BACKGROUND: Alveolar echinococcosis (AE) is a severe helminth disease affecting humans, which is caused by the fox tapeworm Echinococcus multilocularis. AE represents a serious public health issue in larger regions of China, Siberia, and other regions in Asia. In Europe, a significant increase in prevalence since the 1990s is not only affecting the historically documented endemic area north of the Alps but more recently also neighbouring regions previously not known to be endemic. The genetic diversity of the parasite population and respective distribution in Europe have now been investigated in view of generating a fine-tuned map of parasite variants occurring in Europe. This approach may serve as a model to study the parasite at a worldwide level. METHODOLOGY/PRINCIPAL FINDINGS: The genetic diversity of E. multilocularis was assessed based upon the tandemly repeated microsatellite marker EmsB in association with matching fox host geographical positions. Our study demonstrated a higher genetic diversity in the endemic areas north of the Alps when compared to other areas. CONCLUSIONS/SIGNIFICANCE: The study of the spatial distribution of E. multilocularis in Europe, based on 32 genetic clusters, suggests that Europe can be considered as a unique global focus of E. multilocularis, which can be schematically drawn as a central core located in Switzerland and Jura Swabe flanked by neighbouring regions where the parasite exhibits a lower genetic diversity. The transmission of the parasite into peripheral regions is governed by a "mainland-island" system. Moreover, the presence of similar genetic profiles in both zones indicated a founder event.

El presente trabajo se llevó a cabo con el propósito de determinar la fauna de vérmenes parásitos gastrointestinales y su control en gallinas de patio, en las condiciones climático ecológicas del municipio de El Sauce, León. Para el control de los parásitos se utilizaron productos botánicos. Se realizaron muestreos en dos épocas del año (seca y lluvíosa) y dos categorías (pollos y gallinas adultas). La muestra estuvo constituida por 200 animales. El tamaño de las sub-muestras fue de 50 animales por categoria y por epoca. Para la identificacion de los parasitos se uso la helmintologia de Ruff y Norton (1984)Reid y Mac Deugald (1984)Ach y Orichel (1987)EL examen post- morten se realizó de acuerdo a la metodología de Fowler ( 1990). Durante el desarrollo del experimento se identificaron I5 especies de helmintos gastrointestinales. La medía de parásitos, en la categoría de gallina en la época lluvia fue de 4.46 (:t 10.41), y en la época de seca 5.87 (:1: 13.59). En la categoría de pollo, en la época de lluviosa, la media fue de 10.6 (±17.12) y en la época de seca 12.04 (:t 25.6). Los parásitos gastrointestinales de mayor prevalencia independientemente de la categoría y de la época del año fueron los nematodes: Ascaridia galli, Heterakis gallinarum, Tetramere americana; y el cestode Railletina tetragona.Los cestodes Railletina williansi, Hymenolepis cantaniana tienen mayor aparición en época lluviosa .. Al relacionar la prevalencia e intensidad de los parásitos gastrointestinales entre épocas y categorías, se observó mayor aparición durante la época seca que en la epoca lluviosa y más en pollos que en gallinas. Las soluciones acuosas de Neem y madero negro tienen efecto signíficativo en el control de los parásitos gastrointestinales, al igual que el producto químico Amebendazol.

This study concentrated on the reproductive biology of the small pelagic cyprinid Rastrineobola argentea. The results indicate that this fish is an inshore spawning species, which agrees with other recent studies. It was also found that in areas where fishing intensity was likely to be relatively high, the size at first maturity of R. argentea was reduced, which is likely to be an effect of the fish altering its reproductive strategy according to life history theory. The CPUE results showed a general trend of decreasing with distance from shore, however areas less than one kilometer from the shore were not sampled. Evidence was also found suggesting that the cestode parasite, Ligula intestinalis had an adverse effect on the maturation and fecundity of R. argentea. Some management options concerning the findings in this study are also briefly discussed. (PDF has 82 pages)

