The paper presents bighead carp broodstock management, an integral part of hatchery techniques as it will determine the quality of resulting fry and fingerlings. Proper methods on the selection and management of broodstock are discussed.

As a prelude to strain selection for domestication and future marker assisted selection, genetic variation revealed by microsatellite DNA was evaluated in yellow perch, Perca flavescens, from four wild North American populations collected in 2003-2004 (Maine, New York, North Carolina, and Pennsylvania,), and two captive populations (Michigan and Ohio). For the loci examined, levels of heterozygosity ranged from H-e=0.04 to 0.88, genetic differentiation was highly significant among all population pairs, and effective migration ranged from low (N(e)m=0.3) to high (N(e)m=4.5). Deviation from Hardy-Weinberg equilibrium was regularly observed indicating significant departures from random mating. Instantaneous measures of inbreeding within these populations ranged from near zero to moderate (median F=0.16) and overall inbreeding levels averaged F-IS=0.18. Estimates of genetic diversity, Phi(ST), and genetic distance were highest between Michigan and all other broodstock groups and lowest between New York and Ohio. Genetic differentiation among groups did not correlate with geographic distance. Overall, the patterns of variation exhibited by the captive (Michigan and Ohio) populations were similar to patterns exhibited by the other wild populations, indicating that spawning and management practices to date have not significantly reduced levels of genetic variation. A stressful environment induces cortisol that might affect fish breeding and reproduction. In the present work, which aimed to mimic aquacultural conditions of the jundia (Rhamdia quelen) hatcheries in southern South America, females were submitted to normal or stressful handling and the effects of cortisol on serum levels of 17β-estradiol (E2) and testosterone (T) were determined. In addition, the effect of stress on reproductive parameters such as eggs and swim-up fry production was also measured. Eight females from a group submitted to stressful handling (SH) conditions and eight females from a group with normal handling (NH) conditions were captured for blood sampling at D 0 and at D 1, 10, 20, 30, and 40 of the experiment. A typical cortisol response was observed in the SH females group in that they presented higher cortisol level in contrast to the NH female group, in all days sampled, except at D 0. In the 10th and 20th d, the E2 levels were lower in SH females, but cortisol levels were higher, suggesting an effect of cortisol on E2 production and/or release. Stressful handling appeared to affect both the number and the quality of the gametes because a lower number of oocytes was stripped from SH females, and from SH fertilized eggs, a lower number of viable swim-up fry was obtained to be transferred to earthen larviculture ponds. Taken together, the results indicated that stressful handling of broodstock impairs R. quelen reproduction. © Copyright by the World Aquaculture Society 2008.

Herein, we have developed molecular markers for nuclear genes to use in multiplex-PCR and PCR-RFLP, with the goal of characterising hybrid lines derived from crosses between pintado Pseudoplatystoma corruscans and cachara P. reticulatum. These markers, together with others described previously, were used to perform molecular identification analyses as genetic subsidies for Brazilian aquaculture. These analyses were performed due to the problems of high mortality in the offspring reported by the aquaculturist. From a total of 16 broodstock samples, 13 were genetically identified as hybrids; surprisingly, nine of these hybrids were found to be post-F1 lineages. These data show that the fertility of these animals can seriously affect the cultivated stocks, thus causing financial damage in this aquaculture system. The establishment of PCR-RFLP and multiplex-PCR as molecular techniques allows for both the correct management of these animals and the routine monitoring of production and trade of fish hybrids in aquaculture. Consequently, such tools will enable a sustainable development in the aquaculture industry. © 2012 Blackwell Publishing Ltd.

The effects of dietary lipid levels in the spawning quality has been studied in several cultured fish species. Works like those of Watanabe et al. (1984 a); Mourente et al. (1989); Dhert et al. (1991); Bruce et al. (1993); Navas et al. (1997); Rodriguez et al. (1998); Lavens et al. (1999); Furuita et al. (2002, 2003 b); Mazorra et al. (2003); Fernandez- Palacios (2005) and Aijun et al. (2005) show that lipids and fatty acids are the dietetic components that have more influence in the spawning quality, specially in those species with continuous spawning which display short vitellogenesis periods and are able to incorporate these dietetic components in eggs during the spawning period. Diets for gilthead sea bream (Sparus aurata) broodstock with dietary levels of 2.84% n-3 HUFA, combined with levels of 250 mg/kg vitamine E rasure good spawning quality. Putting so indicative the importance for an effective utilization of essential fatty acids the use of adequate levels of antioxidants.

[EN]The present study aimed to determine the spawning efficacy, egg quality and quantity of captive breed meagre induced with a single gonadotrophin-releasing hormone agonist (GnRHa) injection of 0, 1, 5, 10, 15, 20, 25, 30, 40 or 50 μg kg–1 to determine a recommended optimum dose to induce spawning. The doses 10, 15 and 20 μg kg–1 gave eggs with the highest quality (measured as: percentage of viability, floating, fertilisation and hatch) and quantity (measured as: total number of eggs, number of viable eggs, number of floating eggs, number of hatched larvae and number of larvae that reabsorbed the yolk sac). All egg quantity parameters were described by Gaussian regression analysis with R2 = 0.89 or R2 = 0.88. The Gaussian regression analysis identified that the optimal dose used was 15 μg kg–1. The regression analysis highlighted that this comprehensive study examined doses that ranged from low doses insufficient to stimulate a high spawning response (significantly lower egg quantities, p < 0.05) compared to 15 μg kg–1 through to high doses that stimulated the spawning of significantly lower egg quantities and eggs with significantly lower quality (egg viability). In addition, the latency period (time from hormone application to spawning) decreased with increasing doses to give a regression (R2 = 0.93), which suggests that higher doses accelerated oocyte development that in turn reduced egg quality and quantity. The identification of an optimal dose for the spawning of meagre, which has high aquaculture potential, represents an important advance for the Mediterranean aquaculture industry.
