Pothomorphe umbellata is a native plant widely employed in the Brazilian popular medicine. This plant has been shown to exert a potent antioxidant activity on the skin and to delay the onset and reduce the incidence of UVB-induced skin damage and photoaging. The aim of this work was to optimize the appearance, the centrifuge stability and the permeation of emulsions containing R umbellata (0. 1% 4-nerolidylchatecol). Experimental design was used to study ternary mixtures models with constraints and graphical representation by phase diagrams. The constraints reduce the possible experimental domain, and for this reason, this methodology offers the maximum information while requiring the minimum investment. The results showed that the appearance follows a linear model, and that the aqueous phase was the principal factor affecting the appearance; the centrifuge stability parameter followed a mathernatic quadratic model and the interactions between factors produced the most stable emulsions; skin permeation was improved by the oil phase, following a linear model generated by data analysis. We propose as optimized P. umbellata formulation: 68.4% aqueous phase, 26.6% oil phase and 5.0% of self-emulsifying phase. A new method of poly-beta-hydroxybutyrate (PHB) extraction from recombinant E. coli is proposed, using homogenization and centrifugation coupled with sodium hypochlorite treatment. The size of PHB granules and cell debris in homogenates was characterised as a function of the number of homogenization passes. Simulation was used to develop the PHB and cell debris fractionation system, enabling numerical examination of the effects of repeated homogenization and centrifuge-feedrate variation. The simulation provided a good prediction of experimental performance. Sodium hypochlorite treatment was necessary to optimise PHB fractionation. A PHB recovery of 80% at a purity of 96.5% was obtained with the final optimised process. Protein and DNA contained in the resultant product were negligible. The developed process holds promise for significantly reducing the recovery cost associated with PHB manufacture.

Centrifuge experiments modeling single-phase flow in prototype porous media typically use the same porous medium and permeant. Then, well-known scaling laws are used to transfer the results to the prototype. More general scaling laws that relax these restrictions are presented. For permeants that are immiscible with an accompanying gas phase, model-prototype (i.e., centrifuge model experiment-target system) scaling is demonstrated. Scaling is shown to be feasible for Miller-similar (or geometrically similar) media. Scalings are presented for a more, general class, Lisle-similar media, based on the equivalence mapping of Richards' equation onto itself. Whereas model-prototype scaling of Miller-similar media can be realized easily for arbitrary boundary conditions, Lisle-similarity in a finite length medium generally, but not always, involves a mapping to a moving boundary problem. An exception occurs for redistribution in Lisle-similar porous media, which is shown to map to spatially fixed boundary conditions. Complete model-prototype scalings for this example are derived.

The settling characteristics of cell debris and inclusion bodies prior to, and following, fractionation in a disc-stack centrifuge were measured using Cumulative Sedimentation Analysis (CSA) and Centrifugal Disc photosedimentation (CDS). The impact of centrifuge feedrate and repeated homogenisation on both cell debris and inclusion body collection efficiency was investigated. Increasing the normalised centrifuge feedrate (Q/Sigma) from 1.32 x 10(-9) m s(-1) to 3.97 x 10(-9) m s(-1) leads to a 36% increase in inclusion body paste purity. Purity may also be improved by repeated homogenisation. Increasing the number of homogeniser passes results in smaller cell debris size whilst leaves inclusion body size unaltered. At a normalised centrifuge feedrate of 2.65 x 10(-9) m s(-1), increasing the number of homogeniser passes from two (2) to ten (10) improved overall inclusion body paste purity by 58%. Grade-efficiency curves for both the cell debris and inclusion bodies have also been generated in this study. The data are described using an equation developed by Mannweiler (1989) with parameters of k = 0.15-0.26 and n = 2.5-2.6 for inclusion bodies, and k = 0.12-0.14 and n = 2.0-2.2 for cell debris. This is the first accurate experimentally-determined grade efficiency curve for cell debris. Previous studies have simply estimated debris grade efficiency curves using an approximate debris size distribution and grade efficiency curves determined with 'ideal particles' (e.g. spherical PVA particles). The findings of this study may be used to simulate and optimise the centrifugal fractionation of inclusion bodies from cell debris.

