Na busca de novos recursos genéticos capazes de produzir enzimas celulolíticas a baixas/médias temperaturas, o continente Antártico vem demonstrando ser um ambiente bastante promissor. Neste contexto, o objetivo do presente trabalho foi avaliar a influência de diferentes fatores na produção de celulases por fungos filamentosos isolados de amostras da Antártica visando otimização do processo possível aplicação das mesmas na produção de etanol de segunda geração. Foram utilizados os fungos L1-1 e E5B da Central de Recursos Microbianos da UNESP (CRM-UNESP) os quais foram previamente selecionados devido ao potencial celulolítico. O delineamento experimental foi utilizado para analisar a influência de variáveis independentes na produção enzimática. A quantificação da celulase foi realizada pelo método do ácido dinitrosalicílico (ADNS). Antes de iniciar à aplicação dos planejamentos experimentais, foram adotadas estratégias para tentar minimizar e otimizar ao máximo o potencial dos isolados, as quais resultaram no estabelecimento da melhor agitação e a temperatura para a produção de celulase em 150 rpm e 20°C, para os dois isolados estudados. Inicialmente os fungos E5B e L1-1 apresentavam suas produções enzimáticas em 0,233U/mL e 0,342U/mL, respectivamente (antes da aplicação dos desenhos experimentais). Durante a condução do planejamento experimental do tipo Plackett&Burman(PB), foi verificada a preferência dos isolados pela fonte de carbono glicose, com efeito significativo na produção de celulases para os dois isolados. Tendo em vista o seu elevado custo comercial, foram realizados estudos com a sacarose, uma fonte de carbono alternativa e mais barata, bem como indutores enzimáticos. A reversed-phase high performance liquid chromatography method was validated for the determination of cefazolin sodium in lyophilized powder for solution for injection to be applied for quality control in pharmaceutical industry. The liquid chromatography method was conducted on a Zorbax Eclipse Plus C18 column (250 x 4.6 mm, 5 μm), maintained at room temperature. The mobile phase consisted of purified water: acetonitrile (60: 40 v/v), adjusted to pH 8 with triethylamine. The flow rate was of 0.5 mL min-1 and effluents were monitored at 270 nm. The retention time for cefazolin sodium was 3.6 min. The method proved to be linear (r2 =0.9999) over the concentration range of 30-80 µg mL-1. The selectivity of the method was proven through degradation studies. The method demonstrated satisfactory results for precision, accuracy, limits of detection and quantitation. The robustness of this method was evaluated using the Plackett–Burman fractional factorial experimental design with a matrix of 15 experiments and the statistical treatment proposed by Youden and Steiner. Finally, the proposed method could be also an advantageous option for the analysis of cefazolin sodium, contributing to improve the quality control and to assure the therapeutic efficacy

A fast, simple and environmentally friendly ultrasound-assisted dispersive liquid-liquid microextraction (USA-DLLME) procedure has been developed to preconcentrate eight cyclic and linear siloxanes from wastewater samples prior to quantification by gas chromatography-mass spectrometry (GC-MS). A two-stage multivariate optimization approach has been developed employing a Plackett-Burman design for screening and selecting the significant factors involved in the USA-DLLME procedure, which was later optimized by means of a circumscribed central composite design. The optimum conditions were: extractant solvent volume, 13 µL; solvent type, chlorobenzene; sample volume, 13 mL; centrifugation speed, 2300 rpm; centrifugation time, 5 min; and sonication time, 2 min. Under the optimized experimental conditions the method gave levels of repeatability with coefficients of variation between 10 and 24% (n=7). Limits of detection were between 0.002 and 1.4 µg L−1. Calculated calibration curves gave high levels of linearity with correlation coefficient values between 0.991 and 0.9997. Finally, the proposed method was applied for the analysis of wastewater samples. Relative recovery values ranged between 71–116% showing that the matrix had a negligible effect upon extraction. To our knowledge, this is the first time that combines LLME and GC-MS for the analysis of methylsiloxanes in wastewater samples.

