This study evaluated the spirometry and respiratory static pressures in 17 young women, twice a week for three successive ovulatory menstrual cycles to determine if such variables changed across the menstrual, follicular, periovulatory, early-tomid luteal and late luteal phases. The factors phases of menstrual cycle and individual cycles had no significant effect on the spirometry variables except for peak expiratory flow (PEF) and respiratory static pressures. Significant weak positive correlations were found between the progesterone:estradiol ratio and PEF and between estrogen and tidal volume (r = 0.37), inspiratory time (r = 0.22), expiratory time (r = 0.19), maximal inspiratory pressure (r = 0.25) and maximal expiratory pressure (r = 0.20) and for progesterone and maximal inspiratory pressure (r = 0.32) during the early-to-mid luteal phase. Although most parameters of the spirometry results did not change during the menstrual cycle, the correlations observed between sexual hormones and respiratory control variables suggest a positive influence of sexual female hormones controlling the thoracic pump muscles in the luteal phase

STUDY OBJECTIVES: To determine whether cerebral metabolite changes may underlie abnormalities of neurocognitive function and respiratory control in OSA. DESIGN: Observational, before and after CPAP treatment. SETTING: Two tertiary hospital research institutes. PARTICIPANTS: 30 untreated severe OSA patients, and 25 age-matched healthy controls, all males free of comorbidities, and all having had detailed structural brain analysis using voxel-based morphometry (VBM). MEASUREMENTS AND RESULTS: Single voxel bilateral hippocampal and brainstem, and multivoxel frontal metabolite concentrations were measured using magnetic resonance spectroscopy (MRS) in a high resolution (3T) scanner. Subjects also completed a battery of neurocognitive tests. Patients had repeat testing after 6 months of CPAP. There were significant differences at baseline in frontal N-acetylaspartate/choline (NAA/Cho) ratios (patients [mean (SD)] 4.56 [0.41], controls 4.92 [0.44], P = 0.001), and in hippocampal choline/creatine (Cho/Cr) ratios (0.38 [0.04] vs 0.41 [0.04], P = 0.006), (both ANCOVA, with age and premorbid IQ as covariates). No longitudinal changes were seen with treatment (n = 27, paired t tests), however the hippocampal differences were no longer significant at 6 months, and frontal NAA/Cr ratios were now also significantly different (patients 1.55 [0.13] vs control 1.65 [0.18] P = 0.01). No significant correlations were found between spectroscopy results and neurocognitive test results, but significant negative correlations were seen between arousal index and frontal NAA/Cho (r = -0.39, corrected P = 0.033) and between % total sleep time at SpO(2) < 90% and hippocampal Cho/Cr (r = -0.40, corrected P = 0.01). CONCLUSIONS: OSA patients have brain metabolite changes detected by MRS, suggestive of decreased frontal lobe neuronal viability and integrity, and decreased hippocampal membrane turnover. These regions have previously been shown to have no gross structural lesions using VBM. Little change was seen with treatment with CPAP for 6 months. No correlation of metabolite concentrations was seen with results on neurocognitive tests, but there were significant negative correlations with OSA severity as measured by severity of nocturnal hypoxemia. The antitumour antibiotic, adriamycin, inhibited oxidative phosphorylation in freshly prepared mitochondria from the heart, liver and kidney of the rat. It abolished respiratory control and stimulated ATPase activity. Sccinate oxidation by heart mitochondria was extremely sensitive to the drug when hexokinase was present in the reaction medium. The sensitive site has been identified to lie in the region between the succinate dehydrogenase flavoprotein and ubiquinone of the respiratory chain.

