A data set of a commercial Nellore beef cattle selection program was used to compare breeding models that assumed or not markers effects to estimate the breeding values, when a reduced number of animals have phenotypic, genotypic and pedigree information available. This herd complete data set was composed of 83,404 animals measured for weaning weight (WW), post-weaning gain (PWG), scrotal circumference (SC) and muscle score (MS), corresponding to 116,652 animals in the relationship matrix. Single trait analyses were performed by MTDFREML software to estimate fixed and random effects solutions using this complete data. The additive effects estimated were assumed as the reference breeding values for those animals. The individual observed phenotype of each trait was adjusted for fixed and random effects solutions, except for direct additive effects. The adjusted phenotype composed of the additive and residual parts of observed phenotype was used as dependent variable for models' comparison. Among all measured animals of this herd, only 3160 animals were genotyped for 106 SNP markers. Three models were compared in terms of changes on animals' rank, global fit and predictive ability. Model 1 included only polygenic effects, model 2 included only markers effects and model 3 included both polygenic and markers effects. Bayesian inference via Markov chain Monte Carlo methods performed by TM software was used to analyze the data for model comparison. Two different priors were adopted for markers effects in models 2 and 3, the first prior assumed was a uniform distribution (U) and, as a second prior, was assumed that markers effects were distributed as normal (N). Higher rank correlation coefficients were observed for models 3_U and 3_N, indicating a greater similarity of these models animals' rank and the rank based on the reference breeding values. Model 3_N presented a better global fit, as demonstrated by its low DIC. The best models in terms of predictive ability were models 1 and 3_N. Differences due prior assumed to markers effects in models 2 and 3 could be attributed to the better ability of normal prior in handle with collinear effects. The models 2_U and 2_N presented the worst performance, indicating that this small set of markers should not be used to genetically evaluate animals with no data, since its predictive ability is restricted. In conclusion, model 3_N presented a slight superiority when a reduce number of animals have phenotypic, genotypic and pedigree information. It could be attributed to the variation retained by markers and polygenic effects assumed together and the normal prior assumed to markers effects, that deals better with the collinearity between markers. Validity of comparisons between expected breeding values obtained from best linear unbiased prediction procedures in genetic evaluations is dependent on genetic connectedness among herds. Different cattle breeding programmes have their own particular features that distinguish their database structure and can affect connectedness. Thus, the evolution of these programmes can also alter the connectedness measures. This study analysed the evolution of the genetic connectedness measures among Brazilian Nelore cattle herds from 1999 to 2008, using the French Criterion of Admission to the group of Connected Herds (CACO) method, based on coefficients of determination (CD) of contrasts. Genetic connectedness levels were analysed by using simple and multiple regression analyses on herd descriptors to understand their relationship and their temporal trends from the 19992003 to the 20042008 period. The results showed a high level of genetic connectedness, with CACO estimates higher than 0.4 for the majority of them. Evaluation of the last 5-year period showed only a small increase in average CACO measures compared with the first 5 years, from 0.77 to 0.80. The percentage of herds with CACO estimates lower than 0.7 decreased from 27.5% in the first period to 16.2% in the last one. The connectedness measures were correlated with percentage of progeny from connecting sires, and the artificial insemination spread among Brazilian herds in recent years. But changes in connectedness levels were shown to be more complex, and their complete explanation cannot consider only herd descriptors. They involve more comprehensive changes in the relationship matrix, which can be only fully expressed by the CD of contrasts.

Bei der Kommissionierung im Warendistributionszentrum müssen die heterogenen Produkte mit den unterschiedlichsten Eigenschaften stabil gestapelt und dürfen während des Transportes nicht beschädigt werden. Für eine automatische Kommissionieranlage wurde das neue Optimierungsprogramm UNIT_OrderPacking für die Planung und Optimierung der Ladeeinheiten eingesetzt. Nach der Problemstellung werden die Einflussfaktoren analysiert und definiert. Die Strategien zur Berücksichtigung solcher Faktoren im Optimierungsverfahren werden vorgestellt. Das gesamte Optimierungsverfahren wird anschließend in Teilproblemen dargestellt. Für jedes Teilproblem werden die Lösungsstrategien nach der Problemanalyse durch Verallgemeinerung der Lösungsvorgehensweise abgeleitet und zusammengefasst. Folgende Teilprobleme werden relativ ausführlich behandelt: * die Bewertung und Selektion eines Packobjektes für jeden Packschritt hinsichtlich der Tragfähigkeit, des Gewichtes und der Warengruppe, * die Kraftübertragung der Packobjekte von oben nach unten und die „Relationship Matrix“ der Packobjekte sowie * die Strategien und Prioritäten für das Anordnen und Stapeln der Packobjekte im Packraum.

