Roles for the transcription factor RFL in rice axillary meristem development were studied. Its regulatory effects on LAX1, CUC1, and OsPIN3 reveal its functions in axillary meristem specification and outgrowth.Axillary meristems (AMs) are secondary shoot meristems whose outgrowth determines plant architecture. In rice, AMs form tillers, and tillering mutants reveal an interplay between transcription factors and the phytohormones auxin and strigolactone as some factors that underpin this developmental process. Previous studies showed that knockdown of the transcription factor gene RFL reduced tillering and caused a very large decrease in panicle branching. Here, the relationship between RFL, AM initiation, and outgrowth was examined. We show that RFL promotes AM specification through its effects on LAX1 and CUC genes, as their expression was modulated on RFL knockdown, on induction of RFL:GR fusion protein, and by a repressive RFL-EAR fusion protein. Further, we report reduced expression of auxin transporter genes OsPIN1 and OsPIN3 in the culm of RFL knockdown transgenic plants. Additionally, subtle change in the spatial pattern of IR4 DR5:GFP auxin reporter was observed, which hints at compromised auxin transport on RFL knockdown. The relationship between RFL, strigolactone signalling, and bud outgrowth was studied by transcript analyses and by the tillering phenotype of transgenic plants knocked down for both RFL and D3. These data suggest indirect RFL-strigolactone links that may affect tillering. Further, we show expression modulation of the auxin transporter gene OsPIN3 upon RFL:GR protein induction and by the repressive RFL-EAR protein. These modified forms of RFL had only indirect effects on OsPIN1. Together, we have found that RFL regulates the LAX1 and CUC genes during AM specification, and positively influences the outgrowth of AMs though its effects on auxin transport.

通过菲污染诱导动态响应预实验确定了小麦为供试植物,赤霉素(GA3)、细胞分裂素的玉米素和玉米素核苷总含量(Z&ZR)以及脱落酸(ABA)为植物激素供试指标,并且确定试验终点为小麦生长14d。在预实验的基础上,以草甸棕壤为供试土壤,采用室内培养箱盆栽方法,进行多环芳烃菲、芘、荧蒽和苯并(a)芘单一,两-两复合以及三-三复合与3种植物激素的剂量-效应关系研究;3种植物激素对土壤重金属镉、铅、铜和锌单一污染诱导的响应关系研究。并以多环芳烃类的菲和重金属类的镉为代表污染物,进行小麦芽期生长指标及植物激素测定,研究植物激素与生长指标对污染物的响应及敏感性比较。 多环芳烃单一及复合污染诱导实验结果表明:ABA对菲、芘、荧蒽、苯并(a)芘单一及复合污染诱导的响应趋势一致,即随着污染诱导浓度增加,ABA含量显著增加;GA3与四种多环芳烃单一及复合污染诱导存在一定的响应关系,但响应趋势呈多样性变化;对于多环芳烃单一及复合污染诱导,随着污染物浓度增加,Z&ZR含量出现了先增加后减少的趋势,表现为显著的污染诱导抑制效应。ABA和Z&ZR对多环芳烃污染诱导响应敏感,这为其成为土壤污染诊断指标提供了可能性。 在不同的镉、铅、铜、锌污染处理中,小麦体内ABA含量均出现明显增加趋势,与多环芳烃污染胁迫响应一致;GA3含量对镉、铅、铜、锌四种重金属污染胁迫响应趋势相同,均为先增加后减少。对于土壤镉和铅污染胁迫Z&ZR表现为先增加后减少的响应趋势,而对于铜和锌污染胁迫,Z&ZR含量变化与镉、铅胁迫时不同。 植物激素与生长指标对污染胁迫的响应及敏感性比较研究表明,对多环芳烃菲污染诱导,植物激素含量同生长指标相比具有显著的敏感性。对重金属镉污染诱导,内源激素含量和生长指标敏感性都显著,小麦根和芽中ABA含量显著增加,进一步证明了其在植物污染抗逆中的作用机理。

