Case study simulations with idealized tracers have been used to determine the relationship between the dynamics and conceptual representations of different midlatitude frontal systems and the amount, distribution, and time scale of boundary layer ventilation by these systems. The key features of ventilation by a kata– and ana–cold frontal system are found to be quantitatively and also often qualitatively similar to the main ventilation pathways, which are the conveyor belts, cloud head, and other convection. The conveyor belts and cloud head occur within cloud, implying that they can be identified using satellite imagery. Differences in the transport by the two systems can be related to their conceptual representations and include a sensitive dependence on the diurnal cycle for the kata- but not the ana-cold frontal case.

An experiment was designed to test the response of growing pullets to two changes in photoperiod (an increase from 8 to 14 h followed 5 weeks later by the reverse change, or a decrease from 14 to 8 h followed by an increase). The first change was made either at 35 days or at 56 days of age, to test the influence of age on the responses observed. Control groups were kept oil constant 8-h and constant 14-h photoperiods and the responses to appropriate single changes were also tested. Mean age at first egg varied from 111 days for birds given a single increment at 56 days to 166 days for pullets given an increase in photoperiod at 35 days followed by a reduction at 70 days. Responses to the single changes confirmed earlier reports that sensitivity to change in photoperiod varies with age ill a manner that is quantitatively predictable. Responses to the double changes could be explained by Postulating that the initial change altered the 'physiological age' of the bird to all extent that was also quantitatively predictable. An early increase in photoperiod advances sexual development and makes the bird more sensitive to a subsequent decrease than would be expected by reference to its chronological age. An early decrease in photoperiod delays sexual development, which can have the effect of making the bird more or less sensitive to a subsequent increase since, ill layer-strain pullets, sensitivity to an increment in photoperiod normally increases Lip to about 9 weeks of age but decreases thereafter. Mean age at first egg predicted using these concepts was very highly correlated with observed age at first egg. The results provide a rational basis for constructing a model to predict age at first egg for any combination of increases and decreases in photoperiod applied to growing pullets.

Transport of pollution and heatout of streets into the boundary layer above is not currently understood and so fluxes cannot be quantified. Scalar concentration within the street is determined by the flux out of it and so quantifying fluxes for turbulent flow over a rough urban surface is essential. We have developed a naphthalene sublimation technique to measure transfer from a two-dimensional street canyon in a wind tunnel for the case of flow perpendicular to the street. The street was coated with naphthalene, which sublimes at room temperature, so that the vapour represented the scalar source. The transfer velocity wT relates the flux out of the canyon to the concentration within it and is shown to be linearly related to windspeed above the street. The dimensionless transfer coefficient wT/Uδ represents the ventilation efficiency of the canyon (here, wT is a transfer velocity,Uδ is the wind speed at the boundary-layer top). Observed values are between 1.5 and 2.7 ×10-3 and, for the case where H/W→0 (ratio of buildingheight to street width), values are in the same range as estimates of transfer from a flat plate, giving confidence that the technique yields accurate values for street canyon scalar transfer. wT/Uδ varies with aspect ratio (H/W), reaching a maximum in the wake interference regime (0.3 < H/W < 0.65). However, when upstream roughness is increased, the maximum in wT/Uδ reduces, suggesting that street ventilation is less sensitive to H/W when the flow is in equilibrium with the urban surface. The results suggest that using naphthalene sublimation with wind-tunnel models of urban surfaces can provide a direct measure of area-averaged scalar fluxes.

