A Terra atua como um grande magneto esférico, cujo campo assemelha-se àquele gerado por um dipolo magnético. Este campo apresenta mudanças de intensidade que variam com a localização e a hora local. A parte principal do campo geomagnético se origina no interior da Terra através de processos eletromagnéticos. Extensivos estudos mostraram ainda que existem contribuições de origem externa ao planeta, principalmente de origem solar. Dentre estas fontes há anomalias do campo magnético que surgem a partir de um aumento diurno da corrente elétrica em uma estreita faixa da ionosfera, de direção leste-oeste, centrada no equador magnético e denominada Eletrojato Equatorial (EEJ). Ocasionalmente estas correntes podem apresentar reversões de fluxo, sendo denominadas Contra-Eletrojato (CEJ). Vários autores têm estudado os efeitos do EEJ e CEJ sobre as observações geoeletromagnéticas. Eles estão interessados no efeito combinado do EEJ e estruturas geológicas condutivas 1-D e 2-D. Nestes trabalhos a estrutura 2-D sempre se apresentava paralela ao eletrojato, o que é uma hipótese bastante restritiva ao se modelar ambientes geológicos mais realistas, em que corpos bidimensionais podem ter qualquer strike em relação ao EEJ. Neste trabalho apresentamos a solução deste problema sem esta restrição. Assim, mostramos os campos geoeletromagnéticos devidos a estruturas bidimensionais que possuam strike oblíquo em relação ao EEJ, através de perfis dos campos elétrico e magnéticos calculados na superfície e formando direção arbitrária à heterogeneidade condutiva 2-D. Com esta resposta avaliamos ainda qual a influência que estruturas bidimensionais exercem sobre a resposta magnetotelúrica, sob influência do Eletrojato Equatorial. Durante o desenvolvimento deste trabalho, utilizamos o método de elementos finitos, tendo por fonte eletromagnética o EEJ e o CEJ, que por sua vez foram representados por uma combinação de distribuições gaussianas de densidade de corrente. Estas fontes foram decompostas nas direções paralela e perpendicular à estrutura 2-D, resultando nos modos de propagação TE1 e TE2 e TM acoplados, respectivamente. Resolvemos o modo acoplado aplicando uma Transformada de Fourier nas equações de Maxwell e uma Transformada Inversa de Fourier na solução encontrada. De acordo com os experimentos numéricos realizados em um modelo interpretativo da Anomalia Condutiva da Bacia do Parnaíba, formado por uma enorme estrutura de 3000 ohm-m dentro de um corpo externo condutivo (1 ohm-m), concluímos que a presença do CEJ causa uma inversão na anomalia, se compararmos com o resultado do EEJ. Concluímos também que para as frequências mais altas as componentes do campo elétrico apresentam menor influência da parte interna do corpo 2-D do que da parte externa. Já para frequências mais baixas