Las respuestas de los componentes del rendimiento y de la calidad del aceite a la temperatura aún no han sido estudiadas en olivo ni en otras oleaginosas que acumulan aceite principalmente en mesocarpo (cerca de 95 por ciento del total en el caso de olivo). La información disponible en olivo se basa en correlaciones ligadas a variaciones en ubicación geográfica, altitud y años en los que pudieron covariar con la temperatura otros factores (radiación, disponibilidad hídrica, nutrición, etc). El objetivo de esta tesis fue evaluar el rol de la temperatura durante el crecimiento de frutos de olivo sobre el peso seco de los mismos, su contenido y concentración de aceite y las proporciones de ácidos grasos del aceite. Se realizaron experimentos manipulativos aplicados a ramas fructíferas creciendo a campo, utilizando cámaras transparentes con control y registro de temperatura. Los rangos de temperaturas medias alcanzados (entre 15-32 ºC) fueron mucho mayores a los rangos de 1 y 4 ºC logrados en estudios correlativos y permitieron generar relaciones funcionales para el peso seco del fruto, su concentración de aceite, y la proporción de ácidos grasos en el aceite. El contraste entre aplicaciones de temperaturas mayores o menores al ambiente durante períodos largos (114 días) y cortos (30 días) demostraron que tanto la proporción de aceite del fruto como la de ácido oleico en el aceite de fruto entero (mesocarpo + semilla) disminuye al aumentar la temperatura. Si bien en ambas estructuras del fruto la temperatura tuvo un efecto negativo sobre la concentración de aceite, las respuestas a la temperatura de la proporción de ácidos grasos de la semilla y el mesocarpo, medidos en forma separada, difirieron entre sí. Los patrones de respuesta de las proporciones de ácidos grasos en el aceite de semilla mostraron similitudes con las conocidas para especies oleaginosas de semilla como girasol. Por contraste, en el aceite de mesocarpo de olivo la proporción de ácido oleico disminuyó con el aumento de la temperatura, patrón opuesto a lo manifestado en semillas de olivo y en las de oleaginosas anuales. En esta tesis también se evaluó el efecto de la temperatura mínima nocturna sobre el crecimiento de los frutos y la calidad del aceite. Los resultados sugieren que la temperatura mínima nocturna y la amplitud térmica diarias son las dimensiones del régimen térmico diario con los que mejor se asociaron los cambios en el peso seco del fruto, la proporción de aceite y las proporciones de ácidos grasos en el mismo. Los resultados de esta tesis sirven para guiar la selección de sitios de plantación de nuevos olivares basados en los registros térmicos zonales, y para explicar los desajustes de la proporción de ácidos grasos del aceite de oliva producido en el NOA y la normativa del Consejo Oleícola Internacional (COI). Las relaciones funcionales entre las variables respuesta y la temperatura definidas podrán incorporarse en modelos de simulación del rendimiento y calidad que se desarrollen en el futuro. Tomados en conjunto con la tendencia al aumento de la temperatura, producto del calentamiento global, los resultados permitirán estimar la magnitud de los impactos negativos que podrían afectar el rendimiento del cultivo y la calidad del aceite.