The dielectric properties of (Ba, Sr)TiO3 films were found to be remarkably sensitive to the postannealing treatment atmosphere. This study demonstrates that postannealing in an oxygen atmosphere increases the dielectric relaxation phenomenon and that postannealing in a nitrogen atmosphere produces a slight dielectric relaxation. Such dependence of the dielectric relaxation was related both to oxygen vacancies and to the presence of negatively charged oxygen, trapped at the grain boundary and/or at the electrode/dielectric film interface. A highly sensitive amperometric biosensor for determination of carbamate pesticides directly in water, fruit and vegetable samples has been evaluated, electrochemically characterized and optimized. The biosensor strip was fabricated in screen printed technique on a ceramic support using silver-based paste for reference electrode, and platinum-based paste for working and auxiliary electrodes. The working electrode was modified by a layer of carbon paste mixed with cobalt(II) phthalocyanine and acetylcellulose. Cholinesterase (ChE) enzymes with low enzymatic charge were immobilized on this layer. The operational simplicity of the biosensor consists in that a small drop (similar to 50 mu l) of substrate or sample is deposited on a horizontally positioned biosensor strip representing the microelectrochemical cell. The working potential of the biosensor was 370 mV versus Ag/AgI on a ship reference electrode preventing the interference of electroactive species which are oxidable at more positive potentials. The biosensor was applied to investigate the degradation of two reference ChE inhibitors in freeze dried water under different storage conditions and for direct determination of some N-methylcarbamates (NMCs) in fruit and vegetable samples at ppb concentration levels without any sample pretreatment. dThe detection of aromatic compounds from pesticides and industrial wastewater has become of great interest, since these compounds withstand chemical oxidation and biological degradation, accumulating in the environment. In this work, a highly sensitive biosensor for detecting catechol was obtained with the immobilization of Cl-catechol 1,2-dioxygenase (CCD) in nanostructured films. CCD layers were alternated with poly(amidoamine) generation 4 (PAMAM G4) dendrimer using the electrostatic layer-by-layer (LbL) technique. Circular dichroism (CD) measurements indicated that the immobilized CCD preserved the same conformation as in solution. The thickness of the very first CCD layers in the LbL films was estimated at ca. 3.6 nm, as revealed by surface plasmon resonance (SPR). PAMAM/CCD 10-bilayer films were employed in detecting diluted catechol solutions using either an optical or electrical approach. Due to the mild immobilization conditions employed, especially regarding the pH and ionic strength of the dipping solutions, CCD remained active in the films for periods longer than 3 weeks. The optical detection comprised absorption experiments in which the formation of cis-cis muconic acid, resulting from the reaction between CCD and catechol, was monitored by measuring the absorbance at 260 nm after film immersion in catechol solutions. The electrical detection was carried out using LbL films deposited onto gold-interdigitated electrodes immersed in aqueous solutions at different catechol concentrations. Using impedance spectroscopy in a broad frequency range (1Hz-1kHz), we could detect catechol in solutions at concentrations as low as 10(-10) M. (c) 2005 Elsevier B.V. All rights reserved.

Humic acids (HAs), naturally occurring biomacromolecules, were incorporated into nanostructured polymeric films using the layer-by-layer (LbL) technique, in which HA layers were alternated with layers of poly(allylamine hydrochloride) (PAH). Atomic force microscopy (AFM) revealed very smooth films, with mean roughness varying from 0.89 to 1.19 nm for films containing 5 and 15 PAH/HA bilayers, respectively. The films displayed electroactivity, with the presence of only one reduction peak at ca. 0.675 V (vs Ag/AgCl). Such a well-defined electroactivity allowed the films to be used as highly sensitive pesticide sensors, with detection of pentachlorophenol (PCP) in solutions at concentrations as low as 10(-9) mol L(-1).

The movement of sensitive stamens in flowers of the Plains Prickly Pear (Opuntia polyacantha) is described in detail along with the external and internal filament anatomy. The goals of this investigation were: (1) to provide a synthesis of floral phenology and determine whether this rather unique stamen movement is nastic or a tropism and (2) to conduct macro- and micro-morphological analyses of filaments to determine if there are anatomical traits associated with this movement. To better understand the internal and external structure in sensitive filaments of O. polyacantha, we performed comparative anatomical analyses in two additional species from the Opuntioideae with stamens lacking such sensitivity. The consistent unidirectional movement of stamens, independent of the area stimulated, indicates a thigmonastic response. This movement serves multiple purposes, from enhancing pollen presentation to facilitating cross-pollination, protecting pollen and preventing insects from robbing pollen. Anatomically, the sensitive and non-sensitive filaments exhibit different tissue organization. Cuticle thickness, presence of capsular structures, two layers of curved cells, and more and larger intercellular spaces are characteristic of sensitive filaments. A thin unicellular epidermal layer is characteristic in sensitive filaments versus 2-3 epidermal layers in non-sensitive filaments. Another striking feature in sensitive filaments is the presence of papillae and capsular structures. We believe that these elements are related to water mobility with subsequent contraction during the thigmonastic response. Capsular structures might have a role in fluid mobility according to the stimulus of the filaments. We hypothesize that the thigmonastic response is controlled by cells with elastic properties, as evidenced by the plasmolyzed curved and contracted cells in the filaments and the fact that the movement is activated by changes in cell turgor followed by contraction as a result of plasmolysis. © 2013 Elsevier GmbH.