este comportamento se observa com as componentes do campo magnético. Com relação à frequência, vimos os efeitos do “skin-depth”, principalmente nas respostas magnéticas. Além disso, quando a estrutura 2-D está paralela ao eletrojato, o campo elétrico é insensível à estrutura interna do modelo para todos os valores de frequência utilizados. Com respeito ao ângulo θh entre a heterogeneidade e a fonte, vimos que o modo TM se manifesta naturalmente quando θh é diferente de 0°. Neste caso, o modo TE é composto por uma parte devido à componente da fonte paralela à heterogeneidade e a outra devido à componente da fonte perpendicular, que é acoplada ao modo TM. Assim, os campos calculados têm relação direta com o valor de θh. Analisando a influência do ângulo entre a direção do perfil dos campos e o strike da heterogeneidade verificamos que, à medida que θh se aproxima de 90°, os campos primários tornam-se variáveis para valores de θp diferentes de 90°. Estas variações causam uma assimetria na anomalia e dão uma idéia da inclinação da direção do perfil em relação aos corpos. Finalmente, concluímos que uma das influências que a distância entre o centro do EEJ e o centro da estrutura 2-D, causa sobre as componentes dos campos está relacionado às correntes reversas do EEJ e CEJ, pois a 500 km do centro da fonte estas correntes têm máxima intensidade. No entanto, com o aumento da distância, as anomalias diminuem de intensidade. Nas sondagens MT, nós também usamos o EEJ e o CEJ como fonte primária e comparamos nossos resultados com a resposta da onda plana. Deste modo observamos que as componentes do campo geoeletromagnético, usadas para calcular a impedância, têm influência do fator de acoplamento entre os modos TE2 e TM. Além disso, esta influência se torna maior em meios resistivos e nas frequências mais baixas. No entanto, o fator de acoplamento não afeta os dados magnetotelúricos em frequências maiores de 10-2 Hz. Para frequências da ordem de 10-4 Hz os dados MT apresentam duas fontes de perturbação: a primeira e mais evidente é devido à presença fonte 2-D (EEJ e CEJ), que viola a hipótese da onda plana no método MT; e a segunda é causada pelo acoplamento entre os modos TE2 e TM, pois quando a estrutura bidimensional está obliqua à fonte 2-D temos correntes elétricas adicionais ao longo da heterogeneidade. Concluimos assim, que o strike de uma grande estrutura condutiva bidimensional relativamente à direção do EEJ ou CEJ tem de fato influência sobre o campo geomagnético. Por outro lado, para estudos magnetotelúricos rasos (frequências maiores de 10-3 Hz) o efeito do ângulo entre a estrutura geológica 2-D e a direção do EEJ não é tão importante. Contudo, em estudos de litosfera frequências menores de 10-3 Hz) o acoplamento entre os modos TE2 e TM não pode ser ignorado.

