There is in the literature a noteworthy lack of sampling methods to be employed in the selection of papaya fruits for genetical improvement purposes. In the present experiment methods applied to melon and watermelon were extended to papaya with good results. The authors tried to correlate the solids content-as determined by a hand refractometer-with total sugars in the fruit. The correlation does exist being significant and positive. A regression equation was worked out; it permits to calculate the total sugar content provided the Brix value determined by the hand refractometer is known. The equation is as follows: a = 0.57 s where a = total amount of sugar as glucose, and s = total solids.

The present paper deals with sampling methods to be employed in the determination of the total sugar content obtained from brix measurements in Carica papaya L., given by Zeiss hand refractometer. The results obtained led to the conclusion that, rather than using several samples per fruit, it is better to get a large number of fruits, taking just one sample per fruit. The sample in each fruit should be taken always from the same region.

L’oxydoréduction de monocouches auto-assemblées (SAMs) de ferrocénylalcanethiolates à la surface d’or (FcRSAu) a été étudiée en temps réel par la spectroscopie de résonance de plasmons de surface couplée avec l’électrochimie (E-SPR). La sensibilité de cette technique permet de déterminer des changements d’épaisseur de couche l’ordre de quelques angström résultant d’un changement de structure de la SAM. Plusieurs études antérieures ont proposé que l’oxydation électrochimique d’une SAM de FcRSAu induit une réorientation moléculaire. L’E-SPR est utilisé pour identifier l’origine de ce changement structurel. D’abord, une calibration du réfractomètre SPR utilisé a été effectuée afin de trouver une équation de conversion du signal SPR obtenu en pixel en angle d’incidence pour que l’on puisse calculer le changement d’épaisseur de monocouche à partir du changement d’angle de résonance avec le modèle de Fresnel. Par la suite, une caractérisation approfondie des SAMs de FcCnSAu (où n = 6, 8, 12, 14) en contact avec du NaClO4 acidifié a été réalisée par électrochimie, éllipsométrie, spectroscopie infrarouge et microscopie à force atomique. Les résultats obtenus montrent que l’augmentation de la longueur des chaînes alkyles donne des SAMs de ferrocènes plus épaisses et moins désordonnées. L’analyse par l’E-SPR de ces SAMs pures montre que le changement d’épaisseur induit par l’électro-oxydation dépend linéairement du nombre de méthylènes sur la chaîne alkyle. En appliquant la déconvolution mathématique aux voltampérogrammes cycliques enregistrés pour les SAM mixtes (FcC12SAu/C11SAu) de différentes compositions, on arrive à la conclusion qu’il y a un redressement des chaînes alkyles dans les domaines des ferrocènes agrégés mais la réorientation des têtes de ferrocène dans les domaines de ferrocènes agrégés ou dispersés ne peut pas être exclue. Enfin, l’effet de l’anion électrolytique sur le changement d’épaisseur de la SAM mesuré par l’E-SPR a été étudié. L’analyse électrochimique montre que la capacité de pairage d’anions avec les ferrocéniums décroit comme suit : PF6- > ClO4- > BF4- > NO3-. Tandis que l’épaisseur de la SAM donnée par le changement d’angle de résonance suit la tendance suivante : NO3- ≥ ClO4- > PF6- ≈ BF4-. Des études plus approfondies seront nécessaire pour clarifier cette tendance observée par E-SPR.