In the field of organic thin films, manipulation at the nanoscale can be obtained by immobilization of different materials on platforms designed to enhance a specific property via the layer-by-layer technique. In this paper we describe the fabrication of nanostructured films containing cobalt tetrasulfonated phthalocyanine (CoTsPc) obtained through the layer-by-layer architecture and assembled with linear poly(allylamine hydrochloride) (PAH) and poly(amidoamine) dendrimer (PAMAM) polyelectrolytes. Film growth was monitored by UV-vis spectroscopy following the Q band of CoTsPc and revealed a linear growth for both systems. Fourier transform infrared (FTIR) spectroscopy showed that the driving force keeping the structure of the films was achieved upon interactions of CoTsPc sulfonic groups with protonated amine groups present in the positive polyelectrolyte. A comprehensive SPR investigation on film growth reproduced the deposition process dynamically and provided an estimation of the thicknesses of the layers. Both FTIR and SPR techniques suggested a preferential orientation of the Pc ring parallel to the substrate. The electrical conductivity of the PAH films deposited on interdigitated electrodes was found to be very sensitive to water vapor. These results point to the development of a phthalocyanine-based humidity sensor obtained from a simple thin film deposition technique, whose ability to tailor molecular organization was crucial to achieve high sensitivity.

The layer-by-layer (LbL) technique combined with field-effect transistor (FET) based sensors has enabled the production of pH-sensitive platforms with potential application in biosensors. A variation of the FET architecture, so called separative extended gate FET (SEGFET) devices, are promise as an alternative to conventional ion sensitive FET (ISFET). SEGFET configuration exhibits the advantage of combining the field-effect concept with organic and inorganic materials directly adsorbed on the extended gate, allowing the test of new pH-sensitive materials in a simple and low cost way. In this communication, poly(propylene imine) dendrimer (PPI) and TiO2 nanoparticles (TiO2-np) were assembled onto gold-covered substrates via layer-by-layer technique to produce a low cost SEGFET pH sensor. The sensor presented good pH sensitivity, ca. 57 mV pH(-1), showing that our strategy has potential advantages to fabricate low cost pH-sensing membranes. (C) 2012 Elsevier B.V. All rights reserved.

Ultra-thin (thicknesses of 50-90 nm) nanocomposite films of cobalt ferrite nanoparticles (np-CoFe2O4, 18 nm in diameter) and polyelectrolytes (doped polyaniline-PANI, poly-3,4-ethylenedioxy thiophene: polystyrene sulfonic acid-PEDOT:PSS, and sulfonated lignin-SL) are assembled layer-by-layer onto interdigitated microelectrodes aiming at to create novel nanostructured sensoactive materials for liquid media chemical sensors. The nanocomposites display a distinctive globular morphology with nanoparticles densely-packed while surrounded by polyelectrolytes. Due to the presence of np-CoFe2O4 the nanocomposites display low electrical conductivity according to impedance data. On the other hand, this apparent shortcoming turns such nanocomposites much more sensitive to the presence of ions in solution than films made exclusively of conducting polyelectrolytes. For example, the electrical resistance of np-CoFe2O4/PEDOT:PSS and PANI/SL/np-CoFe2O4/SL architectures has a 10-fold decrease when they are immersed in 20 mmol. L-1 NaCl solution. Impedance spectra fitted with the response of an equivalent circuit model suggest that the interface created between nanoparticles and polyelectrolytes plays a major role on the nanocomposites electrical/dielectrical behavior. Since charge transport is sensitive to nanoparticle-polyelectrolyte interfaces as well as to the physicochemical conditions of the environment, the np-CoFe2O4-based nanocomposites can be used as sensing elements in chemical sensors operated under ac regime and room temperature.