A hidatidose policística é uma zoonose causada pelo cestóide Echinococcus vogeli, amplamente distribuído no norte do Brasil. Os hospedeiros definitivos são Speothos venaticus (cachorro-vinagre) e Canis familiaris (cães domésticos), enquanto Agouti paca (paca) é hospedeiro intermediário. Tanto as pacas quanto o homem (hospedeiro acidental) desenvolvem a forma larvar (metacestóide), principalmente na superfície e no interior do fígado. Esta tese tem como objetivo geral estudar as características parasitológicas e histopatológicas de metacestóides de E. vogeli, originários de pacas e humanos da região norte do Brasil, visto o conhecimento insuficiente ou mesmo o seu desconhecimento. Os fígados e mesentérios foram obtidos de oito pacientes com hidatidose policística durante ato cirúrgico na Fundação Hospital Estadual do Acre. Pacas foram capturadas no Município de Bujari, Floresta Estadual do Antimary, Acre. Durante a necropsia das pacas, foram observadas lesões macroscópicas (massas esbranquiçadas ou amareladas, semelhantes a bolhas na superfície dos fígados). Para a análise parasitológica foram aplicadas as microscopias de luz, contraste interferencial de Normaski (DIC) e varredura laser confocal. A análise morfométrica foi realizada com o auxílio do Programa Image Pro Plus Media Cybernetics. Os órgãos de pacas e humanos foram submetidos à análise histopatológica. Os pequenos e grandes ganchos rostelares apresentaram polimorfismo morfológico, enquanto a organização dos protoescólices acompanhou o padrão descrito para Echinococcus sp. Todas as pacas apresentavam cistos hepáticos, porém em apenas duas encontramos líquido hidático, comprovados pela presença dos ganchos e protoescólices. A análise histopatológica dos tecidos hepáticos das pacas confirmou a hidatidose policística e evidenciou, pela presença de agrupamentos de ovos, a coinfecção com Calodium hepaticum. As características morfológicas dos ganchos rostelares dos casos humanos não diferiram do descrito para as pacas, entretanto, os ganchos dos helmintos provenientes do fígado foram maiores que os mesentéricos. Já em relação aos protoescólices, os mesentéricos foram maiores do que os hepáticos. Cistos mesentéricos e hepáticos apresentaram protoescólices em diferentes estágios de desenvolvimento, com coroas de ganchos, formadas por grandes e pequenos ganchos, e dois pares de ventosas, além dos corpúsculos calcários. Os cistos hepáticos apresentaram as três membranas características (adventícia, anista e germinativa), enquanto os cistos mesentéricos não apresentaram a membrana adventícia, sendo a anista aquela que mais se destacou neste órgão. No mesentério, as células mononucleares foram os principais constituintes do infiltrado leucocitário, cuja intensidade foi relacionada à capacidade proliferativa da membrana germinativa. Além de cistos hepáticos típicos de fase crônica, dependendo da resposta inflamatória, foram observados cistos na fase aguda e sub-aguda. Foi encontrado um caso de coinfecção com vírus (HIV, HCB e HCV) e outro com envolvimento da vesícula biliar. Em suma, confirma-se hidatidose policística em pacas no Acre e são apresentados novos casos de infecção humana no Acre e Amazonas. Pela primeira vez, é demonstrado polimorfismo, padrão diferente de desenvolvimento dos cistos de acordo com o órgão, coinfecção e envolvimento da vesícula biliar