The design and development of a fibre optic evanescent wave refractometer for the detection of trace amounts of paraffin oil and palm oil in coconut oil is presented. This sensor is based on a side-polished plastic optical fibre. At the sensing region, the cladding and a small portion of the core are removed and the fibre nicely polished. The sensing region is fabricated in such a manner that it sits perfectly within a bent mould. This bending of the sensing region enhances its sensitivity. The oil mixture of different mix ratios is introduced into the sensing region and we observed a sharp decrease in the output intensity. The observed variation in the intensity is found to be linear and the detection limit is 2% (by volume) paraffin oil/palm oil in coconut oil. The resolution of this refractometric sensor is of the order of 10−3. Since coconut oil is consumed in large volumes as edible oil in south India, this fibre optic sensor finds great relevance for the detection of adulterants such as paraffin oil or palm oil which are readily miscible in coconut oil. The advantage of this type of sensor is that it is inexpensive and easy to set up. Another attraction of the side-polished fibre is that only a very small amount of analyte is needed and its response time is only 7 s.

The shrimp farming industry is the most profitable area of the aquaculture at Rio Grande do Norte (RN) state, which is one of the largest producers in Brazil. However the infections that affect the shrimp cause major economic losses. The infection is a result of the interaction between the shrimp, the environment and pathogen. The change of these factors may lead to a condition of stress and susceptibility to opportunistic infections. One of these infections caused by Infectious Hypodermal and Hematopoietic Necrosis Virus (IHHNV) is widely distributed in several countries and affects a wide range of hosts. To optimize conditions for production of Litopenaeus vannamei shrimp, the more species cultivated in Brazil, it is necessary to understand the effects of environmental factors in the susceptibility of this species to infections. The aim of this study was to determine the IHHNV prevalence and to investigate the influence of environmental factors as salinity, temperature, stocking density, dissolved oxygen and rainfall in the IHHNV incidence in L. vannamei grown in farms, in the RN state. To determine the IHHNV prevalence were used 1089 samples of L. vannamei collected in seven farms. To perform the study about the influence of environmental factors, 525 samples of L. vannamei shrimp were collected in eight farms located in regions of low (0-1 ), medium (21-30 ) and high (38-57 ) salinity, using extensive (≤15 shrimp/m2 ), semi-intensive (18-33 shrimp/m2) or intensive (>36 shrimp/m2) stocking density systems. The IHHNV infection was determined in pleopod and hemolymph using the polymerase chain reaction (PCR). The environmental factors were recorded during the collection of animals, using a refractometer to measure the salinity and a multi-parameter meter to measure the temperature and concentration of dissolved oxygen in the water. The IHHNV prevalence in RN was 43% (468 infected shrimp out of 1089), varying on different farms. On the seven farms studied, IHHNV prevalence ranged from 18.6% to 54.8%. The infection rates in the shrimp cultured in low, medium and high salinity were respectively 43.10% (125/290), 31.2% (15/48) and 24.6% (46/187) and was significantly higher in shrimp grown in low salinity (P<0.001). The infection rates in ponds of extensive, semi-intensive and intensive systems were respectively, 28.7%, 28.28% and 47.84%, and was significantly higher in high stocking densities (P<0.001). This study indicated a high IHHNV prevalence and a significant effect of salinity and stocking density, but not of the temperature, rainfall and dissolved oxygen on the IHHNV infection rate in the L. vannamei shrimp cultured in the northeastern Brazil

