Intrahepatic cholestasis of pregnancy (ICP) is the most common cholestatic liver disease during pregnancy. The reported incidence varies from 0.4 to 15% of full-term pregnancies. The etiology is heterogeneous but familial clustering is known to occur. Here we have studied the genetic background, epidemiology, and long-term hepatobiliary consequences of ICP. In a register-based nation-wide study (n=1 080 310) the incidence of ICP was 0.94% during 1987-2004. A slightly higher incidence, 1.3%, was found in a hospital-based series (n=5304) among women attending the University Hospital of Helsinki in 1992-1993. Of these 16% (11/69) were familial and showed a higher (92%) recurrence rate than the sporadic (40%) cases. In the register-based epidemiological study, advanced maternal age and, to a lesser degree, parity were identified as new risk factors for ICP. The risk was 3-fold higher in women >39 years of age compared to women <30 years. Multiple pregnancy also associated with an elevated risk. In a genetic study we found no association of ICP with the genes regulating bile salt transport (ABCB4, ABCB11 and ATP8B1). The livers of postmenopausal women with a history of ICP tolerated well the short-term exposure to oral and transdermal estradiol, although the doses used were higher than those in routine clinical use. The response of serum levels of sex hormone-binding globulin (SHBG) to oral estradiol was slightly reduced in the ICP group. Transdermal estradiol had no effect on C-reactive protein (CRP) or SHBG. A number of liver and biliary diseases were found to be associated with ICP. Women with a history of ICP showed elevated risks for non-alcoholic liver cirrhosis (8.2 CI 1.9-36), cholelithiasis and cholecystitis (3.7 CI 3.2-4.2), hepatitis C (3.5 CI 1.6-7.6) and non-alcoholic pancreatitis (3.2 CI 1.7-5.7). In conclusion, ICP complicates around 1% of all full-term pregnancies in Finland and its incidence has remained unchanged since 1987. It is familial in 16% of cases with a higher recurrence rate. Although the cause remains unknown, several risk factors, namely advanced maternal age, parity and multiple pregnancies, can be identified. Both oral and transdermal regimens of postmenopausal hormone therapy (HT) are safe for women with a history of ICP when liver function is considered. Some ICP patients are at risk of other liver and biliary diseases and, contrary to what has been thought, a follow-up is warranted.

We investigated a 100 × 100 km high-salinity region of the North Atlantic subtropical gyre during the Sub-Tropical Atlantic Surface Salinity Experiment/Salinity Processes in the Upper-ocean Regional Study (STRASSE/SPURS) cruise from August 21, 2012, to September 9, 2012. Results showed great variability in sea surface salinity (SSS; over 0.3 psu) in the mesoscale, over 7 cm of total evaporation, and little diapycnal mixing below 36 m depth, the deepest mixed layers encountered. Strong currents in the southwestern part of the domain, and the penetration of freshwater, suggest that advection contributed greatly to salinity evolution. However, it was further observed that a smaller cyclonic structure tucked between the high SSS band and the strongest currents contributed to the transport of high SSS water along a narrow front. Cross-frontal transport by mixing is also a possible cause of summertime reduction of SSS. The observed structure was also responsible for significant southward salt transport over more than 200 km.

