Argentina es el principal productor mundial de limones en el mundo y el 90 por ciento de esta producción se sitúa en la provincia de Tucumán. Las pérdidas económicas ocasionadas por las enfermedades de poscosecha representan uno de los principales problemas de la citricultura mundial, siendo el 80 por ciento atribuíbles a infecciones fúngicas. El moho verde causado por Penicillium digitatum (Pers.)Sacc. (PD), es la enfermedad de mayor incidencia y severidad durante la exportación de frutos de limón. Actualmente el control de estas enfermedades se realiza con el uso de fungicidas de síntesis química. Debido a las severas restricciones impuestas a estos productos, por regulaciones ambientales y de la salud, hay una fuerte necesidad de métodos alternativos de control. El objetivo de este trabajo de tesis fue evaluar alternativas de bajo impacto ambiental : bacterias lácticas, extractos vegetales, Serenade® (Bacillus subtilis QST 713)y bicarbonato de sodio; con actividad antifúngica directa o a través de la inducción de mecanismos de defensa, para el control de PD en condiciones in vitro e in vivo (frutos de limón). De un total de 33 cepas de bacterias lácticas evaluadas, se seleccionaron ocho con propiedades antifúngicas frente a PD. Se identificaron los metabolitos antifúngicos como ácidos orgánicos (ácido láctico, ácido acético y ácido fenil láctico)y se determinó la combinación óptima con mayor actividad inhibitoria. Los productos alternativos: extractos vegetales, Serenade® y bicarbonato de sodio, presentaron actividad antifúngica in vitro a PD con porcentajes de inhibición entre 2,1 - 16 por ciento. En los frutos de limón, los tratamientos alternativos no fueron efectivos por sí solos para controlar en forma directa la enfermedad. Estos tratamientos redujeron significativamente la severidad de la enfermedad, excepto el Serenade® cuando fue aplicado simultáneamente con el inóculo. Esta reducción fue más marcada cuando se dejaron transcurrir 18 h entre inoculación y aplicación de los tratamientos. El bicarbonato de sodio al 3 por ciento y en menor medida el Serenade® al 1 por ciento indujeron una respuesta de defensa de los frutos y controlaron la enfermedad en un 96,7 por ciento y 56,7 por ciento, respectivamente. En cromatografía de capa fina y fluorometría se detectó un aumento de la fitoalexina escoparona, sugiriendo que este efecto se produciría como resultado de la inducción de mecanismos de defensa, mientras que la umbeliferona no fue detectada. Estos resultados indican que los productos biológicos actuaron como una barrera al avance de la enfermedad, retrasando la severidad de la misma e induciendo de una respuesta de defensa en los frutos.

Gibberellic acid and potassium nitrate did not promote the germination of myrtle seeds when tested at 20/30degreesC (16/8h). Germination was promoted considerably by alternating temperatures. The results of an investigation on a two-dimensional temperature gradient plate showed that myrtle seeds germinated most rapidly (within 14 days) and fully (all viable seeds) at 35/22.5degreesC (16/8 h) and similar regimes. Tests on five seed lots of Lagerstroemia speciosa and L. floribunda showed the efficacy of the alternating temperature regime of 35/20degreesC (16/8 h) in promoting germination. Thus we recommend myrtle seeds be tested for germination in this regime for 28 days.

The endosperm of seeds of Sesbania virgata (Cav.) Pers. accumulates galactomannan as a cell wall storage polysaccharide. It is hydrolysed by three enzymes, one of them being alpha-galactosidase. A great amount of protein bodies is found in the cytoplasm of endospermic cells, which are thought to play the major role as a nitrogen reserve in this seed. The present work aimed at understanding how the production of enzymes that degrade storage compounds is controlled. We performed experiments with addition of inhibitors of transcription (actinomycin-d and alpha-amanitin) and translation (cycloheximide) during and after germination. In order to follow the performance of storage mobilisation, we measured fresh mass, protein contents and alpha-galactosidase activity. All the inhibitors tested had little effect on seed germination and seedling development. Actinomycin-d and cycloheximide provoked a slight inhibition of the storage protein degradation and concomitantly lead to an elevation of the alpha-galactosidase activity. Although alpha-amanitin showed some effect on seedling development at latter stages, it presented the former effect and did not change galactomannan degradation performance. Our data suggest that some of the proteases may be synthesised de novo, whereas alpha-galactosidase seems to be present in the endosperm cells probably as an inactive polypeptide in the protein bodies, being probably activated by proteolysis when the latter organelle is disassembled. These evidences suggest the existence of a connection between storage proteins and carbohydrates mobilisation in seeds of S. virgata, which would play a role by assuring a balanced afflux of the carbon and nitrogen to the seedling development.

A 140,0 kDa lectin was purified and characterized from the mushroom Clavaria cristata. The purification procedures from the crude extract of the mushroom comprised gel filtration chromatography on Sephacryl s200 and ion exchange on Resource Q column. The purified lectin agglutinated all types of human erythrocytes with preference for trypsinized type O erythrocytes. The haemagglutinating activity is dependent of Ca 2+ ions and was strongly inhibited by the glycoprotein bovine submaxillary mucin (BSM) up to the concentration of 0, 125 mg/mL. The C. cristata lectin (CcL) was stable in the pH range of 2,5-11,5 and termostable up to 80 °C. CcL molecular mass determined by gel filtration on a Superose 6 10 300 column was approximately 140,3 kDa. SDS polyacrilamide gel electrophoresis revealed a single band with a molecular mass of approximately 14,5 kDa, when the lectin was heated at 100 ⁰C in the presence or absence of β-mercaptoethanol. CcL induced activation of murine peritoneal macrophages in vitro resulting in the release of nitric oxide (NO), reaching the maximum production at 24 h. In experimental paw oedema model in mice, CcL showed proinflammatory activity being able to induce oedema formation. Cell viability of HepG2, MDA 435 e 3T3 cell lines was examined after 72 h of incubation with CcL in different concentrations (0,5-50 μg/mL). CcL inhibited HepG2 cells growth with an IC50 value of 50 μg/mL. In the present work, the observed immunomodulatory and antiproliferative effects indicate CcL as a possible immunomodulator compound, interfering in the macrophages immune response, taking possible anti-parasitic, anti-tumoral effects or diagnostic and/or therapeutic

A taxonomic survey of representatives of the genus Geastrum took place during the rainy season from 2006 to 2009 in four districts in the state of Rio Grande do Norte: Parque Estadual Dunas do Natal (Atlantic Forest), Mata do Jiqui EMPARN (Atlantic Forest) and Estação Ecológica do Seridó (Caatinga). Fourteen species were recorded: G. entomophilum, G. fimbriatum, G. hirsutum, G. javanicum, G. lageniforme, G. lloydianum, G. minimum, G. morganii, G. ovalisporum, G. pectinatum, G. saccatum, G. schweinitzii, G. setiferum and G. triplex. Of these species, eleven occurred in the Atlantic Forest and six in the Caatinga. A new species has been recorded to science, G. entomophilum, other as a first record for Brazil, G. morganii, six new records for the Northeast, and ten new records for Rio Grande do Norte. The material was tumbled in the Herbarium UFRN. Additionally, a survey of the species of the genus deposited in the Herbarium UFRN was accomplished, resulting in 244 herbarium specimens belonging to thirty-three species. Of these ones, twenty-three were collected in Brazil and ten are from Czech Republic, Europe, as donation from the VZ Herbarium