Olive (Olea europaea L.), one of the main crops in the Mediterranean basin, is mainly propagated by cuttings, a classical propagation method that relies on the ability of the cuttings to form adventitious roots. While some cultivars are easily propagated by this technique, some of the most interesting olive cultivars are considered difficult-to-root which poses a challenge for their preservation and commercialization. Therefore, increasing the current knowledge on adventitious root formation is extremely important for species like olive. This research focuses on evaluating the role of free auxins and oxidative enzymes on adventitious root formation of two olive cultivars with different rooting ability - ‘Galega vulgar’ (difficult-to-root) and ‘Cobrançosa’ (easy-to-root). In this context, free auxin levels and enzyme activities were determined in in vitro-cultured ‘Galega vulgar’ microshoots and in semi-hardwood cuttings of cvs. ‘Galega vulgar’ and ‘Cobrançosa’. To attain this goal, an analytical method for the quantification of free indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) was developed, which is based on dispersive liquid-liquid microextraction followed by microwave derivatization (DLLME-MAD) and gas chromatography-mass spectrometry (GC/MS) analysis. The developed method was validated in terms of linearity, recovery, limit of detection (LOD) and limit of quantification (LOQ) and proved to be useful in the analysis of two very different types of plant tissues. The results from auxin quantification in olive samples point at a relationship between free auxin levels and rooting ability of both microshoots and semihardwood cuttings. A defective IBA-IAA conversion, resulting in a peak of free IAA during initiation phase, seems to be associated with low rooting ability. Likewise, differences in the activity of oxidative enzymes also appear to be related with rooting ability. Higher polyphenol oxidases (PPO) activity is likely related with an easyto- root behavior, while the opposite is true for peroxidases (POX) (including IAA oxidase (IAAox)) activity. A possible hypothesis for adventitious root formation in olive microcuttings is presented herein for the first time. Free auxins, oxidative enzymes, alternative oxidase (AOX) and reactive oxygen species (ROS) are some of the factors that may be involved in this highly complex physiological process. Interestingly, while temporal changes in auxin levels were similar between microshoots and semihardwood cuttings, the conclusions obtained from enzyme activity results in microshoots didn’t translate to semi-hardwood tissues, showing the emerging need for adaptation of classical agronomical research studies to modern techniques; Resumo: Procurando compreender o papel das auxinas e enzimas oxidativas na formação de raízes adventícias em cultivares de oliveira (Olea europaea L.) A oliveira (Olea europaea L.) é uma das principais culturas da bacia Mediterrânica e é propagada maioritariamente por estacaria, um processo altamente dependente da capacidade das estacas para formar raízes adventícias. Enquanto algumas cultivares são fáceis de propagar desta forma, algumas das cultivares de oliveira mais interessantes são consideradas difíceis de enraizar, o que dificulta a sua preservação e comercialização e torna extremamente importante aprofundar o conhecimento sobre o enraizamento adventício desta espécie. Este trabalho foca-se na avaliação do papel das auxinas livres e das enzimas oxidativas na formação de raízes adventícias em duas cultivares de oliveira com diferente capacidade de enraizamento - ‘Galega vulgar’ (difícil de enraizar) e ‘Cobrançosa’ (fácil de enraizar). Neste contexto, determinaram-se os níveis de auxinas livres e as actividades de enzimas oxidativas em microestacas de ‘Galega vulgar’ cultivadas in vitro bem como em estacas semi-lenhosas das cvs. ‘Galega vulgar’ e ‘Cobrançosa’. Para tal foi necessário desenvolver uma metodologia analtica para a quantificação de ácido indol-3-acético (IAA) e ácido indol-3-butírico (IBA), baseada em microextracção dispersiva lquido-lquido (DLLME) seguida de derivatização em microondas (MAD) e análise por cromatografia gasosa acoplada a espectrometria de massa (GC/MS). O método desenvolvido foi validado em termos de linearidade, recuperação, limite de detecção (LOD) e limite de quantificação (LOQ), e mostrou-se eficaz na análise de dois tipos de tecidos vegetais bastante diferentes. Os resultados da análise de auxinas em amostras de oliveira apontam para uma possível relação entre os níveis de auxinas livres e a capacidade de enraizamento, tanto em microestacas como em estacas semi-lenhosas. Uma conversão IBA-IAA deficiente, que resulta num pico de IAA durante a fase de iniciação, parece estar associada à baixa capacidade de enraizamento. Por outro lado, a capacidade de enraizamento também parece estar relacionada com diferenças na actividade de enzimas oxidativas. Comportamentos fáceis de enraizar estão associados a actividade mais elevada das polifenoloxidases (PPO), enquanto o oposto é verdade para a actividade das peroxidases (POX) (incluindo a IAA oxidase (IAAox)). Neste trabalho propõe-se pela primeira vez uma possível explicação para o enraizamento adventício em microestacas de oliveira. Auxinas livres, enzimas oxidativas, oxidase alternativa (AOX) e espécies reactivas de oxigénio (ROS) são alguns dos factores envolvidos neste processo fisiolgico altamente complexo. Curiosamente, enquanto as alterações temporais nos níveis de auxinas foram semelhantes entre microestacas e estacas semi-lenhosas, o mesmo não se observou relativamente à actividade enzimática, o que mostra a necessidade de adaptação dos estudos agronómicos tradicionais às técnicas correntes.