Structural health monitoring (SHM) refers to the procedure of assessing the structure conditions continuously so it is an alternative to conventional nondestructive evaluation (NDE) techniques [1]. With the growing developments in sensor technology acoustic emission (AE) technology has been attracting attention in SHM applications. AE are characterized by waves produced by the sudden internal stress redistribution caused by the changes in the internal structure, such as fatigue, crack growth, corrosion, etc. Piezoelectric materials such as Lead Zirconate Titanate (PZT) ceramic have been widely used as sensor due to its high electromechanical coupling factor and piezoelectric d coefficients. Because of the poor mechanical characteristic and the lack in the formability of the ceramic, polymer matrix-based piezoelectric composites have been studied in the last decade in order to obtain better properties in comparison with a single phase material. In this study a composite film made of polyurethane (PU) and PZT ceramic particles partially recovered with polyaniline (PAni) was characterized and used as sensor for AE detection. Preliminary results indicate that the presence of a semiconductor polymer (PAni) recovering the ceramic particles, make the poling process easier and less time consuming. Also, it is possible to observe that there is a great potential to use such type of composite as sensor for structure health monitoring.

En la presente Tesis se realizó un análisis numérico, usando el código comercial Ansys-Fluent, de la refrigeración de una bola de combustible de un reactor de lecho de bolas (PBR, por sus siglas en inglés), ante un escenario de emergencia en el cual el núcleo sea desensamblado y las bolas se dejen caer en una piscina de agua, donde el mecanismo de transferencia de calor inicialmente sería la ebullición en película, implicando la convección y la radiación al fluido. Previamente se realizaron pruebas de validación, comparando los resultados numéricos con datos experimentales disponibles en la literatura para tres geometrías diferentes, lo cual permitió seleccionar los esquemas y modelos numéricos con mejor precisión y menor costo computacional. Una vez identificada la metodología numérica, todas las pruebas de validación fueron ampliamente satisfactorias, encontrándose muy buena concordancia en el flujo de calor promedio con los datos experimentales. Durante estas pruebas de validación se lograron caracterizar numéricamente algunos parámetros importantes en la ebullición en película con los cuales existen ciertos niveles de incertidumbre, como son el factor de acoplamiento entre convección y radiación, y el factor de corrección del calor latente de vaporización. El análisis térmico de la refrigeración de la bola del reactor por ebullición en película mostró que la misma se enfría, a pesar del calor de decaimiento, con una temperatura superficial de la bola que desciende de forma oscilatoria, debido al comportamiento inestable de la película de vapor. Sin embargo, la temperatura de esta superficie tiene una buena uniformidad, notándose que las áreas mejor y peor refrigeradas están localizadas en la parte superior de la bola. Se observó la formación de múltiples domos de vapor en diferentes posiciones circunferenciales, lo cual causa que el área más caliente de la superficie se localice donde se forman los domos más grandes. La separación entre los domos de vapor fue consistente con la teoría hidrodinámica, con la adición de que la separación entre domos se reduce a medida que evolucionan y crecen, debido a la curvatura de la superficie. ABSTRACT A numerical cooling analysis of a PBR fuel pebble, after an emergency scenario in which the nucleus disassembly is made and the pebbles are dropped into a water pool, transmitting heat by film boiling, involving convection and radiation to the fluid, is carried out in this Thesis. First, were performed validation tests comparing the numerical results with experimental works available for three different geometries, which allowed the selection of numerical models and schemes with better precision and lower computational cost. Once identified the numerical methodology, all validation tests were widely satisfactory, finding very good agreement with experimental works in average heat flux. During these validation tests were achieved numerically characterize some important parameters in film boiling with which there are certain levels of uncertainty, such as the coupling factor between convection and radiation, and the correction factor of the latent heat of vaporization. The thermal analysis of pebble cooling by film boiling shows that despite its decay heat, cooling occurs, with pebble surface temperature descending from an oscillatory manner, due to the instability of the vapor film. However, the temperature of this surface has a good uniformity, noting that the best and worst refrigerated area is located at the top of the pebble. The formation of multiple vapor domes at different circumferential positions is observed, which cause that the hottest area of the surface was located where biggest vapor domes were formed. The separation between vapor domes was consistent with the hydrodynamic theory, with the addition that the separation is reduced as the vapor dome evolves and grows, due to the surface curvature.

Most chloroplast genes in vascular plants are organized into polycistronic transcription units, which generate a complex pattern of mono-, di-, and polycistronic transcripts. In contrast, most Chlamydomonas reinhardtii chloroplast transcripts characterized to date have been monocistronic. This paper describes the atpA gene cluster in the C. reinhardtii chloroplast genome, which includes the atpA, psbI, cemA, and atpH genes, encoding the α-subunit of the coupling-factor-1 (CF1) ATP synthase, a small photosystem II polypeptide, a chloroplast envelope membrane protein, and subunit III of the CF0 ATP synthase, respectively. We show that promoters precede the atpA, psbI, and atpH genes, but not the cemA gene, and that cemA mRNA is present only as part of di-, tri-, or tetracistronic transcripts. Deletions introduced into the gene cluster reveal, first, that CF1-α can be translated from di- or polycistronic transcripts, and, second, that substantial reductions in mRNA quantity have minimal effects on protein synthesis rates. We suggest that posttranscriptional mRNA processing is common in C. reinhardtii chloroplasts, permitting the expression of multiple genes from a single promoter.