Cork is a natural and renewable material obtained as a sustainable product from cork oak (Quercus suber L.) during the tree’s life. Cork formation is a secondary growth derived process resulting from the activity of cork cambium. However, despite its economic importance, only very limited knowledge is available about the molecular mechanisms underlying the regulation of cork biosynthesis and differentiation. The work of this PhD thesis was focused on the characterization of an R2R3-MYB transcription factor, the QsMYB1, previously identified as being putatively involved in the regulatory network of cork development. The first chapter introduces cork oak and secondary growth, with special emphasis on cork biosynthesis. Some findings concerning transcriptional regulation of secondary growth are also described. The MYB superfamily and the R2R3-MYB family (in particular) of transcription factors are introduced. Chapter II presents the complete QsMYB1 gene structure with the identification of two alternative splicing variants. Moreover, the results of QsMYB1 expression analysis, done by real-time PCR, in several organs and tissues of cork oak are also reported. Chapter III is dedicate to study the influence of abiotic stresses (drought and high temperature) and recovery on QsMYB1 expression levels. The effects of exogenous application of phytohormones on the expression profile of QsMYB1 gene are evaluated on Chapter IV. Chapter V describes the reverse genetic approach to obtain transgenic lines of Populus tremula L. x tremulóides Michx. overexpressing the QsMYB1 gene. Finally, in Chapter VI the final conclusions of this PhD thesis are presented and some future research directions are pointed based on the obtained results.

Senescence represents the final developmental act of the leaf, during which the leaf cell is dismantled in a coordinated manner to remobilize nutrients and to secure reproductive success. The process of senescence provides the plant with phenotypic plasticity to help it adapt to adverse environmental conditions. Here, we provide a comprehensive overview of the factors and mechanisms that control the onset of senescence. We explain how the competence to senesce is established during leaf development, as depicted by the senescence window model. We also discuss the mechanisms by which phytohormones and environmental stresses control senescence, as well as the impact of source-sink relationships on plant yield and stress tolerance. In addition, we discuss the role of senescence as a strategy for stress adaptation and how crop production and food quality could benefit from engineering or breeding crops with altered onset of senescence.

The phytohormones gibberellin (GA) and abscisic acid (ABA) regulate important developments events in germinating seeds. Specifically, GA induces the expression of hyrolase genes, like the α-amylase gene Amy32b, which mobilizes starch reserves to be used by the embryo, and ABA suppresses this induction. Recent advancements identified ABA and GA receptors and key components in the signaling pathways, however, the mechanism of crosstalk between the hormones remains largely unknown. To further elucidate the mechanism of ABA suppression of GA-induced genes, we focused on the transcription factor TaABF1, a member of the ABA response element binding factor family. TaABF1 has been shown to physically interact with the SnRK2 kinase PKABA1 and overexpression of TaABF1 or PKABA1 can suppress Amy32b. We carried out particle bombardment experiments to investigate how TaABF1 suppresses Amy32b and how TaABF1 is activated by ABA. The role of TaABF1 in ABA-mediated suppression of Amy32b is more complicated than hypothesized. Unlike PKABA1, overexpression of TaABF1 did not cause a decrease of GAMyb expression and in fact resulted in an increase of GAMyb expression. When TaABF1 and GAMyb were simultaneously overexpressed in aleurone, the GAMyb induction of Amy32b was unaffected, indicating that the target of TaABF1 action must be upstream of GAMyb. Furthermore, TaABF1 and ABA demonstrated an additive effect on the suppression of Amy32b. Based on our findings, we propose a model in which PKABA1 activates two separate targets, one being TaABF1 which then modifies an unknown target upstream of GAMyb and the other being an unknown transcription factor that suppresses GAMyb transcription.

The objective of this work was to evaluate the effect of different artemisinin concentrations in the flowering induction of A. annua. Two genotypes of A. annua (CPQBA 239x1V and 3MxPOP) were atomized with four different artemisinin concentrations (0, 500, 5000, and 10000 mg L-1). The application of artemisinin didn't induce the flowering of both genotypes tested, in none of the used concentrations.

The objective of this work was to evaluate the effects of the inoculation with plant growth-promoting bacteria on the formation of sugarcane pre-sprouted seedlings, originated from individual buds, and to quantify the initial growth of these seedlings in association with nitrogen application, in a low fertility soil. Two experiments were carried out: one in greenhouse conditions, with a time span of 50 days, and the other in vases under field conditions, with a time span of 180 days. In both experiments, a randomized complete block design was used with a factorial arrangement of 2x3, in the first experiment - with or without inoculation, and with three quantities of bud reserve -, and of 2x2x4, in the second one - with or without inoculation, with or without nitrogen, evaluated in four times: at 45, 90, 135, and 180 days. The inoculant had effect on the initial growth of the pre-sprouted seedlings, increasing sprouting speed and dry matter accumulation on roots and shoots, regardless of the bud reserve amount. In the second experiment, the inoculant increased the initial growth of shoots and root system up to 180 days after transplantation, increasing height, tillering, stalk diameter, dry matter production of stalks and straw, and root length, regardless of nitrogen application. The inoculant has a positive physiological effect on plant growth.

