O conhecimento da variabilidade genética e da relação entre diferentes acessos de Stylosanthes guianensis é importante para maximizar o uso dos recursos genéticos disponíveis. Este trabalho teve como objetivo avaliar a variabilidade genética em 20 acessos dessa espécie, pertencentes ao banco de germoplasma da Embrapa Gado de Corte, usando marcadores RAPD (DNA Polimórfico Amplificado ao Acaso). Foram selecionados 40 primers randômicos, que produziram um total de 210 bandas; destas, 82 polimórficas (39,05%). Os dados obtidos foram utilizados para gerar uma matriz de similaridade genética usando o coeficiente de Jaccard. A similaridade genética variou de 0,747 a 0,945, o que indicou uma variabilidade genética pouco acentuada entre os acessos estudados. Os acessos com menor similaridade genética foram SG01 e SG14. Os resultados das análises de agrupamento com os métodos UPGMA (Agrupamento com Média Aritmética Não Ponderada) e Tocher foram similares. Apesar da baixa variabilidade genética detectada entre os 20 acessos, estes foram separados em nove grupos distintos pelo método UPGMA e seis grupos pelo método de Tocher.

O objetivo deste trabalho foi comparar a quantidade e qualidade do DNA isolado de folhas de S. guianensis pelos métodos de Bonato et al. (2002), de Faleiro et al. (2003) e um método baseado no de Bonato et al. (2002), mas com modificações que resultem um DNA mais puro e em quantidade suficiente para execução de diversas análises moleculares, incluindo amplificação, clonagem e sequenciamento.

The distribution of the holothurian H. KH.) scabra indicated its availability all along coastal areas on Palk Bay from Rameswavam to Mallipattinam and along the Gulf of Hannah coast from Pamban to Ervadi and Tuticmhin,'at 4~2O m depth.The major fishing for holmthurians was done by skin diving at all the centres. The tallu valai was operated at Tuticorin and Vedalai and trawlevs were operated at Rameswaram.The fmod of H. KN.) scabra consists of ovganic matter which contains mud, sand, shell debris, bivalves and algae. Obsehvatinns indicated the species seems to be a n0n—se1ective feeder. The assimilation efficiency from sediment to faeces indicated that the faecal pellets of H. KH.) scabra are semidigested.A multiple relationship was fitted between total length, total weight, gutted weight, gonad weight and maturity stages were found significant.The fishing season for holothurians commences from October to March along Gulf of Manner coast and from March to October along Palk Bay coast.The percentage of catches recorded by skin diving, trawlere and tallu valai were 80.04%, 10.27% and 9.69% respectively. Skin diving contributes to maximum catch.The holothurians landed all along the Gulf of Manner and Palk Bay coasts constitute 25.6% and 74.4% respectively. This showed that Palk Bay coast is more productive.

The objective of this research was to determine the effect of two sowing times on phytomass production of two varieties of Slylosanthes guianensis (var. pauciflora and var. vulgaris). Two experimental periods were studied (1: January - May/1998 and 2: November/1998 - March/1999) using a completely randomized factorial design 2 x 2 x 14 (two periods, two varieties and fourteen ages of evaluation), with four replications. The results showed a difference between the periods concerning the growth and development of Stylosanthes, and that period 2 was the most favourable to this forage plant. There was, also, different adaptability between the two varieties concerning the sowing times. The var. pauciflora was more adapted in period 1, and the var. vulgaris, in period 2. The data showed the possibility of selecting Stylosanthes cultivars adapted to different seasonal conditions.

O objetivo do presente trabalho foi estudar os efeitos de diferentes níveis de cálcio, no desenvolvimento de plantas de Stylosanthes guyanensis (Aubl.) Sw. cv. Cook, através de alguns parâmetros que compõem a análise fisiológica de crescimento. Para tal foram empregados 4 tratamentos a saber: T1 (200 mg de cálcio/litro); T2 (133,33 mg de cálcio/litro); T3 (66,66 mg de cálcio/litro) e T4 (omisso em cálcio). O trabalho foi montado em cultivo hidropônico, utilizando-se a solução nutritiva n° 1 de Hoagland & Arnon e conduzido em casa de vegetação. Foram realizadas observações em intervalos entre 5 coletas, distribuidas da seguinte maneira: intervalo I (entre 24° e 38° dia pós-germinação; intervalo II ( do 38° ao 52° dia); intervalo III (do 52° ao 66° dia) e intervalo IV (do 66° ao 80° dia). Os parâmetros estudados foram: a. taxa assimilatória líquida (TAL); b. taxa de crescimento relativo (TCR) e c. razão alfa. Dos resultados obtidos pode-se concluir que soluções com 200 e 133,33 mg de cálcio/litro, foram mais eficientes em promover o desenvolvimento de plantas de estilosantes.