A eficiência econômica da bovinocultura leiteira está relacionada à utilização de animais que apresentem, concomitantemente, bom desempenho quanto à produção, reprodução, saúde e longevidade. Nisto, o índice de seleção configura-se como ferramenta importante ao aumento da lucratividade nesse sistema, visto que permite a seleção de reprodutores para várias características simultaneamente, considerando a relação entre elas bem como a relevância econômica das mesmas. Com a recente disponibilidade de dados genômicos tornou-se ainda possível expandir a abrangência e acurácia dos índices de seleção por meio do aumento do número e qualidade das informações consideradas. Nesse contexto, dois estudos foram desenvolvidos. No primeiro, o objetivo foi estimar parâmetros genéticos e valores genéticos (VG) para características relacionadas à produção e qualidade do leite incluindo-se a informação genômica na avaliação genética. Foram utilizadas medidas de idade ao primeiro parto (IPP), produção de leite (PROD), teor de gordura (GOR), proteína (PROT), lactose, caseína, escore de células somáticas (ECS) e perfil de ácidos graxos de 4.218 vacas bem como os genótipos de 755 vacas para 57.368 polimorfismos de nucleotídeo único (SNP). Os componentes de variância e VG foram obtidos por meio de um modelo misto animal, incluindo-se os efeitos de grupos de contemporâneas, ordem de lactação, dias em lactação e os efeitos aditivo genético, ambiente permanente e residual. Duas abordagens foram desenvolvidas: uma tradicional, na qual a matriz de relacionamentos é baseada no pedigree; e uma genômica, na qual esta matriz é construída combinando-se a informação de pedigree e dos SNP. As herdabilidades variaram de 0,07 a 0,39. As correlações genéticas entre PROD e os componentes do leite variaram entre -0,45 e -0,13 enquanto correlações altas e positivas foram estimadas entre GOR e os ácidos graxos. O uso da abordagem genômica não alterou as estimativas de parâmetros genéticos; contudo, houve aumento entre 1,5% e 6,8% na acurácia dos VG, à exceção de IPP, para a qual houve uma redução de 1,9%. No segundo estudo, o objetivo foi incorporar a informação genômica no desenvolvimento de índices econômicos de seleção. Neste, os VG para PROD, GOR, PROT, teor de ácidos graxos insaturados (INSAT), ECS e vida produtiva foram combinados em índices de seleção ponderados por valores econômicos estimados sob três cenários de pagamento: exclusivamente por volume de leite (PAG1); por volume e por componentes do leite (PAG2); por volume e componentes do leite incluindo INSAT (PAG3). Esses VG foram preditos a partir de fenótipos de 4.293 vacas e genótipos de 755 animais em um modelo multi-característica sob as abordagens tradicional e genômica. O uso da informação genômica influenciou os componentes de variância, VG e a resposta à seleção. Entretanto, as correlações de ranking entre as abordagens foram altas nos três cenários, com valores entre 0,91 e 0,99. Diferenças foram principalmente observadas entre PAG1 e os demais cenários, com correlações entre 0,67 e 0,88. A importância relativa das características e o perfil dos melhores animais foram sensíveis ao cenário de remuneração considerado. Assim, verificou-se como essencial a consideração dos valores econômicos das características na avaliação genética e decisões de seleção.

Background: Nitric oxide (NO) is a major regulator of cardiovascular homeostasis and has anti-atherogenic properties. Reduced NO formation is associated with endothelial dysfunction and with cardiovascular risk factors. Although NO downregulates the expression and activity of the pro-atherogenic enzyme matrix metalloproteinase-9 (MMP-9), no previous clinical study has examined whether endogenous NO formation is inversely associated with the circulating levels of pro-MMP-9, which are associated with cardiovascular events. We examined this hypothesis in 175 healthy male subjects who were non-smokers. Methods: To assess NO bioavailability, the plasma concentrations of nitrite, nitrate, and cGMP were determined using an ozone-based chemiluminescence assay and an enzyme immunoassay. Pro-MMP-9 and pro-MMP-2 levels were measured in plasma samples by gelatin zymography. Results: We found significant negative correlations between pro-MMP-9 levels and plasma nitrite (P=0.035, rs=-0.159), nitrate (P=0.040, rs=-0.158), and cGMP (P=0.011, rs=-0.189) concentrations. However, no significant correlations were found between pro-MMP-2 levels and the plasma concentrations of markers of NO bioavailability (all P>0.05). Conclusions: There is an inverse relationship between markers of NO formation and plasma MMP-9 levels. This finding may shed some light on the possible mechanisms involved in the increased cardiovascular risk of apparently healthy subjects with low NO bioavailability or high circulating levels of pro-MMP-9. Purpose: The phenotype of vascular smooth muscle cells (SMCs) is altered in several arterial pathologies, including the neointima formed after acute arterial injury. This study examined the time course of this phenotypic change in relation to changes in the amount and distribution of matrix glycosaminoglycans. Methods: The immunochemical staining of heparan sulphates (HS) and chondroitin sulphates (CS) in the extracellular matrix of the arterial wall was examined at early points after balloon catheter injury of the rabbit carotid artery. SMC phenotype was assessed by means of ultrastructural morphometry of the cytoplasmic volume fraction of myofilaments. The proportions of cell and matrix components in the media were analyzed with similar morphometric techniques. Results: HS and CS were shown in close association with SMCs of the uninjured arterial media as well as being more widespread within the matrix. Within 6 hours after arterial injury, there was loss of the regular pericellular distribution of both HS and CS, which was associated with a significant expansion in the extracellular space. This preceded the change in ultrastructural phenotype of the SMCs. The glycosaminoglycan loss was most exaggerated at 4 days, after which time the HS and CS reappeared around the medial SMCs. SMCs of the recovering media were able to rapidly replace their glycosaminoglycans, whereas SMCs of the developing neointima failed to produce HS as readily as they produced CS. Conclusions: These studies indicate that changes in glycosaminoglycans of the extracellular matrix precede changes in SMC phenotype after acute arterial injury. In the recovering arterial media, SMCs replace their matrix glycosaminoglycans rapidly, whereas the newly established neointima fails to produce similar amounts of heparan sulphates.

