The effects of salt stress on carbohydrate metabolism in Microcoleus vaginatus Gom., a cyanobacterium isolated from desert algal crusts, were investigated in the present study. Extracellular total carbohydrates and exopolysaccharides (EPS) in the culture medium produced by M. vaginatus increased significantly during the growth phase and reached a maximum during the stationary phase. The production of extracellular carbohydrates also significantly increased under higher salt concentrations, which was attributed to an increase in low molecular weight carbohydrates. In the presence of NaCl, the production of cellular total carbohydrates decreased and photosynthetic activity was impaired, whereas cellular reducing sugars, water-soluble sugars and sucrose content and sucrose phosphate synthase activity increased, reaching a maximum in the presence of 200 mmol/L NaCl. These parameters were restored to original levels when the algae were transferred to a non-saline medium. Sodium and K+ concentrations of stressed cells decreased significantly and H+-ATPase activity increased after the addition of exogenous sucrose or EPS. The results suggest that EPS and sucrose are synthesized to maintain the cellular osmotic equilibrium between the intra- and extracellular environment, thus protecting algal cells from osmotic damage, which was attributed to the selective exclusion of cellular Na+ and K+ by H+-ATPase.

The aims of this work were to deepen the knowledge on the physiology of bract abscission in Bougainvillea spectabilis ‘Killie Campbell’ plants, in what relates to respiration and carbon balance. Using the effects induced by Silver Thiosulphate (STS) and/or Naphtalene Acetic Acid (NAA, at high concentration: 500 mg.l-1) on bract abscission under interior conditions, the relationship between bract survival time (longevity) and, respiration rate or carbohydrate levels, was investigated. Treatments that included NAA were the ones that reduced significantly bract abscission. Unexpectedly, the higher the levels of bract soluble and total carbohydrates, measured at day 10 postproduction (PP), the higher the abscission of bracts. These results show, for the first time, that abscission can positively correlate with non structural carbohydrates levels in the organ that abscise. Bract respiration rate was significantly affected by treatment and postproduction day (PP). Treatments that had higher bract respiration rates (WATER and STS) also had higher levels of non structural carbohydrates in the bracts. Bract respiration rate decreased from day 10 to day 17 PP by approximately 50% (on average of all treatments) and was negatively correlated with bract survival time. In the carbon balance per gram of bract dry weight, the treatments WATER and STS, showed the largest decrease in the content of total carbohydrates and had the highest consumption of carbohydrates through respiration. So, these were the bracts that needed to import a higher amount of carbohydrates per gram of bract dry weight. In the carbon balance for the whole mass of bracts and adjacent stems in an average plant, the treatments WATER and STS continued to allow for the largest decreases in total carbohydrate during postproduction. However, and contradicting the results per gram of bract dry weight, the highest total consumption of carbohydrates by respiration was obtained for the NAA and STS+NAA treatments. It makes sense that bracts that last longer have lower individual carbon consumption while, at the plant level, the increased number of remaining bracts causes a higher overall expenditure. Respiration rate has been used as an indicator of flower longevity, this correlation is here extended for the flower+bract system. Plants that had higher bract respiration rates, most probably, had a higher flow of carbohydrates through the bracts (and flowers), which, in the end, was sensed as a higher carbohydrate level.

This thesis Entitled Haematological responses of penaeus monodon to environmental alterations and pathogenic invasion. Thesis concluded from the present study that stress is accompanied by alterations in haemolymph metabolic variables and immune responses that influences the susceptibility of P. monodon to infection. Acute salinity variations were proved to be a stress condition that enhances the susceptibility of P. monodon to V. harveyi and WSSV infection. Ambient Cu at 0.1 mg 1" and ambient Zn at 1.0 mg 1" proved immunostimulatory in increasing the immunocompetence of P. monodon to WSSV infection and higher concentrations of Cu and Zn proved immunosuppressive. Haemolymph total protein, total carbohydrates and total lipids showed the highest relation with immune responses. THC, PO, ACP and ALP that greatly correlated with the survival rate proposed as reliable biomarkers of health in P. monodon. The study highlights the need for proper management practices and regular health monitoring to be adopted to avoid mass mortality in shrimp culture ponds.