Uma das maiores causas de degradação dos revestimentos é a presença de sais solúveis, tanto nas zonas costeiras como nas zonas continentais. Estes sais podem ter origens distintas, tais como: humidade ascensional, nevoeiro salino, inundações ou ainda estarem presentes nos próprios materiais, como é o caso da utilização de areias mal lavadas. O local onde os sais cristalizam é dependente do tipo de transporte entre a alvenaria e o revestimento, assim como da severidade da envolvente ambiental (temperatura e humidade relativa). Com esta tese pretende-se desenvolver revestimentos de substituição compatíveis, eficazes e duráveis para alvenarias antigas sujeitas à ação severa da água (humidade ascensional através das fundações com elevada concentração de NaCl); desenvolver ensaios de envelhecimento acelerado baseados em ciclos de dissolução e cristalização que permitam simular a ação severa da água; perceber a influência de revestimentos com caraterísticas extremas de permeabilidade ao vapor de água aplicados nas duas faces do mesmo suporte (argamassas de cimento e resina versus argamassas de cal), e sujeitos à ação severa da água; perceber a influência de vários fatores, tais como : rasgos verticais contínuos executados no emboço e a utilização de hidrófugo no reboco no desempenho dos vários sistemas de revestimento desenvolvidos, quando sujeitos à ação severa da água;; utilizar a remoção eletrocinética de sais, em provetes simulando a influência da alvenaria e dos revestimentos, com o objetivo de reduzir a sua concentração de NaCl, para que no futuro possa ser aplicado de forma eficaz em paredes de edifícios antigos, enquanto ação de manutenção, e desta forma aumentar a durabilidade dos revestimentos. Para atingir os objetivos descritos, foram considerados: i) o desenvolvimento de provetes que permitam considerar os sistemas de revestimento desenvolvidos nesta tese e o suporte, e que simulem uma alvenaria revestida em ambas as faces, e com os quais seja possível a simulação da ação severa da água em laboratório e, ii) o desenvolvimento ciclos de dissolução e cristalização numa parede de grandes dimensões existente em laboratório, que permitam simular a ação severa da água em condições tão reais quanto possível. O objetivo final deste estudo é o desenvolvimento de um revestimento de substituição denominado “emboço ventilado” para paredes de edifícios antigos com revestimentos degradados devido à presença de humidade ascensional e sais solúveis. Pretende-se que este sistema de revestimento, composto por duas camadas de revestimento (camada base e reboco), seja compatível, durável e eficaz, e que funcione como um sistema de acumulação no qual os sais cristalizem na camada base do sistema de revestimento (executada com rasgos verticais) e não na alvenaria ou na camada exterior.

In the 1960s North Atlantic sea surface temperatures (SST) cooled rapidly. The magnitude of the cooling was largest in the North Atlantic subpolar gyre (SPG), and was coincident with a rapid freshening of the SPG. Here we analyze hindcasts of the 1960s North Atlantic cooling made with the UK Met Office’s decadal prediction system (DePreSys), which is initialised using observations. It is shown that DePreSys captures—with a lead time of several years—the observed cooling and freshening of the North Atlantic SPG. DePreSys also captures changes in SST over the wider North Atlantic and surface climate impacts over the wider region, such as changes in atmospheric circulation in winter and sea ice extent. We show that initialisation of an anomalously weak Atlantic Meridional Overturning Circulation (AMOC), and hence weak northward heat transport, is crucial for DePreSys to predict the magnitude of the observed cooling. Such an anomalously weak AMOC is not captured when ocean observations are not assimilated (i.e. it is not a forced response in this model). The freshening of the SPG is also dominated by ocean salt transport changes in DePreSys; in particular, the simulation of advective freshwater anomalies analogous to the Great Salinity Anomaly were key. Therefore, DePreSys suggests that ocean dynamics played an important role in the cooling of the North Atlantic in the 1960s, and that this event was predictable.

KCNQ1 (Kv7.1), together with its KCNE β subunits, plays a pivotal role both in the repolarization of cardiac tissue and in water and salt transport across epithelial membranes. Nedd4/Nedd4-like (neuronal precursor cell-expressed developmentally downregulated 4) ubiquitin-protein ligases interact with the KCNQ1 potassium channel through a PY motif located in the C terminus of KCNQ1. This interaction induces ubiquitylation of KCNQ1, resulting in a reduced surface density of the channel. It was reported recently that the epithelial sodium channel is regulated by the reverse process-deubiquitylation-mediated by USP2 (ubiquitin-specific protease 2).