Diante da grande quantidade de glicerol bruto gerado na síntese do biodiesel e seu baixo valor comercial, torna-se fundamental encontrar formas alternativas para converter este substrato em produtos com valor agregado. Neste contexto, este trabalho teve como objetivo avaliar diferentes leveduras oleaginosas capazes de metabolizar o glicerol bruto, gerado como coproduto na síntese de biodiesel, visando produzir biomassa como fonte de lipídios. Todos os cultivos foram realizados em frascos agitados, em condições estabelecidas de acordo com cada etapa do trabalho, sendo obtidos dados relativos ao crescimento celular e à produção de lipídios, tratados estatisticamente conforme o propósito. Lipomyces lipofer NRRL Y-1155 apresentou diferenças significativas em relação às outras leveduras oriundas de banco de cultura, atingindo 57,64% de lipídios na biomassa. Estas leveduras apresentarem perfis de ácidos graxos diferenciados, semelhantes aos dos principais óleos vegetais utilizadas na síntese de biodiesel, com predominância de ácidos graxos poli-insaturados, especialmente ácido linoleico (68,3% na levedura Rhodotorula glutinis NRRL YB-252). O ácido gama-linolênico, um ácido graxo essencial ω6, foi detectado em todas as leveduras analisadas, sendo que na biomassa de Candida cylindracea NRRL Y-17506 chegou a 23,1%. Através de um planejamento experimental Plackett-Burman, verificou-se que as variáveis concentração de extrato de levedura e de MgSO4.7H20 demonstraram maior influência na produção de lipídios por uma linhagem silvestre de Rhodotorula mucilaginosa. Para esta levedura, a partir da análise de efeitos foi possível estabelecer a seguinte condição para a produção de lipídios: 30,0 g.L-1 glicerol; 5,0 g.L-1 KH2PO4; 1,0 g.L-1 Na2HPO4; 3,0 g.L-1 MgSO4.7H2O; 1,2 g.L-1 extrato de levedura; pH inicial 4,5; temperatura 25°C. Nestas condições conseguiu-se um teor de lipídios de 59,96% e lipídios totais produzidos de 5,51 g.L-1 . Também foi possível observar aumento no teor de lipídios da biomassa ao longo do tempo de cultivo, bem como o aumento do teor relativo do ácido linoleico, que atingiu 52%. Dentre as leveduras isoladas a partir de amostras ambientais do Extremo Sul do Brasil, a levedura identificada como Cryptococcus humicola se destacou das demais, apresentando proporção de 23,5% de ácidos graxos saturados, 14,8% de ácidos graxos monoinsaturados e 54,9% de ácidos graxos poli-insaturados, destacando-se o ácido linoleico. O planejamento Plackett-Burman foi também utilizado para esta levedura, sendo que as variáveis concentração de extrato de levedura e glicerol bruto demonstraram maior influência na produção de lipídios. Posteriormente, um delineamento composto central rotacional (DCCR) foi proposto visando à otimização da produção de lipídios. Os modelos empíricos preditivos obtidos para biomassa máxima e lipídios totais permitiram estabelecer para a produção de lipídios por Cryptococcus humicola a seguinte condição otimizada: 100,0 g.L-1 glicerol; 5,0 g.L-1 KH2PO4; 1,0 g.L-1 Na2HPO4; 4,8 g.L-1 extrato de levedura; pH inicial 4,5; temperatura 25°C. Esta condição representou um incremento de cerca de 2 vezes nos lipídios totais em relação à melhor condição estabelecida pelo planejamento Plackett-Burmann e um acréscimo de cerca de 4,8 vezes em relação às condições testadas inicialmente, atingindo 37,61% de lipídios e 8,85 g.L-1 de lipídios totais. Deste modo, os propósitos de valorização de um coproduto oriundo da síntese de biodiesel, bem como a produção de um óleo com potencial para a produção de biodiesel, foram cumpridos.

The flow rates of drying and nebulizing gas, heat block and desolvation line temperatures and interface voltage are potential electrospray ionization parameters as they may enhance sensitivity of the mass spectrometer. The conditions that give higher sensitivity of 13 pharmaceuticals were explored. First, Plackett-Burman design was implemented to screen significant factors, and it was concluded that interface voltage and nebulizing gas flow were the only factors that influence the intensity signal for all pharmaceuticals. This fractionated factorial design was projected to set a full 2(2) factorial design with center points. The lack-of-fit test proved to be significant. Then, a central composite face-centered design was conducted. Finally, a stepwise multiple linear regression and subsequently an optimization problem solving were carried out. Two main drug clusters were found concerning the signal intensities of all runs of the augmented factorial design. p-Aminophenol, salicylic acid, and nimesulide constitute one cluster as a result of showing much higher sensitivity than the remaining drugs. The other cluster is more homogeneous with some sub-clusters comprising one pharmaceutical and its respective metabolite. It was observed that instrumental signal increased when both significant factors increased with maximum signal occurring when both codified factors are set at level +1. It was also found that, for most of the pharmaceuticals, interface voltage influences the intensity of the instrument more than the nebulizing gas flowrate. The only exceptions refer to nimesulide where the relative importance of the factors is reversed and still salicylic acid where both factors equally influence the instrumental signal. Compiler optimizations help to make code run faster at runtime. When the compilation is done before the program is run, compilation time is less of an issue, but how do on-the-fly compilation and optimization impact the overall runtime? If the compiler must compete with the running application for resources, the running application will take more time to complete. This paper investigates the impact of specific compiler optimizations on the overall runtime of an application. A foldover Plackett and Burman design is used to choose compiler optimizations that appear to contribute to shorter overall runtimes. These selected optimizations are compared with the default optimization levels in the Jikes RVM. This method selects optimizations that result in a shorter overall runtime than the default O0, O1, and O2 levels. This shows that careful selection of compiler optimizations can have a significant, positive impact on overall runtime.