The activity of oxidative enzymes and the levels of free auxins were determined during adventitious root formation in olive explants. Rooting trials were performed both with in vitro-cultured micro shoots of the cultivar ‘Galega Vulgar’, treated with indole-3-butyric acid (IBA) and with salicylhydroxamic acid(SHAM) + IBA, as well as with semi-hardwood cuttings of the cultivars ‘Galega Vulgar’ (difficult-to-root)and ‘Cobrançosa’ (easy-to-root), treated with IBA. The auxin (IBA) was used in all experiments as a rooting promoter, while SHAM was used in micropropagation trials as rooting inhibitor, providing a negative control. Free indole-3-acetic acid (IAA) and IBA concentrations were determined in microshoots, as well as in semi-hardwood cuttings, throughout the rooting period at pre-established time-points. At the sametime-points, the enzymatic activity of polyphenol oxidases (PPO), peroxidases (POX), and IAA oxidase(IAAox) was evaluated in the microshoots. Microshoots treated with SHAM + IBA revealed higher POX and IAAox activity, as well as lower PPO activity, than those treated only with IBA. IAA levels were higher in IBA-treated microshoots during induction phase, but lower during early initiation phase. Incontrast, free IBA levels were higher in microshoots treated with SHAM + IBA during induction, but lower during initiation. A similar pattern of free auxin levels was observed in semi-hardwood cuttings of the two contrasting cultivars under evaluation. The similarities found on the auxin patterns of microshoots treated with SHAM and those of semi-hardwood cuttings of the difficult-to-root olive cultivar allow considering SHAM a reliable control for when simulation of a difficult-to-root behavior is necessary. The inhibitory effect of SHAM in root formation could be related with 1) the inhibition of alternative oxidase(AOX), leading to a down regulation of phenylpropanoid biosynthetic pathways, which would decrease the concentration of phenolic substrates for PPO; 2) an increase in IAAox activity resulting in lower free IAA levels or; 3) a defective conversion of IBA into IAA.

A oliveira tem sido multiplicada ao longo dos tempos por métodos convencionais de propagação vegetativa como a enxertia e a estacaria lenhosa e semilenhosa. No entanto, estes métodos revelam-se lentos ou ineficientes para determinadas cultivares. No caso da cv. ‘Galega Vulgar’, ainda com grande expressão no olival português, e de difícil enraizamento por estacaria semilenhosa, tem sido usada a micropropagação de modo a contornar essas limitações e assim obter um elevado número de plantas em curto período de tempo. O custo final de produção por este processo ainda é elevado, podendo comprometer a sua aplicação a nível comercial. Grande parte dos custos estão relacionados com a fase de enraizamento in vitro que carece de ambiente estéril e condições de assépsia para a sua execução. Com vista a uma redução de custos associados a esta fase de produção, pretendeu-se com este trabalho testar a viabilidade do enraizamento ex vitro, na ausência de condições de assépsia. Este método poderá permitir uma significativa redução da mão-de-obra, ao mesmo tempo que facilitará a aclimatização das plantas e a obtenção de um sistema radicular de melhor qualidade. Compararam-se as taxas de enraizamento in vitro (controlo), com as obtidas ex vitro. Foram utilizados explantes provenientes de dois clones da cv. ‘Galega Vulgar’, (cl. 1441 e cl. 2022) cultivados e mantidos in vitro há vários anos no Laboratório de Melhoramento e Biotecnologia da Universidade de Évora. Para alm do clone foi avaliada a influência do tipo de estaca (basal e apical), da hormona de enraizamento (AIB e ANA), da sua concentração (540 e 3000 ppm) e ainda de dois substratos, Preformas Jiffy® e pastilhas de fibra coco. Os melhores resultados foram obtidos com o clone 1441 em pastilhas de fibra de coco prensada, com o uso de estacas basais. Quanto à auxina, não se observaram diferenças significativas entre a utilização de ANA na concentração de 540 ppm e AIB na concentração de 3000 ppm. A aclimatização das plantas foi conseguida com taxas elevadas de sucesso, independentemente do tratamento utilizado. Conclui-se que a aplicação do método de enraizamento ex vitro simplifica procedimentos e mantém taxas de enraizamento elevadas, conduzindo assim a uma efetiva redução de tempo e custos associados; Simplifying procedures for in vitro propagation of olive “Olea europaea L.” Abstract: The olive tree has been multiplied