The increasing use of nanomaterials in several products in different areas such as electronics, cosmetics, food and drugs has attracted the attention of the scientific community due to unknown properties of these materials, such as the life cycle and the interaction with receptor organisms once they reach the environment. In this context, the present work shows an updated scenario of nanomaterials applications, characterization methods and toxicity evaluation, also addressing important aspects related to the development and application of nanotechnology under the environmental point of view.

Azo dyes constitute the largest group of colorants used in industry and can pass through municipal waste water plants nearly unchanged due to their resistance to aerobic treatment, which potentially exposes humans and local biota to adverse effects. Unfortunately, little is known about their environmental fate. Under anaerobic conditions, some azo dyes are cleaved by microorganisms forming potentially carcinogenic aromatic amines. In the present study, the azo dye Disperse Orange 1, widely used in textile dyeing, was tested using the comet, Salmonella/microsome mutagenicity, cell viability, Daphnia similis and Microtox (R) assays. The human hepatoma cell line (HepG2) was used in the comet assay and for cell viability. In the mutagenicity assay. Salmonella typhimurium strains with different levels of nitroreductase and o-acetyltransferase were used. The dye showed genotoxic effects with respect to HepG2 cells at concentrations of 0.2, 0.4, 1.0, 2.0 and 4.0 mu g/mL. In the mutagenicity assay, greater responses were obtained with the strains TA98 and YG1041, suggesting that this compound mainly induces frameshift mutations. Moreover, the mutagenicity was greatly enhanced with the strains overproducing nitroreductase and o-acetyltransferase, showing the importance of these enzymes in the mutagenicity of this dye. In addition, the compound induced apoptosis after 72 h in contact with the HepG2 cells. No toxic effects were observed for either D. similis or Vibrio fischeri. (C) 2011 Elsevier B.V. Polymeric particulate-systems are of great relevance due to their possible biomedical applications, among them as carriers for the nano- or microencapsulation of drugs. However, due to their unique specific properties, namely small size range, toxicity issues must be discarded before allowing its use on health-related applications. Several polymers, as poly(methyl methacrylate) (PMMA), have proved to be suitable for the preparation of particulate-systems. However, a major drawback of its use refers to incomplete drug release from particles matrix. Recent strategies to improve PMMA release properties mention the inclusion of other acrylic polymers as Eudragit (EUD) on particles formulation. Though PMMA and EUD are accepted by the FDA as biocompatible, their safety on particle composition lacks sufficient toxicological data. The main objective of this thesis was to evaluate the biological effects of engineered acrylic particulate-systems. Preparation, physicochemical characterization and in vitro toxicity evaluation were assessed on PMMA and PMMA-EUD (50:50) particles. The emulsification-solvent evaporation methodology allowed the preparation of particles with spherical and smooth surfaces within the micrometer range (±500 nm), opposing surface charges and different levels of hydrophobicity. It was observed that particles physicochemical properties (size and charge) were influenced by biological media composition, such as serum concentration, ionic strength or pH. In what concerns to the in vitro toxicological studies, particle cellular uptake was observed on different cell lines (macrophages, osteoblasts and fibroblasts). Cytotoxicity effects were only found after 72 h of cells exposure to the particles, while no oxidative damage was observed neither on osteoblasts nor fibroblasts. Also, no genotoxicity was found in fibroblast using the comet assay to assess DNA damage. This observation should be further confirmed with other validated genotoxicity assays (e.g. Micronucleus Assay). The present study suggests that the evaluated acrylic particles are biocompatible, showing promising biological properties for potential use as carriers in drug-delivery systems.

Great effort is put into developing reliable, predictive, high-throughput, and low-cost screening approaches for the toxicity evaluation of ambient and manufactured nanoparticles (NP). These tests often consider oxidative reactivity, as oxidative stress is a well-documented pathway in particle toxicology. Based on a panel of six carbonaceous and five metal/metal oxide (Me/MeOx) nanoparticles, we: (i) compared the specifications (linearity, detection limits, repeatability) of three acellular reactivity tests using either dithiothreitol (DTT assay), dichlorofluorescein (DCFH assay), or ascorbic acid (AA-assay) as the reducing agent; and (ii) evaluated which physicochemical properties were important for explaining the observed reactivity. The selected AA assay was found to be neither sensitive nor robust enough to be retained. For the other tests, the surface properties of carbonaceous NP were of utmost importance for explaining their reactivity. In particular, the presence of "strongly reducing" surface functions explained most of its DCFH reactivity and a large part of its DTT reactivity. For the selected Me/MeOx, a different picture emerged. Whereas all particles were able to oxidize DCFH, dissolution and complexation processes could additionally influence the measured reactivity, as observed using the DTT assay. This study suggests that a combination of the DTT and DCFH assays provides complementary information relative to the quantification of the oxidative capacity of NP.

