O objetivo deste trabalho foi determinar os diferentes níveis de resistência à ferrugem-da-folha em genótipos de aveia, e avaliar seu efeito sobre componentes do rendimento de grãos. Os experimentos foram conduzidos na Estação Experimental Agronômica da Universidade Federal do Rio Grande do Sul, em 1995 e 1996. Foram avaliados dez genótipos de aveia com reações diferenciadas à moléstia. A ferrugem-da-folha causou prejuízos no rendimento de grãos, em diferentes densidades de cultivo da aveia, e o peso de grãos foi o componente mais afetado. Porém, alguns genótipos, como o UFRGS 910906, não foram afetados, apesar de apresentarem níveis intermediários de severidade da moléstia, o que evidencia a possibilidade de combinar resistência parcial e potencial de rendimento num mesmo genótipo.

Vinte e dois isolados que diferem quanto aos genes de avirulência/virulência de Puccinia triticina foram inoculados em 55 genótipos de trigo (Triticum aestivum) selecionados entre os recomendados e em experimentação na região sul e centro sul do Brasil nos anos de 1996 e/ou 1997 e em linhas monogênicas para genes (Lr) de resistência à ferrugem da folha, no estádio de plântula. Os tipos de infecção dos genótipos portadores, cada um de distinto gene Lr conhecido foram comparados com os dos genótipos brasileiros, para determinar a existência de possíveis genes de resistência nestes. Os genes de plântula identificados com mais freqüência nas cultivares e linhagens brasileiras foram: Lr26, Lr23, Lr10 e Lr24. O gene Lr16, que confere resistência moderada à maioria das raças, foi encontrado em apenas dois genótipos. Muitos dos trigos analisados possuem um ou mais genes Lr provavelmente ainda não descritos.

An outdoor experiment was conducted to increase understanding of apical leaf necrosis in the presence of pathogen infection. Holcus lanatus seeds and Puccinia coronata spores were collected from two adjacent and otherwise similar habitats with differing long-term N fertilization levels. After inoculation, disease and necrosis dynamics were observed during the plant growing seasons of 2003 and 2006. In both years high nutrient availability resulted in earlier disease onset, a higher pathogen population growth rate, earlier physiological apical leaf necrosis onset and a reduced time between disease onset and apical leaf necrosis onset. Necrosis rate was shown to be independent of nutrient availability. The results showed that in these nutrient-rich habitats H. lanatus plants adopted necrosis mechanisms which wasted more nutrients. There was some indication that these necrosis mechanisms were subject to local selection pressures, but these results were not conclusive. The findings of this study are consistent with apical leaf necrosis being an evolved defence mechanism.

Grassland ecosystems comprise a major portion of the earth’s terrestrial surface, ranging from high-input cultivated monocultures or simple species mixtures to relatively unmanaged but dynamic systems. Plant pathogens are a component of these systems with their impact dependent on many interacting factors, including grassland species population dynamics and community composition, the topics covered in this paper. Plant pathogens are affected by these interactions and also act reciprocally by modifying their nature. We review these features of disease in grasslands and then introduce the 150-year long-term Park Grass Experiment (PGE) at Rothamsted Research in the UK. We then consider in detail two plant-pathogen systems present in the PGE, Tragopogon pratensis-Puccinia hysterium and Holcus lanata-Puccinia coronata. These two systems have very different life history characteristics: the first, a biennial member of the Asteraceae infected by its host-specific, systemic rust; the second, a perennial grass infected by a host-non-specific rust. We illustrate how observational, experimental and modelling studies can contribute to a better understanding of population dynamics, competitive interactions and evolutionary outcomes. With Tragopogon pratensis-Puccinia hysterium, characterised as an “outbreak” species in the PGE, we show that pathogen-induced mortality is unlikely to be involved in host population regulation; and that the presence of even a short-lived seed-bank can affect the qualitative outcomes of the host-pathogen dynamics. With Holcus lanata-Puccinia coronata, we show how nutrient conditions can affect adaptation in terms of host defence mechanisms, and that co-existence of competing species affected by a common generalist pathogen is unlikely.

This article documents the addition of 171 microsatellite marker loci and 27 pairs of single nucleotide polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Bombus pauloensis, Cephalorhynchus heavisidii, Cercospora sojina, Harpyhaliaetus coronatus, Hordeum vulgare, Lachnolaimus maximus, Oceanodroma monteiroi, Puccinia striiformis f. sp. tritici, Rhea americana, Salmo salar, Salmo trutta, Schistocephalus solidus, Sousa plumbea and Tursiops aduncus. These loci were cross-tested on the following species: Aquila heliaca, Bulweria bulwerii, Buteo buteo, Buteo swainsoni, Falco rusticolus, Haliaeetus albicilla, Halobaena caerulea, Hieraaetus fasciatus, Oceanodroma castro, Puccinia graminis f. sp. Tritici, Puccinia triticina, Rhea pennata and Schistocephalus pungitii. This article also documents the addition of 27 sequencing primer pairs for Puffinus baroli and Bulweria bulwerii and cross-testing of these loci in Oceanodroma castro, Pelagodroma marina, Pelecanoides georgicus, Pelecanoides urinatrix, Thalassarche chrysostoma and Thalassarche melanophrys.

Abundant, generally well-preserved radiolarians from Sites 737, 744, 745, 746, 747, 748, and 751 were used in stratigraphic analysis of Neogene, and particularly middle Miocene to Holocene, Kerguelen Plateau sediments. The composite Kerguelen section is more complete than the Weddell Sea sections recovered by Leg 113, and the radiolarians are better preserved. Leg 113 radiolarian zonations of Weddell Sea sites were applicable with only slight modification, and three new zones - Siphonosphaera vesuvius, Acrosphaeral labrata, and Amphymenium challengerae - are proposed for the latest Miocene. Geologic age estimates are given for all radiolarian zones used. Major hiatuses affecting most sites were seen within the middle Miocene, in the latest Miocene, and latest Pliocene. Five new species are described: Acrosphaera? labrata, Acrosphaera? mercurius, Siphonosphaera vesuvius, Actinomma? magnifenestra, and Helotholus? haysi.