O presente trabalho buscou conhecer a variação das características físicas e químicas do abacaxi comercializado no Entreposto Terminal de São Paulo (ETSP), da Companhia de Entrepostos e Armazéns Gerais de São Paulo (CEAGESP), no período de setembro de 2005 a março de 2006. As avaliações foram realizadas na CEAGESP e nos laboratórios do Departamento de Agroindústria, Alimentos e Nutrição da Escola Superior de Agricultura Luiz de Queiroz - ESALQ/USP. Foram realizadas coletas em intervalos de duas semanas, em três atacadistas, com as amostras recolhidas de forma aleatória, padronizadas em frutos tipo 10 (caixa contendo 10 frutos), compostas de dez frutos por atacadista/origem. As amostras foram coletadas nos lotes oferecidos para comercialização pelos atacadistas. Foram avaliadas as seguintes características físicas e químicas: peso do fruto inteiro, peso da coroa, avaliação visual da coloração da casca, densidade, coloração da polpa, pH, teor de sólidos solúveis (SS), acidez titulável (AT), além da relação SS/AT. Para a análise estatística, foi utilizado o Programa SAS. Pelos resultados, observou-se que, dentre as regiões produtoras de abacaxi, destacaram-se, com relação à regularidade de oferta para os três atacadistas, os pólos de Canápolis ('Smooth Cayenne'), Sapé e Miracema do Tocantins ('Pérola'). As diferentes regiões produtoras de abacaxi 'Pérola' apresentaram uma grande variação em relação ao peso médio dos frutos comercializados, indicando diferenças na tecnologia de produção adotada. Dentre as regiões produtoras de abacaxi, houve variação estatística no teor de sólidos solúveis ao longo do período analisado nos pólos de Canápolis ('Smooth Cayenne') e Itaberaba, Sapé e Miracema do Tocantins ('Pérola'). Dentre os parâmetros químicos analisados, a determinação do teor de sólidos solúveis por amostragem com uso do refratômetro manual, associado à maturação aparente (cor da casca), pode representar um avanço significativo na indicação da qualidade dos frutos comercializados, o que permite concluir ser desnecessário o uso de etefon na fase de pré-colheita.

The objective of this study was to evaluate the accumulation of matter, dry matter content of Brix and two varieties of sugar cane, influenced by the phosphorus cultivated on "Zona da Mata" region, State of Alagoas. The experiment, a factorial 2 x 6, was composed of two varieties of cane; RB867515 and RB92579 and six phosphorus levels: zero, 30, 60, 90, 120 and 150 kg ha(-1), with the source of the phosphorus triple superphosphate. Was also used nitrogen and potassium in doses equivalent to 100 and 200 kg ha(-1) and K, respectively. The soil of the experimental area was set to raise the base saturation to 60%. At the beginning of the first rainfall of the rainy season of 2006 was performed with micro fertilization in coverage, in doses equivalent to 6,0; 6,0 and 7,0 kg ha(-1) of Cu, Zn and Mn, respectively. Was The chemical control of weeds. Every month, from August to October 2006, samples were collected in the cane plant. There was initially the number of plants m(2), followed by weighing the material. It was subsequently withdrawn a subsample containing ten plants for weighing. After weighing the material was passed in horsemanship and homogenized, again drew up a sample of approximately 300g, leading them to forced ventilation oven at 65 degrees C, toobtain constant weight. Resulting in the production of natural (MN) and dry matter (DM). In July, we analyzed the levels of Brix% juice in the refractometer Brix. The average values of production of natural (MN), dry matter (DM) and contents of Brix% were subjected to analysis of variance and regression for variables. The accumulation of dry matter and were influenced by fertilization. The contents of Brix% in two varieties did not suffer significant influence of fertilization.

The purpose of this study was to evaluate the accuracy of hand-held refractometer in determining plasma protein concentrations in South American snake-necked turtle (Hydromedusa tectifera) as compared with the standard biuret method. The results indicated that plasma protein values may be accurately determined in snake- necked turtle with a hand-held refractometer.

The purpose of this study was to evaluate the accuracy of hand-held refractometer in determining serum protein concentrations in ring-necked pheasants (Phasianus colchicus) as compared with the standard biuret method. Coastal cities attract a large number of tourists for their beautiful landscape and recreational activities, increasing the municipalities' income source. Thus, a need is clear for the establishment of beach quality monitoring programs to ensure bathers health. Although there is an effective monitoring program for recreational waters in Brazil there are no programs to certify the quality of beach sands. In this sense, the aim of this work was to analyze the density of bacteria from the genus Enterococcus in both sand and water from two beaches from São Vicente, São Paulo (Brazil) and correlate these densities to abiotic parameters such as: temperature, salinity, particle size, organic matter and tides). Water and sand samples were collected during February 2006 on the beaches of Gonzaguinha e Ilha Porchat and bacterial densities were determined by membrane filter technique. Temperature and salinity were measured in situ with a thermometer and a refractometer while particle size and organic matter were determined according to methods described by Suguio and Dean. There were significant differences between densities found in water and sand (p=0.004), being approximately 20 times higher in Gonzaguinha's beach sands. Similar results were found for Ilha Porchat beach samples, being the densities found in sand 3 times greater than those found in water. Both beaches showed a negative correlation between bacterial densities and salinity and temperature, suggesting a deleterious effect of these parameters on the bacterial community. On the other hand, no correlation was found between organic matter and particle size and bacterial densities. This work demonstrates that there is a great necessity for monitoring beach sand since the evaluation of beach quality is an important part of integrated coastal management programs.