1. a-p-Chlorophenoxyisobutyric acid, the ethyl ester of which is widely used as an antihypercholesterolaemic drug, is an inhibitor of energy-transfer reactions in isolated rat liver mitochondria. 2. The compound at lower concentrations (<4.0mmol/mg of mitochondrial protein) inhibits state 3 oxidation, stimulates state 4 oxidation, abolishes respiratory control and stimulates the latent adenosine triphosphatase activity of mitochondria. The inhibition imposed on state 3 oxidation is relieved by dinitrophenol. 3. At higher concentrations it inhibits coupled phosphorylation as well as dinitrophenol-stimulated adenosine triphosphatase activity. The inhibition of state 3 oxidation under these conditions is not reversed by uncouplers. 4. The three coupling sites of phosphorylation exhibit differential susceptibility to inactivation by this compound. Coupled phosphorylation at the first site is abolished at a drug concentration of 3.0mmol/mg of protein. The third site is inactivated when the concentration of the drug reaches 5.0mmol/mg of protein. The second site is the most refractory and drug concentrations of the order of 10.0mmol/mg of protein are required effectively to inhibit phosphorylation at this site. 5. The compound also inhibits ATP-dependent reversal of electron transport as well as the adenosine triphosphatase activity in submitochondrial particles. 6. The oxidation of NADH and succinate in these particles is not inhibited. 7. These properties indicate that the compound acts as an `inhibitory uncoupler' of energy-transfer reactions in isolated mitochondria.

Liver mitochondria isolated from vanadate-administered rats showed increased (20-25%) rates of oxidation of both NAD(+)-linked substrates and succinate. Respiratory control index and ADP/O were unaffected by the treatment. Dormant and uncoupler-stimulated ATPase activity also was not affected by vanadate administration. Membrane-bound, electron-transport-linked dehydrogenase activities (both NAD(+)- and succinate-dependent) increased by 15-20% on vanadate treatment. Mitochondrial alpha-glycerophosphate dehydrogenase activity increased by 50% on vanadate administration. The above effects of vanadate on oxidoreductase activities could be prevented by the prior administration of antagonists to alpha-adrenergic receptors. Substrate-dependent H2O2 generation by mitochondria also showed an increase on vanadate administration.

Addition of ferrous sulfate, but not ferric chloride, in micromolar concentrations to rat liver mitochondria induced high rates of consumption of oxygen. The oxygen consumed was several times in excess of the reducing capacity of ferrous-iron (O: Fe ratios 5�8). This occurred in the absence of NADPH or any exogenous oxidizable substrate. The reaction terminated on oxidation of ferrous ions. Malondialdehyde (MDA), measured as thiobarbituric acid-reacting material, was produced indicating peroxidation of lipids. The ratio of O2: MDA was about 4: 1. Pretreatment of mitochondria with ferrous sulfate decreased the rate of oxidation (state 3) with glutamate (+malate) as the substrate by about 40% but caused little damage to energy tranduction process as represented by ratios of ADP: O and respiratory control, as well as calcium-stimulated oxygen uptake and energy-dependent uptake of [45Ca]-calcium. Addition of succinate or ubiquinone decreased ferrous iron-induced lipid peroxidation in intact mitochondria. In frozen-thawed mitochondria, addition of succinate enhanced lipid peroxidation whereas ubiquinone had little effect. These results suggest that ferrous-iron can cause peroxidation of mitochondrial lipids without affecting the energy transduction systems, and that succinate and ubiquinone can offer protection from damage due to such ferrous-iron released from the stores within the cells.

The present study deals with the in vitro and in vivo effects of methyl isocyanate (MIC) on rat brain mitochondrial function. Addition of MIC to tightly coupled brain mitochondria in vitro resulted in a mild stimulation of state 4 respiration, abolition of respiratory control, decrease in ADP/0 ratio, and inhibition of state 3 oxidation. The oxidation of NAD+-linked substrates (glutamate + malate) was more sensitive (fourfold) to the inhibitory action of MIC than succinate while cytochrome oxidase was unaffected. Administration of MIC subcutaneously at a lethal dose affected respiration only with glutamate + malate as the substrate (site I) and caused a 20% decrease in state 3 oxidation leading to a significant decrease in respiratory control index while state 4 respiration and ADP/O ratio remained unaffected. As both the malondialdehyde and iron contents of brain mitochondria were not altered, it may be inferred that the observed in vivo inhibition of state 3 oxidation is induced by MIC through systemic stagnant hypoxia leading to ischemia of brain, which further contributes to the cerebral hypoxia.