Transposable elements (TEs) account for a large portion of plant genomes, particularly in grasses, in which they correspond to 50%-80% of the genomic content. TEs have recently been shown to be a source of new genes and new regulatory networks. The most striking contribution of TEs is referred as "molecular domestication", by which the element coding sequence loses its movement capacity and acquires cellular function. Recently, domesticated transposases known as mustang and derived from the Mutator element have been described in sugarcane. In order to improve our understanding of the function of these proteins, we identified mustang genes from Sorghum bicolor and Zea mays and performed a phenetic analysis to assess the diversity and evolutionary history of this gene family. This analysis identified orthologous groups and showed that mustang genes are highly conserved in grass genomes. We also explored the transcriptional activity of sugarcane mustang genes in heterologous and homologous systems. These genes were found to be ubiquitously transcribed, with shoot apical meristem having the highest expression levels, and were downregulated by phytohormones. Together, these findings suggest the possible involvement of mustang proteins in the maintenance of hormonal homeostasis.

The direct induction of adventitious buds and somatic embryos from explants is a morphogenetic process that is under the influence of exogenous plant growth regulators and its interactions with endogenous phytohormones. We performed an in vitro histological analysis in peach palm (Bactris gasipaes Kunth) shoot apexes and determined that the positioning of competent cells and their interaction with neighboring cells, under the influence of combinations of exogenously applied growth regulators (NAA/BAP and NAA/TDZ), allows the pre-procambial cells (PPCs) to act in different morphogenic pathways to establish niche competent cells. It is likely that there has been a habituation phenomenon during the regeneration and development of the microplants. This includes promoting the tillering of primary or secondary buds due to culturing in the absence of NAA/BAP or NAA/TDZ after a period in the presence of these growth regulators. Histological analyses determined that the adventitious roots were derived from the dedifferentiation of the parenchymal cells located in the basal region of the adventitious buds, with the establishment of rooting pole, due to an auxin gradient. Furthermore, histological and histochemical analyses allowed us to characterize how the PPCs provide niches for multipotent, pluripotent and totipotent stem-like cells for vascular differentiation, organogenesis and somatic embryogenesis in the peach palm. The histological and histochemical analyses also allowed us to detect the unicellular or multicellular origin of somatic embryogenesis. Therefore, our results indicate that the use of growth regulators in microplants can lead to habituation and to different morphogenic pathways leading to potential niche establishment, depending on the positioning of the competent cells and their interaction with neighboring cells. The aim of the present thesis was to better understand the physiological role of the phytohormones jasmonates (JAs) and abscisic acid (ABA) during fruit ripening in prospect of a possible field application of JAs and ABA to improve fruit yield and quality. In particular, the effects of exogenous application of these substances at different fruit developmental stages and under different experimental conditions were evaluated. Some aspects of the water relations upon ABA treatment were also analysed. Three fruit species, peach (Prunus persica L. Batsch), golden (Actinidia chinensis) and green kiwifruit (Actinidia deliciosa), and several of their cvs, were used for the trials. Different experimental models were adopted: fruits in planta, detached fruit, detached branches with fruit, girdled branches and micropropagated plants. The work was structured into four sets of experiments as follows: (i) Pre-harvest methyl jasmonate (MJ) application was performed at S3/S4 transition under field conditions in Redhaven peach; ethylene production, ripening index, fruit quality and shelf-life were assessed showing that MJ-treated fruit were firmer and thus less ripe than controls as confirmed by the Index of Absorbance Difference (IAD), but exhibited a shorter shelf-life due to an increase in ethylene production. Moreover, the time course of the expression of ethylene-, auxin- and other ripening-related genes was determined. Ripening-related ACO1 and ACS1 transcript accumulation was inhibited though transiently by MJ, and gene expression of the ethylene receptor ETR2 and of the ethylene-related transcription factor ERF2 was also altered. The time course of the expression of several auxin-related genes was strongly affected by MJ suggesting an increase in auxin biosynthesis, altered auxin conjugation and release as well as perception and transport; the need for a correct ethylene/auxin balance during ripening was confirmed. (ii) Pre- and post-harvest ABA applications were carried out under field conditions in Flaminia and O’Henry peach and Stark Red Gold nectarine fruit; ethylene production, ripening index, fruit quality and shelf-life were assessed. Results show that pre-harvest ABA applications increase fruit size and skin color intensity. Also post-harvest ABA treatments alter ripening-related parameters; in particular, while ethylene production is impaired in ABA-treated fruit soluble solids concentration (SSC) is enhanced. Following field ABA applications stem water potential was modified since ABA-treated peach trees retain more water. (iii) Pre- and post-harvest ABA and PDJ treatments were carried out in both kiwifruit species under field conditions at different fruit developmental stages and in post-harvest. Ripening index, fruit quality, plant transpiration, photosynthesis and stomatal conductance were assessed. Pre-harvest treatments enhance SSC in the two cvs and flesh color development in golden kiwifruit. Post-harvest applications of either ABA or ABA plus PDJ lead to increased SSC. In addition, ABA reduces gas exchanges in A. deliciosa. (iv) Spray, drench and dipping ABA treatments were performed in micropropagated peach plants and in peach and nectarine detached branches; plant water use and transpiration, biomass production and fruit dehydration were determined. In both plants and branches ABA significantly reduces water use and fruit dehydration. No negative effects on biomass production were detected. The present information, mainly arising from plant growth regulator application in a field environment, where plants have to cope with multiple biotic and abiotic stresses, may implement the perspectives for the use of these substances in the control of fruit ripening.