The fractionation of major sea-water ions, or deviation in their relative concentrations from Standard Mean Ocean Water ratios, has been frequently observed in sea ice. It is generally thought to be associated with precipitation of solid salts at certain eutectic temperatures. The variability found in bulk sea-ice samples indicates that the fractionation of ions depends on the often unknown thermal history of sea ice, which affects the structure of pore networks and fate of solid salts within them. Here we investigate the distribution of ions in Arctic sea ice that is a few weeks old with a reconstructible thermal history. We separate the centrifugable (interconnected) and entrapped (likely disconnected) contributions to the ice salinity and determine their ion fractionation signatures. The results indicate that differential diffusion of ions, rather than eutectic precipitation of cryohydrates, has led to significant ion fractionation. The finding emphasizes the role of coupled diffusive–convective salt transport through complex pore networks in shaping the biogeochemistry of sea ice.

Direct observations, satellite measurements and paleo records reveal strong variability in the Atlantic subpolar gyre on various time scales. Here we show that variations of comparable amplitude can only be simulated in a coupled climate model in the proximity of a dynamical threshold. The threshold and the associated dynamic response is due to a positive feedback involving increased salt transport in the subpolar gyre and enhanced deep convection in its centre. A series of sensitivity experiments is performed with a coarse resolution ocean general circulation model coupled to a statistical-dynamical atmosphere model which in itself does not produce atmospheric variability. To simulate the impact of atmospheric variability, the model system is perturbed with freshwater forcing of varying, but small amplitude and multi-decadal to centennial periodicities and observational variations in wind stress. While both freshwater and wind-stress-forcing have a small direct effect on the strength of the subpolar gyre, the magnitude of the gyre's response is strongly increased in the vicinity of the threshold. Our results indicate that baroclinic self-amplification in the North Atlantic ocean can play an important role in presently observed SPG variability and thereby North Atlantic climate variability on multi-decadal scales.

The Atlantic subpolar gyre (SPG) is one of the main drivers of decadal climate variability in the North Atlantic. Here we analyze its dynamics in pre-industrial control simulations of 19 different comprehensive coupled climate models. The analysis is based on a recently proposed description of the SPG dynamics that found the circulation to be potentially bistable due to a positive feedback mechanism including salt transport and enhanced deep convection in the SPG center. We employ a statistical method to identify multiple equilibria in time series that are subject to strong noise and analyze composite fields to assess whether the bistability results from the hypothesized feedback mechanism. Because noise dominates the time series in most models, multiple circulation modes can unambiguously be detected in only six models. Four of these six models confirm that the intensification is caused by the positive feedback mechanism.

An ensemble of new, high-resolution records of surface ocean hydrography from the Indian-Atlantic oceanic gateway, south of Africa, demonstrates recurrent and high-amplitude salinity oscillations in the Agulhas Leakage area during the penultimate glacial-interglacial cycle. A series of millennial-scale salinification events, indicating strengthened salt leakage into the South Atlantic, appear to correlate with abrupt changes in the North Atlantic climate and Atlantic Meridional Overturning Circulation (AMOC). This interhemispheric coupling, which plausibly involved changes in the Hadley Cell and midlatitude westerlies that impacted the interocean transport at the tip of Africa, suggests that the Agulhas Leakage acted as a source of negative buoyancy for the perturbed AMOC, possibly aiding its return to full strength. Our finding points to the Indian-to-Atlantic salt transport as a potentially important modulator of the AMOC during the abrupt climate changes of the Late Pleistocene.

