The microbiological flora of 108 water samples was explored to evaluate the role of recreational waters as a possible source of human diseases in Araraquara, S.P., Brazil. These waters included six swimming pools and three lakes with beaches. The number of total and fecal coliforms, Escherichia coli, fecal streptococci, Pseudomonas aeruginosa, Staphylococcus aureus, Candida albicans, and heterotrophic organisms was determined. As was the occurrence of Salmonella, Shigella, Yersinia, enteropathogenic E. coli (EPEC), enteroinvasive E. coli (EIEC), Mycobacteria, yeasts in general and dermatophytes. Shigella, Yersinia, EIEC and dermatophytes were not isolated. Other organisms or groups of microorganisms were found in variable proportions. From this study it is concluded that recreational waters used by the population of Araraquara, may be contaminated with potentially pathogenic microorganism and this may serve as a source of human diseases.

The present study evaluated in vitro susceptibility testing of dermatophytes isolates from healthy cattle and soil samples against three antifungal agents and three topical veterinarian drugs. Itraconazole and terbinafine showed a higher in vitro fungicidal activity than fluconazole. The veterinarian drugs LEPECID (R) and iodine 5% were more active in vitro than the UNGUENTO (R) spray. All drugs showed fungicidal activity against Microsporum gypseum, and they may be considered as efficient agents for the topical treatment of dermatophytoses in cattle.

In this communication, we show that the growth of isolate H6 of the dermatophyte Trichophyton rubrum on non-buffered medium and under saturating phosphate conditions is dependent on the initial growth pH, with an apparent optimum at pH 4.0. In addition, irrespective of the initial growth pH, the pH of the medium altered during cultivation reaching values that ranged from 8.3 to 8.9. Furthermore, this isolate synthesized and secreted almost the same levels of an alkaline phosphatase with an apparent optimum pH ranging from 9.0 to 10.0 when grown on both low- and high-phosphate medium. Also, this alkaline phosphatase is activated by Mg2+ and is EDTA-sensitive. On the other hand, the very low levels of the enzyme retained by the mycelium grown on buffered medium at pH 5.0-5.2 suggest that this enzyme is encoded by an alkaline gene, i.e., a gene responsive to ambient pH signaling.

Dermatophytosis on skin, hair and nails are the most common infectious process in the world. A total of 94 individuals from Public Institutions from the city of Araraquara - Sao Paulo/Brazil, with suspected of dermatophytic lesions were examined in order to determine the incidence and etiology of dermatophytosis. 105 specimens were collected from August to December of 2001 in the Mycology Laboratory of the Faculdade de Ciências Farmacêuticas. It was observed that 47 samples were positive for dermatophytes. Trichophyton rubrum was the prevalent specie (59.6%), followed by Microsporum canis (17%), T. tonsurans (10.6%), T. mentagrophytes (8.5%) and Epidermophyton floccosum (4.3%). T. rubrum was the most frequent in interdigital lesions (81.5%) and M. canis was the main dermatophyte involved in scalp lesions (58.3%). Therefore, it was observed a predominance of antropophilic and zoophilic species, respectively. These results are in agreement with statistical data from South and Southeast regions of Brazil, as well as from other parts of the world in which these fungi were the most frequently isolates from tinea pedis and tinea capitis. In this study, it was also observed a high percentage of T. tonsurans (41.7%) in tinea capitis and this result was different from the statistical data collected until now in our region.