throughout the ages by conventional methods of vegetative propagation such as grafting and wood or softwood cuttings. These propagation methods are somehow inefficient for certain cultivars. For the CV. ‘Galega Vulgar‘, still with great expression in Portuguese olive orchards, propagation has been attempted by in vitro culture in order to circumvent these limitations and so obtain a large number of plants in short time period. The final production fees associated to this process are still high which may compromise its application to a commercial level. Most of this process fees are related to the in vitro rooting phase which lacks sterile and aseptic conditions for its implementation. Aiming to reduce the costs associated with this production phase, this work tested the feasibility of the ex vitro rooting in the absence of aseptic conditions, which can allow a significant reduction of the manpower involved and an easier plant acclimatization due to its transplant with a balled-root system. In vitro rooting rates (control) were compared with those obtained with the ex vitro experiments. Explants from two clones of the cv. ‘Galega Vulgar‘ (cl. 1441 and cl. 2022), grown and maintained in vitro for several years in the Laboratory of Biotechnology and Plant Breeding of the University of Évora, were used in the trials. In addition to the clone, the effect of the cutting type (basal and apical), the rooting hormone (AIB and ANA), their concentration (540 and 3000 ppm) and two substrates, Preformas Jiffy ® and pressed coco fiber pellets, were also evaluated. The best results were obtained with the clone 1441, when rooted in pressed coco fiber pellets, using basal cuttings. Under this conditions no significant differences were observed between the use of ANA at 540 ppm or AIB in the 3000 ppm. Acclimatization of plants was achieved with high rates of success, regardless of the treatment used. It can be concluded that the application of the ex vitro rooting method allows to maintain high rooting rates, contributing for an effective reduction of time and fees of the rooting process.

大车前(Plantago major L. "Giant Turkish.")不仅有很高的药用价值,在生态学研究方面也是重要模式植物。大车前的组织培养工作,目前报道很少。对其组织培养体系的建立,为筛选大车前耐盐突变体和基因转化建立高效的体外再生系统和实验平台体系。通过愈伤组织诱导和直接不定芽再生途径, 建立了大车前(Plantago major L. "Giant Turkish.")的快速高效再生系统。叶片外植体在含有1.0 mg/L NAA的MS培养基中培养3周后,形成愈伤组织,愈伤组织在含4.0 mg/L 6-BA的MS培养基中成功再生,得到完整植株。种子外植体在含0.2 mg/L IAA和1.0 mg/L TDZ的MS培养基中培养4周后产生大量的丛生芽,对9株再生植株进行RAPD检测表明,部分植株在DNA水平上发生了变异。 植物抵御盐胁迫的一个重要机制是在液泡中积累Na+,从而使细胞质内Na+保持在较低水平,并且降低细胞渗透势。Na+运输到液泡是由液泡Na+/H+逆向转运蛋白完成的。本实验室已从盐生植物盐角草(Salicornia europaea)和番杏(Tetragonia tetragonioides)中分别克隆得到SeNHX1和TtNHX1基因。本文研究了SeNHX1和TtNHX1基因在酵母突变体里的作用。TtNHX1和SeNHX1蛋白在缺陷型酵母菌株里的表达能够提高这些菌株对NaClLiCl潮霉素的抗性,提高到与野生型相当的抗性水平。说明TtNHX1和SeNHX1有着与酵母ScNHX1相似的细胞定位和作用机制,是ScNHX1的功能类似蛋白。

After detachment from benthic habitats, the epibiont assemblages on floating seaweeds undergo substantial changes, but little is known regarding whether succession varies among different seaweed species. Given that floating algae may represent a limiting habitat in many regions, rafting organisms may be unselective and colonize any available seaweed patch at the sea surface. This process may homogenize rafting assemblages on different seaweed species, which our study examined by comparing the assemblages on benthic and floating individuals of the fucoid seaweeds Fucus vesiculosus and Sargassum muticum in the northern Wadden Sea (North Sea). Species richness was about twice as high on S. muticum as on F. vesiculosus, both on benthic and floating individuals. In both seaweed species benthic samples were more diverse than floating samples. However, the species composition differed significantly only between benthic thalli, but not between floating thalli of the two seaweed species. Separate analyses of sessile and mobile epibionts showed that the homogenization of rafting assemblages was mainly caused by mobile species. Among these, grazing isopods from the genus Idotea reached extraordinarily high densities on the floating samples from the northern Wadden Sea, suggesting that the availability of seaweed rafts was indeed limiting. Enhanced break-up of algal rafts associated with intense feeding by abundant herbivores might force rafters to recolonize benthic habitats. These colonization processes may enhance successful dispersal of rafting organisms and thereby contribute to population connectivity between sink populations in the Wadden Sea and source populations from up-current regions.

