Background and objectives Interleukin 18 (IL-18) is a pleiotropic cytokine involved in rheumatoid arthritis (RA) pathogenesis. This study was carried out to evaluate the effi cacy of IL-18 binding protein (IL-18BP) gene therapy in the rat adjuvant- induced arthritis (AIA) model and to decipher the mechanisms by which IL-18BP delivery lessens bone destruction.Materials and methods Arthritis was induced in female Lewis rat by Mycobacterium butyricum and the mRNA expression of IL-18 and IL-18BP was determined in the joints. In a preventive study, rats were divided into an adenovirus producing IL-18BP-Fc (AdmIL-18BP-Fc) group (n=8) and an adenovirus producing green fl uorescent protein (AdGFP) group (n=7). On day 8 after AIA induction, adenoviruses were injected. Clinical parameters were assessed. At day 18, during maximal arthritis, the rats were euthanized, ankles were collected and x-rays were performed. mRNA and protein were extracted from joints for analysis by quantitative reverse transcriptase-PCR, multiplex, Western blot and zymography.Results The authors observed a decrease in the (IL-18BP/ IL-18) ratio from day 7 to 45. Administration of AdmIL-18BPd-Fc decreased clinical parameters and prevented bone and joint destruction compared to AdGFP administration. IL-18BP delivery reduced the (receptor activator of nuclear factor κB ligand (RANKL)/osteoprotegerin (OPG)) ratio by 70%, the matrix metalloproteinase 9 (MMP9) level by 33% and the tartrate-resistant acid phosphatase (TRAP) level by 44% in the joint homogenates from AdmIL-18BPd-Fc compared to AdGFP treated rats.Conclusions In rat AIA, a decrease in the (IL-18BP/IL-18) ratio was observed. IL-18BP delivery prevented joint and bone destruction by downregulating MMP9, (RANKL/OPG) and TRAP, suggesting a potential benefi t of a similar therapy in RA.Abstract topics Towards novel therapeutic strategies.

Se efectuaron mediciones de TS (Fuerza de Blanco, Target Strenght) durante los 108 lances de comprobación de ecotrazos realizados por el BIC Humboldt en el marco del Crucero 9808-09 de Evaluación Hidroacústica de Recursos Pelágicos. Se consideran para el análisis únicamente aquellos lances donde más del 90% de la captura perteneció a una sola especie. Los factores b20 de la ecuación de TS (TS=20 log L - b20) que han sido determinados, las cuales deberán ser considerados como provisionales, son las siguientes: Pez cinta (Trichiurus lepturus) = 70,95 (120 kHz); Vinciguerria o pez linterna (Vinciguerria lucetia pacifici) = 83,29 (120 kHz); Samasa (Anchoa nasus) = 86,57 (120 kHz); Caballa (Scomber japonicus) = 83,09 (120 kHz); Pez cinta (Trichiurus lepturus) = 71,41 (38 kHz); y Vinciguerria o pez linterna (Vinciguerria lucetia pacifici)= 82,04 (38kHz).

Se realizaron mediciones de la Fuerza de Blanco (TS) del krill (Euphausia superba) durante los 29 lances efectuados en el Crucero de Evaluación Hidroacústica utilizando la ecosonda SIMRAD EK 500 a bordo del BIC Humboldt entre los días 12 y 24 de enero de 1998 a lo largo del Estrecho de Bransfield y alrededores de la Isla Elefante. Se derivaron los valores de b20 a partir de le ecuación de TS de FOOTE (1990), de las longitudes promedios de los individuos capturados durante cada uno de los lances y de las tablas de TS generadas por la ecosonda, determinándose una ecuación de TS para el krill en un rango de longitud comprendido entre 2,1 y 5,3 cm de la siguiente forma: TS = 20 log L - 89,26. Se discute el posible sesgo de la ecuación debido, entre otros aspectos a que no se consideraron los estadíos sexuales del krill.

Se consideraron para el análisis únicamente aquellos lances donde más del 90% de la captura perteneció a una determinada especie y que, además, se haya observado en ellos una adecuada correlación entre los histogramas de tallas y de TS. Los factores b20 de la ecuación de TS (TS = 20 log L - b20) que han sido determinados, las cuales serán consideradas como provisionales, corresponden a anchoveta (2) y a bregmaceros (1). Estas son las siguientes: anchoveta, b20 = 81, 8 (para ejemplares entre 6 y 8,5 cm); anchoveta, b20 = 78,5 (para ejemplares entre 11,5 y 14,5 cm); bregmaceros, b20 = 80,25 (para ejemplares entre 5,5 y 8 cm).

Los factores b20 de la ecuación de TS (TS=20 log L - b20) que han sido determinados, las cuales serán consideradas como provisionales, corresponden a anchoveta (2) y a pota (1). Estas son las siguientes: anchoveta a 120 kHz, b20=82,2 (para ejemplares entre 5,5 y 9,5 cm); anchoveta 38 kHz, b20=81,2 (para ejemplares entre 5,5 y 10,5 cm); pota a 38 kHz, b20=87,2 (para ejemplares entre 12 y 24 cm de longitud de manto).