PURPOSE: The combination of embolic beads with a multitargeted tyrosine kinase inhibitor that inhibits tumor vessel growth is suggested as an alternative and improvement to the current standard doxorubicin-eluting beads for use in transarterial chemoembolization. This study demonstrates the in vitro loading and release kinetics of sunitinib using commercially available embolization microspheres and evaluates the in vitro biologic efficacy on cell cultures and the resulting in vivo pharmacokinetics profiles in an animal model. MATERIALS AND METHODS: DC Bead microspheres, 70-150 µm and 100-300 µm (Biocompatibles Ltd., Farnham, United Kingdom), were loaded by immersion in sunitinib solution. Drug release was measured in saline in a USP-approved flow-through apparatus and quantified by spectrophotometry. Activity after release was confirmed in cell culture. For pharmacokinetics and in vivo toxicity evaluation, New Zealand white rabbits received sunitinib either by intraarterial injection of 100-300 µm sized beads or per os. Plasma and liver tissue drug concentrations were assessed by liquid chromatography-tandem mass spectroscopy. RESULTS: Sunitinib loading on beads was close to complete and homogeneous. A total release of 80% in saline was measured, with similar fast-release profiles for both sphere sizes. After embolization, drug plasma levels remained below the therapeutic threshold (< 50 ng/mL), but high concentrations at 6 hours (14.9 µg/g) and 24 hours (3.4 µg/g) were found in the liver tissue. CONCLUSIONS: DC Bead microspheres of two sizes were efficiently loaded with sunitinib and displayed a fast and almost complete release in saline. High liver drug concentrations and low systemic levels indicated the potential of sunitinib-eluting beads for use in embolization.

As plantas medicinais são ampla e culturalmente utilizadas de forma empírica na Amazônia no tratamento de diversas doenças. Grande parte dessas plantas ainda não foi investigada cientificamente sobretudo quanto aos aspectos relacionados as atividades biológicas. A espécie selecionada neste trabalho é Vatairea guianensis, utilizada na medicina tradicional para tratar infecções de pele como as micoses cutâneas. Este trabalho avaliou a atividade antibacteriana in vitro dos extratos hidroetanólico, henicosânico, clorofórmico e metanólico obtidos das sementes de Vatairea guianensis pelo método da microdiluição em caldo para obtenção da Concentração Inibitória Mínima (CIM) e Concentração Bactericida Mínima (CBM), frente a bactérias Gram-positivas (Staphylococus aureus e Enterecoccus faecalis) e Gram-negativas (Pseudomonas aeruginosa e Salmonella sp). Avaliou-se também a atividade cicatrizante sobre feridas cutâneas abertas em ratos pela aplicação tópica do extrato hidroetanólico, representados pelos grupos G1 controle positivo (fibrinase); G2 controle negativo (sol salina); e grupos experimentais, (G3 dose 500mg/kg; G4 250mg/kg; G5 100mg/kg) por sete dias e a toxicidade aguda por via oral. A análise histológica do processo cicatricial foi avaliada, por meio de técnica convencional incluindo coloração HE e Picrossírius para observação das características histomorfológicas da reação inflamatória e análise descritiva da deposição de colágeno,respectivamente. Todos os extratos demonstraram atividade antimicrobiana contra todos os microrganismos testados com CIM que variaram de 3,12μg/mL a 50μg/mL e CBM com valores 6,25μg/mL a 100μg/mL. A análise histológica mostrou que o extrato hidroetanólico diminuiu a intensidade da reação inflamatória nos grupos G3 e G1, estimulou a síntese de colágeno tipo III em G1, G3 e G4 e aumentou a síntese de colágeno em G2 e G5. O experimento com extrato hidroetanólico obtido das sementes de Vatairea guianensis pareceu retardar o processo cicatricial nas doses de 500 e 250mg/kg em feridas abertas o que pode ser um efeito positivo por impedir a formação de cicatriz hipertrófica, sugerindo assim um efeito modulatório por parte do extrato. A avaliação da toxicidade aguda em camundongos revelou que o extrato hidroetanólico apresentou baixa toxicidade por via oral.