OBJECTIVE: This study investigated the effect of different ferrule heights on endodontically treated premolars. MATERIAL AND METHODS: Fifty sound mandibular first premolars were endodontically treated and then restored with 7-mm fiber post (FRC Postec Plus #1 Ivoclar-Vivadent) luted with self-polymerized resin cement (Multilink, Ivoclar Vivadent) while the coronal section was restored with hybrid composite core build-up material (Tetric Ceram, Ivoclar-Vivadent), which received all-ceramic crown. Different ferrule heights were investigated: 1-mm circumferential ferrule without post and core (group 1 used as control), a circumferential 1-mm ferrule (group 2), non-uniform ferrule 2-mm buccally and 1-mm lingually (group 3), non-uniform ferrule 3-mm buccally and 2-mm lingually (group 4), and finally no ferrule preparation (group 5). The fracture load and failure pattern of the tested groups were investigated by applying axial load to the ceramic crowns (n=10). Data were analyzed statistically by one-way ANOVA and Tukey's post-hoc test was used for pair-wise comparisons (α=0.05). RESULTS: There were no significant differences among the failure load of all tested groups (P<0.780). The control group had the lowest fracture resistance (891.43±202.22 N) and the highest catastrophic failure rate (P<0.05). Compared to the control group, the use of fiber post reduced the percentage of catastrophic failure while increasing the ferrule height did not influence the fracture resistance of the restored specimens. CONCLUSIONS: Within the limitations of this study, increasing the ferrule length did not influence the fracture resistance of endodontically treated teeth restored with glass ceramic crowns. Insertion of a fiber post could reduce the percentage of catastrophic failure of these restorations under function.

Testing contexts have been shown to critically influence experimental results in psychophysical studies. One of these contexts that show important modulation of the behavioral effects of different stimulatory conditions is the separate (blocked) or mixed presentation of these stimulatory conditions. The study presents evidence that the apparent discriminabilities of two target stimuli can change according to which of these two testing contexts is used. A cross inside a ring and a vertical line inside a ring were presented as go stimuli in a go/no-go reaction time task. In one experiment, each of these stimuli was presented to a different group of volunteers and in another experiment they were presented to the same group of volunteers, randomly mixed in the blocks of trials. Similar reaction times were obtained for the two stimuli in the first experiment, and different reaction times (faster for the cross) in the second experiment. The latter result indicates that the two stimuli have different discriminabilities from the no-go stimulus; the cross having greater discriminability. This difference is however masked, presumably by the adoption of specific compensatory attentional sets, in a separate testing context.

This paper aims to formulate and investigate the application of various nonlinear H(infinity) control methods to a fiee-floating space manipulator subject to parametric uncertainties and external disturbances. From a tutorial perspective, a model-based approach and adaptive procedures based on linear parametrization, neural networks and fuzzy systems are covered by this work. A comparative study is conducted based on experimental implementations performed with an actual underactuated fixed-base planar manipulator which is, following the DEM concept, dynamically equivalent to a free-floating space manipulator. The procedure for online process control by attributes consists of inspecting a single item at every m produced items. It is decided on the basis of the inspection result whether the process is in-control (the conforming fraction is stable) or out-of-control (the conforming fraction is decreased, for example). Most articles about online process control have cited the stoppage of the production process for an adjustment when the inspected item is non-conforming (then the production is restarted in-control, here denominated as corrective adjustment). Moreover, the articles related to this subject do not present semi-economical designs (which may yield high quantities of non-conforming items), as they do not include a policy of preventive adjustments (in such case no item is inspected), which can be more economical, mainly if the inspected item can be misclassified. In this article, the possibility of preventive or corrective adjustments in the process is decided at every m produced item. If a preventive adjustment is decided upon, then no item is inspected. On the contrary, the m-th item is inspected; if it conforms, the production goes on, otherwise, an adjustment takes place and the process restarts in-control. This approach is economically feasible for some practical situations and the parameters of the proposed procedure are determined minimizing an average cost function subject to some statistical restrictions (for example, to assure a minimal levelfixed in advanceof conforming items in the production process). Numerical examples illustrate the proposal.

Nearest–neighbour balance is considered a desirable property for an experiment to possess in situations where experimental units are influenced by their neighbours. This paper introduces a measure of the degree of nearest–neighbour balance of a design. The measure is used in an algorithm which generates nearest–neighbour balanced designs and is readily modified to obtain designs with various types of nearest–neighbour balance. Nearest–neighbour balanced designs are produced for a wide class of parameter settings, and in particular for those settings for which such designs cannot be found by existing direct combinatorial methods. In addition, designs with unequal row and column sizes, and designs with border plots are constructed using the approach presented here.