There are many methods for the analysis and design of embedded cantilever retaining walls. They involve various different simplifications of the pressure distribution to allow calculation of the limiting equilibrium retained height and the bending moment when the retained height is less than the limiting equilibrium value, i.e. the serviceability case. Recently, a new method for determining the serviceability earth pressure and bending moment has been proposed. This method makes an assumption defining the point of zero net pressure. This assumption implies that the passive pressure is not fully mobilised immediately below the excavation level. The finite element analyses presented in this paper examine the net pressure distribution on walls in which the retained height is less, than the limiting equilibrium value. The study shows that for all practical walls, the earth pressure distributions on the front and back of the wall are at their limit values, Kp and K-a respectively, when the lumped factor of safety F-r is less than or equal to2.0. A rectilinear net pressure distribution is proposed that is intuitively logical. It produces good predictions of the complete bending moment diagram for walls in the service configuration and the proposed method gives results that have excellent agreement with centrifuge model tests. The study shows that the method for determining the serviceability bending moment suggested by Padfield and Mair(1) in the CIRIA Report 104 gives excellent predictions of the maximum bending moment in practical cantilever walls. It provides the missing data that have been needed to verify and justify the CIRIA 104 method.

The exact description of the thermodynamics of solutions has been used to describe, without approximation, the distribution of all the components of an incompressible solution in a centrifuge cell at sedimentation equilibrium. Thermodynamic parameters describing the interactions between solute components of known molar mass can be obtained by direct analysis of the experimental data. Interpretation of the measured thermodynamic parameters in terms of molecular interactions requires that an arbitrary distinction be made between nonassociative forces, like hard-sphere volume-exclusion and mean-field electrostatic repulsion or attraction, and specific short-range forces of association that give rise to the formation of molecular aggregates. Provided the former can be accounted for adequately, the effects of the latter can be elucidated in the form of good estimates of the equilibrium constants for the reactions of aggregation.

O presente trabalho, elaborado entre Fevereiro e Julho de 2011, pretendeu analisar o impacto de algumas estratégias de optimização, adoptadas no âmbito da gestão de processo. As soluções estudadas tiveram como objectivo fazer face a alguns problemas de funcionamento da estação de tratamento de águas residuais (ETAR) de Crestuma, com recurso aos meios existentes. Para esse efeito foi realizada uma análise ao histórico dos registos de exploração referentes ao período de Janeiro de 2008 a Julho de 2011, pois foi considerado que este seria o período necessário para a análise e apuramento de resultados conclusivos. A ETAR fica situada na freguesia de Crestuma, actualmente com 2619 habitantes, localizada na confluência do regato de Vessadas com o rio Douro. Destina-se a tratar as águas residuais provenientes do sistema de drenagem de três localidades, S. Fioso, S. Picoto e S. Vessada, contribuindo para a despoluição do rio Douro. Genericamente, a ETAR tem apresentado um desempenho bastante satisfatório, respeitando os valores limites de emissão previstos na legislação actual (Decreto-Lei 152/97). A permanência de grandes quantidades de lama no processo, associada a uma elevada idade das lamas, está na origem da sua má sedimentabilidade, dificultando, por sua vez, a sua remoção do sistema. Com objectivo de se conseguir remover as lamas biológicas de uma forma mais eficiente, foi adoptada uma estratégia que compreendeu duas etapas. Com a finalidade de melhorar a sedimentabilidade, numa primeira etapa foi introduzido no processo um produto biológico, composto essencialmente por fungos, comercialmente conhecido por Optibiom 7450L. Com vista a obter-se uma maior eficiência na desidratação das lamas, a segunda etapa consistiu na substituição do polímero usado na lama espessada, por outro, de composição diferente, comercialmente conhecido por Easy 6040. Com as alterações introduzidas, foi possível obter um melhor controlo do processo e convergir para parâmetros de funcionamento mais próximos dos valores referidos na bibliografia. As alterações mais relevantes foram verificadas nos valores de sólidos suspensos totais (SST) no reactor biológico e na recirculação, tendo-se obtido valores médios de 3846 e 9716 mg/L, respectivamente (correspondendo a valores anteriores ao tratamento de 5301 e 7430 mg/L). Assim, a razão entre as concentrações de SST na corrente de recirculação e no reactor biológico passou de 1,4 para 2,6. A idade das lamas diminuiu de 42 para 36 dias com a adição do polímero Easy 6040 e o índice volumétrico de lamas apresentou uma redução, de 134 mL/g para 100 mL/g, o que se traduziu numa melhor sedimentabilidade das lamas. Desta forma, obteve-se uma diminuição de 33,3% no volume de lama produzida por m3 de afluente tratado, bem como uma redução de 27% no número de horas de funcionamento da centrífuga e um decréscimo ligeiro no consumo global de energia da ETAR (0,89%). Conclui-se que as alterações introduzidas no processo, adição de um produto biológico composto por fungos (Optibiom 7450L) e substituição do polímero usado na lama espessada por outro de composição diferente (Easy 6040), conduziram a uma melhoria no funcionamento do tratamento biológico da ETAR de Crestuma.