Background Epidemiological studies have shown a reduced incidence of cardiovascular disease in the Mediterranean population attributed to the consumption of dietary olive oil rich in antioxidants. This has lead to increased interest in the antioxidant properties of other phenolic compounds of olive tree products. It has been suggested that olive leaf extract may also have health benefits due to its antioxidant and anti-inflammatory activities. Antioxidants can prevent the effects of oxidative metabolism by scavenging free radicals and decreasing the hyperactivity of platelets associated with the development of occlusive thrombosis. No studies to date have investigated the effects of olive leaf extract on platelet function to our knowledge. Improved understanding of the antioxidant properties of olive leaf extract and its effect on platelet function could lead to improved cardiovascular health. Objective The current study used an olive leaf extract prepared from the Olea europaea L. tree. The aim was to determine if polyphenols in olive leaf extract would reduce platelet activity and, to establish an optimal dose in vitro that would reduce platelet aggregation and ATP release. Design Eleven subjects with normal platelet counts (150–400 x 109/L) were recruited for the current in vitro study. Olive leaf extract was added to citrated whole blood to obtain five concentrations ranging from 5.4 ug/mL to 54.0 ug/mL for a dose response curve. Baseline samples, without olive leaf extract were used as a negative control for each subject. After 2 hours incubation with olive leaf extract samples were analyzed for platelet aggregation and ATP release from platelets stimulated by the addition of collagen. Results Whole blood analysis (n=11) showed a clear dose-dependant reduction in platelet aggregation with the increasing olive leaf extract concentrations (p<0.0001). There was also a similar decrease in ATP release from collagen stimulated platelets (p=0.02). Conclusion In the current study the olive leaf extract obtained from Olea europaea L. inhibited platelet aggregation and ATP release from collagen stimulated platelets in vitro. This study suggests olive leaf extract may prevent occlusive thrombosis by reducing platelet hyperactivity.

This work reports the investigation of tolerance to salt in two year-old olive trees (Olea europaea L.) of three varieties, Arbequina, Cobrançosa and Galega vulgar. Plants were grown in 10 L plastic pots containing approximately 9 Kg of a sandy granitic soil, on a greenhouse. For 3 months (from the beginning of February to the end of April 2012), they were subjected to three levels of salinity in the irrigation water, 0 mM, 80 mM and 200 mM NaCl (6 plants per salinity level in a total of 18 plants of each variety),. Stomatal conductance (gs) and relative leaf chlorophyll content were assessed on each plant in February, March and April. Mid-day leaf water potential () and soil salinity were measured at the end of the experiment (April). On average, concerning all treatments and dates of determination, stomatal conductance of Arbequina and Galega vulgar was quite similar, around 40 mmol m-2 s-1, but Cobrançosa had a value of gs 36% higher, almost 50% higher (61 mmol m-2 s-1) when compared with the controls (0 mM salt) of the other two varieties. In percentage of controls, there was little difference in gs between varieties and between salinities during February and March. In contrast, in April, after about 90 days of exposure to salt, there was a clear decrease in gs with salt irrigation, proportional to salt concentration. Compared with controls, plants irrigated with 200 mM salt showed around 80% (Arbequina) or 85% (Cobrançosa and Galega vulgar) decrease in gs. Chlorophyll content of leaves showed less than 5% difference between varieties on the average of all treatments and dates of determination. During the course of this experiment, the salinity levels used did not show any relevant effect on chlorophyll content. Overall, at the end of the experimental period (April), leaf water potential () at midday was significantly higher in Cobrançosa (-1,4 MPa) than in Galega vulgar (-1,7 MPa) or Arbequina (-1,8 MPa), and salt decreased  of control plants (-1,25 MPa) by an average 30% (with 80 mM) and 65% (with 200 mM). At the end of the experiment, salinity in the soil irrigated with 0 mM, 80 mM or 200 mM NaCl was, on average of all varieties, 0,2 mS, 1,0 mS or 2,0 mS, respectively. Soil salinity was quite similar in Arbequina and Galega vulgar but about 35% lower in the pots of Cobrançosa, on average of all salt-irrigation levels. Plants of Cobrançosa had higher stomatal conductance, however they showed higher water potential and lower salinity in the soil. These apparently contradictory results seem to suggest that Cobrançosa responds to salt differently from the other two varieties. This issue needs further investigation.