A sediment core from the western tropical Atlantic covering the last 21,000 yr has been analysed for centennial scale reconstruction of sea surface temperature (SST) and ice volume-corrected oxygen isotopic composition of sea water (delta18O(ivc-sw)) using Mg / Ca and delta18O of the shallow dwelling planktonic foraminifer Globigerinoides ruber (white). At a period between 15.5 and 17.5 kyr BP, the Mg / Ca SST and delta18O(ivc-sw), a proxy for sea surface salinity (SSS), reveals a warming of around 2.5 °C along with an increase in salinity. A second period of pronounced warming and SSS increase occurred between 11.6 and 13.5 kyr BP. Within age model uncertainties, both warming intervals were synchronous with air temperature increase over Antarctica and ice retreat in the southern South Atlantic and terminated with abrupt centennial scale SSS decrease and slight SST cooling in conjunction with interglacial reactivation of the meridional overturning circulation (MOC). We suggest that during these warm intervals, production of saline and warm water of the North Brazil Current resulted in pronounced heat and salt accumulation, and was associated with warming in the southern Atlantic, southward displacement of the intertropical convergence zone and weakened MOC. At the termination of the Younger Dryas and Heinrich event 1, intensification of cross-equatorial heat and salt transport caused centennial scale cooling and freshening of the western tropical Atlantic surface water. This study shows that the western tropical Atlantic served as a heat and salt reservoir during deglaciation. The sudden release of accumulated heat and salt at the end of Younger Drays and Heinrich event 1 may have contributed to the rapid reinvigoration of the Atlantic MOC.

Mutations in the sister of P-glycoprotein (Spgp) or bile salt export pump (BSEP) are associated with Progressive Familial Intrahepatic Cholestasis (PFIC2). Spgp is predominantly expressed in the canalicular membranes of liver. Consistent with in vitro evidence demonstrating the involvement of Spgp in bile salt transport, PFIC2 patients secrete less than 1% of biliary bile salts compared with normal infants. The disease rapidly progresses to hepatic failure requiring liver transplantation before adolescence. In this study, we show that the knockout of spgp gene in mice results in intrahepatic cholestasis, but with significantly less severity than PFIC2 in humans. Some unexpected characteristics are observed. Notably, although the secretion of cholic acid in mutant mice is greatly reduced (6% of wild-type), total bile salt output in mutant mice is about 30% of wild-type. Also, secretion of an unexpectedly large amount of tetra-hydroxylated bile acids (not detected in wild-type) is observed. These results suggest that hydroxylation and an alternative canalicular transport mechanism for bile acids compensate for the absence of Spgp function and protect the mutant mice from severe cholestatic damage. In addition, the spgp−/− mice display a significant increase in the secretion of cholesterol and phospholipids into the bile. This latter observation in spgp−/− mice suggests that intrahepatic, rather than intracanalicular, bile salts are the major driving force for the biliary lipid secretion. The spgp−/− mice thus provide a unique model for gaining new insights into therapeutic intervention for intrahepatic cholestasis and understanding mechanisms associated with lipid homeostasis.

Using a coupled model of intermediate complexity the sensitivity of the last glacial maximum (LGM) Atlantic meridional overturning circulation (AMOC) to the strength of surface wind-stress is investigated. A threshold is found below which North Atlantic deep water formation (DWF) takes place south of Greenland and the AMOC is relatively weak. Above this threshold, DWF occurs north of the Greenland-Scotland ridge, leading to a vigorous AMOC. This nonlinear behavior is explained through enhanced salt transport by the wind-driven gyre circulation and the overturning itself. Both pattern and magnitude of the Nordic Sea's temperature difference between strong and weak AMOC states are consistent with those reconstructed for abrupt climate changes of the last glacial period. Our results thus point to a potentially relevant role of surface winds in these phenomena.

Abnormal “polymer-in-salt” conduction behavior is observed in a solid electrolyte composed of lithium iodide (LiI) and 3-hydroxypropionitrile (HPN). Based on comprehensive investigations by X-ray diffraction (XRD) and Raman and infrared spectroscopy, this abnormal conduction behavior is attributed to the formation of new ionic associates [Lim +In−]· · ·N C (m> n) and the reinforced hydrogen bonding of I· · ·HO in the electrolyte at high LiI concentrations.