O presente experimento teve como finalidade estudar os efeitos de níveis de cálcio no desenvolvimento de plantas de Stylosanthes guyanensis (Aubl.) Sw. cv Cook, em sua fase vegetativa, através dos parâmetros que compõem a análise fisiológica de crescimento e de que forma estes parâmetros se alteram, em função dos tratamentos utilizados. Delinearam-se quatro tratamentos, a saber: T1 (200 mg de cálcio/litro); T2 (133,33 mg de cálcio/litro); T3 (66,66 mg de cálcio/litro); T4 (omisso em cálcio). O experimento foi instalado em cultivo hidropônico, empregando-se solução nutritiva n° 1 de Hoagland & Arnon (1950) e conduzido em casa de vegetação. O experimento, inteiramente casualizado, obedeceu ao delineamento de parcelas sub-divididas, considerando-se as cinco coletas, realizadas a intervalos de 14 dias como parcelas, sendo os tratamentos empregados definidos como sub-parcelas. A influência dos diferentes tratamentos, foi avaliada através da área foliar (AF), área foliar específica (AFE), razão de massa foliar (RMF) e razão de área foliar (RAF). Dos resultados obtidos, pôde-se concluir que plantas de estilosantes, durante a fase de crescimento vegetativo, alcançaram melhor desenvolvimento em níveis de 200 mg de cálcio/litro de solução nutritiva.

O Stylosanthes spp. cv. Campo Grande é uma leguminosa recomendada para a recuperação de áreas de pastagens degradadas, entretanto são escassas as informações sobre seu desenvolvimento em solos compactados. Assim, objetivou-se com o presente trabalho avaliar o desenvolvimento do Stylosanthes cv. Campo Grande em diferentes níveis de densidade de um Latossolo Vermelho eutroférrico (LVef). O experimento foi desenvolvido no período entre setembro de 2007 e março de 2008, em cultivo protegido. O delineamento experimental utilizado foi inteiramente casualizado. Os tratamentos consistiram de cinco níveis de compactação do solo em sub-superfície (1,0; 1,2; 1,4; 1,6 e 1,8 Mg m-3) com quatro repetições. As unidades experimentais foram compostas por vasos montados a partir de anéis de PVC sobrepostos. No solo estudado (LVef) o máximo crescimento do Stylosanthes cv. Campo Grande ocorre com a densidade de solo em torno de 1,27 Mg m-3; e densidades de solo acima de 1,00 Mg m-3 limitam o desenvolvimento radicular desta leguminosa.

Effect of water potential on germination of seeds of Slylosanthes guianensis (Aubl.) Sw. To evaluate the water potential effect on germination of S. guianensis two experiments were performed.The first one used osmotic pre-treatment in the imbibition phase and after this period (14 h) the seeds were germinated on filter paper moistened with distilled water. In the second experiment, besides the imbibition phase, seeds were kept in a range of water potentials during all the process. The potentials ranged from 0 to -18 bars, with 3 bars increments, induced by mannitol or by polyethylene glycol. Each treatment was replicated 3 times with 100 seeds per replication. The seeds pre-treated during imbibition had high germination percentage, the highest being the ones in polhyetylene glycol. In the second experiment the polyethylene glycol solutions reduced dramatically the germination percentage in relation to mannitol. From -12 bars on germination ceased in the polyethylene glycol treatments, while in mannitol solution there was 52,67% of germination, in the same water potential.