Kolumnare Apfelbäume (Malus x domestica) stellen aufgrund ihres auffälligen Phänotyps eine ökonomisch interessante Wuchsform dar. Diese extreme Form des Kurztriebwuchses zeichnet sich durch einen insgesamt sehr schlanken, säulenförmigen Habitus aus, welcher eine dichte Pflanzung und damit einhergehend Ertragssteigerungen im Vergleich zu normalwüchsigen Bäumen ermöglicht. Verursacht wird der Phänotyp durch die Anwesenheit eines einzelnen, dominanten Allels des Columnar (Co)-Gens. Bis auf die approximative Lokalisation des Gens auf Chromosom 10 ist über mögliche Identität und Funktion bislang nichts bekannt.rnIn der vorliegenden Arbeit wurde ein erster Versuch unternommen, mit Hilfe von Next Generation Sequencing (NGS) Technologien und RNA-Seq Einblicke in das Transkriptom des Sprossapikalmeristems (SAM) kolumnarer Apfelbäume zu gewinnen. So konnte gezeigt werden, dass unabhängig vom Zeitpunkt der Entnahme des Materials mehrere hundert Gene differentiell reguliert werden. Diese lassen sich funktional in mehrere überrepräsentierte Kategorien gruppieren, von denen sich einige wiederum mit dem kolumnaren Phänotyp assoziieren lassen. Durch den Einsatz weiterer Expressionsstudien (Microarrays, qRT-PCR) konnten frühere Ergebnisse bezüglich des Hormonhaushalts auf Genebene bestätigt und neue Erkenntnisse gewonnen werden, die eine mögliche Erklärung für den Phänotyp darstellen. Weiterhin ergab der Vergleich aller durchgeführten Expressionsstudien eine Anreicherung signifikant differentiell regulierter Gene auf Chromosom 10, was auf einen „selective sweep“ hindeutet. Eine potentielle epigenetische Regulation dieser Gene durch das Genprodukt von Co könnte daher möglich sein. Mehr als die Hälfte dieser Gene lassen sich darüber hinaus aufgrund ihrer Funktion direkt mit dem kolumnaren Phänotyp assoziieren.rnDiese Ergebnisse zeigen, dass die Anwesenheit des Co-Allels massive Veränderungen in der Genregulation des SAMs mit sich bringt, wobei einige dieser differentiell regulierten Gene mit großer Wahrscheinlichkeit an der Etablierung des kolumnaren Phänotyps beteiligt sind. Auch wenn die Funktion des Co-Genproduktes nicht abschließend geklärt werden konnte, sind doch anhand der Resultate schlüssige Hypothesen diesbezüglich möglich.rn

Nowadays microalgae are studied, and a number of species already mass-cultivated, for their application in many fields: food and feed, chemicals, pharmaceutical, phytoremediation and renewable energy. Phytoremediation, in particular, can become a valid integrated process in many algae biomass production systems. This thesis is focused on the physiological and biochemical effects of different environmental factors, mainly macronutrients, lights and temperature on microalgae. Microalgal species have been selected on the basis of their potential in biotechnologies, and nitrogen occurs in all chapters due to its importance in physiological and applicative fields. There are 5 chapters, ready or in preparation to be submitted, with different specific matters: (i) to measure the kinetic parameters and the nutrient removal efficiencies for a selected and local strain of microalgae; (ii) to study the biochemical pathways of the microalga D. communis in presence of nitrate and ammonium; (iii) to improve the growth and the removal efficiency of a specific green microalga in mixotrophic conditions; (iv) to optimize the productivity of some microalgae with low growth-rate conditions through phytohormones and other biostimulants; and (v) to apply the phyto-removal of ammonium in an effluent from anaerobic digestion. From the results it is possible to understand how a physiological point of view is necessary to provide and optimize already existing biotechnologies and applications with microalgae.
