Drug induced liver injury is one of the frequent reasons for the drug removal from the market. During the recent years there has been a pressure to develop more cost efficient, faster and easier ways to investigate drug-induced toxicity in order to recognize hepatotoxic drugs in the earlier phases of drug development. High Content Screening (HCS) instrument is an automated microscope equipped with image analysis software. It makes the image analysis faster and decreases the risk for an error caused by a person by analyzing the images always in the same way. Because the amount of drug and time needed in the analysis are smaller and multiple parameters can be analyzed from the same cells, the method should be more sensitive, effective and cheaper than the conventional assays in cytotoxicity testing. Liver cells are rich in mitochondria and many drugs target their toxicity to hepatocyte mitochondria. Mitochondria produce the majority of the ATP in the cell through oxidative phosphorylation. They maintain biochemical homeostasis in the cell and participate in cell death. Mitochondria is divided into two compartments by inner and outer mitochondrial membranes. The oxidative phosphorylation happens in the inner mitochondrial membrane. A part of the respiratory chain, a protein called cytochrome c, activates caspase cascades when released. This leads to apoptosis. The aim of this study was to implement, optimize and compare mitochondrial toxicity HCS assays in live cells and fixed cells in two cellular models: human HepG2 hepatoma cell line and rat primary hepatocytes. Three different hepato- and mitochondriatoxic drugs (staurosporine, rotenone and tolcapone) were used. Cells were treated with the drugs, incubated with the fluorescent probes and then the images were analyzed using Cellomics ArrayScan VTI reader. Finally the results obtained after optimizing methods were compared to each other and to the results of the conventional cytotoxicity assays, ATP and LDH measurements. After optimization the live cell method and rat primary hepatocytes were selected to be used in the experiments. Staurosporine was the most toxic of the three drugs and caused most damage to the cells most quickly. Rotenone was not that toxic, but the results were more reproducible and thus it would serve as a good positive control in the screening. Tolcapone was the least toxic. So far the conventional analysis of cytotoxicity worked better than the HCS methods. More optimization needs to be done to get the HCS method more sensitive. This was not possible in this study due to time limit.

In order to design a High Temperature Superconducting (HTS) machine that is able to operate safely and reliably, studies on the characterization of Second Generation (2G) HTS tapes are of paramount importance. This paper presents an experimental setup to measure critical current of 2G HTS tapes in high DC magnetic fields (up to 5 Tesla) with an AC current ripple superimposed, as well as various temperatures ranging from 25 K to 77 K. The 2G tape measured is the SGS12050 coated conductor made by SuperPower. The critical current is measured by a flux vector with reference to the widest sample face from 0 to 90 degrees in 10 degree steps. Smaller steps are required close to 0 . A Variable Temperature Insert (VTI) is utilized to control temperature change. © 2010 IEEE.