Hidatidose policística é uma zoonose negligenciada causada pela larva do cestóide, Echinococcus vogeli, em habitantes de florestas úmidas da região neotropical. Caçar e alimentar cães com vísceras da caça são hábitos relatados nos indivíduos suspeitos ou com esta parasitose comprovada. O diagnóstico é baseado em dados clínicos, resultado de análise de imagens e detecção de anticorpos pelo teste de Immunoblot o que tem sido descrito para pacientes com a forma crônica da doença. Apenas nesta fase, os pacientes procuram por assistência médica. Este trabalho tem como objetivo avaliar a reatividade sorológica para hidatidose policística, caracterizar os aspectos epidemiológicos e avaliar a presença de enteroparasitoses nos indivíduos moradores de duas regiões de floresta em Sena Madureira (Acre). Os indivíduos concordaram em participar após apreciação do Termo de Consentimento Livre e Esclarecido. Por meio de questionário foram obtidas informações sobre identificação, endereço, sexo, idade e hábitos de caça. Foram coletadas amostras de sangue em 125 habitantes da unidade de conservação (Floresta Estadual do Antimary) e outra unidade de não conservação ambiental em Sena Madureira, onde foram obtidas 207 amostras de sangue. Os soros foram processados pelo teste de Immunoblot para a pesquisa de IgG, nas diluições 1:100 e 1:10, no Serviço de Referência Nacional em Hidatidose-IOC-Fiocruz. Na Unidade de conservação, 39,2% dos indivíduos apresentaram sorologia positiva e 17,4% na outra área. Demonstrou-se que existe diferença significativa entre as áreas, embora a correlação seja baixa. A maioria dos indivíduos reativos em ambas as áreas apresentou as bandas de 40kDA e 28kDa. Nas duas áreas de estudo não houve diferença significativa na reatividade em relação a faixa etária. O hábito de caçar entre os reativos foi de 100% dos indivíduos praticantes da caça na Unidade de conservação e na Unidade de não conservação foi de 88,9% dos indivíduos. A presença de um a três cães foi predominante nas duas áreas. A prática da caça e a presença de cães não teve associação significativa com a reatividade sorológica em ambas as áreas. A ocupação dos indivíduos reativos nas duas regiões foi predominantemente agricultores e estudantes. Em relação ao exame parasitológico não foi encontrado nenhum parasito nos indivíduos reativos que pudesse ter reação cruzada no teste imunológico. Conclui-se que existe diferença de reatividade entre as áreas, sendo que a Unidade de conservação tem maior número de indivíduos reativos. Não foi verificada associação entre a reatividade, o gênero, a faixa etária, prática de caça e presença de cães. Os indivíduos reativos também possuíam enteroparasitoses, porém estas não estão relacionadas com a possível reação cruzada no teste de Immunoblot. A presença das bandas de 40kDa e 28kDa no teste de Immunoblot podem ser uma característica do perfil sorológico de indivíduos de área endêmica que já entraram em contato com o parasito.

Some aspects of association and development of the caryophyllid cestode Lytocestus indicus Moghe, 1925 infecting the catfish Clarias batrachus (Linn.) from the Kailla Beel of Mymensingh, Bangladesh were studied. About 33.14% of Clarias batrachus were infected with a mean intensity of 3.75, mean density 1.25. The infection was not found throughout the year. Two seasonal occurrence of this cestode were observed, one in April-May and the other in August-September. However, maturation period of the worm coincided with the maturation of the host. The worm was found attached to the wall of the intestine of the host. At the site of attachment tissue layers were compressed due to mechanical injuries. Prevalence and mean intensity of infection increased with length groups. No variation in infection was significantly observed in different sexes of the host examined.

Three nematodes, two acanthocephalans and three cestodes were collected by dissecting 15 species of fish. Of these parasites only Hedruris sp. a larval nematode is responsible directly for the death of fish, since it is found in predatory species a reduction of these might prove of considerable consequence to fish, production. Bothriocephalus gowleonensis, a cestode probably introduced from China with Chinese carp, was recorded in an indigenous carp Puntius sarana. This parasite causes heavy mortality in grass carp in China and is a potential danger to local species. A single crustacean parasite was recorded. It is likely that they are widespread. No protozoa were recorded but there can be little doubt that they occur in local freshwater species and may be of considerable importance in fisheries. Several predators (insects, fish and other vertebrates) were recorded and are doubtless of considerable importance. However, the role of parasites and of predators (especially fishes) in relation to the productivity of fisheries can hardly be assessed at present and needs further study. A study of the fauna found at the shallow edges of 21 irrigation reservoirs showed an abundance of fauna in the low-country reservoirs whilst the up-country reservoirs had a poor fauna. The fauna showed insects, crustacea and mollusca in this order of abundance. The presence of insect and molluscs feeding fish in our fauna shows that these rich sources of food are being utilised.