Previous work has shown that irrespective of the route of exposure methyl isocyanate (MIC) caused acute lactic acidosis in rats (Jeevaratnam et al., Arch. Environ. Contam. Toxicol. 19, 314�319, 1990) and the hypoxia was of stagnant type due to tissue hypoperfusion resulting from hypovolemic hypotension in rabbits administered MIC subcutaneously (Jeevarathinam et al., Toxicology 51, 223�240, 1988). The present study was designed to investigate whether MIC could induce histotoxic hypoxia through its effects on mitochondrial respiration. Male Wistar rats were used for liver mitochondrial and submitochondrial particle (SMP) preparation. Addition of MIC to tightly coupled mitochondria in vitro resulted in stimulation of state 4 respiration, abolition of respiratory control, decrease in ADP/O ratio, and inhibition of state 3 oxidation. The oxidation of NAD+-linked substrates (glutamate + malate) was more sensitive (fiveto sixfold) to the inhibitory action of MIC than succinate while cytochrome oxidase remained unaffected. MIC induced twofold delay in the onset of anerobiosis, and cytochrome b reduction in SMP with NADH in vitro confirms inhibition of electron transport at complex I region. MIC also stimulated the ATPase activity in tightly coupled mitochondria while lipid peroxidation remained unaffected. As its hydrolysis products, methylamine and N,N?-dimethylurea failed to elicit any change in vitro; these effects reveal that MIC per se acts as an inhibitor of electron transport and a weak uncoupler. Administration of MIC sc at lethal dose caused a similar change only with NAD+-linked substrates, reflecting impairment of mitochondrial respiration at complex I region and thereby induction of histotoxic hypoxia in vivo.

Effects of fluctuations in habitat temperature (18-30 degrees) on mitochondrial respiratory behavior and oxidative metabolic responses in the euryhaline ectotherm Scylla serrate are not fully understood. In the present study, effects of different temperatures ranging from 12 to 40 degrees C on glutamate and succinate mediated mitochondrial respiration, respiratory control ratio (RCR), ATP generation rate, ratio for the utilization of phosphate molecules per atomic oxygen consumption (P/O), levels of lipid peroxidation and H2O2 in isolated gill mitochondria of S. serrata are reported. The pattern of variation in the studied parameters was similar for the two substrates at different temperatures. The values recorded for RCR ( >= 3) and P/O ratio (1.4-2.7) at the temperature range of 15-25 degrees C were within the normal range reported for other animals (3-10 for RCR and 1.5-3 for P/O). Values for P/O ratio, ATP generation rate and RCR were highest at 18 degrees C when compared to the other assay temperatures. However, at low and high extreme temperatures, i.e. at 12 and 40 degrees C, states III and IV respiration rates were not clearly distinguishable from each other indicating that mitochondria were completely uncoupled. Positive correlations were noticed between temperature and the levels of both lipid peroxidation and H2O2. It is inferred that fluctuations on either side of ambient habitat temperature may adversely influence mitochondria respiration and oxidative metabolism in S. serrata. The results provide baseline data to understand the impacts of acute changes in temperature on ectotherms inhabiting estuarine or marine environments. (C) 2014 Elsevier Ltd. All rights reserved.