The present work is focussed mainly on the utilization of this weed-biomass on a biochemical and biotechnological basis. Before designing scientific and systematic utilization of any given biomass, the detailed analysis of its chemical componets is essential. Hence, as the preliminary part of the experimental works, samples of Salvinia were analysed for its chemical constituents.Before designing scientific and systematic utilization of any given biomass, the detailed analysis of its chemical componets is essential .The composition of the substrate contributes much to the nutritive value of mushrooms. Hence, alterations in the nutritive value of mushrooms (in terms of total carbohydrates, proteins, lipids and minerals) in response to Salvinia as substrate were analyzed.Substrate after mushroom harvest (spent substrate) can be utilized for various purposes such as cattle feed, as a source of degradative enzymes, as a substrate for other mushrooms and as garden manure. But studies are limited with regard to the utilization of Pleurotus spent substrate as garden manure. So the value of spent substrate as an organic supplement and its multidimensional impacts on soil chemical status, soil microbial population dynamics and plant growth (Amhurium andreanum) were carried out.Major findings of this work have got much relevance in designing measures to utilize different types of plant biomass, especially aquatic weeds, with the aid of a powerful biological tool, the lignocellulolytic fungus, Pleurorus

Influence of acute salinity stress on the immunological and physiological response of Penaeus monodon to white spot syndrome virus (WSSV) infection was analysed. P. monodon maintained at 15‰ were subjected to acute salinity changes to 0‰ and 35‰ in 7 h and then challenged orally with WSSV. Immune variables viz., total haemocyte count, phenol oxidase activity (PO), nitroblue tetrazolium salt (NBT) reduction, alkaline phosphatase activity (ALP), acid phosphatase activity (ACP) and metabolic variables viz., total protein, total carbohydrates, total free amino acids (TFAA), total lipids, glucose and cholesterol were determined soon after salinity change and on post challenge days 2 (PCD2) and 5 (PCD5). Acute salinity change induced an increase in metabolic variables in shrimps at 35‰ except TFAA. Immune variables reduced significantly (Pb0.05) in shrimps subjected to salinity stress with the exception of ALP and PO at 35‰ and the reduction was found to be more at 0‰. Better performance of metabolic and immune variables in general could be observed in shrimps maintained at 15‰ that showed significantly higher post challenge survival following infection compared to those under salinity stress. Stress was found to be higher in shrimps subjected to salinity change to lower level (0‰) than to higher level (35‰) as being evidenced by the better immune response and survival at 35‰. THC (Pb0.001), ALP (Pb0.01) and PO (Pb0.05) that together explained a greater percentage of variability in survival rate, could be proposed as the most potential health indicators in shrimp haemolymph. It can be concluded from the study that acute salinity stress induces alterations in the haemolymph metabolic and immune variables of P. monodon affecting the immunocompetence and increasing susceptibility to WSSV, particularly at low salinity stress conditions

The individual contribution of four β-galactosidases present in Bifidobacterium bifidum NCIMB 41171 towards galactooligosaccharides (GOS) synthesis was investigated. Although the β-galactosidase activity of the whole cells significantly decreased as a function of temperature (40 to 75 °C), GOS yield was at its maximum at 65 °C. Native-PAGE electrophoresis of the whole cells showed that the contribution of BbgIII and BbgIV towards GOS synthesis increased as the temperature increased. Moreover, BbgIII and BbgIV were found to be more temperature stable and to produce a higher GOS yield than BbgI and BbgII, when used in their free form. The GOS yield using BbgIV was 54.8 % (% of total carbohydrates) and 63.9 % (% lactose converted to GOS) at 65 °C from 43 % w/w lactose. It was shown that BbgIV is the most important β-galactosidase in B. bifidum NCIMB 41171 and can be used for GOS synthesis at elevated temperatures.

