Los servicios portuarios son responsables de la mayor parte del coste que se produce en el paso de la mercancía por un puerto, particularmente el servicio de manipulación de mercancías. La prestación de éstos de forma fiable y eficiente resulta clave en un sector en el que existe una gran opacidad. Con el estudio realizado se dota a la Administración responsable, la Autoridad Portuaria, de una herramienta que le ayude a objetivar la toma de decisiones tanto a la hora de otorgar las preceptivas licencias como durante el periodo de prestación del servicio. Además se proponen una serie de medidas cuya aplicación mejoraría las condiciones de prestación del servicio así como una reducción de costes al paso de la mercancía por el puerto. Port services are responsible for most of the cost for the passing of goods through the port, especially the cargo handling service. Reliability and efficiency in the provision of them are key in a sector where there is a high opacity. With this study, we provide the responsible Administration, the Port Authority, with a tool that will help to objectify the decision making process, both at the time of granting the required licenses and during the period of the service provision. Also, proposes a number of measures whose implementation would improve the conditions of the service delivery and will reduce the total cost of transporting goods through the port.

El desarrollo de actividades de carga y descarga son parte de la esencia de la naturaleza funcional de un puerto, de las cuales derivan en gran medida los ingresos del mismo y la eficiencia de la cadena logística en su conjunto. Las oscilaciones en el interior de una dársena y en un línea de atraque disminuyen la calidad de la estancia de las embarcaciones en puerto, reducen el rendimiento de la estiba de los buques y solicitan y fatigan las estructuras y los cuerpos flotantes amarrados. Si los parámetros que definen la agitación local se aproximan a regiones de fallo 0 parada, el subsistema pierde rendimiento, fiabilidad y finalmente se paralizan las operaciones, produciéndose de este modo tiempos de inactividad. Estas paradas operativas conllevan pérdidas económicas para la terminal y, consecuentemente, para el puerto. Hoy día se dispone vastas redes de monitorización destinadas a la caracterización del medio físico en el entorno de los puertos. Paralelamente, las operaciones de manipulación de cargas en las terminales se están dirigiendo hacia modelos de automatización o semi automatización, que permiten no sólo la sistematización de procesos, sino también un profundo conocimiento del flujo de tareas. En este contexto hay un déficit de información sobre cómo afectan los diferentes forzadores del medio físico al rendimiento, la seguridad funcionalidad del proceso de manipulación de carga y descarga. Esto se debe en gran medida a la falta de registros dilatados en el tiempo que permitan correlacionar todos los aspectos mencionados de un modo particularizado para cada línea de atraque y amarre de un puerto. En esta tesis se desarrolla una metodología de vídeo monitorización no intrusiva y de bajo coste basada en la aplicación de técnicas "pixel tool' y la obtención de los parámetros extrínsecos de una observación monofocal. Con ello pretende poner en valor las infraestructuras de vídeo vigilancia de los puertos y de los laboratorios de experimentación a escala reducida, con el objeto de facilitar el estudio los umbrales operativos de las áreas de atraque y amarre. The development of loading and unloading activities is an essential part of he functional nature of a port, which derive largely from he same income and the efficiency of he supply chain as a whole. The oscillations inside a dock and a mooring line diminish he quality of the stay of vessels in port reducing the performance of the stowage of ship and asking and fatigued structures and moored floating bodies. If the parameters defining the local al agitation regions are close to areas of failure or shutdown, he subsystem looses performance, reliability and eventually paralyzes the operations, thereby producing downtime. These operational stops entail economic 1osses to the terminal and, consequently for the port. Today vast networks of monitoring, aimed at he characterization of the physical environment in the vicinity of he ports, are available. In parallel, the cargo handling operations at terminals are moving towards automation or semi-automation models that allow not only the systematization of processes, but also a deep understanding of he workflow. In this context, there is a lack of information about how the different forcing agents of the physical environment affect the performance and he functional safety of the loading and unloading process. This is due largely to the lack of spread-over-time records which would allow to correlate all aspects mentioned, specifically, for each berthing and mooring of a port. This thesis develops a methodology for non-intrusive and low cost monitoring video based on the application of "pixel tool" techniques and on obtaining the extrinsic parameters of a monofocal observation. It seeks an enhancement of the video monitoring infrastructure at ports and at experimental laboratories of reduced scale, in order to facilitate the study of operational thresholds berthing and mooring areas.

