flow rate of rivers during intervals of higher rainfall.

In der vorliegenden Arbeit wird der Vx-Zyklus und der Ddx-Zyklus unterschiedlicher Pflanzen hinsichtlich ihrer Regulation untersucht. Es konnte an Hand von in vivo Messungen gezeigt werden, dass bei zwei Kieselalgen unterschiedlicher Ordnung (Pennales bzw. Centrales) und einer Haptophyte mit Ddx-Zyklus die Dtx-Epoxidase delta-pH-reguliert ist. Im Gegensatz dazu steht die nicht-regulierte Zx-Epoxidase des Vx-Zyklus einer Raphidophyceae, einer Grünalge und einer aquatischen Höheren Pflanze. Es konnte gezeigt werden, dass der Grund für diese unterschiedliche Regulation der beiden Epoxidasen die verschiedenen Quench-Eigenschaften der Pigmente Dtx bzw. Zx ist. Durch parallele Messungen des NPQ und des De-Epoxidierungsgrads wurde deutlich, dass Zx zum Aufbau eines Quenching direkt den im Licht aufgebauten delta-pH benötigt, während Dtx alleine ausreichend ist, um ein Quenching zu verursachen. Bei diesen in vivo Messungen wurde außerdem deutlich, dass die Aktivitäten der untersuchten Epoxidasen große Unterschiede aufweisen. Diese sind abhängig von der entsprechenden Pigmentierung des jeweiligen Lichtsammelsystems, stehen also in Zusammenhang mit den Carotinoidbiosynthesen. Es konnte gezeigt werden, dass bei allen untersuchten Organismen, die eine Xanthophyll-dominierte Antenne mit Fx als Massenpigment enthielten, die Umsatzraten der Epoxidase sehr hoch waren, im Gegensatz zu Chl-dominierten Antennen. Nach diesen Erkenntnissen wurde die Dtx-Epoxidase weiter untersucht und so erstmalig durch Western-Blotting identifiziert. Es ergaben sich, allerdings erst nach zusätzlicher Proteinstabilisierung, zwei Signale, eins bei 60 kDa, das andere bei 57 kDa. Hierbei ist nach wie vor unklar, warum das Antiserum zwei Signale lieferte und ob es sich dabei um Isoformen, um anderweitige Modifizierungen, oder um eine Kreuzreaktion handelt. Auch der Mechanismus der delta-pH-Regulation der Dtx-Epoxidase konnte trotz in vivo und in vitro durchgeführter Studien nicht endgültig geklärt werden. Allerdings konnten verschiedene Mechanismen, wie z.B. eine direkte pH-Abhängigkeit des Enzyms, eine Regulation durch Reduktion und Oxidation oder durch Phosphorylierung und Dephosphorylierung, auf Grund der Daten falsifiziert werden. Es konnte schließlich die Regulation mit Hilfe eines transmembranen Rezeptors als das einzige, mit allen Daten konsistente Regulationsmodell vorgeschlagen werden.

Due to their relatively high calcium oxide content, industrial mineral oxide wastes are potential candidates for mineral sequestration of carbon dioxide (CO2). Cement kiln dust (CKD), a byproduct of cement manufacturing contains 20-60% CaO making it a possible candidate for CO2 sequestration. In this study, three types of CKD are characterized, before and after carbonation, using environmental scanning electron microscopy and energy dispersive x-ray microanalysis to determine the mineralogical and morphological changes occurring due to carbonation. The reactants, products, and precipitation mechanisms were investigated to enhance understanding of the governing processes and allow better utilization of CKD for CO2 sequestration. The results of multiple independent analyses confirmed the formation of CaCO3 during carbonation. Examinations of the reaction pathways found that CaO and calcium hydroxide (Ca(OH)2) were the major reactants. Three types of CaCO3 precipitation mechanisms were observed: (1) diffusion of CO2 into Ca(OH)2 particles causing precipitation in the pores of the particle and the growth of a CaCO3 ring from the outside inward, (2) precipitation onto existing particles, and (3) precipitation from aqueous solution. The growth of a CaCO3 ring on the outside of a particle may slow further diffusion of CO2 into a particle slowing iv the overall sequestration rate. Additionally, changes caused by carbonation in the solubility of trace metals were studied by mixing pre- and post-carbonated CKD with water and analyzing the solution using inductively coupled plasma mass spectrometry. Decreases in the leaching of chromium, lead, and copper were observed, and is an incentive for use of CKD for CO2 sequestration. Equilibrium modeling using PHREEQC confirmed that CaO and Ca(OH)2 would carbonate readily and form CaCO3.