Um experimento foi realizado com o objetivo de avaliar o aporte de energia, a composição de bactérias e a eficiência microbiana por tourinhos Santa Gertrudes canulados no rúmen e no duodeno alimentados com dietas compostas de feno de capim-marandu e concentrado. Empregou-se o delineamento em quadrado latino 4 ´ 4, no qual os tratamentos foram concentrados ajustados para ganho de peso corporal (GPC) diário de 0,5 e 1 kg/animal e potencial de fermentação microbiana (y) de 9,5 e 11 g de PB microbiana/MJ energia metabolizável fermentável. Houve diferença para as concentrações de nutrientes digestíveis totais (NDT) e energia metabolizável fermentável (EMFe) ingeridos e para a composição em matéria orgânica e mineral das bactérias ruminais para as dietas ajustadas para diferentes GPC. Houve interação significativa GPC ´ y para a ingestão de NDT como porcentagem do peso corporal e a composição em PB e carboidratos totais das bactérias ruminais. Não foram encontradas diferenças para os potenciais de fermentação microbiana. A eficiência de síntese microbiana também não diferiu entre as dietas e apresentou valores de 12,7 g PB microbiana/100 g NDT e 9,2 g PB microbiana/MJ EMFe. As diferenças encontradas não justificaram o balanceamento dos concentrados para os diferentes potenciais de fermentação microbiana avaliados.

The effect of inoculation of Aspergillus flavus, Fusarium verticillioides, and Penicillium sp. in Dystrophic Red Latosol (DRL) and Eutroferric Red Latosol (ERL) soils with or without glucose on the total carbohydrate content and the dehydrogenase and amylase activities was studied. The fungal growth and spore production in culture medium with and without glucose were also evaluated. A completely randomized design with factorial arrangement was used. The addition of glucose in the culture medium increased the growth rate of A. flavus and Penicillium sp. but not of F. verticillioides. The number of spores increased 1.2 for F. verticillioides and 8.2 times for A. flavus in the medium with glucose, but was reduced 3.5 times for Penicillium sp. The total carbohydrates contents reduced significantly according to first and second degree equations. The consumption of total carbohydrates by A. flavus and Penicillium sp. was higher than the control or soil inoculated with F. verticillioides. The addition of glucose to soils benefited the use of carbohydrates, probably due to the stimulation of fungal growth. Dehydrogenase activity increased between 1.5 to 1.8 times (p <0.05) in soils with glucose and inoculated with the fungi (except F. verticillioides), in relation to soil without glucose. Amylase activity increased 1.3 to 1.5 times due to the addition of glucose in the soil. Increased amylase activity was observed in the DRL soil with glucose and inoculated with A. flavus and Penicillium sp. when compared to control.

Sistemas autossustentáveis favorecem as populações microbianas devido à conservação e ao aumento da matéria orgânica no solo. Além disso, as plantas que fazem parte desses sistemas promovem o efeito rizosférico, por meio da zona de influência das raízes, que resulta no aumento da atividade e na modificação da população microbiana. O objetivo deste trabalho foi avaliar o efeito da rotação de culturas de inverno sobre sequências de verão, em sistema de semeadura direta, nos atributos bioquímicos (amilase, urease, celulase e protease) e químicos (carbono orgânico total - COT, carboidratos totais e proteínas totais) em solo rizosférico (SR) e não rizosférico (SNR). Este estudo foi constituído de três culturas de inverno: milho (Zea mays L.), girassol (Helianthus anuus L.) e guandu (Cajanus cajan (L.) Millsp), que estavam em rotação sobre três sequências de verão: soja/soja (Glycine max L.), milho/milho e soja/milho, e duas posições no solo: solo aderido às raízes das plantas (SR) e solo da entrelinha de plantio (SNR). As atividades da amilase, celulase, protease e urease no SR foram 16, 85, 62 e 100 % maiores do que no SNR; para COT e proteínas totais a diferença foi de 21 %. Das culturas de inverno, o milho foi a que mais estimulou as atividades das enzimas amilase, celulase, urease e protease no SR, bem como a atividade das enzimas amilase, urease e protease no SNR. de modo geral, os teores de proteínas totais não foram influenciados pelas culturas de inverno e pelas sequências de verão; os carboidratos totais foram influenciados pelas culturas de inverno milho e girassol. Para o COT houve influência apenas da sequência de verão milho/milho. Os atributos bioquímicos e químicos avaliados neste estudo podem ser utilizados como indicadores das alterações no solo promovidas pelas culturas de inverno e pelas sequências de verão.