Soil salinization is a problem in the Mediterranean region. This paper reports a research on the response to salt in two year-old olive trees (Olea europaea L.) of three Iberian varieties: Arbequina, Cobrançosa and Galega Vulgar. Plants were grown in plastic pots containing approximately 9 Kg of a sandy granitic soil, on a greenhouse at the University of Évora since February 2010. The experiment went from February to April 2012. As a rule, plants were watered every other day alternating salt solution (0 mM, 80 mM or 200 mM NaCl) or tap water. After three months irrigation with the different NaCl solutions, soil electric conductivity and soil water content were significantly higher on salt-irrigated pots. Salt also decreased significantly stomatal conductance (gs) and mid-day leaf water potential (), Cobrançosa having in general higher gs and but lower SLA than the two other varieties. Chlorophyll content of leaves was not affected by salt after this three months exposure to NaCl but was significantly higher on Arbequina and lower on Cobrançosa. In general, hyperspectral reflectance indexes did not show significant correlations with salt irrigation, except for the Photochemical Reflectance Index (PRI) which was clearly lower on plants of all three varieties irrigated with salt. Interestingly, Cobrançosa showed frequently vegetation indexes different from the other two varieties.

No presente trabalho desenvolveram-se estudos visando a valorização do coberto vegetal da Ilha de Porto Santo, através de duas metodologias de investigação complementares: a) preservação e reintrodução na Ilha de uma espécie endémica e em risco do Arquipélago da Madeira (Olea maderensis) e de uma espécie naturalizada (Olea europaea ssp. europaea var. sylvestris), recorrendo para o efeito a técnicas de biotecnologia (micropropagação); b) análise da percepção da comunidade local e visitante sobre o fenómeno da desertificação e a valorização do coberto vegetal bem como a sua aceitação relativamente à aplicação de técnicas de biotecnologia (para micropropagar e reintroduzir espécies de oliveira na Ilha) para minimização do processo de desertificação. A dissertação estrutura-se em quatro partes principais. A Parte I caracteriza a Ilha de Porto Santo em termos geográficos, geolgicos, climáticos, sócio-economicos e do uso do solo. Enquadra, ainda, o problema da desertificação, através da caracterização/evolução do coberto vegetal ao longo dos anos. Finalmente apresentam-se os objectivos gerais deste estudo. A Parte II centra-se no desenvolvimento de metodologias no âmbito da biotecnologia vegetal para propagação de espécies de O. maderensis e O. europaea ssp. europaea var. sylvestris. em larga escala. Esta parte está dividida em seis capítulos. O Capítulo II.1 aborda a distribuição geográfica das espécies de oliveira e faz uma revisão bibliográfica dos aspectos mais importantes da micropropagação de oliveira (O. europaea L.), principalmente através da micropropagação por estimulação de gomos axilares. No final deste capítulo apresentam-se os objectivos específicos desta investigação. No Capítulo II.2 faz-se a caracterização genética de genótipos de O. maderensis do Arquipélago da Madeira através da análise da ploidia e do conteúdo em DNA por citometria de fluxo (FCM) e através da detecção de polimorfismos por análise de microssatélites (SSR). Nesta análise usaram-se