Die bedeutendste Folge der Luftverschmutzung ist eine erhöhte Konzentration an Ozon (O3) in der Troposphäre innerhalb der letzten 150 Jahre. Ozon ist ein photochemisches Oxidationsmittel und ein Treibhausgas, das als wichtigste Vorstufe des Hydroxyradikals OH die Oxidationskraft der Atmosphäre stark beeinflusst. Um die Oxidationskraft der Atmosphäre und ihren Einfluss auf das Klima verstehen zu können, ist es von großer Bedeutung ein detailliertes Wissen über die Photochemie des Ozons und seiner Vorläufer, den Stickoxiden (NOx), in der Troposphäre zu besitzen. Dies erfordert das Verstehen der Bildungs- und Abbaumechanismen von Ozon und seiner Vorläufer. Als eine für den chemischen Ozonabbau wichtige Region kann die vom Menschen weitgehend unberührte marine Grenzschicht (Marine boundary layer (MBL)) angesehen werden. Bisher wurden für diese Region jedoch kaum Spurengasmessungen durchgeführt, und so sind die dort ablaufenden photochemischen Prozesse wenig untersucht. Da etwa 70 % der Erdoberfläche mit Ozeanen bedeckt sind, können die in der marinen Granzschicht ablaufenden Prozesse als signifikant für die gesamte Atmosphäre angesehen werden. Dies macht eine genaue Untersuchung dieser Region interessant. Um die photochemische Produktion und den Abbau von Ozon abschätzen zu können und den Einfluss antrophogener Emissionen auf troposphärisches Ozon zu quantifizieren, sind aktuelle Messergebnisse von NOx im pptv-Bereich für diese Region erforderlich. Die notwendigen Messungen von NO, NO2, O3, JNO2, J(O1D), HO2, OH, ROx sowie einiger meteorologischer Parameter wurden während der Fahrt des französischen Forschungsschiffes Marion-Dufresne auf dem südlichen Atlantik (28°S-57°S, 46°W-34°E) im März 2007 durchgeführt. Dabei sind für NO und NO2 die bisher niedrigsten gemessenen Werte zu verzeichnen. Die während der Messcampagne gewonnen Daten wurden hinsichtlich Ihrer Übereinstimmung mit den Bedingungen des photochemischen stationären Gleichgewichts (photochemical steady state (PSS)) überprüft. Dabei konnte eine Abweichung vom PSS festgestellt werden, welche unter Bedingungen niedriger NOx-Konzentrationen (5 bis 25pptv) einen unerwarteten Trend im Leighton-Verhältnis bewirkt, der abhängig vom NOx Mischungsverhältnis und der JNO2 Intensität ist. Signifikante Abweichungen vom Verhältnis liegen bei einer Zunahme der JNO2 Intensität vor. Diese Ergebnisse zeigen, dass die Abweichung vom PSS nicht beim Minimum der NOx-Konzentrationen und der JNO2 Werte liegt, so wie es in bisherigen theoretischen Studien dargelegt wurde und können als Hinweis auf weitere photochemische Prozesse bei höheren JNO2-Werten in einem System mit niedrigem NOx verstanden werden. Das wichtigste Ergebnis dieser Untersuchung, ist die Verifizierung des Leighton-Verhältnisses, das zur Charakterisierung des PSS dient, bei sehr geringen NOx-Konzentrationen in der MBL. Die bei dieser Doktorarbeit gewonnenen Erkenntnisse beweisen, dass unter den Bedingungen der marinen Granzschicht rein photochemischer Abbau von Ozon stattfindet und als Hauptursache hierfür während des Tages die Photolyse gilt. Mit Hilfe der gemessenen Parameter wurde der kritische NO-Level auf Werte zwischen 5 und 9 pptv abgeschätzt, wobei diese Werte im Vergleich zu bisherigen Studien vergleichsweise niedrig sind. Möglicherweise bedeutet dies, dass das Ozon Produktion/ Abbau-Potential des südlichen Atlantiks deutlich stärker auf die Verfügbarkeit von NO reagiert, als es in anderen Regionen der Fall ist. Im Rahmen der Doktorarbeit wurde desweiteren ein direkter Vergleich der gemessenen Spezies mit dem Modelergebnis eines 3-dimensionalen Zirkulationsmodel zur Simulation atmosphären chemischer Prozesse (EMAC) entlang der exakten Schiffsstrecke durchgeführt. Um die Übereinstimmung der Messergebnisse mit dem bisherigen Verständnis der atmosphärischen Radikalchemie zu überprüfen, wurde ein Gleichgewichtspunktmodel entwickelt, das die während der Überfahrt erhaltenen Daten für Berechungen verwendet. Ein Vergleich zwischen der gemessenen und der modellierten ROx Konzentrationen in einer Umgebung mit niedrigem NOx zeigt, dass die herkömmliche Theorie zur Reproduktion der Beobachtungen unzureichend ist. Die möglichen Gründe hierfür und die Folgen werden in dieser Doktorarbeit diskutiert.