Die hohen Leistungsansprüche an moderne Lagersysteme führen zu immer kürzeren Taktzeiten beim Warenumschlag, die auch von Regalbediengeräten (RBG) realisiert werden müssen. Um trotz der kurzen Umschlagzeiten eine schnelle und sichere Gutübergabe zu gewährleisten, muss innerhalb der Ausschwingzeit die Amplitude der Schwingungen des Lastaufnahmemittels vor dem Ein- beziehungsweise Auslagerungsvorgang weitgehend abgeklungen sein. Diese Wartezeiten beeinflussen die Taktzeit und damit die Umschlagleistung des RBG negativ. Ein möglicher Ansatz, die Ausschwingzeiten zu verkürzen, ist der Einsatz hochdämpfender Materialien, wie etwa Verbundwerkstoffe. Durch die erreichte Reduzierung der Schwingungsamplituden bei gleichzeitiger Erhöhung des Leichtbaugrades ist eine deutliche Effizienzsteigerung von RBG zu erwarten.

Pancreatic β-cells are highly sensitive to suboptimal or excess nutrients, as occurs in protein-malnutrition and obesity. Taurine (Tau) improves insulin secretion in response to nutrients and depolarizing agents. Here, we assessed the expression and function of Cav and KATP channels in islets from malnourished mice fed on a high-fat diet (HFD) and supplemented with Tau. Weaned mice received a normal (C) or a low-protein diet (R) for 6 weeks. Half of each group were fed a HFD for 8 weeks without (CH, RH) or with 5% Tau since weaning (CHT, RHT). Isolated islets from R mice showed lower insulin release with glucose and depolarizing stimuli. In CH islets, insulin secretion was increased and this was associated with enhanced KATP inhibition and Cav activity. RH islets secreted less insulin at high K(+) concentration and showed enhanced KATP activity. Tau supplementation normalized K(+)-induced secretion and enhanced glucose-induced Ca(2+) influx in RHT islets. R islets presented lower Ca(2+) influx in response to tolbutamide, and higher protein content and activity of the Kir6.2 subunit of the KATP. Tau increased the protein content of the α1.2 subunit of the Cav channels and the SNARE proteins SNAP-25 and Synt-1 in CHT islets, whereas in RHT, Kir6.2 and Synt-1 proteins were increased. In conclusion, impaired islet function in R islets is related to higher content and activity of the KATP channels. Tau treatment enhanced RHT islet secretory capacity by improving the protein expression and inhibition of the KATP channels and enhancing Synt-1 islet content.

Stingless bees collect plant resins and make it into propolis, although they have a wider range of use for this material than do honey bees (Apis spp.). Plebeia spp. workers employ propolis mixed with wax (cerumen) for constructing and sealing nest structures, while they use viscous (sticky) propolis for defense by applying it onto their enemies. Isolated viscous propolis deposits are permanently maintained at the interior of their colonies, as also seen in other Meliponini species. Newly-emerged Plebeia emerina (Friese) workers were observed stuck to and unable to escape these viscous propolis stores. We examined the division of labor involved in propolis manipulation, by observing marked bees of known age in four colonies of P. emerina from southern Brazil. Activities on brood combs, the nest involucrum and food pots were observed from the first day of life of the marked bees. However, work on viscous propolis deposits did not begin until the 13th day of age and continued until the 56th day (maximum lifespan in our sample). Although worker bees begin to manipulate cerumen early, they seem to be unable to handle viscous propolis till they become older.