Radiolarians are abundant and well preserved in the Neogene of the Kerguelen Plateau. They are common and moderately to well preserved in the Oligocene sequences of Site 738, where the Eocene/Oligocene boundary was observed for the first time in subantarctic sediments, and Site 744. Radiolarians are absent from all glacial sediments from Prydz Bay. Classical Neogene stratigraphic markers were tabulated at all sites. Correlations with paleomagnetic ages were made at Sites 745 and 746 for 26 Pliocene-Pleistocene radiolarian events. Many Miocene to Holocene species are missing from Sites 736 and 737, which were drilled in shallow water (less than 800 m). The missing species are considered to be deepliving forms. Occurrences and relative abundances of morphotypes at six sites are reported. Two new genera (Eurystomoskevos and Cymaetron) and 17 new species (Actinomma kerguelenensis, A. campilacantha, Prunopyle trypopyrena, Stylodictya tainemplekta, Lithomelissa cheni, L. dupliphysa, Lophophaena(?) thaumasia, Pseudodictyophimus galeatus, Lamprocyclas inexpectata, L. prionotocodon, Botryostrobus kerguelensis, B. rednosus, Dictyoprora physothorax, Eucyrtidium antiquum, E.(?) mariae, Eurystomoskevos petrushevskaae, and Cymaetron sinolampas) are described from the middle Eocene to Oligocene sediments at Sites 738 and 744. Twenty-seven stratigraphic events are recorded in the middle to late Eocene of Site 738, and 27 additional stratigraphic datums are recorded, and correlated to paleomagnetic stratigraphy, in the early Oligocene at Sites 738 and 744. Eight radiolarian events are recorded in the late Oligocene at Site 744. New evolutionary lineages are proposed for Calocyclas semipolita and Prunopyle trypopyrena.

A dihaploid mapping population comprising 65 lines was developed between barley parent varieties Tallon and Kaputar and used to construct a genetic linkage map. This map, comprising 195 amplified fragment length polymorphism and 38 simple sequence repeat markers, was used to identify markers linked to the net form of net blotch (Pyrenophora teres f.sp. teres) and to stripe rust (Puccinia striiformis f.sp. hordei) in barley. The population was screened with five pathotypes of net blotch at the seedling stage in the glasshouse and subjected to a natural inoculation in Hermitage, Queensland. Stripe rust screening was conducted at the adult plant stage in Toluca, Mexico. Analyses of the markers were performed using Mapmanager and Qgene software. One region on chromosome 6H was highly significantly associated with resistance to the net blotch (R2 = 79%). This association was consistent for all pathotypes studied. One region on chromosome 5H was found to be highly significantly associated with resistance to stripe rust (R2= 65%). There are a number of very closely linked markers showing strong associations in these regions, and these markers present an opportunity for marker assisted selection of these traits in barley breeding programs.

Devido a grande importância da cultura de Eucalyptus no Brasil, empresas do setor florestal têm buscado através de programas de melhoramento genético, reduzir as perdas de produção e atender a demanda do mercado de papel e celulose. Um exemplo, é a busca por genes de resistência a doenças, principalmente a ferrugem causada por Puccinia psidii Winter, que resulta em redução da produtividade em plantas altamente suscetíveis. No presente trabalho, mudas de Eucalyptus pertencentes a uma geração F1, provenientes do cruzamento controlado entre parentais híbridos E. grandis X E. urophylla, sendo eles resistente e suscetível, foram inoculadas com Puccinia psidii em casa de vegetação e acompanhadas até o aparecimento dos sintomas da ferrugem. Foram classificadas, em dois grupos: resistentes (ausência de sintomas) e suscetíveis (presença de sintomas e esporulação). As amostras de DNA foram comparadas com o uso de marcadores moleculares associado ao método de BSA (Bulked Segregant Analysis). O polimorfismo entre os grupos foi geneticamente relacionado ao loco que determina a característica de resistência ou sucetibilidade. Dentre os 720 "primers" testados, 19 foram polimórficos, porém, apenas o marcador AK 01 manteve-se presente, quando testado em todos os indivíduos da população, mostrando-se a uma distância genética estimada de 20 cM em repulsão ao gene de resistência.

Eudarluca caricis is a common hyperparasite of rusts. A total of 100 cultures were isolated from six Puccinia species or forms growing on 10 species of British grasses at two sites approximately 3 km apart. 82 isolates collected in 2005 were partially sequenced at the ITS locus, and amplified fragment length polymorphism profiles generated for 86 isolates from 2005 and 12 from 2007. Partial ITS sequences of most isolates grouped closely, in a clade with previously reported graminaceous Puccinia isolates and a number of Melampsora isolates. A second clade was very distinct and contained mostly isolates from P. poarum on Poa trivialis. All isolates had distinct AFLP haplotypes. The P. poarum isolates were very distinct from isolates collected from other rusts at the same site. Isolates from P. brachypodii f. sp. arrehenatheri growing on Arrhenatherum elatius in 2005 and 2007 at the same location were distinct (P < 0.001). Isolates from each rust or grass in one year and site were more similar than expected from overall variation between isolates (P<0.001). Isolates from P. coronata on different grasses clustered together (with isolates from P. brachypodii f. sp. poae-nemoralis), suggesting partial host rust specialisation in E. caricis.
