The mechanisms of action of Cu 2+ in improving the longevity of cut flowers and foliage have not been elucidated. Possible antimicrobial action of Cu 2+ against stem end and vase solution colonising bacteria was investigated using Cu 2+ treatments optimised for cut Acacia holosericea A. Cunn. ex G. Don foliage stems. These treatments were a 5h pulse with 2.2mM Cu 2+ or a 0.5mM Cu 2+ vase solution versus a deionised water (no Cu 2+) control. Bacterial growth over time was assessed by a standard plate count agar technique and with scanning electron microscopy. Cu 2+ treatments significantly extended the cut foliage vase life. However, they did not have sustained antibacterial activity against stem end or vase solution colonising bacteria. Also, regular recutting of 1-2cm from the stem ends did not substantially improve either cut stem water relations or longevity. The positive effects of Cu 2+ treatments were unaffected by the repeated stem end recutting. It was concluded that the primary mechanism of Cu 2+ was not antibacterial. Moreover, naturally growing vase solution and stem end microbial populations had relatively insignificant effects on cut A. holosericea vase life. Research into alternative mechanisms of Cu 2+ is required. © 2012 Elsevier B.V.

Microbes and their exopolysaccharides (EPS) can block xylem vessels, thereby increasing the hydraulic resistance and decreasing the vase life of cut flowers and foliage. Scanning electron microscopy (SEM) provides a powerful tool for investigation of bacteria-induced xylem occlusion. However, conventional preparation protocols for SEM involving chemicals can cause loss of hydrated EPS material, and thereby damage the bacterial biofilms during dehydration. A modified chemical fixation protocol involving pre-fixation with 75 mM lysine plus 2.5% glutaraldehyde followed by the normal fixation in 3% glutaraldehyde was, therefore, tested for improved preservation of bacterial biofilm at the stem-ends of cut Acacia holosericea foliage stems. Stem-end segments with different stages of bacterial growth were obtained from stems stood into water. The lysine-based protocol was compared with four other processing protocols of critical point drying (CPD) without fixation (control), freeze-drying (FD), conventional chemical fixation followed by drying with hexamethyldisilazane (HMDS), and conventional chemical fixation with CPD. The non-fixed control. FD and the glutaraldehyde fixation with HMDS drying gave poor preservation of hydrated material, including bacterial EPS. Conventional glutaraldehyde fixation followed by CPD was superior to these three methods in terms of better preserving the EPS. However, this fourth method gave condensation of biofilms during dehydration. In contrast, the modified lysine-based protocol resulted in superior preservation of EPS and biofilm structure. Thus, this fifth method was the most appropriate for examination of bacterial stem-end blockage in cut ornamentals. (C) 2012 Elsevier B.V. All rights reserved.

Ptilotus nobilis (Lindl.) F. Muell. has potential in the floriculture industries as a cut flower crop. Ethylene production and respiration rates, fresh weight changes and volatile scent production from cut inflorescences of P. nobilis cultivars Passion (dark pink flowers) and Purity (white-green flowers) were measured during vase life. Inflorescence weight loss was significant (P<0.001) during vase life with wilting and colour loss being the primary reasons for loss of vase life. Inflorescences ready for the cut market stored and at 22 degrees C had vase lives of >12 d. Ethylene production by inflorescences was low to negligible. Treatment with silverthiosulphate (STS) and ethylene had no effects on vase life. Evidently, ethylene did not play a role in determining the postharvest longevity of cut P. nobilis flowers. Respiration rates of inflorescences were high at harvest (>700 mg CO2 kg(-1) FW h(-1)) and declined gradually there-after during vase life. Total volatile emissions followed a similar pattern. For Passion, respiration rates of immature florets were significantly greater (P=0.02) than florets from other developmental stages while the calyx produced the most CO2. For Purity, respiration rates of florets of different maturities did not differ and the reproductive tissue produced the most CO2. Only fully opened mature florets with their stigma and anthers revealed, emitted significant quantities of volatiles (P<0.001) and primarily from the calyx tissue for both cultivars. The individual volatiles differed somewhat for the two cultivars. However, both produced significant quantities of benzaldehyde, 3,5-dimethoxytoluene and benzyl alcohol. Many authors have noted that consumer confidence in buying fresh flowers is strongly related to their perceived value in that quality and vase life must be high and consistent over time for consumers to repeat buy. Growers, wholesalers, exporters and retailers seek practical information about recommended handling and treatments at the harvest and postharvest stages, including that relating to flowers native to Australia and South Africa ("wildflowers"). This information is essential for products to be of high quality with an acceptable vase life for the end consumer, especially if exported. Published postharvest manuals generally focus on traditional flower crops and so rarely include many, or any, wildflowers. A manual entitled Postharvest Handling of Australian flowers from Native Plants and Related Species was published in 2002 and addressed this gap, but required updating. This situation presented an opportunity to provide in-depth information to compliment the Australian wildflower quality specifications (see accompanying paper in the same volume), and to assemble the latest knowledge on wildflower quality and postharvest issues. The resultant manual contains extensive information about harvesting, quality issues and recommended postharvest care focussed on wildflowers. Much of the information is documented for the first time, being based on the most up to date research and development (R&D) as well as practical experience of the floral supply chain, researchers and other technical experts. The manual provides practical and detailed information on postharvest treatment of fresh wildflowers for growers, florists, wholesalers and exporters to use on a daily basis. It discusses the many unique features of wildflowers that must be understood and managed in order to maximise their quality and vase life after marketing and export. The manual also includes postharvest advice for 16 flower- and foliage lines for which quality specifications were not produced. This advice is presented according to the same template as the specifications.