Objective: To characterize physically and chemically, non-sweetened orange juices and orange nectars sweetened with sucrose or sweet flavoring agents, with respect to their pH, titratable acidity (TA) and total soluble solids content (TSSC), as well as to evaluate the correlation of the latter property with the others. Method: Aliquots of three lots of two orange juices and two orange nectars containing sucrose and two containing sweet flavoring agents were evaluated. Mineral water was used as a control. The TSSC was determined using an Abbe refractometer. The pH was recorded using a digital pH meter, while TA was quantified by titrating samples of the beverages with 0.1 M NaOH until reaching pHs 5.5 and 7.0. Data were subjected to Pearson's correlation test, regression analysis, analysis of variance and Tukey's test (α=0.05). Results: TSSC values presented a strong correlation with TA, and these properties exhibited a quadratic relationship. Although the pH values were not dependent on the presence of sucrose or sweet flavoring agents, a significantly greater amount of base was necessary to reach pHs 5.5 and 7.0 in the beverages without sucrose or flavoring agents. Conclusion: Orange juices and nectars presented similar pH values, which was not associated with the presence of sucrose or sweet flavoring agents in the beverages. Higher TA values were obtained for the juice and lower for the nectars, regardless of containing sucrose or sweet flavoring agents. The increase of TSSC did not implicate in decrease of TA in the beverages.

Usamos uma metodologia experimental para investigar propriedades ópticas e térmicas da oleína de palma (OP), a fração líquida do azeite de palma (Elaies guineensis) obtida por fracionamento natural, com propriedades físicas que diferem apreciavelmente do azeite. Em uma primeira abordagem, estudamos os espectros de absorção e emissão monitorando a modificação dos espectros com o aumento da concentração β-caroteno misturado com OP, o que permitiu encontrar o coeficiente de absorção molar do β-caroteno (ε = 920,802 mol -1.cm-1.L). Este valor é muito baixo em relação aos conhecidos para o β-caroteno diluído em um solvente puro, como o benzeno ou o n-hexano. Experimentos feitos com β-caroteno diluído em hexano nos permitiram medir ε = 117.900 L.mol-1.cm-1, que representa aproximadamente 18% abaixo dos valores reportados na literatura. Em uma segunda abordagem, os espectros de absorção foram usados para medir o coeficiente de absorção da OP, A = 0,028 cm-1 em 532 nm, o comprimento de onda de um laser usado como fonte de excitação no estudo das propriedades térmicas da OP usando a técnica de lente térmica. Neste sentido foi medido também o coeficiente térmico do índice de refração da OP, dn/dt = - 3,821 x 10 -4 °C -1. Estes resultados, associados ao modelo aberrante para estudos de espectroscopia de lente térmica, levaram à medição do coeficiente de difusão, D = 2,19 ± 0,11 x 10-4 cm2/s, e da condutividade térmica, k = 5,96 ± 0,08 W/m°C. Embora as medidas das propriedades térmicas da OP não sejam ainda conhecidas na literatura, observa-se que elas têm as mesmas ordens de grandeza daquelas já conhecidas para outros óleos vegetais que foram relatadas na literatura.