With the development of seismic exploration, the target becomes more and more complex, which leads to a higher demand for the accuracy and efficiency in 3D exploration. Fourier finite-difference (FFD) method is one of the most valuable methods in complex structure exploration, which keeps the ability of finite-differenc method in dealing with laterally varing media and inherits the predominance of the phase-screen method in stablility and efficiency. In this thesis, the accuracy of the FFD operator is highly improved by using simulated annealing algorithm. This method takes the extrapolation step and band width into account, which is more suitable to various band width and discrete scale than the commonely-used optimized method based on velocity contrast alone. In this thesis, the FFD method is extended to viscoacoustic modeling. Based on one-way wave equation, the presented method is implemented in frequency domain; thus, it is more efficient than two-way methods, and is more convenient than time domain methods in handling attenuation and dispersion effects. The proposed method can handle large velocity contrast and has a high efficiency, which is helpful to further research on earth absorption and seismic resolution. Starting from the frequency dispersion of the acoustic VTI wave equation, this thesis extends the FFD migration method to the acoustic VTI media. Compared with the convetional FFD method, the presented method has a similar computational efficiency, and keeps the abilities of dealing with large velocity contrasts and steep dips. The numerical experiments based on the SEG salt model show that the presented method is a practical migration method for complex acoustical VTI media, because it can handle both large velocity contrasts and large anisotropy variations, and its accuracy is relatively high even in strong anisotropic media. In 3D case, the two-way splitting technique of FFD operator causes artificial azimuthal anisotropy. These artifacts become apparent with increasing dip angles and velocity contrasts, which prevent the application of the FFD method in 3D complex media. The current methods proposed to reduce the azimuthal anisotropy significantly increase the computational cost. In this thesis, the alternating-direction-implicit plus interpolation scheme is incorporated into the 3D FFD method to reduce the azimuthal anisotropy. By subtly utilizing the Fourier based scheme of the FFD method, the improved fast algorithm takes approximately no extra computation time. The resulting operator keeps both the accuracy and the efficiency of the FFD method, which is helpful to the inhancements of both the accuracy and the efficiency for prestack depth migration. The general comparison is presented between the FFD operator and the generalized-screen operator, which is valuable to choose the suitable method in practice. The percentage relative error curves and migration impulse responses show that the generalized-screen operator is much sensiutive to the velocity contrasts than the FFD operator. The FFD operator can handle various velocity contrasts, while the generalized-screen operator can only handle some range of the velocity contrasts. Both in large and weak velocity contrasts, the higher order term of the generalized-screen operator has little effect on improving accuracy. The FFD operator is more suitable to large velocity contrasts, while the generalized-screen operator is more suitable to middle velocity contrasts. Both the one-way implicit finite-difference migration and the two-way explicit finite-differenc modeling have been implemented, and then they are compared with the corresponding FFD methods respectively. This work gives a reference to the choosen of proper method. The FFD migration is illustrated to be more attractive in accuracy, efficiency and frequency dispertion than the widely-used implicit finite-difference migration. The FFD modeling can handle relatively coarse grids than the commonly-used explicit finite-differenc modeling, thus it is much faster in 3D modeling, especially for large-scale complex media.

In this paper we base on the anisotropic theory and Zoeppritz function of the transmission theory and the law of amplitude versus offset simplify seismic reflection coefficient of different media, analyze the characteristic of the gas or oil saturated stratum or the VTI and HTI models. Discuss the P wave reflection relationship and the meanings of the different parameters. We use measured parameters of a reservoir to simulate the characteristic of the reservoir, study the different effects of stratum saturated with gas or oil and analyze the characteristic of the seismic response of different models which change with different incident angles and different azimuths. Using the field data of logs ,analyze the rock property parameters, build the relationship of logs and parameters by Gassmann theory or empirical function. Calculate the density and the shear modulus and bulk modulus, reconstruct the log curves, calculate shear wave logs and correlate the logs affected by mud and other environmental factors. Finally perform the relationship of the seismic data log of saturated stratum and enhance the ability and reliability in reservoir prediction. Our aim is by the prestack seismic processing to get high solution and amplitude preserved seismic data. Because in incident angle gathers or azimuthal gathers, the low signal to noise ratio and low different covers affect the result of the prestack reservoir prediction. We apply prestack noise erase, cell regularization process and relatively amplitude preservation in the high solution seismic process routine to preserve the characteristic of stratum response, and erase the effects of the noise. In this paper we finished prestack invertion in the BYT survey and fractured reservoir depiction in MB survey. By the invertion and multiple attributes crossplot. we can get the stratum profiles and oil indicator profiles which can predict the distribution of the reservoir and oil. In the MB survey, we get orientation and density of fractured reservoir by the azimuthal seismic amplitude and depict the potential oil and gas reservoir. Prestak invertion works better in distinguishing oil and reservoir.

Aiming at solving the seismic imaging difficulty in complex area, the static correction methods and the migration imaging techniques taking the anisotropy into account are studied in this dissertation. To solve the static correction problems, a new tomography inversion approach is presented which takes use of the apparent slowness and apparent velocity and inverts both head and diving waves over the complete offset ranges. This approach is also taken practice to the practical seismic data processing of south areas of China and gets ideal effects. There are obvious differences between the actual statics and the statics based on the surface consistency hypothesis. In this dissertation, the exact differences formula is derived. The wave-eqation datuming method based on a single shot gather and the hybrid zero-offset wave-equation datuming algorithm based on f-x domain and f-k domain are presented at the same time. Further more, some forward modelings are made and tested. These methods are also put into practical seismic data processing and good results are made. In this dissertation, the true amplitude Kirchhoff pre-stack time migration fomula in VTI media is presented. The high-dense bispectral scanning technique based on the anelliptical time-shifted hyperbola and the geostatistical filtering are adopted to extract the anellipticity parameter. Simultaneously, combined with the practical seismic data imaging, the anisotropic pre-stack time migration flow is proposed and good processing results are made.