ainda outros genótipos, nomeadamente: O. europaea ssp. europaea var. sylvestris, O. cerasiformes e O. europaea ssp. europaea var. europaea. Este estudo contribuiu para uma melhor caracterização desta espécie e permitiu a detecção de um nível de ploidia novo no género Olea (tetraploidia). O Capítulo II.3 descreve a optimização das condições de cultura in vitro (e.g. desinfecção, meio de cultura e enraizamento) para propagar e preservar a O. maderensis. Avalia-se ainda a “performance” dos rebentos in vitro (taxas de crescimento, avaliação da aparência das folhas e estudos fisiolgicos), de modo a confirmar a optimização das condições de propagação. Neste capítulo define-se um meio novo (OMG) para propagação desta espécie endémica. O Capítulo II.4 descreve dois protocolos de micropropagação e aclimatização de ambas as espécies (O. maderensis e O. europaea ssp. europaea var. sylvestris) e a qualidade das plantas (“true-to-type”) é avaliada através da possível ocorrência de variabilidade genética através de FCM (ploidia) e SSRs. O Capítulo II.5 descreve um protocolo eficiente de aclimatização ao campo de O. maderensis e avalia a “performance” das plantas micropropagadas no campo através da análise de parâmetros fisiolgicos durante o processo. O Capítulo II.6 apresenta os estudos em curso relativamente às plantas de O. maderensis em aclimatização no campo, bem como a introdução de plantas micropropagadas num outro local da Ilha com um maior grau de degradação dos solos. Estas estratégias estão a ser aplicadas juntamente com a DRFRAM, no âmbito de programas de florestação em curso. Finalmente é realada a necessidade de estudos semelhantes com outras espécies nativas. Na Parte III, são apresentados os estudos sobre a percepção da comunidade local relativamente à valorização do coberto vegetal para a minimização dos processos de degradação dos solos/desertificação. A introdução faz o enquadramento teórico sobre o fenómeno da desertificação, particularmente na Ilha de Porto Santo e sobre a percepção social da desertificação. São ainda apresentados os objectivos específicos desta investigação. A metodologia adoptada recorreu à aplicação de inquéritos por questionário à população residente e aos visitantes da Ilha de Porto Santo e ainda a realização de inquéritos por entrevista a algumas entidades e especialistas. Estes estudos permitiram verificar que existe uma nítida consciência da situação de risco da ilha, das medidas tomadas e a tomar e da premência da resolução do problema. Face ao recurso de estratégias alternativas envolvendo biotecnologia, e apesar de existir algum desconhecimento, concluiu-se ainda que a população manifesta aceitação, desde que essas estratégias valorizem o coberto vegetal e, assim, ajudem a combater a degradação biofísica dos solos. Finalmente são apresentadas as conclusões e algumas recomendações. Na Parte IV apresentam-se as conclusões gerais e perspectivas futuras, onde o potencial ambiental destas oliveiras bravas micropropagadas é destacado, bem como é considerado o alargamento da aplicação destas estratégias a outras espécies indígenas em risco, nesta Ilha (e noutros locais). São ainda resumidas as principais visões da população e das entidades e dos especialistas que poderão contribuir para apoiar a elaboração de medidas de mitigação e prevenção no combate ao processo de degradação dos solos/desertificação em curso.