We have begun to take a genetic approach to study chloroplast protein import in Chlamydomonas reinhardtii by creating deletions in the transit peptide of the γ-subunit of chloroplast ATPase-coupling factor 1 (CF1-γ, encoded by AtpC) and testing their effects in vivo by transforming the altered genes into an atpC mutant, and in vitro by importing mutant precursors into isolated C. reinhardtii chloroplasts. Deletions that removed 20 or 23 amino acid residues from the center of the transit peptide reduced in vitro import to an undetectable level but did not affect CF1-γ accumulation in vivo. The CF1-γ transit peptide does have an in vivo stroma-targeting function, since chimeric genes in which the stroma-targeting domain of the plastocyanin transit peptide was replaced by the AtpC transit peptide-coding region allowed plastocyanin to accumulate in vivo. To determine whether the transit peptide deletions were impaired in in vivo stroma targeting, mutant and wild-type AtpC transit peptide-coding regions were fused to the bacterial ble gene, which confers bleomycin resistance. Although 25% of the wild-type fusion protein was associated with chloroplasts, proteins with transit peptide deletions remained almost entirely cytosolic. These results suggest that even severely impaired in vivo chloroplast protein import probably does not limit the accumulation of CF1-γ.

Studies of recombination-dependent replication (RDR) in the T4 system have revealed the critical roles played by mediator proteins in the timely and productive loading of specific enzymes onto single-stranded DNA (ssDNA) during phage RDR processes. The T4 recombination mediator protein, uvsY, is necessary for the proper assembly of the T4 presynaptic filament (uvsX recombinase cooperatively bound to ssDNA), leading to the recombination-primed initiation of leading strand DNA synthesis. In the lagging strand synthesis component of RDR, replication mediator protein gp59 is required for the assembly of gp41, the DNA helicase component of the T4 primosome, onto lagging strand ssDNA. Together, uvsY and gp59 mediate the productive coupling of homologous recombination events to the initiation of T4 RDR. UvsY promotes presynaptic filament formation on 3′ ssDNA-tailed chromosomes, the physiological primers for T4 RDR, and recent results suggest that uvsY also may serve as a coupling factor between presynapsis and the nucleolytic resection of double-stranded DNA ends. Other results indicate that uvsY stabilizes uvsX bound to the invading strand, effectively preventing primosome assembly there. Instead, gp59 directs primosome assembly to the displaced strand of the D loop/replication fork. This partitioning mechanism enforced by the T4 recombination/replication mediator proteins guards against antirecombination activity of the helicase component and ensures that recombination intermediates formed by uvsX/uvsY will efficiently be converted into semiconservative DNA replication forks. Although the major mode of T4 RDR is semiconservative, we present biochemical evidence that a conservative “bubble migration” mode of RDR could play a role in lesion bypass by the T4 replication machinery.

The performance of circular, rectangular and cross irises for the coupling of microwave power to rectangular waveguide cavity resonators is discussed. For the measurement of complex permittivity of materials using cavity perturbation techniques, rectangular cavities with high Q-factors are required. Compared to the conventional rectangular and circular irises, the cross Iris coupling structure provides very high loaded quality factor for all the resonant frequencies. The proposes cross iris coupling structure enhances the accuracy of complex permittivity measurements.

The possibility that the QCD coupling constant (alpha(s)) has an infrared finite behavior (freezing) has been extensively studied in recent years. We compare phenomenological values of the frozen QCD running coupling between different classes of solutions obtained through non-perturbative Schwinger-Dyson Equations. With these solutions were computed QCD predictions for the asymptotic pion form factor which, in turn, were compared with experiment.