Realizou-se pesquisa objetivando determinar o melhor entre quatro pontos de colheita da haste floral de crisântemos de maço do tipo pompom cv. White Polaris a concentração mais adequada de sacarose para tratamento de pulsing dessas hastes: estas foram colhidas em estufa de produção comercial, nos pontos preestabelecidos e transportadas para laboratório, onde foram totalmente imersas em água de torneira, à sombra, durante três horas. Selecionaram-se as hastes pela uniformidade do seu desenvolvimento e cortaram-nas sob água na base do caule, entre 50 e 60 cm, e identificadas, o que permitiu avaliar as mudanças morfológicas associadas às inflorescências individuais. As hastes foram distribuídas e mantidas nos diferentes tratamentos de pulsing durante 24 horas, à luz branca contínua de 1.500 lux, 60 a 90% de umidade relativa do ar e temperatura ambiente de 25 ± 2°C. Após o tratamento de pulsing, as hastes foram transferidas para água destilada, permanecendo por 10 horas sob luz branca contínua, nas mesmas condições de laboratório citadas. A vida floral em vaso começou a ser avaliada na instalação do experimento, após o tratamento de pulsing, e terminou quando as folhas e pétalas perderam a turgescência e o valor decorativo. As hastes colhidas em estádio de botão (25 e 50% de abertura das inflorescências apicais) não alcançaram o ponto de abertura adequado em nenhuma das seis concentrações de sacarose (0 a 146,07 mol/m³); as concentrações de 116,9 146,1 mol/m³, porém, estimularam, em geral, a abertura de botões. Os pontos de colheita 1 e 2 (100 e 75% de abertura das inflorescências apicais respectivamente) apresentaram bons resultados em todas as concentrações de sacarose.

0 objetivo do trabalho foi determinar o melhor tratamento pós-colheita para manutenção floral e aumento da longevidade de crisântemo de maço do tipo pompom (Dendranthema grandiflorum (Ramat.) S. Kitamura) cv. White Polaris. Estabeleceu-se como ponto de colheita o momento em que as hastes apresentavam três inflorescências apicais com as pétalas externas em ângulo de 45° em relação à horizontal. Durante o ensaio em laboratório, as hastes, colhidas em estufa de produção comercial, após totalmente imersas em água de torneira, à sombra, durante três horas, foram cortadas sob água na base do caule entre 50 e 60 cm. As hastes foram distribuídas nos diferentes tratamentos de pulsing durante 24 horas, com luz contínua de 1.500 lux, 60 a 90% de umidade relativa do ar e temperatura ambiente de 25 ± 2°C. No primeiro experimento, testou-se a eficiência de 8-hidroxiquinolina (8-HQ) e tiabendazole (TBZ) como germicidas de manutenção da qualidade na solução de pulsing; testaram-se, também, dois reguladores de crescimento, a saber: ácido giberélico (GA3), 6-benzilaminopurina (6-BA) ou a mistura dos dois, com o objetivo de preservar a cor e a turgidez da folhagem. Os melhores resultados foram com 8-HQ (0,69 mol/m³) e GA3 (0,058 mo1/m³). No segundo experimento, avaliaram-se os seguintes inibidores de etileno: tiossulfato de prata (STS), nitrato de prata (AgNO3) e cloreto de cobalto (COC1(2)). A melhor resposta foi obtida com AgNO3 (2,9 e 4,4 mo1/m³).

O objetivo deste trabalho foi estudar alguns aspectos da fisiologia pós-colheita de inflorescências de sorvetão cultivadas no Submédio São Francisco. Hastes florais recém-colhidas foram submetidas a diferentes tratamentos (água destilada; 75mg L-1 de nitrato de prata - AgNO3; 1000mg L-1 de cloreto de cobalto - CoCl2; 5mg L-1 de ácido giberélico - GA3 - Progibb® e 10mg L-1 de 6-Benzilaminopurina - BAP), em ambiente com temperatura e umidade controlada por 15 dias. A vida pós-colheita foi acompanhada a partir da escala de notas, da massa da matéria fresca e do consumo da solução conservante. O tratamento com AgNO3 em hastes de sorvetão, foi o mais eficiente na manutenção da vida de vaso de sorvetão, porém, devido a sua toxidez, recomenda-se o uso de GA3.