Aims: The clinical benefits of angiotensin II type 1 (AT1) receptor blockers (ARB) in heart failure (HF) include cardiac anti-remodeling and improved ventricular function. However, the cellular mechanisms underlying the benefits of ARB on ventricular function need to be better clarified. In the present manuscript, we evaluated the effects of AT1 receptor blockade on the net balance of Ca(2+) handling proteins in hearts of mice lacking alpha(2A) and alpha(2C) adrenoceptors (alpha(2A)/alpha(2C)ARKO), which develop sympathetic hyperactivity (SH) induced-HF. Main methods: A cohort of male wild-type (WT) and congenic alpha(2A)/alpha(2C)ARKO mice in a C57BL6/J genetic background (5-7 mo of age) was randomly assigned to receive either placebo or ARB (Losartan, 10 mg/kg for 8wks). Ventricular function (VF) was assessed by echocardiography, and cardiac myocyte width and ventricular fibrosis by a computer-assisted morphometric system. Sarcoplasmic reticulum Ca(2+) ATPase (SERCA2), phospholamban (PLN), phospho-Ser(16)-PLN, phospho-Thr(17)-PLN, phosphatase 1 (PP1), Na(+)-Ca(2+) exchanger (NCX), Ca(2+)/calmodulin-dependent protein kinase 11 (CaMKII) and phospho-Thr(286)-CaMKII were analyzed by Western blot. Key findings: alpha(2A)/alpha(2C)ARKO mice displayed ventricular dysfunction, cardiomyocyte hypertrophy and cardiac fibrosis paralleled by decreased SERCA2 and increased phospho-Thr(17)-PLN, CaMKII, phospho-Thr(286)-CaMKII and NCX levels. ARB induced anti-cardiac remodeling effect and improved VF in alpha(2A)/alpha(2C)ARKO associated with increased SERCA2 and phospho-Ser(16)-PLN levels, and SERCA2:NCX ratio. Additionally, ARB decreased phospho-Thr(17)-PLN levels as well as reestablished NCX, CaMKII and phospho-Thr(286)-CaMKII toward WT levels. Significance: Altogether, these data provide new insights on intracellular Ca(2+) regulatory mechanisms underlying improved ventricular function by ARB therapy in HF. (c) 2011 Elsevier Inc. Previous studies show that exercise training and caloric restriction improve cardiac function in obesity. However, the molecular mechanisms underlying this effect on cardiac function remain unknown. Thus, we studied the effect of exercise training and/or caloric restriction on cardiac function and Ca(2+) handling protein expression in obese rats. To accomplish this goal, male rats fed with a high-fat and sucrose diet for 25 weeks were randomly assigned into 4 groups: high-fat and sucrose diet, high-fat and sucrose diet and exercise training, caloric restriction, and exercise training and caloric restriction. An additional lean group was studied. The study was conducted for 10 weeks. Cardiac function was evaluated by echocardiography and Ca(2+) handling protein expression by Western blotting. Our results showed that visceral fat mass, circulating leptin, epinephrine, and norepinephrine levels were higher in rats on the high-fat and sucrose diet compared with the lean rats. Cardiac nitrate levels, reduced/oxidized glutathione, left ventricular fractional shortening, and protein expression of phosphorylated Ser(2808)-ryanodine receptor and Thr(17-)phospholamban were lower in rats on the high-fat and sucrose diet compared with lean rats. Exercise training and/or caloric restriction prevented increases in visceral fat mass, circulating leptin, epinephrine, and norepinephrine levels and prevented reduction in cardiac nitrate levels and reduced: oxidized glutathione ratio. Exercise training and/or caloric restriction prevented reduction in left ventricular fractional shortening and in phosphorylation of the Ser(2808)-ryanodine receptor and Thr(17)-phospholamban. These findings show that exercise training and/or caloric restriction prevent cardiac dysfunction in high-fat and sucrose diet rats, which seems to be attributed to decreased circulating neurohormone levels. In addition, this nonpharmacological paradigm prevents a reduction in the Ser(2808)-ryanodine receptor and Thr(17-)phospholamban phosphorylation and redox status. (Hypertension. 2010;56:629-635.)

We investigated potential Ca(2+) abnormalities in skeletal muscles comprising different fiber compositions and investigated whether aerobic exercise training would improve muscle function in a genetic model of sympathetic hyperactivity-induced HF. A cohort of male 5-mo-old wild-type (WT) and congenic alpha(2A)/alpha(2C) adrenoceptor knockout (ARKO) mice in a C57BL/6J genetic background were randomly assigned into untrained and trained groups. Exercise training consisted of a 8-wk running session of 60 min, 5 days/wk (from 5 to 7 mo of age). After completion of the exercise training protocol, exercise tolerance was determined by graded treadmill exercise test, muscle function test by Rotarod, ambulation and resistance to inclination tests, cardiac function by echocardiography, and Ca(2+) handling-related protein expression by Western blot. alpha(2A)/alpha(2C)ARKO mice displayed decreased ventricular function, exercise intolerance, and muscle weakness paralleled by decreased expression of sarcoplasmic Ca(2+) release-related proteins [alpha(1)-, alpha(2)-, and beta(1)-subunits of dihydropyridine receptor (DHPR) and ryanodine receptor (RyR)] and Ca(2+) reuptake-related proteins [sarco(endo) plasmic reticulum Ca(2+)-ATPase (SERCA) 1/2 and Na(+)/Ca(2+) exchanger (NCX)] in soleus and plantaris. Aerobic exercise training significantly improved exercise tolerance and muscle function and reestablished the expression of proteins involved in sarcoplasmic Ca(2+) handling toward WT levels. We provide evidence that Ca(2+) handling-related protein expression is decreased in this HF model and that exercise training improves skeletal muscle function associated with changes in the net balance of skeletal muscle Ca(2+) handling proteins.
