Background: Peroxiredoxins have diverse functions in cellular defense-signaling pathways. 2-Cys-peroxiredoxins (2-Cys-Prx) reduce H2O2 and alkyl-hydroperoxide. This study describes the purification and characterization of a genuine 2-Cys-Prx from Vigna unguiculata (Vu-2-Cys-Prx). Methods: Vu-2-Cys-Prx was purified from leaves by ammonium sulfate fractionation, chitin affinity and ion exchange chromatography. Results: Vu-2-Cys-Prx reduces H2O2 using NADPH and DTT. Vu-2-Cys-Prx is a 44 kDa (SDS-PAGE)/46 kDa (exclusion chromatography) protein that appears as a 22 kDa molecule under reducing conditions, indicating that it is a homodimer linked intermolecularly by disulfide bonds and has a pI range of 4.56-4.72; its NH2-terminal sequence was similar to 2-Cys-Prx from Phaseolus vulgaris (96%) and Populus tricocarpa (96%). Analysis by ESI-Q-TOF MS/MS showed a molecular mass/pI of 28.622 kDa/5.18. Vu-2-Cys-Prx has 8% alpha-helix, 39% beta-sheet, 22% of turns and 31% of unordered forms. Vu-2-Cys-Prx was heat stable, has optimal activity at pH 7.0, and prevented plasmid DNA degradation. Atomic force microscopy shows that Vu-2-Cys-Prx oligomerized in decamers which might be associated with its molecular chaperone activity that prevented denaturation of insulin and citrate synthase. Its cDNA analysis showed that the redox-active Cys(52) residue and the amino acids Pro(45), Thr(49) and Arg(128) are conserved as in other 2-Cys-Prx. General significance: The biochemical and molecular features of Vu-2-Cys-Prx are similar to other members of 2-Cys-Prx family. To date, only one publication reported on the purification of native 2-Cys-Prx from leaves and the subsequent analysis by N-terminal Edman sequencing, which is crucial for construction of stromal recombinant 2-Cys-Prx proteins. The cowpea (Vigna unguiculata L. Walp) is a major food crops in northeastern Brazil. In Rio Grande do Norte, the cowpea, vigna beans or cowpea, as it is known, has great socioeconomic importance as a source of nutrients in food, with great emphasis among agricultural products. To improve productivity and resistance to pests, two cultivars were developed exclusively by EMPARN (Agricultural Research Corporation of Rio Grande do Norte), for breeding. The samples were provided by EMPARN, two improved (Potiguar and Laugh year) and two landraces (Rib of beef and Canapu). The seeds were ground and made into flour samples and the determination of moisture and ash by graviméticos methods, lipids by Soxhlet extraction, fibers with determiner fiber, carbohydrates by difference and minerals by ICP-OES were performed except the match analyzed by UVvis. The results showed a high fiber content (55.55% and 55.32% and 50.04% improved samples and 50.32% creole samples) and protein (25.52% and 25.27% improved and 27 samples, 23% and 24.99% creole samples). Comparing the results of the mineral content, the improved cultivars stood out in relation to Ca, Co, P, Mg, Mo and Na. Creole cultivars showed better results for Cu, Cr (low), Fe, Mn, Ni, K and Zn. The results will be important in future nutritional research and to build a table of Brazilian chemical composition of foods.

O feijão-de-metro é um importante alimento, rico em proteína, vitaminas, fibras e minerais. Muito consumido na região Norte do país, principalmente no estado do Pará, onde é conhecido como feijão-verde. Os materiais utilizados atualmente são os mesmos introduzidos no início do século passado. São comercializadas sementes de uma única cultivar registrada no Registro Nacional de Cultivares (RNC) e de outra que não tem registro. Este trabalho foi realizado com o objetivo de avaliar linhagens de feijão-de-metro em Be©m-PA. Foram avaliados 66 acessos no delineamento de blocos de Federer, com duas testemunhas comuns. O experimento foi conduzido na Embrapa Amazônia Oriental, em delineamento de Blocos Federer. Foram avaliados caracteres qualitativos e quantitativos. Com base na produtividade e nos caracteres cor, comprimento e aspecto geral da vagem, importantes para a aceitação comercial, destacaram-se com produtividade superior à 1,6 kg/2m2 os seguintes acessos: China town (PI 487501), TVO 2141 (MNC 1618), CP-74 (PI 487550), Dagupan pangasinan (PI 487486).