BACKGROUND: Superficial fungal infections are caused by dermatophytes, yeasts or filamentous fungi. They are correlated to the etiologic agent, the level of integrity of the host immune response, the site of the lesion and also the injured tissue. OBJECTIVE: The purpose of this study is to isolate and to identify onychomycosis agents in institutionalized elderly (60 years old +). METHODS: The identification of the fungi relied upon the com-bined results of mycological examination, culture isolation and micro cultures observation under light microscopy from nail and interdigital scales, which were collected from 35 elderly with a clinical suspicion of onychomycosis and a control group (9 elderly with healthy interdigital space and nails). Both groups were insti-tutionalized in two nursing homes in Sao Bernardo do Campo, SP, Brazil. RESULTS: The nail scrapings showed 51.40% positivity. Of these, dermatophytes were found in 44.40% isolates, 27.78% identified as Trichophyton rubrum and 5.56% each as Trichophyton tonsurans, Trichophyton mentagrophytes and Microsporum gypseum. The second more conspicuous group showed 38.89% yeasts: 16.67% Candida guilliermondii, 11.11% Candida parapsilosis, 5.56% Candida glabrata, and 5.56% Trichosporon asahii. A third group displayed 16.70% filamen-tous fungi, like Fusarium sp, Aspergillus sp and Neoscytalidium sp (5.56% each). The interdigital scrapings pre-sented a positivity rate of 14.29%. The agents were coincident with the fungi that caused the onychomycosis. In the control group, Candida guilliermondii was found at interdigital space in one person. CONCLUSION: Employing a combination of those identification methods, we found no difference between the etiology of the institutional-ized elderly onychomycosis from that reported in the literature for the general population. © 2013 by Anais Brasileiros de Dermatologia.

A tinea pedis, comumente chamada de “pé de atleta”, é uma das dermatofitoses mais comuns, sendo causada principalmente por fungos antropofílicos: Trichophyton rubrum e Trichophyton mentagrophytes. Diversos fatores influenciam a prevalência de tinea pedis incluindo: clima, estilo de vida, banheiros públicos, piscinas, saunas, sapato fechado, falta de cuidados com a higiene dos pés. Além disso, estudos mostram que a infecção geralmente ocorre quando os pés dos indivíduos são expostos a esses patógenos. As formas clínicas de tinea pedis são: a) interdigital; b) hiperceratósica e c) vesicular. O objetivo do nosso estudo foi avaliar a freqüência, epidemiologia e aspectos clínicos de tinea pedis, em Belém do Pará, Brazil, no período de 1999 a 2004. Este estudo foi realizado no ambulatório do Serviço de Dermatologia do Departamento de Patologia Tropical do Centro de Ciências da Saúde da UFPA. No período de Janeiro de 1999 a Junho de 2004 foram feitos 810 exames micológicos de pacientes com suspeita clínica de tinea pedis e todos os exames positivos (195) foram incluídos no estudo. O material coletado foi submetido ao exame microscópico usando KOH, cultura em ágar Sabouraud e microcultivo em lâmina. Trichophyton rubrum (40%) e Trichophyton mentagrophytes (26%) foram as espécies mais frequentemente isoladas. Correlação entre sexo, idade, local de residência, variação sazonal e agentes etiológicos são considerados neste estudo. Melhores condições de higiene e diagnóstico precoce são ferramentas importantes para o controle e redução da incidência de dermatofitoses em Belém-PA.

As dermatofitoses são infecções superficiais capazes de produzir lesões em tecidos queratinizados como pele, pêlo e unhas e estão entre as micoses mais freqüentes que acometem o homem. Quanto ao hospedeiro preferencial e habitat natural, os hermatófitos são classificados em antropofílicos, zoofílicos e geofílicos e são conhecidos três gêneros: Trichophyton, Microsporum e Epidermophyton. A tinea capitis é uma doença infecto-contagiosa causada por dermatófitos dos gêneros Trichophyton e Microsporum, de ocorrência predominante na infância e rara após a adolescência. Pode ser dividida em: tinha favosa, favus ou favo, tinha tricofítica e tinha microspórica. O presente trabalho teve como objetivo avaliar a prevalência de tinea capitis em pacientes atendidos no Serviço de Dermatologia do Departamento de Patologia Tropical / Centro de Ciências da Saúde / Universidade Federal do Pará, no período de janeiro de 1999 a junho de 2004. Todos os pacientes com suspeita clínica da doença foram encaminhados ao Setor de Micologia para realização do exame micológico direto usando clarificador KOH 20% e cultura em Ágar Sabouraud. Foram observados 324 casos de tinea capitis, e analisados apenas 308. Destes, o sexo mais acometido foi o feminino, com 222 casos (72,1%), a faixa etária mais acometida foi de 0 a 12 anos (270 casos), com predomínio estatisticamente significante do sexo feminino em 69,6% dos pacientes em idade infantil. Entre os adultos, o sexo mais acometido foi também o feminino com 34 casos (89,5%). O agente etiológico mais freqüente foi o Trichophyton tonsurans (75,8%), seguido do Microsporum canis isolado em 17,7% dos casos. Os resultados demonstraram o predomínio de tinea capitis em crianças em idade escolar e que a espécie antropofilica Trichophyton tonsurans foi o agente mais comum de lesões no couro cabeludo.