O desequilíbrio nutricional no início da vida leva ao desenvolvimento da obesidade, diabetes e doenças cardiovasculares na idade adulta. Este estudo teve como objetivo analisar os efeitos a longo prazo da hiperalimentação na lactação por meio do modelo de redução da ninhada na hemodinâmica e bioenergética cardíaca. Vinte e quatro camundongos machos Swiss adultos foram divididos em dois grupos (controle e hiperalimentado) submetidos a duas condições (linha de base e isquemia/reperfusão) formando quatro grupos no total: grupo controle linha de base (GCLB), grupo controle isquemia/reperfusão (GCIR), grupo hiperalimentado linha de base (GHLB) e o grupo hiperalimentado isquemia/reperfusão (GHIR), todos com seis camundongos/grupo. As alterações cardíacas foram analisadas por meio da hemodinâmica cardíaca, da respiração mitocondrial e da biologia molecular. Os parâmetros hemodinâmicos analisados foram a velocidade de contração (Max dP/dt), a velocidade de relaxamento (Min dP/dt), o tempo de relaxamento cardíaco isovolumétrico (Tau) e os batimentos por minuto (BPM). A respiração mitocondrial foi avaliada por meio da razão do controle respiratório (RCR) na oxidação de carboidratos e ácidos gordos, e finalmente, a biologia molecular, através de proteínas-chave como a proteína quinase B (AKT), a proteína quinase ativada por adenosina monofosfato (AMPK), a carnitina palmitoil transferase 1 (CPT-1), a proteína desacopladora 2 (UCP2), o 4-hidroxinonenal (4-HNE) e a gliceraldeído-3-fosfato desidrogenase (GAPDH). Os camundongos do GH desenvolveram maior peso corporal (30,95%, P<0,001), gordura epididimal (68,64%, P<0,001), gordura retroperitoneal (109,38%, P<0,01) e glicemia de jejum (19,52%, P<0,05) comparados aos do GC. Os parâmetros Max dP/dt e BPM apresentaram diminuição no GHIR quando comparado ao GHLB (P<0,001 e P<0,05). O parâmetro Min dP/dt apresentou-se reduzido no GCIR e GHIR quando comparado aos grupos GCLB e GCLB (P<0,05; P<0,0001 respectivamente). Camundongos do GHIR apresentaram redução do Tau quando comparado aos grupos GCIR e GHLB (P<0,0001). Estes desequilíbrios na hemodinâmica cardíaca foram associados a função mitocondrial, uma vez que, o GHLB apresenta a RCR reduzida para oxidação de ácidos graxos e carboidratos (P<0,05 e P<0,01, respectivamente) e o GHIR apenas na oxidação dos ácidos graxos (P<0,01). Além disso, o GHIR apresentou diversas alterações nas proteínas-chave do metabolismo energético cardíaco, como diminuição do conteúdo de AKT (P<0,05) e aumento do conteúdo de CPT-1 (P<0,05), 4-HNE (P<0,05) e GAPDH (P<0,05) quando comparado ao CGIR. Finalmente, a expressão do mRNA para CPT1, GAPDH e UCP2 foi aumentada no GHIR quando comparado aos GCIR (P<0,05) e GHLB (P<0,05). A expressão de mRNA para UCP2 e CPT-1 foi reduzida no GCIR quando comparado ao GCLB (P<0,01 e P<0,05, respectivamente). O estudo apresenta resultados consistentes, demonstrando efeitos deletérios sobre o metabolismo cardíaco adulto resultante de alterações nutricionais durante a lactação.

A disfunção mitocondrial tem sido associada a várias doenças, incluindo a colestase hepática, caracterizada pela ativação de células de Kupffer e células fibrogênicas, as quais produzem matriz extracelular excessiva. O acúmulo de sais biliares tóxicos no parênquima hepático leva à lesão crônica com dano mitocondrial, redução da síntese de ATP, aumento de espécies reativas de oxigênio (ROS) e apoptose, resultando em comprometimento da função hepática. Trabalhos anteriores do nosso grupo mostraram o efeito positivo do transplante de células mononucleares da medula óssea (CMMO) na resolução da fibrose hepática e recuperação da função hepática em ratos com colestase, induzida pela ligadura de ducto biliar (LDB). Assim, o presente estudo teve como objetivo analisar a bioenergética mitocondrial após o transplante de CMMO no fígado de ratos com fibrose induzida por ligadura de ducto bilar (LDB). Ratos Wistar machos foram divididos em quatro grupos: animais normais, animais com fibrose hepática após 14 e 21 dias de LDB (F14d e F21d, respectivamente), e animais que após 14 dias de LDB receberam 1x107 CMMO, e foram eutanasiados sete dias após. Nossos dados demonstraram aumento do conteúdo de colágeno tipo I no grupo F21d em relação ao grupo normal, indicativo de fibrose, e sua diminuição após o transplante de CMMO. A análise da fisiologia mitocondrial do fígado mostrou diminuição significativa da taxa respiratória máxima estimulada por ADP (estado 3) nos grupos F14d e F21d, indicando redução da capacidade de oxidação de carboidratos e ácidos graxos. Além disso, a razão do controle respiratório (RCR), indicativa de acoplamento da fosforilação oxidativa com a produção de ATP, apresentou-se significativamente diminuída nos grupos F14d e F21d, sugerindo desacoplamento mitocondrial. No entanto, o transplante de CMMO aumentou significativamente nestes grupos tanto a capacidade oxidativa quanto o acoplamento mitocondrial a níveis semelhantes aos do grupo normal. Estes resultados foram confirmados por análise de western blotting, que mostrou aumento significativo no conteúdo de UCP2 e diminuição do conteúdo de PGC-1α no grupo F21d, com consequente restauração após o transplante de CMMO. Além disso, os resultados mostraram um aumento significativo do conteúdo de 4-HNE no grupo F14d com redução após o transplante CMMO. Assim, podemos concluir que o transplante de CMMO tem um efeito positivo sobre a bioenergética mitocondrial de fígados de ratos com colestase, com aumento da capacidade oxidativa e redução do estresse oxidativo, o que, por sua vez, contribui para a recuperação da função hepática.

