In this work a simple and sensitive procedure to extract organic mercury from water and sediment samples, using methylene chloride in acidic media followed by CVAFS quantification has been developed. The method was evaluated for possible interferents, using different inorganic mercury species and humic acid, no effects being observed. The detection limit for organic mercury was 160 pg and 396 pg for water and sediment samples respectively. The accuracy of the method was evaluated using a certified reference material of methylmercury (BCR-580, estuarine sediment). Recovery tests using methylmercury as surrogate spiked with 1.0 up to 30.0 ng L-1 ranged from 90 up to 109% for water samples, whereas for sediments, recoveries ranged from 57 up to 97%.

Mercury distribution and geochemical support on the Continental Margin was evaluated at the Campos Basin, Rio de Janeiro state, Brazil. The average concentrations for all analyzed elements were, respectively, 20 ± 5 ng g-1 (Hg); 30 ± 14 mg g-1 (Al); 16 ± 6 mg g-1 (Fe), and 254 ± 83 µg g-1 (Mn). Silt and clay content, total carbonate and Hg, and organic carbon increased with depth. Finally, the relationship between Hg and silt clay showed significant positive correlation. Total Hg concentrations are the background level described primarily (~40 ng g-1).

A method for selective determination of tributyltin in the presence of Sn(IV) by combining hydride generation inductively coupled plasma optical emission spectrometry (HG-ICP OES) and solid phase extraction (SPE) using baker's yeast is proposed. The procedure is based on selective retention of TBT by the yeast at pH = 6. Detection limit of 1.9 µg L-1 and quantification limit of 6.3 µg L-1 were obtained. TBT and San(IV) were determined in the range of 0-25 µg L-1 and the proposed approach was applied to analyze river water, sea water and biological extracts, with recoveries of 114, 101 and 86%, respectively.

Mercury distribution and fractionation were determined in sediments from the Paraíba do Sul River – RJ, Brazil. Total mercury concentration ranged from 1 to 158 ng g-1. Hg associated with the weakly bound fraction was dominant in the estuarine areas (main - 60% and secondary - 55%); followed by fluvial end member (48%) and mangrove (18%). These results reinforce the mercury availability to fluvial and estuarine areas and emphasize the key role played by mangroves as an efficient biogeochemical barrier. In conclusion, the continuous reduction of the mangrove ecosystem around the world can exacerbate the damage resulting from the mercury accumulation.

Foram efetuadas reações entre o heterometálico [WCl(CO)3(bipy)(HgCl)], (bipy = 2,2'-bipiridina) e tiouréias que levaram à formação de compostos do tipo [WCl(CO)3(bipy)(HgCl)L] [L = tiouréia (tu); N-metiltiouréia (mtu); N,N-dimetiltiouréia (dmtu)]. Os espectros de absorção no infravermelho mostram que a esfera de coordenação do tungstênio mantêm-se inalterada e que as tiouréias encontram-se ligadas ao átomo de mercúrio através do átomo de enxofre.

This paper discusses a rapid and sensitive method developed to determine trace levels of mercury in natural water samples by cold vapor atomic absorption spectrometry using a preconcentration system composed by mini-column packed with 100 mg of 2-aminothiazol modified silica gel (SiAT) coupled on-line with the spectrometer's cold vapor generator system. The optimum preconcentration conditions are also described here. The preconcentrated Hg(II) ions were eluted directly from the column to the spectrometer's cold vapor generator system using 100 µL of 2 mol L-1 hydrochloric acid and the retention efficiency achieved exceeded 95%. The enrichment factors determined were 29, 38 and 46 using 3, 4 and 5 mL of preconcentrated aqueous solutions containing 400 ng L-1 of Hg. The detection limit calculated was 5 ng L-1. The preconcentration procedure was applied to determine trace level mercury in spiked river water samples.

Atualmente, grupamento de anastomose (AG) de Rhizoctonia sp. em crisântemo e ocorrência deste fungo em gipsófila ainda não foram relatados no Brasil. Assim, realizou-se teste de patogenicidade normal e cruzada e sequenciamento da região ITS-5.8S rDNA para identificar o AG de isolado obtido de plantas de crisântemo (Papiro Branco) e de gipsófila, ambas originárias de Holambra / São Paulo, Brasil. Após os testes, relata-se pela primeira vez a ocorrência de R. solani AG-4 HG I em crisântemo (Papiro Branco e Amarelo) e R. solani AG-4 HG III em gipsófila, no estado de São Paulo, Brasil, e, também, a sua patogenicidade cruzada.

Objective: Io evaluate the expression of p16INK4a and p53 biomarkers in conization specimens from patients with high grade cervical intraepithelial neoplasia (HG-CIN), correlating them with the ability to predict the recurrence. Methods : we conducted a retrospective study of patients with HG-CIN in cervical biopsy treated with conization between January 1999 and January 2006 who had a minimum follow-up of 18 months. The expression of the p16 and p53 was assessed by tissue microarrays and correlated with disease recurrence. For analysis, we used the test of proportions (chi-square), considering value p<0.05, 95% CI and calculations of sensitivity, specificity and accuracy of these immunomarkers in predicting recurrence. Results : the series comprised 83 patients aged between 16 and 86 years (35±11.7), divided into two groups: 30 with HG-CIN recurrence (study group) and 53 without recurrence (control group). Mean age, parity, smoking and conization technique were similar in both groups. The p53 expression was present in 43% of the study group and 57% of the control group, and the p16 was present in 43% of the study group and in 57% of the control group (p>0.05). p53 had a positive predictive value (PPV) of 42% and negative predictive value (NPV) of 73%, sensitivity 70%, specificity of 47% and accuracy of 59%. The p16, PPV 42%, NPV 72%, sensitivity 66%, specificity of 49% and accuracy of 56%. Conclusion : immunohistochemistry expression of p53 and p16 showed low sensitivity and low specificity as predictors of HG-CIN recurrence after conization treatment.

Um novo método espectrofotométrico simples, sensível e com boa seletividade é apresentado para a determinação rápida de mercúrio (II) em nível de traços com 2-mercapto-5-metilbenzilimidazol (MMBI) como um novo reagente espectrofotométrico (lambda max = 320 nm), em um meio aquoso ligeiramente alcalino (tampão Britton-Robinson (BR) 0,04 M, pH 8,0). A reação é instantânea e a absorbância permanece estável por mais de 24 horas. O coeficiente de absorção molar encontrado foi de 2,71 x 10(4) L mol-1 cm-1. A composição estequiométrica do complexo é de 1:1 (Hg:MMBI). Considerável excesso dos íons cobre, zinco, chumbo e cádmio não interfere na determinação. O método desenvolvido permitiu a determinação de mercúrio na faixa de 2 x 10-6 a 4 x 10-5 mol.L-1 com boa precisão e exatidão, e o limite de detecção para Hg foi de 9,9 x 10-7 mol.L-1. O método foi aplicado com sucesso a amostras de peixe e os resultados foram avaliados com o clássico método de espectrometria de absorção atômica (EAA). Os desvios padrão relativos para as amostras analisadas foram de 7,2 e 33% (n = 5), enquanto os erros foram de 1,63 e 11,6. O método mostrou-se sensível, seletivo e foi aplicado para a determinação de mercúrio em amostras de peixe com resultados satisfatórios.