In this work we investigate a possible magnetic moment generation for massive neutral particles with spins-1 and -2 coupled non-minimally, in a specific way, to an external electromagnetic field. It is found that, in the nonrelativistic limit, these particles present g = 1. This result, worked out in the framework of Relativistic Quantum Mechanics, seems to suggest that g = 1 for all massive and neutral particles of any spin ≤ 2. We also compare with the results obtained for massive charged particles of spins-1 and -2, in the same regime (nonrelativistic), in order to investigate the role played by the spin separetely from the charge. The g-factor is a constant which connects the magnetic moment $vec{mu}$ of a charged particle, of charge q and mass m, with its angular momentum $vec{J}$. Thus, the magnetic moment can be writen $ vec{mu}_J=g_Jfrac{q}{2m}vec{J}$. The g-factor for a free particle of spin s=1/2 should take the value g=2. But due to quantum electro-dynamical effects it deviates from this value by a small amount, the so called g-factor anomaly $a_e$, which is of the order of $10^{-3}$ for the free electron. This deviation is even bigger if the electron is exposed to high electric fields. Therefore highly charged ions, where electric field strength gets values on the order of $10^{13}-10^{16}$V/cm at the position of the bound electron, are an interesting field of investigations to test QED-calculations. In previous experiments [H"aff00,Ver04] using a single hydrogen-like ion confined in a Penning trap an accuracy of few parts in $10^{-9}$ was obtained. In the present work a new method for precise measurement of magnetic the electronic g-factor of hydrogen-like ions is discussed. Due to the unavoidable magnetic field inhomogeneity in a Penning trap, a very important contribution to the systematic uncertainty in the previous measurements arose from the elevated energy of the ion required for the measurement of its motional frequencies. Then it was necessary to extrapolate the result to vanishing energies. In the new method the energy in the cyclotron degree of freedom is reduced to the minimum attainable energy. This method consist in measuring the reduced cyclotron frequency $nu_{+}$ indirectly by coupling the axial to the reduced cyclotron motion by irradiation of the radio frequency $nu_{coup}=nu_{+}-nu_{ax}+delta$ where $delta$ is, in principle, an unknown detuning that can be obtained from the knowledge of the coupling process. Then the only unknown parameter is the desired value of $nu_+$. As a test, a measurement with, for simplicity, artificially increased axial energy was performed yielding the result $g_{exp}=2.000~047~020~8(24)(44)$. This is in perfect agreement with both the theoretical result $g_{theo}=2.000~047~020~2(6)$ and the previous experimental result $g_{exp1}=2.000~047~025~4(15)(44).$ In the experimental results the second error-bar is due to the uncertainty in the accepted value for the electron's mass. Thus, with the new method a higher accuracy in the g-factor could lead by comparison to the theoretical value to an improved value of the electron's mass. [H"af00] H. H"affner et al., Phys. Rev. Mutations of the VHL tumor suppressor gene occur in patients with VHL disease and in the majority of sporadic clear cell renal carcinomas (VHL−/− RCC). Loss of VHL protein function is associated with constitutive expression of mRNAs encoding hypoxia-inducible proteins, such as vascular endothelial growth factor. Overproduction of angiogenic factors might explain why VHL−/− RCC tumors are so highly vascularized, but whether this overproduction is sufficient for oncogenesis still remains unknown. In this report, we examined the activity of transforming growth factor-α (TGF-α), another VHL-regulated growth factor. We show that TGF-α mRNA and protein are hypoxia-inducible in VHL−/− RCC cells expressing reintroduced VHL. In addition to its overexpression by VHL−/− RCC cells, TGF-α can also act as a specific growth-stimulatory factor for VHL−/− RCC cells expressing reintroduced wild-type VHL, as well as primary renal proximal tubule epithelial cells, the likely site of origin of RCC. This role is in contrast to those of other growth factors overexpressed by VHL−/− RCC cells, such as vascular endothelial growth factor and TGF-β1, which do not stimulate RCC cell proliferation. A TGF-α-specific antisense oligodeoxynucleotide blocked TGF-α production in VHL−/− RCC cells, which led to the dependence of those cells on exogenous growth factors to sustain growth in culture. Growth of VHL−/− RCC cells was also significantly reduced by a drug that specifically inhibits the epidermal growth factor receptor, the receptor through which TGF-α stimulates proliferation. These results suggest that the generation of a TGF-α autocrine loop as a consequence of VHL inactivation in renal proximal tubule epithelial cells may provide the uncontrolled growth stimulus necessary for the initiation of tumorigenesis.