O excesso ou a privação de nutrientes em períodos específicos do desenvolvimento, tais como a lactação, estimulam alterações no metabolismo celular, por exemplo. Estas modificações perpetuam-se ao longo da vida e em conseqüência tornam o organismo mais suscetível ao aparecimento de patologias na idade adulta (Programação Metabólica). Estudamos a influência da grelina na secreção de insulina em camundongos Swiss de 120 dias submetidos à hiperalimentação na lactação. Para induzir a hiperalimentação as ninhadas foram reduzidas a 3 filhotes machos por lactante no 3o dia de vida pós-natal. As ninhadas controle foram ajustadas para 9 filhotes machos por lactante. Na idade adulta os animais hiperalimentados (AH) exibiram em comparação aos animais controle (AC) um incremento de 20% no peso corporal, maior índice de Lee (1705,63 g/mm + 29,3 vs 1374,10 g/mm + 54,9; p< 0,001), elevação da gordura corporal (31,0% + 4,6 vs 21,5% + 3,6; p< 0,01), aumento da gordura retroperitoneal (0,79 g + 0,1 vs 0,44 g + 0,1; p< 0,001), hiperglicemia de jejum (151,83 mg/ dl + 8,3 vs 118,0 mg/ dl + 1,0; p< 0,001), hiperinsulinemia de jejum (54,06 UI/ml + 2,3 vs 19,28 UI/ml + 1,53; p< 0,001) e hipogrelinemia de jejum (98,64 pg/ml + 56,5 vs 201,14 pg/ml + 46,4; p< 0,05). Os AH apresentaram maior secreção de insulina in vitro em presença de glicose aos 10 minutos (209,66 UI/ml + 46,5; p< 0,05), 30 minutos (441,88 UI/ml + 30,2; p< 0,05) e 60 minutos (214,34 UI/ml + 29,8) em comparação aos AC, respectivamente 86,90 UI/ml + 9,5; 74,31 UI/ml + 7,7 vs 27,45 UI/ml + 6,1; p< 0,05. As ilhotas pancreáticas dos AH adultos demonstraram em relação aos AC diminuição do consumo de O2 (1,76 pmols O2/ s. ilhota-1 + 0,4 vs 4,85 pmols O2/ s. ilhota-1 + 1,5; p< 0,001) e elevação do conteúdo do receptor de grelina GHSR1A (3,05 % + 2,13 vs 0,95 % + 0,1; p< 0,05). A grelina acilada estimulou a secreção de insulina in vitro dos AC aos 30 minutos (controle com grelina: 208,50 UI/ml + 40,85 vs controle sem grelina: 74,31 UI/ml + 7,7; p< 0,05) e diminuiu a razão do controle respiratório (controle com grelina: 1,45 + 0,2 vs controle sem grelina: 2,51 + 0,7; p< 0,05). Nos AH, a grelina acilada elevou o conteúdo de GLUT2 nas ilhotas pancreáticas em relação aos AC (hiperalimentados com grelina: 2,07 % + 0,5 vs controle com grelina: 0,85 % + 0,4; p< 0,01); entretanto a grelina não foi capaz de estimular a secreção de insulina nestes animais. Concluímos que a hiperalimentação na lactação associou-se ao aumento da gordura corporal e elevou a secreção de insulina na fase tardia do desenvolvimento. A grelina acilada estimulou a secreção de insulina somente nos AC adultos.