Objetivou-se avaliar o efeito da substituição parcial do milho pela casca de soja e pelo farelo de gérmen de milho sobre a produção e eficiência de síntese de proteína microbiana e sobre as taxas de diluição e passagem ruminal. Foram utilizados três novilhos de corte, mestiços, canulados no rúmen e no duodeno, distribuídos em dois delineamentos em quadrado latino 3 x 3. As dietas experimentais, à base de silagem de milho (60%), apresentavam como fonte de proteína farelo de girassol e, como fonte de energia, milho (MI) ou sua substituição parcial pela casca de soja (CS) ou pelo farelo de gérmen de milho (FGM). Para o isolamento da massa microbiana, foram coletadas amostras de conteúdo ruminal às 3, 6, 9 e 12 horas após a alimentação. Utilizaram-se RNA como marcador microbiano e dicromato de sódio e Co-EDTA como indicadores das taxas de passagem e de diluição, respectivamente. A composição dos microrganismos ruminais não foi influenciada pelas dietas experimentais ou pelos horários de coleta. Não houve diferenças significativas no fluxo de matéria orgânica, carboidratos totais, nitrogênio e nitrogênio microbiano para o duodeno e na eficiência de síntese de proteína microbiana. As taxas de diluição foram semelhantes entre as dietas, com média de 13,4%/h. A taxa de passagem da silagem de milho mordentada foi menor, com tendência de menor taxa de passagem também para o farelo de girassol na dieta MI. A casca de soja e o farelo de gérmen de milho podem substituir parcialmente o milho, proporcionando ambiente ruminal adequado ao desenvolvimento da flora microbiana e conseqüente produção de proteína microbiana ruminal em novilhos confinados.

The objective of this study was to evaluate the chemical characteristics of the forage and ingestive behavior of Nellore heifers fed hydrolyzed sugarcane in different periods of storage. Twenty-four heifers with initial body weight of 119.6±8.1 kg were utilized. The experimental design was completely randomized, in which the treatments were diets with fresh sugarcane and hydrolyzed sugarcane (5 g of lime kg-1 of chopped sugarcane) stored for 24, 48 or 72 hours as the only roughage. The addition of lime to sugarcane associated with its storage up to 72 hours provided an increase of 20% of the potentially degradable cell wall of carbohydrates, from 382.4 to 458.8 g kg-1 of total carbohydrates. The in vitro digestibility of dry matter was altered by the storage of hydrolyzed sugarcane, increasing 7.08% when the storage time was increased from 24 to 72 hours. Heifers fed fresh sugarcane remained more time consuming compared with heifers fed other diets. The time used for water intake was not influenced by the diet. The rumination time presented a quadratic variation in relation to storage time of the hydrolyzed sugarcane, with higher values for the of hydrolyzed sugarcane diets stored for 48 hours. Heifers fed hydrolyzed sugarcane spent more time on other activities than those fed fresh sugarcane. The supply of hydrolyzed sugarcane stored up to 72 hours in the proportion of 600 g kg-1 of dry matter in the diet, alters the intake patterns, reducing the feed intake in Nellore heifers.