A determinação de As por espectrometria de absorção atômica com geração de hidretos (HG-AAS) constitui um método simples, sensível, preciso e de baixo custo. Entretanto, essa técnica requer a pré-redução das espécies de As(V), o que se obtém através do uso de agentes redutores como o KI. Em extratos contendo agentes oxidantes, a pré-redução do As é comprometida, como acontece em extratos obtidos pela aplicação do método BCR (acrônimo francês para Community Bureau of Reference) para a extração sequencial de As em sedimentos. O objetivo deste trabalho foi avaliar as condições de redução do As(V) a As(III), de modo a permitir o uso da HG-AAS para a quantificação do arsênio em extratos obtidos a partir do método BCR. Foram avaliadas condições reacionais utilizando KI, L-cisteína e ácido ascórbico. Para cada uma das etapas de extração do método BCR, diferentes condições de pré-redução possibilitaram a detecção quantitativa do As presente. O uso do método BCR para a extração de arsênio em amostras de sedimentos contaminadas e a aplicação das condições de pré-redução do As(V) selecionadas, seguida pela detecção por HG-AAS, forneceram percentagens de recuperação entre 91 e 99 %.

In urban communities, there are often limited amounts of right-of-way available for establishing a large setback distance from the curb for fixed objects. Urban communities must constantly weigh the cost of purchasing additional right-of-way for clear zones against the risk of fixed object crashes. From 2004 to 2006, this type of crash on curbed roads represented 15% of all fatal crashes and 3% of all crashes in the state of Iowa. Many states have kept the current minimum AASHTO recommendations as their minimum clear zone standards; however, other states have decided that these recommendations are insufficient and have increased the required minimum clear zone distance to better suit the judgment of local designers. This report presents research on the effects of the clear zone on urban curbed streets. The research was conducted in two phases. The first phase involved a synthesis of practice that included a literature review and a survey of practices in jurisdictions that have developmental and historical patterns similar to those of Iowa. The second phase involved investigating the benefits of a 10 ft clear zone, which included examining urban corridors in Iowa that meet or do not meet the 10 ft clear zone goal. The results of this study indicate that a consistent fixed object offset results in a reduction in the number of fixed object crashes, a 5 ft clear zone is most effective when the goal is to minimize the number of fixed object c ashes, and a 3 ft clear zone is most effective when the goal is to minimize the cost of fixed object crashes.

Iowa’s traffic safety culture is influenced by laws and policies, enforcement methods, driver education, roadway engineering, and drivers’ behaviors. The Center for Social and Behavioral Research at the University of Northern Iowa was contracted by the Iowa Department of Transportation to conduct a general population survey of adult Iowans. Telephone interviews were conducted with 1,088 adult Iowans from October to December 2011. A dual-frame (cell phone and landline) sampling design was used. The interview covered a wide range of traffic safety topics (e.g., traffic safety policies, enforcement techniques, and distracted driving). Most Iowans said driving in Iowa is about as safe now as it was 5 years ago; however, one-fourth said driving in Iowa is less safe now. There are a number of driving-related behaviors many adult Iowans consider serious threats to traffic safety and never acceptable to do while driving. Yet, many Iowans report often seeing other drivers engaging in these behaviors and admit engaging in some themselves. For example, nearly 1 in 5 adult Iowa drivers said they have sent or read a text message or email while driving in the past 30 days despite this being prohibited since July of 2011. A slight majority said they support using cameras on highways, interstates, and city streets to automatically ticket drivers for speeding, with even stronger support for red light cameras. A comprehensive approach to traffic safety in Iowa is required to encourage protective factors that enhance traffic safety and reduce the impact of detrimental factors.

The aim of this work was to compare the performances of the L'vov and integrated platforms in order to overcome the interferences found on lead determination in zidovudine by ET AAS. Pyrolysis and atomization temperatures found with integrated platform were 700 and 1400°C, respectively. The characteristic masses were 12.7 ± 1.2 pg with integrated platform and 11.1 ± 1.3 pg with grooved platform. The ratio between the slopes of zidovudine and aqueous curves shows a decrease in the interferences when the L'vov platform is used (bAZT/b aq = 0.97 ± 0.10) and the detection limit found was 0.03 µg.g-1. That ratio was 0.85 ± 0.07 with the integrated platform.

Mercury is a toxic metal used in a variety of substances over the course history. One of its more dubious uses is in dental amalgam restorations. It is possible to measure very small concentrations of this metal in the urine of exposed subjects by the cold vapor atomic absorption technique. The present work features the validation as an essential tool to confirm the suitability of the analytical method chosen to accomplish such determination. An initial analysis will be carried out in order to evaluate the environmental and occupational levels of exposure to mercury in 39 members of the auxiliary dental staff at public consulting rooms in the city of Araguaína (TO).