Multi-waves and multi-component get more and more attentions from oil industry. On the basis of existent research results, My research focuses on some key steps of OBC 4C datum processing. OBC datum must be preprocessed quite well for getting a good image. We show a flow chart of preprocess including attenuation of noise on multi-component datum、elimination ghost by summing P and Z and rotation of horizontal components. This is a good foundation for the coming steps about OBC processing. How to get exact converted point location and to analyze velocity are key points in processing reflection seismic converted wave data. This paper includes computing converted point location, analyzing velocity and nonhyperbolic moveout about converted waves. Anisotropic affects deeply the location of converted wave and the nonhyperbolic moveout. Supposed VTI, we research anisotropic effect on converted wave location and the moveout. Since Vp/Vs is important, we research the compute method of Vp/Vs from post-stack data and pre-stack data. It is a part of the paper that inversing anisotropic parameter by traveltime. Pre-stack time migration of converted wave is an focus, using common-offset Kirchhoff migration, we research the velocity model updating in anisotropic media. I have achieved the following results: 1) using continued Fractions, we proposed a new converted point approximate equation, when the offset is long enough ,the thomsen’s 2 order equation can’t approximate to the exact location of converted point, our equation is a good approximate for the exact location. 2) our new methods about scanning nonhyperbolic velocity and Vp/Vs can get a high quality energy spectrum. And the new moveout can fit the middle and long offset events. Processing the field data get a good result. 3) a new moveout equation, which have the same form as Alkhalifah’s long offset P wave moveout equation, have the same degree preciseness as thomsen’s moveout equation by testing model data. 4) using c as a function of the ratio offset to depth, we can uniform the Li’s and thomsen’s moveout equation in a same equation, the model test tell us choice the reasonable function C can improve the exact degree of Li’s and thomsen’s equation. 5) using traveltime inversion ,we can get anisotropic parameter, which can help to flat the large offset event and propose a model of anisotropic parameter which will useful for converted wave pre-stack time migration in anisotropic media. 6)using our pre-stack time migration method and flow, we can update the velocity model and anisotropic parameter model then get good image. At present, in order to image complex structures more accurately, the seismic migration methods has been developed from isotropic media to the anisotropic media. This dissertation develops a prestack time migration algorithm and application aspects for complex structures systematically. In transversely isotropic media with a vertical symmetry axis (VTI media), the dissertation starts from the theory that the prestack time migration is an approximation of the prestack depth migration, based on the one way wave equation and VTI time migration dispersion relation, by combining the stationary-phase theory gives a wave equation based VTI prestack time migration algorithm. Based on this algorithm, we can analytically obtain the travel time and amplitude expression in VTI media, as while conclude how the anisotropic parameter influence the time migration, and by analyzing the normal moveout of the far offset seismic data and lateral inhomogeneity of velocity, we can update the velocity model and estimate the anisotropic parameter model through the time migration. When anisotropic parameter is zero, this algorithm degenerates to the isotropic time migration algorithm naturally, so we can propose an isotopic processing procedure for imaging. This procedure may keep the main character of time migration such as high computational efficiency and velocity estimation through the migration, and, additionally, partially compensate the geometric divergence by adopting the deconvolution imaging condition of wave equation migration. Application of this algorithm to the complicated synthetic dataset and field data demonstrates the effectiveness of the approach. In the dissertation we also present an approach for estimating the velocity model and anisotropic parameter model. After analyzing the velocity and anisotropic parameter impaction on the time migration, and based on the normal moveout of the far offset seismic data and lateral inhomogeneity of velocity, through migration we can update the velocity model and estimate the anisotropic parameter model by combining the advantages of velocity analysis in isotropic media and anisotropic parameter estimation in VTI media. Testing on the synthetic and field data, demonstrates the method is effective and very steady. Massive synthetic dataset、2D sea dataset and 3D field datasets are used for VTI prestack time migration and compared to the stacked section after NMO and prestack isotropic time migration stacked section to demonstrate that VTI prestack time migration method in this paper can obtain better focusing and less positioning errors of complicated dip reflectors. When subsurface is more complex, primaries and multiples could not be separated in the Radon domain because they can no longer be described with simple functions (parabolic). We propose an attenuating multiple method in the image domain to resolve this problem. For a given velocity model, since time migration takes the complex structures wavefield propagation in to account, primaries and multiples have different offset-domain moveout discrepancies, then can be separated using techniques similar to the prior migration with Radon transform. Since every individual offset-domain common-reflection point gather incorporates complex 3D propagation effects, our method has the advantage of working with 3D data and complicated geology. Testing on synthetic and real data, we demonstrate the power of the method in discriminating between primaries and multiples after prestack time migration, and multiples can be attenuated in the image space considerably.