OBJECTIVE To analyze whether the level of institutional and matrix support is associated with better certification of primary healthcare teams.METHODS In this cross-sectional study, we evaluated two kinds of primary healthcare support – 14,489 teams received institutional support and 14,306 teams received matrix support. Logistic regression models were applied. In the institutional support model, the independent variable was “level of support” (as calculated by the sum of supporting activities for both modalities). In the matrix support model, in turn, the independent variables were the supporting activities. The multivariate analysis has considered variables with p < 0.20. The model was adjusted by the Hosmer-Lemeshow test.RESULTS The teams had institutional and matrix supporting activities (84.0% and 85.0%), respectively, with 55.0% of them performing between six and eight activities. For the institutional support, we have observed 1.96 and 3.77 chances for teams who had medium and high levels of support to have very good or good certification, respectively. For the matrix support, the chances of their having very good or good certification were 1.79 and 3.29, respectively. Regarding to the association between institutional support activities and the certification, the very good or good certification was positively associated with self-assessment (OR = 1.95), permanent education (OR = 1.43), shared evaluation (OR = 1.40), and supervision and evaluation of indicators (OR = 1.37). In regards to the matrix support, the very good or good certification was positively associated with permanent education (OR = 1.50), interventions in the territory (OR = 1.30), and discussion in the work processes (OR = 1.23).CONCLUSIONS In Brazil, supporting activities are being incorporated in primary healthcare, and there is an association between the level of support, both matrix and institutional, and the certification result.

Ashby was a keen observer of the world around him, as per his technological and psychiatrical developments. Over the years, he drew numerous philosophical conclusions on the nature of human intelligence and the operation of the brain, on artificial intelligence and the thinking ability of computers and even on science in general. In this paper, the quite profound philosophy espoused by Ashby is considered as a whole, in particular in terms of its relationship with the world as it stands now and even in terms of scientific predictions of where things might lead. A meaningful comparison is made between Ashby's comments and the science fiction concept of 'The Matrix' and serious consideration is given as to how much Ashby's ideas lay open the possibility of the matrix becoming a real world eventuality.

A simple proof is given that a 2 x 2 matrix scheme for an inverse scattering transform method for integrable equations can be converted into the standard form of the second-order scalar spectral problem associated with the same equations. Simple formulae relating these two kinds of representation of integrable equations are established.

Subjects with spinal cord injury (SCI) exhibit impaired left ventricular (LV) diastolic function, which has been reported to be attenuated by regular physical activity. This study investigated the relationship between circulating matrix metalloproteinases (MMPs) and tissue inhibitors of MMPs (TIMPs) and echocardiographic parameters in SCI subjects and the role of physical activity in this regard. Forty-two men with SCI [19 sedentary (S-SCI) and 23 physically-active (PA-SCI)] were evaluated by clinical, anthropometric, laboratory, and echocardiographic analysis. Plasmatic pro-MMP-2, MMP-2, MMP-8, pro-MMP-9, MMP-9, TIMP-1 and TIMP-2 levels were determined by enzyme-linked immunosorbent assay and zymography. PA-SCI subjects presented lower pro-MMP-2 and pro-MMP-2/TIMP-2 levels and improved markers of LV diastolic function (lower E/Em and higher Em and E/A values) than S-SCI ones. Bivariate analysis showed that pro-MMP-2 correlated inversely with Em and directly with E/Em, while MMP-9 correlated directly with LV mass index and LV end-diastolic diameter in the whole sample. Following multiple regression analysis, pro-MMP-2, but not physical activity, remained associated with Em, while MMP-9 was associated with LV mass index in the whole sample. These findings suggest differing roles for MMPs in LV structure and function regulation and an interaction among pro-MMP-2, diastolic function and physical activity in SCI subjects.