The caryophyllaeid cestode Lytocestoides fossilis infects the freshwater catfish Heteropneustes fossilis. The study was conducted for two consecutive years (2004-06) to record the bio-statistical data of the parasite. The incidence, intensity, density and index of infection of the parasite have been recorded. The infection was more during June to September, moderate during February to May and low during October to January. The parasite brought about severe histopathological changes in the stomach of infected fish. The changes observed in the stomach of fish included structural damage of the villi, inflammation, and fibrosis associated with hyperplasia and metaplasia. The hypertrophy of mucous layer led to vacuolation and necrosis. Histochemical changes were noticed with enhanced carbohydrate, protein and lipid contents. The enhanced substrate content in the infected organ might be due to the disfunctioning of the digestive tract, which results in the accumulation of various metabolites. Mucus secretion was triggered as a protective interaction against parasitic invasion. The parasitic infection affects the general metabolic state of the host and as the result, the fish becomes sluggish and moribund.

The helminth fauna of Oreochromis leucosticus (Trewavas) from Lake Naivasha is described. The study was carried out on 1,050 gillnetted fish caught over a period of 24 months at different sites within the lake. Oreochromis leucosticus which is a commercially important fish was observed to be a host of four helminth parasites. These were an acanthocephalan polyacanthorhynchus kenyensis (Schmidt and Canaris), a nematode contracaecum sp.. a trematode clinostomum sp. and a dilepidid cestode Amirthalingamia sp. Seasonal variation in prevalence, intensity of infection, organs infected and the variation in infection with the size, sex and body condition of the host are described. There was no seasonal variation in prevalence but intensity of infection was observed to increase with the age of the fish. Although male fish were observed to be more heavily infected, the parasites did not seem to have any significant effect on the body condition of the fish.

An in vitro assay was used to examine the effect of Bothriocephalus acheilognathi Yamaguti, 1934 (Cestoda: Pseudophyllidea) on the polarization response of pronephric leucocytes of carp, Cyprinus carpio. Leucocytes, isolated from naive, naturally-infected fish and carp injected intraperitoneally with cestode extracts, were exposed to parasite extracts (protein concentrations 0-10.0 mu g ml(-1)), for up to 24 h in the presence or absence of carp serum. In general, polarization responses of the pronephric leucocytes, primarily neutrophils and eosinophils, increased with incubation time although there was no significant difference in the response induced by the different protein concentrations. Differences in the polarization response were, however, observed in naive, naturally infected and injected fish and the cells responded differently in the presence and absence of carp serum. In the absence of carp serum the polarization response of pronephric leucocytes in vitro was significantly reduced with cells obtained from injected and naturally infected fish compared with those obtained from naive carp. This suppression of leucocyte migration was however reduced by the addition of carp serum to the in vitro system. The role of this interaction between the possible suppression of polarization induced by the parasite and stimulation by serum is discussed.

Cyprinid fish, Hemiculter leucisculus, Cultrichthys erythropterus and Culter dabryi, were sampled from Liangzi, Honghu and Tangxun lakes in the flood plain of the Yangtze River. The cestode Bothriocephalus acheilognathi Yamaguti, 1934 was found in the 3 lakes, but C. erythropterus sampled from Liangzi lake was found uninfected due probably to the small sample size. Findings of the cestode in the 3 lakes represent the first record of the parasite in the flood plain of the Yangtze River, indicating that B. acheilognathi may be distributed much wider in China than previously recognized.

