The common occurrence of human derived contaminants like pharmaceuticals, steroids and hormones in surface waters has raised the awareness of the role played by the release of treated or untreated sewage in the water quality along sensitive coastal ecosystems. South Florida is home to many important protected environments ranging from wetlands to coral reefs which are in close proximity to large metropolitan cities. Since large portions of South Florida and most of the Florida Keys population are not served by modern sewage treatment plants and rely heavily on the use of inefficient septic systems; a comprehensive survey of selected human waste contamination markers is needed in these areas to assess water quality with respect to non-traditional micro-constituents. ^ This study reports the development and application of new sensitive and selective analytical methods for the fast screening of multiple wastewater tracers, classified as Emergent Pollutants of Concern (EPOC). Novel methods for the trace analysis of non-traditional markers of human-specific contamination such as aminopropanone were developed and used to assess the potential of non-traditional markers as wastewater tracers. ^ During our investigation, surface water samples collected from near shore environments along the South Florida were analyzed for fifteen hormones and steroids, and five commonly detected pharmaceuticals. The compounds most frequently detected were: coprostanol, cholesterol, estrone, β-estradiol, caffeine, triclosan and DEET. Concentrations of caffeine, bisphenol A and DEET were usually higher and more prevalent than the hormonal steroids. In general, it was found that common pharmaceuticals and steroids are widely present in major coastal environments in South Florida. It is also evident that aquatic bodies in heavily urbanized sectors such as the Miami River and Key Largo Harbor contain higher concentrations of several compounds while relatively open bay waters and agricultural areas show reduced chemical signatures. Concentrations of hormones in the Little Venice area of Marathon Key were above the Lowest Observable Effect Levels (LOELs) for several species, indicating that biological resources in this area are at risk. Water quality issues in some of these coastal water environments go beyond eutrophication, thus EPOC should be the target goal for future mitigation projects. ^

A LLE-GC-MS method was developed to detect PPCPs in surface water samples from Big Cypress National Park, Everglades National Park and Biscayne National Park in South Florida. The most frequently found PPCPs were caffeine, DEET and triclosan with detected maximum concentration of 169 ng/L, 27.9 ng/L and 10.9 ng/L, respectively. The detection frequencies of hormones were less than PPCPs. Detected maximal concentrations of estrone, 17β-estradiol, coprostan-3-ol, coprostane and coprostan-3-one were 5.98 ng/L, 3.34 ng/L, 16.5 ng/L, 13.5 ng/L and 6.79 ng/L, respectively. An ASE-SPE-GC-MS method was developed and applied to the analysis of the sediment and soil area where reclaimed water was used for irrigation. Most analytes were below detection limits, even though some of analytes were detected in the reclaimed water at relatively high concentrations corroborating the fact that PPCPs do not significantly partition to mineral phases. An online SPE-HPLC-APPI-MS/MS method and an online SPE-HPLC-HESI-MS/MS method were developed to analyze reclaimed water and drinking water samples. In the reclaimed water study, reclaimed water samples were collected from the sprinkler for a year-long period at Florida International University Biscayne Bay Campus, where reclaimed water was reused for irrigation. Analysis results showed that several analytes were continuously detected in all reclaimed water samples. Coprostanol, bisphenol A and DEET's maximum concentration exceeded 10 μg/L (ppb). The four most frequently detected compounds were diphenhydramine (100%), DEET (98%), atenolol (98%) and carbamazepine (96%). In the study of drinking water, 54 tap water samples were collected from the Miami-Dade area. The maximum concentrations of salicylic acid, ibuprofen and DEET were 521 ng/L, 301 ng/L and 290 ng/L, respectively. The three most frequently detected compounds were DEET (93%), carbamazepine (43%) and salicylic acid (37%), respectively. Because the source of drinking water in Miami-Dade County is the relatively pristine Biscayne aquifer, these findings suggest the presence of wastewater intrusions into the delivery system or the onset of direct influence of surface waters into the shallow aquifer.