Petroleum and its subproducts are considered a treat for the environmental quality because of the many environmental accidents that may occur during exploitation, transport and storage. A common remediation technique used in the contaminated areas is based on the use of surfactants, mainly the chemical ones, because they have low production costs. In the other hand, some microorganisms have indicate capacities of producing surfactants that emulsify substances and as result, offer a bigger contact surface for the microbiota degradation. This biossurfactants stand out in comparison with the chemical surfactants because they present lower micelar concentration values, are more tolerant for temperature and pH variation, because they are biodegradable, have low toxicity, higher emulsification and hydrocarbon solubilization index. In this way, after the surfactant application, a toxicity evaluation have to be made to identify the treatment effects. In soil, the activity of some microbial enzymes can show the environmental behavior of the contaminant under different treatment conditions. Dehydrogenase is one example of those enzymes that can demonstrate indirectly the effect of the pollutant on the soil microorganisms. The aim of this paper was to evaluate the toxicity after the addition of a surfactant and/or Pseudomonas aeruginosa LBI in soil contaminated by a mineral automotive lubricant. The previous mentioned bacteria are a potential biossurfactant (rhamnolipid) producer. In order to evaluate the toxicity, the dehydrogenase test was run. In this test, trifeniltetrazolium compound (TTC) after utilized as an electron acceptor, turns into trifenil formazan (TPF), that can be indirectly quantified using the absorbance measured by the spectrophotometer UV-visible. In order to improve the ability to link chemical exposure to toxicological and ecological effects, aquatic toxicology will have to move from observing what chemical concentrations induce adverse effects to more explanatory approaches, that are concepts which build on knowledge of biological processes and pathways leading from exposure to adverse effects, as well as on knowledge on stressor vulnerability as given by the genetic, physiological and ecological (e.g., life history) traits of biota. Developing aquatic toxicology in this direction faces a number of challenges, including (i) taking into account species differences in toxicant responses on the basis of the evolutionarily developed diversity of phenotypic vulnerability to environmental stressors, (ii) utilizing diversified biological response profiles to serve as biological read across for prioritizing chemicals, categorizing them according to modes of action, and for guiding targeted toxicity evaluation; (iii) prediction of ecological consequences of toxic exposure from knowledge of how biological processes and phenotypic traits lead to effect propagation across the levels of biological hierarchy; and (iv) the search for concepts to assess the cumulative impact of multiple stressors. An underlying theme in these challenges is that, in addition to the question of what the chemical does to the biological receptor, we should give increasing emphasis to the question how the biological receptor handles the chemicals, i.e., through which pathways the initial chemical-biological interaction extends to the adverse effects, how this extension is modulated by adaptive or compensatory processes as well as by phenotypic traits of the biological receptor.