This study represents the first ß-tubulin sequence from a trematode parasite, namely, the liver fluke, Fasciola hepatica. PCR of genomic DNA showed that at least one ß-tubulin gene from F. hepatica contains no introns. A number of amino acids in the primary sequence of fluke tubulin are different from those described previously in various nematode species and the cestode, Echinococcus multilocularis. ß-Tubulin is an important target for benzimidazole anthelmintics, although (with the exception of triclabendazole) they show limited activity against F. hepatica. The amino acid differences in fluke ß-tubulin are discussed in relation to the selective toxicity of benzimidazoles against helminths and the mechanism of drug resistance.

Neuropeptide F is the most abundant neuropeptide in parasitic flatworms and is analogous to vertebrate neuropeptide Y. This paper examines the effects of neuropeptide F on tetrathyridia of the cestode Mesocestoides vogae and provides preliminary data on the signalling mechanisms employed. Neuropeptide F ( greater than or equal to 10 muM) had profound excitatory effects on larval motility in vitro. The effects were insensitive to high concentrations (I mM) of the anaesthetic procame hydrochloride suggesting extraneuronal sites of action. Neuropeptide F activity was not significantly blocked by a FMRFamide-related peptide analog (GNFFRdFamide) that was found to inhibit GNFFRFamide-induced excitation indicating the occurrence of distinct neuropeptide F and FMRFamide-related peptide receptors. Larval treatment with guanosine 5'-O-(2-thiodiphosphate) trilithium salt prior to the addition of neuropeptide F completely abolished the excitatory effects indicating the involvement of G-proteins and a G-protein coupled receptor in neuropeptide F activity. Addition of guanosine 5'-O-(2-thiodiphosphate) following neuropeptide F had limited inhibitory effects consistent with the activation of a signalling cascade by the neuropeptide. With respect to Ca2+ involvement in neuropeptide F-induced excitation of M. vogae larvae, the L-type Ca2+-channel blockers verapamil and nifedipine both abolished neuropeptide F activity as did high Mg+ concentrations and drugs which blocked sarcoplasmic reticulum Ca2+-activated Ca2+-channels (ryanodine) and sarcoplasmic reticulum Ca2+ pumps (cyclopiazonic acid). Therefore, both extracellular and intracellular Ca2+ is important for neuropeptide F excitation in M. vogae. With resepct to second messengers, the protein kinase C inhibitor chelerythrine chloride and the adenylate cyclase inhibitor MDL-2330A both abolished neuropeptide F-induced excitation. The involvement of a signalling pathway that involves protein kinase C was further supported by the fact that phorbol-12-myristate-13-acetate,known to directly activate protein kinase C, had direct excitatory effects on larval motility. Although neuropeptide F is structurally analogous to neuropeptide Y, its mode-of-action in flatworms appears quite distinct from the common signalling mechanism seen in vertebrates. Using a C-terminally directed pancreatic polypeptide (PP) antiserum and immunocytochemical methods, PP-immunoreactivity (IR) was localized throughout the central (CNS) and peripheral nervous systems (PNS) of the cestode, Moniezia expansa. In the CNS, immunostaining was evident in the paired cerebral ganglia (primitive brain), connecting commissure, and the paired longitudinal nerve cords that are cross-linked by numerous regular transverse connectives. The PNS was seen to consist of a fine anastomosing nerve-net of immunoreactive fibres, many of which were closely associated with reproductive structures. Radioimmunoassay of this peptide IR in acid-alcohol extracts of the worm measured 192.8 ng/g of PP-IR. HPLC analyses of the M. expansa PP-IR identified a single molecular form which was purified to homogeneity. Plasma desorption mass spectrometry (PDMS) of purified parasite peptide resolved a single peptide with a molecular mass of 4599 +/- 10 Da. Automated gas-phase Edman degradation identified a 39-amino acid peptide with a C-terminal phenylalaninamide. Examination of its primary structure shows that it displays significant sequence homology with the vertebrate neuropeptide Y superfamily, suggesting that this platyhelminth-derived peptide is the phylogenetic precursor. Neuropeptide F (M. expansa) is the first regulatory peptide to be fully sequenced from the phylum Platyhelminthes and may represent a member of an important new class of invertebrate neuropeptide.