Coral reefs face unprecedented threats throughout most of their range. Poorly planned coastal development has contributed increased nutrients and sewage contamination to coastal waters, smothering some corals and contributing to overgrowth by macroalgae. My approach to assessing the degree to which coral reef ecosystems have been influenced by terrestrial and anthropogenic organic carbon inputs is through the use of carbon (C) and nitrogen (N) stable isotopes and lipid biomarkers in a marine protected area, the Coral Reef System of Veracruz: Parque Nacional Sistema Arrecifal Veracruzano (PNSAV) in the southwest Gulf of Mexico. Firstly, I used a C and N stable isotope mixing model and a calculated fatty acid (FA) retention factor to reveal the primary producer sources that fuel the coral reef food web. Secondly, I used lipid classes, FA and sterol biomarkers to determine production of terrestrial and marine biogenic material of nutritional quality to pelagic and benthic organisms. Finally, I used coprostanol to determine pollutant loading from sewage in the suspended particulate matter. Results indicate that phytoplankton is the major source of essential metabolite FA for marine fish and that dietary energy from terrestrial sources such as mangroves are transferred to juvenile fish, while seagrass non-essential FA are transferred to the entire food web mainly in the rainy season. Sea urchins may be the main consumers of brown macroalgae, especially in the dry season, while surgeon fish prefer red algae in both dry and rainy seasons. C and N isotopic values and the ratio C:N suggest that fertilizer is the principal source of nitrogen to macroalgae. Thus nitrogen supply also favored phytoplankton and seagrass growth leading to a better nutritional condition and high retention of organic carbon in the food web members during the rainy season when river influence increases. However, the great star coral Montastrea cavernosa nutritional condition decreased significantly in the rainy season. The nearest river to the PNSAV was polluted in the dry season; however, a dilution effect was detected in the rainy season, when some coral reefs were contaminated. In 2013, a new treatment plant started working in the area. I would suggest monitoring δ¹⁵N and the C: N ratio in macroalgae as indicators of the nitrogen input and coprostanol as an indicator of human feces pollution in order to verify the efficiency of the new treatment plant as part of the management program of the PNSAV.

A avaliação do aporte de matéria orgânica no ambiente aquático por atividades antrópicas pode ser realizada através da identificação e quantificação de marcadores moleculares. Diversos estudos apontam à aplicação dos marcadores moleculares com esta finalidade, no entanto, poucos avaliam a variação das concentrações desses compostos ao longo do tempo, registrada nas camadas sedimentares. O presente trabalho realiza um estudo a partir de três classes de marcadores moleculares presentes em perfis sedimentares da região do Complexo Estuarino de Paranaguá (CEP) no Paraná (PR), que nos últimos anos vêm sofrendo com o crescente desenvolvimento de atividades antrópicas. Como objetivo, tem-se identificar as principais fontes de matéria orgânica e estudar o histórico destes aportes em colunas sedimentares do CEP, relacionando as taxas de sedimentação com a deposição de origem natural e antrópica. A legislação vigente para o monitoramento ambiental, no que diz respeito à contaminação por esgoto fecal, sugere a avaliação por indicadores microbiológicos, porém, indicadores químicos como os esteróides fecais são uma alternativa bastante promissora, pois estes são persistentes, sendo menos sensíveis a variações ambientais. Outros dois marcadores moleculares de aportes antrópicos ao ambiente que foram determinados neste estudo são os alquilbenzenos lineares (LABs), presente em detergentes, que indicam aportes antrópicos oriundos de esgoto doméstico e a determinação de cafeína, tendo em vista que os esteróides fecais podem ser originários de fezes de animais de sangue quente, podendo indicar outras fontes. Para o presente trabalho foram coletados 12 testemunhos de até 1 m de profundidade em maio de 2006, totalizando 12 pontos de coleta e um montante de 121 amostras. As análises foram realizadas por cromatografia em fase gasosa com detecção por espectrometria de massas (CG-EM). Os esteróides encontrados em maior concentração foram o β- sitosterol (71,4 µg g-1), estigmasterol (8,7 µg g-1), colestanol (3,6 µg g-1) e o estigmastanol (2,8 µg g-1), todos oriundos de fonte natural, indicando que a maior contribuição para o CEP é por aporte biogênico. O coprostanol, que é um esterol fecal, foi encontrado entre as concentrações de 0,001 e 4,10 µg g-1, outros dois esteróides de origem fecal também foram detectados, coprostanona e epicoprostanol, onde as maiores concentrações foram 3,6 e 0,2 µg g-1, respectivamente, sendo encontrados em regiões próximas a centros urbanos, indicando origem antrópica. As maiores concentrações para o ∑LABs também foram encontradas em regiões próximas às cidades de Antonina e Paranaguá, sendo a maior encontrada no testemunho #3 Gererês (208 ng g-1). Para o último marcador molecular analisado, a cafeína, foi encontrada a maior concentração de 18,41 ng g-1, sendo este ponto localizado longe dos centros urbanos, porém este contaminante é bastante solúvel em água podendo ser transportado na coluna d’água e percorrer grandes distâncias. Através dos compostos analisados, pode-se perceber que a intervenção antropogênica foi mais marcante nos testemunhos coletados no eixo leste-oeste do CEP, ficando registrado nas camadas sedimentares.