This research aimed to describe the frequency of parasites in stool samples in the environment of Ilha da Marambaia, Rio de Janeiro, Brazil. One hundred and five stool samples were collected and processed by the coproparasitological techniques ethyl acetate sedimentation and centrifuge-flotation using saturated sugar solution. Parasites were detected in 81.9% of the samples, hookworm being the most prevalent, followed by Trichuris vulpis. Ascaris sp. eggs were also found. A high level of evolutive forms of parasites with public health risk was found in stool samples of the environment studied. We propose that health education programs, allied to an improvement of human and animal health care, must be employed to reduce the environmental contamination.

This study aims to report the incidence of Calodium hepaticum among dogs and cats, pets or stray animals, captured by the Zoonosis Control Center (CCZ) in Lages, Santa Catarina, Brazil. Fecal samples from 108 pet dogs and eight pet cats, and from 357 stray dogs and 97 stray cats, captured by CCZ, were analyzed within the period from July 2010 to November 2012. Coproparasitological exams were performed by techniques of sedimentation, centrifuge-flotation, and simple flotation. Among 465 fecal samples from dogs and 105 from cats, the overall spurious infections for C. hepaticum eggs were 1.05%. For dogs, this positivity was 0.43% and for cats it was 3.81%. The two positive dogs were stray and out of the four cats, three were stray and one was a pet. Although the occurrence of C. hepaticum eggs was low, these data reveal the existence of infected rodents, especially in public places, since, out of the six infected animals, five (83.33%) were stray. These results are discussed and analyzed with an emphasis on the risk to public health.

El artículo describe los efectos de las aceleraciones positivas (+Gz) sobre el organismohumano. Se consideran los factores determinantes de las aceleraciones: intensidad, velocidad de comienzo, dirección y duración. La fisiopatología describe los factores hidrostáticos,hemodinámicos y de regulación refleja del aparato cardiocirculatorio. La exposición a +Gz produce reacciones adaptativas fisiológicas. Cuando éstas se superan, aparecen patologías de tipocardiocirculatorio, respiratorio, músculoesquelético, nervioso, sensorial,... Se describen los diferentes procedimientos y equipos específicos que protegen al piloto de los efectos de +Gz. Éstos incluyen las maniobras de contracción muscular, los trajes anti-G, los equipos de respiración a presión positiva, así como el entrenamiento en centrífuga.