Clearcutting is a common harvesting practice in many eastern hardwood forests. Among the vegetation strata of these forests, the herbaceous layer is potentially the most sensitive in its response to harvest-mediated disturbances and has the highest species diversity. Thus, it is important to understand the response of herbaceous layer diversity to forest harvesting. Previous work on clearcut and mature stands at the Fernow Experimental Forest (FEF), West Virginia, has shown that, although, harvesting did not alter appreciably herbaceous layer cover, it influenced the relationship of cover to biotic and abiotic factors, such as tree density and soil nutrients, respectively. The purpose of this study was to examine the response of species diversity of the herbaceous layer to harvesting at FEF. Fifteen circular, 0.04 ha sample plots were established in each of four watersheds (60 plots in total) representing two stand age categories: two watersheds with 20 years even-age stands following clearcutting and two watersheds with mature second growth stands. All woody stems ≥2.5 cm diameter at breast height were identified, tallied, and measured for diameter. The herbaceous layer was sampled by identifying all vascular plants ≤1 m in height and estimating cover for each species in each of 10 (1 m2) circular sub-plots per sample plot (600 sub-plots total). Species diversity for each plot was calculated from herbaceous layer data using the ln-based Shannon Index (H′) equation. Ten stand and soil variables also were measured on each plot. Mean herbaceous layer cover for clearcut versus mature stands was 27.2±14.3% versus 20.2±8.1% (P>0.05), respectively and mean H′ was 1.67±0.42 versus 1.55±0.48 (P>0.05), respectively. Herbaceous layer diversity was negatively correlated with cation exchange capacity and extractable Ca and Mg in the mineral soil in clearcut stands. In contrast, herbaceous layer diversity was positively correlated with soil organic matter and clay content. Although, 20 years of recovery after clearcutting did not have significant effects on the species diversity of the herbaceous layer when examining stand age means alone, harvesting did appear to influence the spatial relationships between herbaceous layer diversity and biotic factors (e.g. tree density) and abiotic factors (e.g. soil nutrients).

Layer 2/3 (L2/3) pyramidal neurons are the most abundant cells of the neocortex. Despite their key position in the cortical microcircuit, synaptic integration in dendrites of L2/3 neurons is far less understood than in L5 pyramidal cell dendrites, mainly because of the difficulties in obtaining electrical recordings from thin dendrites. Here we directly measured passive and active properties of the apical dendrites of L2/3 neurons in rat brain slices using dual dendritic-somatic patch-clamp recordings and calcium imaging. Unlike L5 cells, L2/3 dendrites displayed little sag in response to long current pulses, which suggests a low density of I(h) in the dendrites and soma. This was also consistent with a slight increase in input resistance with distance from the soma. Brief current injections into the apical dendrite evoked relatively short (half-width 2-4 ms) dendritic spikes that were isolated from the soma for near-threshold currents at sites beyond the middle of the apical dendrite. Regenerative dendritic potentials and large concomitant calcium transients were also elicited by trains of somatic action potentials (APs) above a critical frequency (130 Hz), which was slightly higher than in L5 neurons. Initiation of dendritic spikes was facilitated by backpropagating somatic APs and could cause an additional AP at the soma. As in L5 neurons, we found that distal dendritic calcium transients are sensitive to a long-lasting block by GABAergic inhibition. We conclude that L2/3 pyramidal neurons can generate dendritic spikes, sharing with L5 pyramidal neurons fundamental properties of dendritic excitability and control by inhibition.

Olfactory glomeruli are the loci where the first odor-representation map emerges. The glomerular layer comprises exquisite local synaptic circuits for the processing of olfactory coding patterns immediately after their emergence. To understand how an odor map is transferred from afferent terminals to postsynaptic dendrites, it is essential to directly monitor the odor-evoked glomerular postsynaptic activity patterns. Here we report the use of a transgenic mouse expressing a Ca(2+)-sensitive green fluorescence protein (GCaMP2) under a Kv3.1 potassium-channel promoter. Immunostaining revealed that GCaMP2 was specifically expressed in mitral and tufted cells and a subpopulation of juxtaglomerular cells but not in olfactory nerve terminals. Both in vitro and in vivo imaging combined with glutamate receptor pharmacology confirmed that odor maps reported by GCaMP2 were of a postsynaptic origin. These mice thus provided an unprecedented opportunity to analyze the spatial activity pattern reflecting purely postsynaptic olfactory codes. The odor-evoked GCaMP2 signal had both focal and diffuse spatial components. The focalized hot spots corresponded to individually activated glomeruli. In GCaMP2-reported postsynaptic odor maps, different odorants activated distinct but overlapping sets of glomeruli. Increasing odor concentration increased both individual glomerular response amplitude and the total number of activated glomeruli. Furthermore, the GCaMP2 response displayed a fast time course that enabled us to analyze the temporal dynamics of odor maps over consecutive sniff cycles. In summary, with cell-specific targeting of a genetically encoded Ca(2+) indicator, we have successfully isolated and characterized an intermediate level of odor representation between olfactory nerve input and principal mitral/tufted cell output.