Além de ser o cogumelo mais consumido no mundo, Agaricus bisporus é um dos cogumelos mais ricos em ergosterol, representando esta molécula quase 90% da sua fração de esteróis. Vários estudos têm atribuído ao ergosterol diferentes bioatividades, incluindo efeitos hipocolesterolémicos semelhantes aos exibidos pelos fitoesteróis. Isto torna o ergosterol uma molécula interessante para ser estudada como composto nutracêutico. Assim, este trabalho teve como objetivo avaliar o potencial de utilização dos extratos de A. bisporus ricos em ergosterol na produção de bebidas lácteas funcionais. Para o efeito, foram realizados testes de incorporação do extrato e do ergosterol puro em iogurtes que se compararam com bebidas lácteas funcionais comerciais (aditivadas com fitoesteróis). As amostras de A. bisporus foram submetidas a uma extração assistida por ultrassons e os extratos obtidos (IEXT), bem como a molécula de ergosterol em diferentes concentrações (IERG1 e IERG2), foram incorporados em iogurtes, e comparadas com amostras controlo (amostras de iogurte sem aditivos) (ICN) e iogurtes comerciais contendo fitoesteróis (ICP). Todas as amostras foram analisadas imediatamente após a incorporação (T0), e após sete dias de armazenagem a 4°C (T1), em relação aos parâmetros nutricionais, atividade antioxidante e propriedades citotóxicas em linhas celulares tumorais humanas e numa cultura primária de células de fígado de porco (não tumoral) para avaliação da toxicidade. O teor de ergosterol incorporado na forma pura, ou presente nos extratos, foi monitorizado por HPLC-UV. Adicionalmente, foi realizado um estudo de microencapsulação utilizando a técnica de coacervação, tendo o quitosano e o isolado proteico de soro como materiais encapsulantes. Num ensaio preliminar determinou-se o pH conducente a um maior rendimento de encapsulação e, seguidamente, verificou-se a influência da razão proteína:quitosano (P/Q) e da temperatura utilizada, no rendimento de encapsulação (Y1), na eficiência de encapsulação (Y2) e na carga (teor de ergosterol nas microesferas) (Y3). Posteriormente, o estudo foi realizado baseando-se nas melhores condições para encapsular ergosterol, sendo também avaliadas as respostas Y1, Y2 e Y3. As bebidas funcionalizadas com o extrato (IEXT) e com ergosterol na mesma concentração existente no extrato (IERG1) revelaram uma atividade antioxidante similar às bebidas comerciais com fitoesteróis. No entanto, as bebidas com ergosterol na mesma concentração do extrato de A. bisporus e de fitoesteróis (IERG2) revelaram uma atividade antioxidante superior. Além disso, apenas IEXT, IERG1 e IERG2 apresentaram um aumento na atividade antioxidante de T0 para T1, com destaque para a atividade exibida por IERG2, significando que o ergosterol e os extratos foram capazes de proteger a bebida láctea da oxidação, aumentando a vida de prateleira do produto. IERG2 foi a amostra que revelou a maior citotoxicidade para as linhas celulares tumorais, enquanto as bebidas com fitoesteróis mostraram a menor atividade, sem diferenças significativas entre T0 e T1. Os estudos de microencapsulação revelaram ainda que a técnica de coacervação permite obter cápsulas de distintos tamanhos e que as condições ótimas do processo ocorrem a pH 5,5, com temperatura de 55ºC e razão P/Q de 0,5, com um menor rendimento de encapsulação, mas com uma maior carga em ergosterol. Este trabalho contribuiu para o estudo do potencial da utilização de extratos de A. bisporus com ergosterol no desenvolvimento de novas bebidas funcionais. Constituiu um primeiro passo que necessita de estudos subsequentes relacionados com a avaliação da viabilidade da sua utilização ao nível industrial e demonstração clara da sua bioatividade in vivo.

Rubus niveus Thunb. plant belongs to Rosaceae family and have been used traditionally to treat wounds, burns, inflammation, dysentery, diarrhea and for curing excessive bleeding during menstrual cycle. The present study was undertaken to investigate the in vivo genotoxicity of Rubus niveus aerial parts extract and its possible chemoprotection on doxorubicin (DXR)-induced DNA damage. In parallel, the main phytochemicals constituents in the extract were determined. The animals were exposed to the extract for 24 and 48h, and the doses selected were 500, 1000 and 2000mg/kg b.w. administered by gavage alone or prior to DXR (30mg/kg b.w.) administered by intraperitoneal injection. The endpoints analyzed were DNA damage in bone marrow and peripheral blood cells assessed by the alkaline alkaline (pH>13) comet assay and bone marrow micronucleus test. The results of chemical analysis of the extract showed the presence of tormentic acid, stigmasterol, quercitinglucoronide (miquelianin) and niga-ichigoside F1 as main compounds. Both cytogenetic endpoints analyzed showed that there were no statistically significant differences (p>0.05) between the negative control and the treated groups with the two higher doses of Rubus niveus extract alone, demonstrating absence of genotoxic and mutagenic effects. Aneugenic/clastogenic effect was observed only at 2000mg/kg dose. On the other hand, in the both assays and all tested doses were observed a significant reduction of DNA damage and chromosomal aberrations in all groups co-treated with DXR and extract compared to those which received only DXR. These results indicate that Rubus niveus aerial parts extract did not revealed any genotoxic effect, but presented some aneugenic/clastogenic effect at higher dose; and suggest that it could be a potential adjuvant against development of second malignant neoplasms caused by the cancer chemotherapic DXR.