Työn tavoitteena on kartoittaa yhdyskuntalietteen käsittelyä lietteenpolttolaitoksen tarpeita ajatellen. Lietteen käsittelytekniikoiden ja kuljetusvaihtoehtojen selvittäminen on siis työn keskeinen tavoite. Lisäksi otetaan selvää näiden tekijöiden kustannusrakenteesta. Yhdyskuntalietteen ominaisuuksien sekä käsittelyyn liittyvien ongelmakohtien valottaminen kuuluu samoin työn tavoitteisiin. Työssä tehdään muun ohella case-tarkastelua Kaakkois-Suomen alueeseen liittyen. Tavoitteena on muodostaa tarkoitukseen soveltuva lietteenkäsittelymalli kullekin tapaukselle. Työn alkuosassa tutustutaan yleisesti lietteeseen sekä polttoaineen että jätteen roolissa. Tarkastelu sisältää tietoja lietteen ominaisuuksista ja muodostuvista määristä sekä lietteenkäsittelyssä olennaisista lainsäädännöllisistä seikoista. Samoin katsastetaan hieman jäteve¬denpuhdistusprosessiin sekä näin ollen lietteen syntyyn. Lietteen esikäsittelyä, mekaanista vedenerotusta, termistä kuivausta ja polttoa tarkastellaan yleisessä valossa. Mekaanisen vedenerotuksen osalta myös eritellään ja vertaillaan laitteita. Etenkin linko, mutta myös suotonauhapuristin osoittautuivat erityisen sopiviksi kunnallisen lietteen käsittelyyn. Työn loppupuoliskolla kiinnitetään huomiota lietteen varastointiin sekä syöttö-ja purkumenetelmiin, lyhyen etäisyyden siirtoon ja pidemmän matkan kuljetukseen. Case-tapauksissa pohditaan Kymenlaakson ja Etelä-Karjalan paikallisia lietteenkäsittelymahdollisuuksia. Mekaanisesti kuivattua lietettä käsitellään kyseisissätapauksissa vuosittain 6000 t ja 15 000 t. Lietteen polton tuottama sähkö- ja lämpöteho näyttävät riippuvan voimakkaasti lietteen kuiva-ainepitoisuudesta, eivät niinkään lietteen muista ominaisuuksista. Lietteenkäsittelykustannukset tiivistetystä lietteestä termiseen kuivaukseen soveltuvaksi polttoaineeksi vaihtelevat10-20 \ lietetonnia kohden, riippuen käsittelyvaiheiden määrästä. Kustannuksia syntyy eniten mekaanisesta vedenerotuksesta ja varastoinnista.

Työn tavoitteena oli tutkia neljän eri koesuotimen soveltuvuutta pigmenttilietteen kiintoainepitoisuuden nostoon. Työssä käytetyt suotimet olivat Larox PF 0,1, Certus-CU-0047, Crossflow-koesuodin ja Steuerung ET06-linko. Suodinväliaineena Larox PF 0,1-suotimessa ja Crossflow-koesuotimessa käytettiin Tamfelt Oyj:n suodinkankaita. Certus-CU-0047-suotimessa suodinväliaineena toimi keraaminen membraani. Työn kirjallisuusosassa tarkasteltiin partikkelien karakterisointia, suodatuksen teoriaa, kakkusuodatusta, suodinväliaineista suodinkankaita ja membraaneja. Lisäksi tarkasteltiin paperin päällystämiseen käytettyjen pigmenttien ominaisuuksia ja niiden suodatusta. Kokeet suoritettiin Larox PF 0,1, Certus-CU-0047 ja Crossflow-koesuotimella vakiopainesuodatuksena yhdellä paineella. Steuerung ET06-lingon kokeet suoritettiin vakiovirtaussuodatuksena käyttäen kolmea eri virtausnopeutta ja kolmea eri lingon ja ruuvin kierrossuhdetta. Työssä suodatettiin neljää erilaista pigmenttilietettä. Suodoksista otettiin näytteitä niiden sameuden määrittämiseksi. Larox Pf 0,1-suotimella ja Steuerung ET06-dekantterilingolla saatiin nostettua pigmenttilietteen kiintoainepitoisuutta erittäin hyvin. Loput kaksi suodinta eivät sovellu tähän tarkoitukseen ollenkaan. Vakiopainesuodatuksissa suodatusajat muodostuivat kuitenkin liian pitkiksi. Suodoksien kiintoainepitoisuudet olivat suurimmaksi osaltaan pieniä lukuun ottamatta tiettyjä suodinkankaita. Lietteen 2 kiintoainepitoisuuden nostaminen oli kaikilla suotimilla erittäin vaikeaa, muut lietteet suodattuivat kohtuullisen hyvin. Näiden kokeiden perusteella voidaan sanoa, että varsinkin vakiovirtaussuodatus soveltuu hyvin ainakin tiettyjen pigmenttilietteiden kiintoainepitoisuuden nostoon.