A durabilidade e a qualidade pós-colheita de flores de corte são atributos fundamentais na sua valoração ao longo da cadeia produtiva e na satisfação dos consumidores. Objetivou-se, nesta pesquisa, verificar o efeito do tiossulfato de prata, associado ou não à sacarose, na manutenção da qualidade pós-colheita de hastes de crisântemos (Chrysanthemum morifolium Ramat cv. Dragon) . O experimento foi conduzido sob delineamento inteiramente casualizado, em esquema fatorial testando soluções de manutenção com tiossulfato de prata (STS), sob cinco níveis (Água destilada; STS a 0,2 mM; STS a 0,2 mM + sacarose a 50 g L-1; STS a 0,4 mM; STS a 0,4 mM + sacarose a 50 g L-1); e data de amostragem, por três níveis (0; 3; 6 dias). Utilizaram-se três repetições com duas hastes florais em cada tratamento. Foram feitas avaliações físicas: coloração, massa fresca e conteúdo relativo de água (CRA); avaliações químicas: açúcares redutores e pigmentos; e avaliações qualitativas: turgescência, cor das flores, e número de botões, flores entreabertas e abertas. O tratamento com 0,2 mM de STS possibilitou melhor manutenção da massa fresca das hastes. A concentração de pigmentos e carboidratos redutores foi maior naqueles tratamentos em que a sacarose foi associada. A coloração e o conteúdo relativo de água foram favorecidos nos tratamentos STS a 0,2 e 0,4mM. A concentração de 0,2 mM de STS foi a que obteve os melhores resultados, prolongando a vida de vaso das hastes. A qualidade das hastes foi superior, tendo as melhores avaliações de conteúdo de água, cor e turgescência.

O desenvolvimento e uso de técnicas que prolonguem a durabilidade das flores, mantendo a qualidade do produto, é imprescindível para redução de perdas pós-colheita. Objetivou-se, neste trabalho, avaliar soluções de manutenção, associadas ou não a sacarose, na manutenção da qualidade pós-colheita de hastes de crisântemos. O experimento foi conduzido sob delineamento inteiramente casualizado, em esquema fatorial, com cinco tratamentos e três repetições, com 2 hastes florais cada uma. Os tratamentos utilizaram água destilada; 8-HQC a 100 mg L-1; 8-HQC a 100 mg L-1 + sacarose a 50 g L-1, 8-HQC a 200 mg L-1; 8-HQC a 200 mg L-1 + sacarose a 50 g L-1. Foram feitas avaliações físicas: coloração, massa fresca e conteúdo relativo de água (CRA); avaliações químicas: açúcares redutores e pigmentos; e avaliações qualitativas: turgescência, cor das flores, e número de botões, flores entreabertas e abertas. A combinação de 8-HQC 200 mg L-1 + sacarose a 50 g L-1 foi a que apresentou melhor desempenho para a manutenção da qualidade das hastes florais, favorecendo a abertura de botões e a turgescência das lígulas. A sacarose contribuiu para uma melhor manutenção de substâncias de reserva na haste, que tiveram a sua vida de vaso estendida.

Alpinia purpurata (Vieill) K. Schum, common name red ginger belongs to Zingiberaceae family, and is a perennial, with nice inflorescences composed of layers of bracts arranged in spirals. The utilization of keeping-quality solutions seeks to prolong vase-life and to maintain the quality of cut flowers. This research was performed to evaluate the effect of seven pulsing solutions (during 24 hours) on water relations, quality (turgor, browning and curvature) and the longevity of cut red ginger. The experiment followed complete randomized design, in factorial arrangement. The following treatments were studied: 1) distilled water (control), 2) sucrose 2% + 8-hidroxyquinoline citrate 200 ppm, 3) sucrose 2%, 4) sucrose 2% + citric acid, 5) benzyladenine 10 μM, 6) benzyladenine 10 μmol + sucrose 2%, 7) quaternary ammonia 0,5 mL/L. There was no significant (P>0.05) reduction of the relative water content (RWC) of the inflorescence bracts, among solutions, in the first period (7 days) and in the second period (14 days) and, between the two periods for all treatments. Significant differences among RWC of some treatments were recorded after 16 days of vase life. The utilization of benzyladenine, benzyladenine plus sucrose and sucrose plus citric acid, in the pulsing solution, allowed to keep the flowers quality for longer periods than the other treatments.