Millions of people and animals suffer from superficial infections caused by a group of highly specialized filamentous fungi, the dermatophytes, which only infect keratinized structures. With the appearance of AIDS, the incidence of dermatophytosis has increased. Current drug therapy used for these infections is often toxic, long-term, and expensive and has limited effectiveness; therefore, the discovery of new anti dermatophytic compounds is a necessity. Natural products have been the most productive source for new drug development. This paper provides a brief review of the current literature regarding the presence of dermatophytes in immunocompromised patients, drug resistance to conventional treatments and new anti dermatophytic treatments.

Onychomychosis, a nail fungus infection is the most frequent nail ailment, constituting about half of all nail disorders. It can be caused by dermatophytes, non-dermatophytes, yeasts and Prothoteca spp. Methods include 5407 samples of patients with suspected onychomycosis, studied from January 2002 to December 2006, by direct mycological examination and fungi culture. The diagnosis of onychomycosis was confirmed in samples from 3822 direct mycological and/or culture positive. The diagnosis was established by culture for fungi. Among the 1.428 identified agents, the dermatophytes were responsible for 68.6% (N = 980) of cases, followed by yeasts with 27.6% (N = 394), non-dermatophytes fungi with 2.2% (N = 31), Prothoteca spp with 0.1% (N = 2), and associations with 1.5% (N = 22). Females were more affected, with 66% (N = 2527) of cases, and the most affected age group ranged from 31 to 60 years of age (median 47 years). Fungal microbiota is often changed in the world, both quantitatively and qualitatively, and is affected by several environmental factors. Thus, the periodic review of the composition of this microbiota is important to evaluate the epidemiology and thus proportion a better therapeutic response.

Dermatophytes are adapted to infect skin, hair and nails by their ability to utilize keratin as a nutrient source. Trichophyton rubrum is an anthropophilic fungus, causing up to 90% of chronic cases of dermatophytosis. The understanding of the complex interactions between the fungus and its host should include the identification of genes expressed during infection. To identify the genes involved in the infection process, representational difference analysis (RDA) was applied to two cDNA populations from T. rubrum, one transcribed from the RNA of fungus cultured in the presence of keratin and the other from RNA generated during fungal growth in minimal medium. The analysis identified differentially expressed transcripts. Genes related to signal transduction, membrane protein, oxidative stress response, and some putative virulence factors were up-regulated during the contact of the fungus with keratin. The expression patterns of these genes were also verified by real-time PCR, in conidia of T. rubrum infecting primarily cultured human keratinocytes in vitro, revealing their potential role in the infective process. A better understanding of this interaction will contribute significantly to our knowledge of the process of dermatophyte infection.

Terbinafine hydrochloride (TerbHCl) is an allylamine derivative with fungicidal action, especially against dermatophytes. Different analytical methods have been reported for quantifying TerbHCl in different samples. These procedures require time-consuming sample preparation or expensive instrumentation. In this paper, electrochemical methods involving capillary electrophoresis with contactless conductivity detection, and amperometry associated with batch injection analysis, are described for the determination of TerbHCl in pharmaceutical products. In the capillary electrophoresis experiments, terbinafine was protonated and analyzed in the cationic form in less than 1 min. A linear range from 1.46 to 36.4 mu g mL(-1) in acetate buffer solution and a detection limit of 0.11 mu g mL(-1) were achieved. In the amperometric studies, terbinafine was oxidized at +0.85 V with high throughput (225 injection h(-1)) and good linear range (10-100 mu mol L-1). It was also possible to determine the antifungal agent using simultaneous conductometric and potentiometric titrations in the presence of 5% ethanol. The electrochemical methods were applied to the quantification of TerbHCl in different tablet samples; the results were comparable with values indicated by the manufacturer and those found using titrimetry according to the Pharmacopoeia. The electrochemical methods are simple, rapid and an appropriate alternative for quantifying this drug in real samples. (C) 2012 Elsevier B.V. All rights reserved.