Extracellular vesicles represent a rich source of novel biomarkers in the diagnosis and prognosis of disease. However, there is currently limited information elucidating the most efficient methods for obtaining high yields of pure exosomes, a subset of extracellular vesicles, from cell culture supernatant and complex biological fluids such as plasma. To this end, we comprehensively characterize a variety of exosome isolation protocols for their efficiency, yield and purity of isolated exosomes. Repeated ultracentrifugation steps can reduce the quality of exosome preparations leading to lower exosome yield. We show that concentration of cell culture conditioned media using ultrafiltration devices results in increased vesicle isolation when compared to traditional ultracentrifugation protocols. However, our data on using conditioned media isolated from the Non-Small-Cell Lung Cancer (NSCLC) SK-MES-1 cell line demonstrates that the choice of concentrating device can greatly impact the yield of isolated exosomes. We find that centrifuge-based concentrating methods are more appropriate than pressure-driven concentrating devices and allow the rapid isolation of exosomes from both NSCLC cell culture conditioned media and complex biological fluids. In fact to date, no protocol detailing exosome isolation utilizing current commercial methods from both cells and patient samples has been described. Utilizing tunable resistive pulse sensing and protein analysis, we provide a comparative analysis of 4 exosome isolation techniques, indicating their efficacy and preparation purity. Our results demonstrate that current precipitation protocols for the isolation of exosomes from cell culture conditioned media and plasma provide the least pure preparations of exosomes, whereas size exclusion isolation is comparable to density gradient purification of exosomes. We have identified current shortcomings in common extracellular vesicle isolation methods and provide a potential standardized method that is effective, reproducible and can be utilized for various starting materials. We believe this method will have extensive application in the growing field of extracellular vesicle research.

A simple preconcentration method of silicon based on coprecipitation with aluminum hydroxide prior to its flame atomic absorption (FAAS) determination was established. The recovery values of analyte ion was higher than 95%. The parameters including types of hydroxide ion source for precipitation, acid type for dissolution step, amount of aluminum ion as collector, pH, temperature, standing and centrifuge time, and sample volume were optimized for the quantitative recovery of the analyte. The influences of matrix ions were also examined. The relative standard deviation was found to be 3.2%. The limit of detection was calculated as (0.1 mg L-1). The preconcentration factor is 100 for (200 mL) solution. The proposed method was successfully applied for the determination of silicon in some water and alloy samples.

This work aimed to adapt the analysis of methemoglobin recommended by Evelyn - Malloy (visible spectrophotometry), in order to facilitate its application in the field, or to analysis in clinical laboratory, of existing sites of diflubenzuron application. The parameters changed included: centrifuge rotation speed; time between the collection of biological sample and analysis, and storage temperature of the samples; and the volume of reagents. The comparison of the rotation speed (rpm) of the reference methodology with the rpm of a "clinical centrifuge" did not reveal a statistically significant difference in the levels of methemoglobin. The time between the collection of biological sample and analysis was extended for a period of up to 48 hours for both conservation by refrigeration and ambient temperature, producing no statistically significant difference when compared to the standard duration of 2 hours. Regarding the reagents, the reference methodology already uses the volume necessary to ensure complete reaction, whereas a wider range from the recommended volume to a 5-fold reduction in comparison to the reference methodology could be used. It was concluded that the proposed changes to the methodology for adapting the analysis are applicable to studies of field / workplace exposure and ensure the reliability of results. The adapted methodology was inter-laboratory validated and the parameters changed can be selected according to the requirements of the laboratory at which the methemoglobin is to be measured.