The crucial role of cell signaling in hemostasis is clearly established by the action of the downstream coagulation protease thrombin that cleaves platelet-expressed G-protein-coupled protease activated receptors (PARs). Certain PARs are cleaved by the upstream coagulation proteases factor Xa (Xa) and the tissue factor (TF)–factor VIIa (VIIa) complex, but these enzymes are required at high nonphysiological concentrations and show limited recognition specificity for the scissile bond of target PARs. However, defining a physiological mechanism of PAR activation by upstream proteases is highly relevant because of the potent anti-inflammatory in vivo effects of inhibitors of the TF initiation complex. Activation of substrate factor X (X) by the TF–VIIa complex is here shown to produce enhanced cell signaling in comparison to the TF–VIIa complex alone, free Xa, or Xa that is generated in situ by the intrinsic activation complex. Macromolecular assembly of X into a ternary complex of TF–VIIa–X is required for proteolytic conversion to Xa, and product Xa remains transiently associated in a TF–VIIa–Xa complex. By trapping this complex with a unique inhibitor that preserves Xa activity, we directly show that Xa in this ternary complex efficiently activates PAR-1 and -2. These experiments support the concept that proinflammatory upstream coagulation protease signaling is mechanistically coupled and thus an integrated part of the TF–VIIa-initiated coagulation pathway, rather than a late event during excessive activation of coagulation and systemic generation of proteolytic activity.

Past studies have shown that epidermal growth factor (EGF) is able to mimic the uterotropic effects of estrogen in the rodent. These studies have suggested a "cross-talk" model in which EGF receptor (EGF-R) signaling results in activation of nuclear estrogen receptor (ER) and its target genes in an estrogen-independent manner. Furthermore, in vitro studies have indicated the requirement for ER in this mechanism. To verify the requirement for ER in an in vivo system, EGF effects were studied in the uteri of ER knockout (ERKO) mice, which lack functional ER. The EGF-R levels, autophosphorylation, and c-fos induction were observed at equivalent levels in both genotypes indicating that removal of ER did not disrupt the EGF responses. Induction of DNA synthesis and the progesterone receptor gene in the uterus were measured after EGF treatment of both ERKO and wild-type animals. Wild-type mice showed increases of 4.3-fold in DNA synthesis, as well as an increase in PR mRNA after EGF treatment. However, these responses were absent in ERKO mice, confirming that the estrogen-like effects of EGF in the mouse uterus do indeed require the ER. These data conclusively demonstrate the coupling of EGF and ER signaling pathways in the rodent reproductive tract.

Two synthetic analogues of murine epidermal. growth factor, [Abu6, 20] mEGF4-48 (where Abu denotes amino-butyric acid) and [G1, M3, K21, H40] mEGF1-48, have been investigated by NMR spectroscopy. [Abu6, 20] mEGF4-48 was designed to determine the contribution of the 6-20 disulfide bridge to the structure and function of mEGF The overall structure of this analogue was similar to that of native mEGF, indicating that the loss of the 6-20 disulfide bridge did not affect the global fold of the molecule. Significant structural differences were observed near the N-terminus, however, with the direction of the polypeptide chain between residues four and nine being altered such that these residues were now located on the opposite face of the main beta-sheet from their position in native mEGF Thermal denaturation experiments also showed that the structure of [Abu6, 20] mEGF4-48 was less stable than that of mEGF. Removal of this disulfide bridge resulted in a significant loss of both mitogenic activity in Balb/c 3T3 cells and receptor binding on A431 cells compared with native mEGF and mEGF4-48, implying that the structural changes in [Abu6, 20] mEGF4-48, although limited to the N-terminus, were sufficient to interfere with receptor binding. The loss of binding affinity probably arose mainly from steric interactions of the dislocated N-terminal region with part of the receptor binding surface of EGF [G1, M3, K21, H40] mEGF1-48 was also synthesized in order to compare the synthetic polypeptide with the corresponding product of recombinant expression. Its mitogenic activity in Balb/c 3T3 cells was similar to that of native mEGF and analysis of its H-1 chemical shifts suggested that its structure was also very similar to native.