The aim of this study was to know the yeast biodiversity from fresh olive (Olea europaea L.) fruits, olive paste (crush olives) and olive pomace (solid waste) from Arbequina and Cornicabra varieties. Yeasts were isolated from fruits randomly harvested at various olive groves in the region of Castilla La Mancha (Spain). Olive paste and pomace, a byproduct of the processing of this raw material, were also collected in sterile flasks from different oil mills. Molecular identification methodology used included comparison of polymerase chain reaction (PCR) amplicons of their 5.8S rRNA gene and internal transcribed spacers ITS1 and ITS2 followed by restriction pattern analysis (RFLP). For some species, sequence analysis of the 5.8S rDNA gene was necessary. The results were compared to sequences held in public databases (BLAST). These techniques allowed to identify fourteen different species of yeasts, belonging to seven different genera (Zygosaccharomyces, Pichia, Lachancea, Kluyveromyces, Saccharomyces, Candida, Torulaspora) from the 108 yeast isolates. Species diversity was thus considerable: Pichia caribbica, Zygosaccharomyces fermentati (Lachancea fermentati) and Pichia holstii (Nakazawaea holstii) were the most commonly isolated species, followed by Pichia mississippiensis, Lachancea sp., Kluyveromyces thermotolerans and Saccharomyces rosinii. The biotechnological properties of these isolates, was also studied. For this purpose, the activity of various enzymes (beta-glucosidase, beta-glucanase, carboxymethylcellulase, polygalacturonase, peroxidase and lipase) was evaluated. It was important that none of species showed lipase activity, a few had cellulase and polygalacturonase activities and the majority of them presented beta-glucanase, beta-glucosidase and peroxidase activities. The Mediterranean Sea is expected to react faster to global change compared to the ocean and is already showing more pronounced warming and acidification rates. A study performed along the Italian western coast showed that porosity of the skeleton increases with temperature in the zooxanthellate (i.e. symbiotic with unicellular algae named zooxanthellae) solitary scleractinian Balanophyllia europaea while it does not vary with temperature in the solitary non-zooxanthellate Leptopsammia pruvoti. These results were confirmed by another study that indicated that the increase in porosity was accompanied by an increase of the fraction of the largest pores in the pore-space, perhaps due to an inhibition of the photosynthetic process at elevated temperatures, causing an attenuation of calcification. B. europaea, L. pruvoti and the colonial non-zooxanthellate Astroides calycularis, transplanted along a natural pH gradient, showed that high temperature exacerbated the negative effect of lowered pH on their mortality rates. The growth of the zooxanthellate species did not react to reduced pH, while the growth of the two non-zooxanthellate species was negatively affected. Reduced abundance of naturally occurring B. europaea, a mollusk, a calcifying and a non-calcifying macroalgae were observed along the gradient while no variation was seen in the abundance of a calcifying green alga. With decreasing pH, the mineralogy of the coral and mollusk did not change, while the two calcifying algae decreased the content of aragonite in favor of the less soluble calcium sulphates and whewellite (calcium oxalate), possibly as a mechanism of phenotypic plasticity. Increased values of porosity and macroporosity with CO2 were observed in B. europaea specimens, indicating reduces the resistance of its skeletons to mechanical stresses with increasing acidity. These findings, added to the negative effect of temperature on various biological parameters, generate concern on the sensitivity of this zooxanthellate species to the envisaged global climate change scenarios.

We present a detailed palaeoclimate analysis of the Middle Miocene (uppermost Badenian-lowermost Sarmatian) Schrotzburg locality in S Germany, based on the fossil macro- and micro-flora, using four different methods for the estimation of palaeoclimate parameters: the coexistence approach (CA), leaf margin analysis (LMA), the Climate-Leaf Analysis Multivariate Program (CLAMP), as well as a recently developed multivariate leaf physiognomic approach based on an European calibration dataset (ELPA). Considering results of all methods used, the following palaeoclimate estimates seem to be most likely: mean annual temperature ~15-16°C (MAT), coldest month mean temperature ~7°C (CMMT), warmest month mean temperature between 25 and 26°C, and mean annual precipiation ~1,300 mm, although CMMT values may have been colder as indicated by the disappearance of the crocodile Diplocynodon and the temperature thresholds derived from modern alligators. For most palaeoclimatic parameters, estimates derived by CLAMP significantly differ from those derived by most other methods. With respect to the consistency of the results obtained by CA, LMA and ELPA, it is suggested that for the Schrotzburg locality CLAMP is probably less reliable than most other methods. A possible explanation may be attributed to the correlation between leaf physiognomy and climate as represented by the CLAMP calibration data set which is largely based on extant floras from N America and E Asia and which may be not suitable for application to the European Neogene. All physiognomic methods used here were affected by taphonomic biasses. Especially the number of taxa had a great influence on the reliability of the palaeoclimate estimates. Both multivariate leaf physiognomic approaches are less influenced by such biasses than the univariate LMA. In combination with previously published results from the European and Asian Neogene, our data suggest that during the Neogene in Eurasia CLAMP may produce temperature estimates, which are systematically too cold as compared to other evidence. This pattern, however, has to be further investigated using additional palaeofloras.