Incident rainfall is a major source of nutrient input to a forest ecosystem and the consequent throughfall and stemflow contribute to nutrient cycling. These rain-based fluxes were measured over 12 mo in two forest types in Korup National Park, Cameroon, one with low (LEM) and one with high (HEM) ectomycorrhizal abundances of trees. Throughfall was 96.6 and 92.4% of the incident annual rainfall (5370 mm) in LEM and HEM forests respectively; stemflow was correspondingly 1.5 and 2.2%. Architectural analysis showed that ln(funneling ratio) declined linearly with increasing ln(basal area) of trees. Mean annual inputs of N, P, K, Mg and Ca in incident rainfall were 1.50, 1.07, 7.77, 5.25 and 9.27 kg ha(-1), and total rain-based inputs to the forest floor were 5.0, 3.2, 123.4, 14.4 and 37.7 kg ha-1 respectively. The value for K is high for tropical forests and that for N is low. Nitrogen showed a significantly lower loading of throughfall and stemflow in HEM than in LEM forest, this being associated in the HEM forest with a greater abundance of epiphytic bryophytes which may absorb more N. Incident rainfall provided c. 35% of the gross input of P to the forest floor (i. e., rain-based plus small litter inputs), a surprisingly high contribution given the sandy P-poor soils. At the start of the wet season leaching of K from the canopy was particularly high. Calcium in the rain was also highest at this time, most likely due to washing off of dry-deposited Harmattan dusts. It is proposed that throughfall has an important `priming' function in the rapid decomposition of litter and mineralization of P at the start of the wet season. The contribution of P inputted from the atmosphere appears to be significant when compared to the rates of P mineralization from leaf litter.

BACKGROUND The human activation peptide of factor XIII (AP-FXIII) comprises the first 37 amino acids of the N-terminus and holds the FXIII in an inactive state. FXIII is activated either proteolytically by cleavage of AP-FXIII by thrombin, or non-proteolytically by high calcium concentrations. OBJECTIVE To investigate the role of AP-FXIII in the expression and stability of FXIII. METHODS We cloned 13 FXIII variants with progressive truncations of AP-FXIII from the N-terminus (delN-FXIII-A), expressed them in mammalian cells, and measured their thermostability, activation, and transglutaminase activity. We also used in silico calculations to analyze the stability of hypothetical delN-FXIII dimers and to identify crucial motifs within AP-FXIII. RESULTS Variants with deletions longer than the first 10 amino acids and an R11Q point mutant were not expressed as proteins. In silico calculations indicated that the sequence (8) FGGR(12) R plays a substantial role in intersubunit interactions in FXIII-A2 homodimers. In agreement with this prediction, the temperature stability of delN-FXIII variants decreased with increasing length of deletion. These results may suggest a role of the N-terminus of AP-FXIII in dimer stability. Substantial sequence homology was found among activation peptides of vertebrate and even invertebrate (crustacean) FXIII-A orthologs, which further supports our conclusion. CONCLUSIONS We conclude that deletion of 11 or more N-terminal amino acids disrupts intersubunit interactions, which may prevent FXIII-A2 homodimer formation. Therefore, AP-FXIII plays an important role in the stability of the FXIII-A2 dimer.

Recent studies of abyssal peridotites (Johnson et al., 1990, doi:10.1029/JB095iB03p02661), mid-ocean-ridge basalts (MORBs) (McKenzie, 1985, doi:10.1016/0012-821X(85)90001-9) and their entrained melt inclusions (Sobolev and Shimizu, 1993, doi:10.1038/363151a0; Humler and Whitechurch, 1988, doi:10.1016/0012-821X(88)90055-6) have shown that fractional melting of the upwelling sub-oceanic mantle produces magmas with a much wider range of compositions than erupted MORBs. In particular, it seems that strongly depleted primary magmas are routinely produced by melting beneath ridges (Johnson et al., 1990, doi:10.1029/JB095iB03p02661). The absence of strongly depleted melts as erupted lavas prompts the question of how long such magmas survive beneath ridges, before their distinctive compositions are concealed by mixing with more enriched magmas. Here we report mineral compositions from a unique suite of oceanic cumulates recovered from DSDP Site 334 (Aumento et al., doi:10.2973/dsdp.proc.37.1977), which indicate that the rocks crystallized from basaltic liquids that were strongly depleted in Na, Ti, Zr, Y, Sr and rare-earth elements relative to any erupted MORB. It thus appears that the magmatic plumbing system beneath the Mid-Atlantic Ridge permitted strongly depleted magmas to accumulate in a magma chamber and remain sufficiently isolated to produce cumulate rocks. Even so, spatial heterogeneity in the compositions of high-calcium pyroxenes suggests that in the later stages of solidification these rocks reacted with infiltrating enriched basaltic liquids.