The topic of this study is about the propagation features of elastic waves in the anisotropic and nonlinear media by numerical methods with high accuracy and stability. The main achievements of this paper are as followings: Firstly, basing on the third order elastic energy formula, principle of energy conservation and circumvolved matrix method, we firstly reported the equations of non-linear elastic waves with two dimensions and three components in VTI media. Secondly, several conclusions about some numerical methods have been obtained in this paper. Namely, the minimum suitable sample stepth in space is about 1/8-1/12 of the main wavelength in order to distinctly reduce the numerical dispersion resulted from the numerical mehtod, at the same time, the higher order conventional finite difference (CFD) schemes will give little contribution to avoid the numerical solutions error accumulating with time. To get the similar accuracy with the fourth order center finite difference method, the half truncation length of SFFT should be no less than 7. The FDFCT method can present with the numerical solutions without obvious dispersion when the paprameters of FCT is suitable (we think they should be in the scope from 0.0001 to 0.07). Fortunately, the NADM method not only can reported us with the higher order accuracy solutions (higher than that of the fourth order finite difference method and lower than that of the sixth order finite difference method), but also can distinctly reduce the numerical dispersion. Thirdly, basing on the numerial and theoretical analysis, we reported such nonlinear response accumulating with time as waveform aberration, harmonic generation and resonant peak shift shown by the propagation of one- and two-dimensional non-linear elasticwaves in this paper. And then, we drew the conclusion that these nonlinear responses are controlled by the product between nonlinear strength (SN) and the amplitude of the source. At last, the modified FDFCT numerical method presented by this paper is used to model the two-dimensional non-linear elastic waves propagating in VTI media. Subsequently, the wavelet analysis and polarization are adopted to investigate and understand the numerical results. And then, we found the following principles (attention: the nonlinear strength presented by this paper is weak, the thickness of the -nonlinear media is thin (200m), the initial energy of the source is weak and the anisotropy of the media is weak too): The non-linear response shown by the elastic waves in VTI media is anisotropic too; The instantaneous main frequency sections of seismic records resulted from the media with a non-linear layer have about 1/4 to 1/2 changes of the initial main frequency of source with that resulted from the media without non-linear layer; The responses shown by the elasic waves about the anisotropy and nonlinearity have obvious mutual reformation, namely, the non-linear response will be stronger in some directions because of the anisotropy and the anisotropic strength shown by the elastic waves will be stronger when the media is nonlinear.