Foram estudados os efeitos da adição de 15% de diferentes aditivos - casca de café, farelo de cacau e farelo de mandioca - à forragem verde de capim-elefante (peso/peso), no momento da ensilagem, sobre a digestibilidade aparente de dietas. Foram usados 20 ovinos machos, não castrados, com média de 22,27±3,24kg de peso corporal, em um delineamento inteiramente ao acaso, com quatro tratamentos e cinco repetições. Os tratamentos foram: T1 = capim-elefante ensilado sem aditivo; T2 = capim-elefante ensilado com 15% de casca de café; T3 = capim-elefante ensilado com 15% de farelo de cacau; T4 = capim-elefante ensilado com 15% de farelo de mandioca. Os animais receberam dieta isoproteica (10% de proteína bruta) em proporção de 60% de volumoso e 40% de concentrado, na base da matéria seca. Utilizou-se o método de coleta total de fezes durante sete dias. Os coeficientes de digestibilidade aparente da matéria seca, proteína bruta, fibra em detergente neutro e fibra em detergente ácido e de digestibilidade verdadeira da proteína bruta foram maiores no capim-elefante ensilado sem aditivo. O coeficiente de digestibilidade do extrato etéreo foi mais alto na silagem sem aditivo e na silagem com farelo de cacau. Os coeficientes de digestibilidade da matéria orgânica e dos carboidratos totais foram mais elevados na silagem sem aditivo e na silagem com farelo de mandioca. Os maiores valores de nutrientes digestíveis totais foram observados nas dietas que continham silagem sem aditivo e silagem com farelo de mandioca. A silagem de capim-elefante com 15% de farelo de mandioca, bem como a silagem sem aditivo apresentaram melhor digestibilidade dos nutrientes.

The nutritive value of elephant grass silages, with increasing levels of dehydrated cashew stalk (DCS) was evaluated. Five addition levels were studied (0: 4: 8: 12 and 16%), using 20 rams in a completely randomized design with five treatments (addition levels) and four replicates (animals). Intake and digestibility of dry matter (DM), organic matter (OM). crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), ether extract (EE), total carbohydrates (TC) and non-fibrous carbohydrates (NFC) were determined, as well as total digestible nutrients (TDN) and silages nitrogen balance (NB) DCS did not affect intakes of DM, OM, NDF, ADF, TC and TDN, neither digestibility of OM, CB, NDF ADF and TC silages. Otherwise. the addition of DCS increased intakes of CP EE and NFC and digestibility of EE, NFC, as well as TON values and nitrogen balance. Inclusion of DCS, showed linear decreasing digestibility of ADF It is concluded that addition of DCS to silages of elephant grass up to 16% on a natural basis can be used as a forage source for ruminants.

Objetivou-se estudar o efeito do tratamento alcalino da cana-de-açúcar com cal virgem ou cal hidratada sobre a composição bromatológica e a digestibilidade in vitro da matéria seca, da fibra em detergente neutro e da fibra em detergente ácido. Utilizou-se um delineamento inteiramente casualizado em esquema fatorial 3 × 3, composto de três formas de processamento da cana (in natura; cana hidrolisada com 0,5% de cal virgem; e cana hidrolisada com 0,5% de cal hidratada) e três tempos de armazenamento (12, 36 e 60 horas). As formas de processamento influenciaram os teores de matéria orgânica, matéria mineral, carboidratos totais e hemicelulose, assim como os teores de fibra em detergente neutro e nutrientes digestíveis totais. Os tempos de armazenamento influenciaram os teores de proteína bruta, matéria orgânica, carboidratos totais e hemicelulose. Entre os minerais, somente o teor de cálcio teve aumento com a inclusão de ambos os tipos de cal em relação à cana-de-açúcar, que não sofreu o processo de hidrólise. Os coeficientes de digestibilidade da matéria seca e da fibra em detergente neutro aumentaram com a hidrólise da cana em comparação à cana in natura. A hidrólise com cal hidratada ou com cal virgem mantém o valor nutricional da cana-de-açúcar, permitindo que possa ser utilizada depois de até 60 horas de armazenamento.