Background:Mechanical ventilation is a critical component of paediatric intensive care therapy. It is indicated when the patient’s spontaneous ventilation is inadequate to sustain life. Weaning is the gradual reduction of ventilatory support and the transfer of respiratory control back to the patient. Weaning may represent a large proportion of the ventilatory period. Prolonged ventilation is associated with significant morbidity, hospital cost, psychosocial and physical risks to the child and even death. Timely and effective weaning may reduce the duration of mechanical ventilation and may reduce the morbidity and mortality associated with prolonged ventilation. However, no consensus has been reached on criteria that can be used to identify when patients are ready to wean or the best way to achieve it.Objectives:To assess the effects of weaning by protocol on invasively ventilated critically ill children. To compare the total duration of invasive mechanical ventilation of critically ill children who are weaned using protocols versus those weaned through usual (non-protocolized) practice. To ascertain any differences between protocolized weaning and usual care in terms of mortality, adverse events, intensive care unit length of stay and quality of life.Search methods:We searched the Cochrane Central Register of Controlled Trials (CENTRAL; The Cochrane Library, Issue 10, 2012), MEDLINE (1966 to October 2012), EMBASE (1988 to October 2012), CINAHL (1982 to October 2012), ISI Web of Science and LILACS. We identified unpublished data in the Web of Science (1990 to October 2012), ISI Conference Proceedings (1990 to October 2012) and Cambridge Scientific Abstracts (earliest to October 2012). We contacted first authors of studies included in the review to obtain further information on unpublished studies or work in progress. We searched reference lists of all identified studies and review papers for further relevant studies. We applied no language or publication restrictions.Selection criteriaWe included randomized controlled trials comparing protocolized weaning (professional-led or computer-driven) versus non-protocolized weaning practice conducted in children older than 28 days and younger than 18 years.Data collection and analysis:Two review authors independently scanned titles and abstracts identified by electronic searching. Three review authors retrieved and evaluated full-text versions of potentially relevant studies, independently extracted data and assessed risk of bias.Main results:We included three trials at low risk of bias with 321 children in the analysis. Protocolized weaning significantly reduced total ventilation time in the largest trial (260 children) by a mean of 32 hours (95% confidence interval (CI) 8 to 56; P = 0.01). Two other trials (30 and 31 children, respectively) reported non-significant reductions with a mean difference of -88 hours (95% CI -228 to 52; P = 0.2) and -24 hours (95% CI -10 to 58; P = 0.06). Protocolized weaning significantly reduced weaning time in these two smaller trials for a mean reduction of 106 hours (95% CI 28 to 184; P = 0.007) and 21 hours (95% CI 9 to 32; P < 0.001). These studies reported no significant effects for duration of mechanical ventilation before weaning, paediatric intensive care unit (PICU) and hospital length of stay, PICU mortality or adverse events.Authors' conclusions:Limited evidence suggests that weaning protocols reduce the duration of mechanical ventilation, but evidence is inadequate to show whether the achievement of shorter ventilation by protocolized weaning causes children benefit or harm.

Neonatal anoxia is a worldwide clinical problem that has serious and lasting consequences. The diversity of models does not allow complete reproducibility, so a standardized model is needed. In this study, we developed a rat model of neonatal anoxia that utilizes a semi-hermetic system suitable for oxygen deprivation. The validity of this model was confirmed using pulse oximetry, arterial gasometry, observation of skin color and behavior and analysis of Fos immunoreactivity in brain regions that function in respiratory control. For these experiments, 87 male albino neonate rats (Rattus norvegicus, lineage Wistar) aged approximate 30 postnatal hours were divided into anoxia and control groups. The pups were kept in an euthanasia polycarbonate chamber at 36 +/- 1 degrees C, with continuous 100% nitrogen gas flow at 3 L/min and 101.7 kPa for 25 min. The peripheral arterial oxygen saturation of the anoxia group decreased 75% from its initial value. Decreased pH and partial pressure of oxygen and increased partial pressure of carbon dioxide were observed in this group, indicating metabolic acidosis, hypoxia and hypercapnia. respectively. Analysis of neuronal activation showed Fos immunoreactivity in the solitary tract nucleus, the lateral reticular nucleus and the area postrema, confirming that those conditions activated areas related to respiratory control in the nervous system. Therefore, the proposed model of neonatal anoxia allows standardization and precise control of the anoxic condition, which should be of great value in indentifying both the mechanisms underlying neonatal anoxia and novel therapeutic strategies to combat or prevent this widespread public health problem.