An improved cultivar, based on 17 genotypes of S. capitata and six of S.macrocephala, was developed at the wEmbrapa Beef Cattle Research Center, Campo Grande, Brazil. The aim was to create durable, race non-specific resistance to anthracnose controlled by polygenic factors. A mass hybridisation technique was employed to produce a high degree of genetic diversity and sizeable quantities of seed of hybrid-derived progenies of Brazilian and Venezuelan genotypes of S. capitata. Outcrossing resulted in a significant improvement in the forage production of progeny of Venezuelan accessions. The multicross was evaluated in multilocational trials, each representing a large tract of country in the Cerrados ecosystem along a north-south transect from lat.6degrees S to 20degrees S. The genetic shift that occurred in S. capitata was a key element in the formation of the new cultivar. It is a complex mixture of two species, and a recombination of much desired forage traits of Brazilian x Venezuelan genotypes, high forage and seed yields coupled with anthracnose resistance. The new cultivar with its diverse genetic make-up has a wide application in the acid-soil savannas of tropical America. It was released by Embrapa for the Cerrados in 2000.

Hybrid stylos (Stylosanthes guianensis var. vulgaris x var. pauciflora) with durable, quantitative resistance to anthracnose, mid-season harvest maturity date (early-July), high DM and seed yields have been selected at the Embrapa Beef Cattle Research Center, Campo Grande, Brazil. The hybrids displayed improved forage traits in Brazil, the native habitat and major center of diversity of the species and its pathogen, as well as in vastly different ecosystems. Dry forage yields and anthracnose resistance of superior selections and their composites were equal, in some instances, significantly better, than those of cv. Mineir (a) over tildeo in multilocational trials situated in the Cerrados from lat. 6degrees S to lat. 20degrees S. Selected hybrids performed well in comparison with the highly successful CIAT 184 (cv.Reyan II) on Hainan Island, China. Composites have also shown good promise in seed multiplication plots in Queensland, Australia. A positive attribute of composite hybrids is their great genetic diversity in contrast to pure-line cultivars with a relatively narrow genetic base. These truly tropical forms of stylo are best adapted to regions with >1500mm average annual rainfall.

Using molecular markers, this work compares the genetic diversity in Colletotrichum gloeosporioides infecting species of the tropical forage legume Stylosanthes at the center of origin in Brazil and Colombia with that of Australia, China, and India, where Srylosanthes spp. have been introduced for commercial use. There was extensive diversity in the pathogen population from Brazil, Colombia, China, and India. The Australian pathogen population was least diverse probably due to its geographical isolation and effective quarantine. The extensive diversity in China and India means that threats from exotic pathogen races to Stylosanthes pastures can potentially come from countries outside the South American center of origin. In Brazil and India, both with native Stylosanthes populations, a high level of genetic differentiation in the pathogen population was associated with sites where native or naturalized host population was widely distributed. There was limited genetic diversity at germplasm evaluation sites, with a large proportion of isolates having identical haplotypes. This contrasts recent pathogenicity results for 78 of the Brazilian isolates that show hot spots of complex races are more common around research stations where host germplasm are tested, but few are found at sites containing wild host populations. For a pathogen in which the same races arise convergently from different genetic backgrounds, this study highlights the importance of using both virulence and selectively neutral markers to understand pathogen population structure.

The objective of this work was to evaluate the herbage availability, nutritive value, dry matter intake and grass and legume percentage in diet of crossbred Holstein-Zebu cows, in pasture with Brachiaria decumbens cv. Basilisk, Stylosanthes guianensis var. vulgaris cv. Mineirão and tree legumes. To estimate the fecal output, it was used 10 g cow -1 day -1 of chromium oxide during ten consecutive days. Extrusa samples were used to determine the chemical composition and in vitro dry matter digestibility. B. decumbens availability varied with climatic conditions, while S. guianensis availability decreased linearly along the experimental period. Dry matter intake was higher in May/2001 (1.9% body weight) and did not differ among other months (1.5% body weight). Low dry matter intake values were related to low in vitro dry matter digestibility coefficients (42.1 % to 48.0%) and high neutral detergent fiber content (70.2% to 79.4%). Dry matter intake was directly related to legume percentage in the pasture. This observation could indicate the potential of mixed pasture for improving nutritive value in dairy cattle diet.