It has been a difficult problem faced by seismologists for long time that how exactly to reconstruct the earth's geometric structure and distribution of physical attributes according to seismic wave's kinematical and dynamic characteristics, obtained in seismological observation. The jointing imaging of seismic reflector and anisotropy attributes in the earth interior is becoming the research hot spot. The limitation of shoot and observation system makes that the obtained seismic data are too scarce to exactly reconstruct the geological objects. It is popular that utilizing only seismic reflection traveltimes or polarizations information make inversion of the earth's velocity distribution by fixing seismic reflector configuration (vice versa), these will lead to the serious non-uniqueness reconstruction due to short of effective data, the non-uniqueness problem of reconstructing anisotropy attributes will be more serious than in isotropy media. Obviously it is not enough to restrict the media structure only by information of seismic reflection traveltimes or polarizations, which even sometimes will lead to distorted images and misinterpretation of subsurface structure. So we try to rebuild seismic reflection structure (geometry) and media anisotropic structure (physics) in the earth interior by jointing data of seismic wave kinematics and dynamics characteristics, we carry out the new experiment step by step, and the research mainly comprises of two parts: one is the reconstruction of P-wave vertical velocity and anisotropic structure(Thomsen parameter s and 8) in the transversely isotropic media with vertical symmetrical axis(VTI) by fixing geometrical structure, and the other is the simultaneous inversion of the reflector surface conformation and seismic anisotropic structure by jointing seismic reflection traveltimes and polarizations data. Simulated annealing method is used to the first research part, linear inversion based on BG theory and Simulated annealing are applied to the second one. All the research methods are checked by model experiments, then applied to the real data of the wide-angle seismic profile from Tunxi, Anhui Province, to Wenzhou, Zhejiang Province. The results are as following The inversion results based on jointing seismic PP-wave or PSV-wavereflection traveltimes and polarizations data are more close to real model than themodels based simply on one of the two data respectively. It is shown that the methodwe present here can effectively reconstruct the anisotropy attributes in the earth'sinterior when seismic reflector structure is fixed. The layer thickness, P-wave vertical velocity and Thomsen anisotropicparameters {s and 8) could be resolved simultaneously by jointing inversion ofseismic reflection traveltimes and polarizations with the linear inversion methodbased on BG theory. The image of the reflector structure, P-wave vertical velocity and theanisotropy parameters in the crust could be obtained from the wide-angle seismicprofile from Tunxi (in Anhui Province), to Wenzhou (in Zhejiang Province). Theresults reveal the difference of the reflector geometrical structure and physicalattributes in the crust between Yangtze block and Cathaysia block, and attempt tounderstand the characteristics of the crustal stress field in the areas.

Wider economic benefits resulting from extended geographical mobility is one argument for investments in high-speed rail. More specifically, the argument for high-speed trains in Sweden has been that they can help to further spatially extend labor market regions which in turn has a positive effect on growth and development. In this paper the aim is to cartographically visualize the potential size of the labor markets in areas that could be affected by possible future high-speed trains. The visualization is based on the forecasts of labor mobility with public transport made by the Swedish national mobility transport forecasting tool, SAMPERS, for two alternative high-speed rail scenarios. The analysis, not surprisingly, suggests that the largest impact of high-speed trains results in the area where the future high speed rail tracks are planned to be built. This expected effect on local labor market regions of high-speed trains could mean that possible regional economic development effects also are to be expected in this area. However, the results, in general, from the SAMPERS forecasts indicaterelatively small increases in local labor market potentials.

We estimate the effect of employment density on wages in Sweden in a large geocoded data set on individuals and workplaces. Employment density is measured in four circular zones around each individual’s place of living. The data contains a rich set of control variables that we use in an instrumental variables framework. Results show a relatively strong but rather local positive effect of employment density on wages. Beyond 5 kilometers the effect becomes negative. This might indicate that the effect of agglomeration economies falls faster with distance than the effects of congestion.

Ageism – A useful concept? Ageism has gained growing attention in Sweden the last decade. Age is even discussed to be included in the discrimination legislation. Still, the concept has not been of much sociological interest. This article is an overview of the concept ageism and how it has been discussed in social gerontology. In the article it is argued that the overall focus has been on overtly expressions such as stereotypes and discriminating behavior while underlying structures and processes of power and power relations have not got enough attention. As a result the concept of ageism has become limited as an analytical tool. Thus, in order to develop the potential of the concept it seems crucial to explicate and theorise power relations. To accomplish this goal, i.e. to focus on how age-based power relations are negotiated, challenged and reproduced in processes and institutionalised practises it is suggested that ageism might have to be complemented with other concepts, such as age coding.