O caupi (Vigna unguiculata (L.) Walp) é um alimento básico das populações do Nordeste brasileiro, devendo merecer atenção com vistas a melhoria da qualidade de grãos, resistência a doenças e pragas e aumento de produtividade. Este trabalho teve por objetivo estudar a variabilidade e o potencial genético de 28 linhagens, escolhidas após uma seleção para cor, tamanho de grãos e resistência a viroses. A produtividade apresentou coeficiente de variação genético de 23,90%, e o valor agronômico, de 3,56%. O número de vagens por pedúnculo apresentou a menor estimativa do coeficiente de determinação genético (4,51%), e o peso de 100 grãos, a maior (81,74%). O coeficiente de determinação genético da produtividade foi de 34,15%. As maiores estimativas de ganho genético foram as do peso de 100 grãos (21,73%) e da produtividade (19,77%). As correlações genotípicas foram superiores às fenotípicas e às de ambiente, destacando-se as correlações entre número de ramos secundários e produtividade (68,13%), e valor agronômico e produtividade (100%). Estes resultados mostram amplas possibilidades de seleção entre as linhagens com relação à maioria dos caracteres estudados.

Estudou-se o nivel de dano economico de Empoasca Kraemeri Ross & Moore em Vigna unguiculata (L.), em condicoes de campo, em Petrolina, Pernambuco. Neste estudo, correlacionou-se a percentagem de reducao de produtividade de V. unguiculata com o nivel medio de infestacao de E. Kraemeri, no periodo e 20 a 60 dias apos o plantio.

This study compared the effect of supplementing maize stover (MS) with cowpea (Vigna unguiculata) haulms or commercial concentrate (CC) on feed intake, nutrient digestibility, live weight gain and carcass yield of male Ethiopian Highland sheep. Two cowpea genotypes, 12688 (forage) and IT96D-774 (dual-purpose), were used. A randomised block design was applied with groups of eight sheep, blocked by weight, allocated to one of six treatments; MS ad libitum either unsupplemented or supplemented daily with 150 or 300g dry matter (DM) of either cowpea or CC. MS contained more neutral detergent fibre (NDF), acid detergent fibre (ADF) and lignin than either cowpeas or CC Crude protein (CP) content of the forage-type cowpeas was higher than either dual-purpose or CC, while MS had the lowest CP content Relative to the negative control group, cowpea at either level significantly (P < 0.01) increased both MS intake and total NDF and lignin. Supplementation significantly (P < 0.01) increased nitrogen (N) intakes relative to the negative control, with N intake for CC and dual-purpose cowpea (high level) being similar to the intakes for cowpeas at 150g. N intake with the forage-type cowpea offered at higher levels was significantly (P < 0.01) greater than the other groups. No significant differences (P > 0.01) in MS intake were identified between cowpeas at either level or CC and, although intake level of CC increased, it did not differ significantly from the negative control group. Supplementation significantly (P < 0.01) improved average daily gain, with the negative control group losing weight over the experimental period, and increased final live weight, carcass cold weight and dressing percentage. Supplementation significantly improved the apparent digestibility of DM, organic matter and NDF, with no significant difference found between cowpeas at either level. N retention was negative for sheep offered only MS, but positive with all supplements, with cowpeas improving N retention to a greater extent than CC. Interestingly, N retention/N intake was higher with cowpeas offered at the lower level suggesting an improvement in utilisation efficiency. The results indicate that the supplementation of MS with cowpea enhanced ruminant production through improvements in digestibility and intake. Further, as production improvements associated with the two levels of supplementation did not differ significantly, it is suggested that where limited quantities of cowpea are available, it may be of greater nutritional benefit to offer smaller quantities over an increased number of animal days.

Seeds sprouts have been used as a good source of basic nutrients and nutraceutical compounds. The high nutritional value of seeds derives from the deposition of compounds during development. However some of these molecules are used in metabolic processes like germination, which leads to a considerable variation in their concentrations once these events are completed. In this work, we investigate the levels of inositols (myo-inositol, D-pinitol and ononitol), soluble carbohydrates and proteins in cotyledons of Phaseolus vulgaris and Vigna unguiculata sprouts. Sprouting increased myo-inositol and glucose content and reduction of raffinose and ononitol was observed. The protein levels increased in P. vulgaris and decreased in V. unguiculata sprouting. The level of sucrose was maintained in both sprouts. D-Pinitol was detected only in quiescent seeds. Our results suggested that bean sprout is an important source of proteins, sucrose, glucose and myo-inositol. Additionally, bean sprouts have low levels of raffinose, an antinutritional compound.
